  • The pharmaceutical characteristics of solid indoprofen inclusion complex such as dissolution, permeation through a cellophane membrane, model analysis of interfacial tranesfer, absorption behaviors in rat intestine, and the plasma concentration of indoprofen after oral administrations to rabbits were examined in comparison with those indoprofen alone. The inclusion complex obtained by freeze-drying method showed the higher dissolution rate and membrane permeability among the test powders, and increased significantly the amont of indoprofen absorbed in rat intestine and the levels of plasma concentration of indoprofen after oral adminstrations to rabbits. The increase of bioavailability of $indoprofen-{\beta}-cyclodextrin$ complex was considered due to the increased solubility and dissolution rate of solid powder form.

  • Effects of siho(Bupleuri Radix) extracts on lipid composition and antioxidant system were investigated in rat fed high fat diet. body weight gain, concentration of plasma triglyceride, total cholesterol, LDL-cholesterol showed a tendency to decrease in siho(Bupleuri Radix) extracts groups. However, the concentration of HDL-cholesterol showed no significantly different among treatments. Plasma and liver TBARS concentration showed a low values in siho(Bupleuri Radix) extracts groups. Activities of SOD, GSH-Px and CAT showed a tendency to increase in siho(Bupleuri Radix) extracts groups.

  • Objective : Effects of Gamibangpungtongseungsan on lipid composition and antioxidant system were investigated in rat fed high fat diet. Methods : We fed control group rats high fat diet and administered normal saline for 8 weeks, and experimental group rats high fat diet and administered extract of Gamibangpungtongseungsan for 8 weeks. Results : Body weight gain, concentration of plasma triglyceride, total cholesterol, LDL-cholesterol showed a tendency to decrease in Gamibangpungtongseungsan groups. However, the concentration of LDL-cholesterol showed a tendency to increase in Gamibangpungtongseungsan groups. Plasma and liver TBARS concentration showed a low values in Gamibangpungtongseungsan groups. Activities of SOD, GSH-Px and CAT showed a tendency to increase in Gamibangpungtongseungsan groups. Conclusions : These results indicate that Gamibangpungtongseungsan has a deep inhibitory effect on lipid composition and antioxidant system.

  • To study the effects of dietry methionine level on lipid peroxidation of rats, rats were fed vitamin E, selenium and methionine - deficient diet or the same diet supplemented with various levels(0.3%, 0.6%, 0.9%) of methionine for 6 weeks. The biochemincal and mophological changes in the rat liver were investigated. Lipid peroxide levels in plasma and hepatic mitochondrial fraction of MF rats were more increased than those of control rats. However, supplementation with 0.6% methionine modified this increment. Catalase activity was decreased in hepatic mitochondrial fraction from rats fed MF diet. Methionine supplementation did not induce this enzyme. The ultrastructural evidence for lipid peroxidation was found in plasma membranes facing sinusoids. The most striking changes in including disruption and loss of microvilli and development of numerous lipid droplets occurred in rats fed MF diet. These changes were not effectively prevented by the same diet supplemented with 0.3% or 0.9% methionine, but supplementation with 0.6% methionine modulated more or less the changes.

  • A high-performance liquid chromatographic method using the liquid extraction procedure was developed for the determination of L -FMAUS. a new L -FMAU derivative, in rat plasma and urine using 3-aminophenyl sulfone as an internal standard. A 100-${\mu}\ell$ aliquot of distilled water containing the L -cysteine (100 mg/$m\ell$) was added to a 100-${\mu}\ell$ aliquot of biological sample. Effects of Protein and Protein Hydrolysate on Nitrogen Metabolism in Rats with Cysteamine-induced Duodenal Ulcer (Cysteamine에 의해 유도된 십이지장 궤양 흰쥐의 체내질소대사에 대한 단백질과 단백질 가수분해물의 섭취효과)

  • This study aimed to verify the nutritional and curative effects of protein hydrolysate in rats with cysteamine-induced duodenal uncer. Duodenal ulcer rat model was established by intraperitoneal injections of cysteamine. Sprague-Dawley, female rats weighing approximately 200g were intraperitoneally injected twice cysteamine(13mg/100g BW) at intervals of 3h per day. This procedure was repeated 3$\times$at intervals of 3d. Animals fed on 10% casein diet for infection periods. After last injection, 4 kinds of diets(10% casein, 20% casein, 10% casein hydrolysate, 20% casein hydrolysate) were given. Gastric montility, trypsin activity in gastrointestinal content, retention rate of nitrogen, plasma total protein, albumin, amino-N, urinary urea nitrogen, creatinine and hydroxyproline were analyzed for nutritional effects of dietary nitrogen levels(10%, 20%) and sources(casein, casein hydrolysate). In duodenal ulcer rat model, there was no differences between 20% casein diet and 20% casein hydrolysate in the view of severeness of ulcer, gastric emptying rate, serum total protein, serum albumin, plasma $\alpha$-amino-N, UUN, creatinine excretion, GFR, nitrogen retention. On the other hand, rats on 10% casein hydrolysate diet group had more curative effect of the ulcer, higher plasma albumin concentration and nitrogen retention than 10% casein diet group. The casein hydrolysate diet group was lower trypsin activity in small intestinal content than the casein diet group, at both nitrogen levels(10%, 20%). The results suggest that protein hydrolysate be applied in diet therapy for the patients with gastrointestinal ulcer.

Simultaneous Determination of Valproic Acid and its Toxic Metabolites, 4-ene-VPA and 2,4-diene-VPA in Rat Plasma using a Gas Chromatographic-mass Spectrometric Method

  • A gas chromatographic-mass spectrometric (GC-MS) method was developed for the simultaneous determination of valproic acid (VPA) and its toxic metabolites, 4-ene-VPA and 2,4-diene-VPA in rat plasma. Extraction was performed in weak acidic condition (pH 5.2) to avoid degradation of 4-ene-VPA and 2,4-diene-VPA. The recoveries for 4-ene-VPA and 2,4-diene-VPA were more than 70% and that for VPA was 33-42%. R value for each compounds exceeded 0.998 in calibration curve during all the analysis. Accuracy and precision ranged from 88.3 to 113.2% and from 2.16 to 14.2%, respectively The method was successfully applied to monitor plasma concentrations of VPA, 4-ene-VPA and 2,4-diene-VPA after intravenous administration of VPA at the dose of 100 mg/kg, suggesting that these toxic metabolites may involved in the hepatotoxicity induced by VPA.

The Effects of Micro Crystalline Chitin and Palm Oil on the Lipid Metabolism in the Rat (미세결정화키틴과 팜유의 수준을 달리한 식이가 흰쥐의 지방대상에 미치는 영향)

  • The effects of MCC and palm oil at different levels on lipid metabolism were assessed in adolescent rat. Dietary fat levels were 20% and 40% (kcal/kcal) and MCC level were 0%, 2%, and 4% (wt/wt). The experimental period took 8 weeks. During the experiment, weight gain and food efficiency ration were not affected by dietary factors. The weight of thymus however, was lower in high fat groups than in middle fat groups. The contents of total lipid and total cholesterol in plasma and HDL-cholesterol in plasma and HDL-cholesterol were significantly affected by dietary factors. The concentrations of cholesterol in LDL and VLDL , and the triglyceride content of VLDL was higher in high fat groups than in middle fat groups. Adding MCC lowered the LDL triglyceride in the high fat groups. The analysis of plasma fatty acids generally reflected the composition of the dietary palm oil. MCC had significant effects on total lipid and triglyceride of feces, but not on total cholesterol . In conclusion, the level of MCC had little hypolipodemic effects on lipid metabolism in rats fed a high fat diet. the middle fat group that contained 4% MCC showed increased contents of fecal triglyceride than the others, indicating that MCC had an effect on lipid absorption. Therefore, the other physiological functions of MCC need to be tested for their useful applications.

  • The effects of application of mustard oil (MO), a small-fiber excitant and inflammatory irritant into the rat maxillary molar tooth pulp on arterial blood pressure and heat race, and electromyographic (EMG) activity of the jaw muscles were assessed in the anesthetized rats. In addition, Evans blue extravasation following pulpal MO application was measured. Application of MO into the tooth pulp produced a significant increase in mean arterial pressure and heat rate which gradually returned to baseline level. Application of MO into the tooth pulp induced a significant and short-lasting increase in EMG activity of digastric masseter and tongue muscle. Application of MO into the tooth pulp significantly increased the plasma extravasation of Evans blue dye. However, Application of mineral oil into the tooth pulp did not produce any significant changes in EMG activity of the digastric, masseter and tongue muscles, mean arterial pressure and heart rate, and plasma extravasation of Evans blue dye. These findings indicate that changes in arterial blood pressure, heart rate, jaw muscle activity and plasma extravasation accompanying noxious tooth pulp stimulation call be utilized as indirect measure of orofacial pain and inflammation.

Analysis of Vasopressin-Induced $Ca^{2+}$ Increase in Rat Hepatocytes

  • To analyze vasopressin-induced $Ca^{2+}$ increase in liver cells, rat hepatocytes were isolated and attached to collagen-coated cover slips. Using fura-2, a $Ca^{2+}$-sensing dye, changes in intracellular $Ca^{2+}$ concentration by vasopressin were monitored. Results in this communication suggested that vasopressin-induced $Ca^{2+}$ increase were composed of both $Ca^{2+}$ release from internal $Ca^{2+}$ stores and influx from the plasma membrane. The $Ca^{2+}$ influx consisted of two distinguishable components. One was dependent on the presence of vasopressin and the other was not. SK&F96365 blocked vasopressin-induced $Ca^{2+}$ influx in a dose-dependent manner. Vasopressin-induced $Ca^{2+}$ release from internal stores diminished in a primary culture of hepatocytes according to the culture time. However, changes in vasopressin-induced $Ca^{2+}$ influx across the plasma membrane differed from changes in the $Ca^{2+}$ release from internal stores, suggesting two separate signalings from receptor activation to internal stores and to the plasma membrane.
