Studies on the Effects of Consecutive Copper-Administration on Testis Weights, No. of Sperm, Motility and Organ Weights and Histological Changes in Rats (연속적인 Cu투여가 Rat의 정소중량, 정자수, 활력, 장기 중량 및 조직변화에 미치는 영향)

  • This study was performed to elucidate the effects of copper poisoning on the reproductive organ of rats. After consecutive oral administrations of copper sulfate, the weights of testis, the numbers and motilities of sperms, organ weights and histological changes of testes were compared between control and experimental groups. 1. Testis weights of 1,000, 2,000 or 4,000 ppm/kg of copper sulfate-administrated rats gradually decreased compared with control group, and the values no significant changes. 2. The sperm numbers of 1,000, 2,000 or 4,000 ppm/kg of copper sulfate-administrated rats were lowered in dose dependent manners than those of control group's and the values no significant changes. 3, The motilities sperms of 1,000, 2,000 or 4:000 ppm/kg of copper sulfate-administrated rats decreased significantly in dose-dependent manners compared with those of control group's. 4. The weights of livers and kidneys of 1,000, 2,000 or 4,000 ppm/kg of copper sulfate-administration rats decreased or increased. 5. Necrosis of hepatocytes around the central veins and infiltrations of fine granules-harboring macrophages in periportal and interstitial tissues were found out in the livers of copper sulfate-administrated rats. The Bowman's capsule and tubules of kidneys were filled with hyaline material.

  • This study was performed to elucidate the effects of lead poisoning on the reproductive organ of rats. After consecutive oral administrations of lead acetate, the weights of testis, the numbers and motilities of sperms and histological changes of organs were compared between control and experimental groups. 1. Testis weights of 1,000, 2,000 or 4,000 ppm/kg of lead acetate-administrated rats decreased compared with control group in dose-dependent manner. 2. The sperm numbers of 1,000, 2,000 or 4,000 ppm/kg of lead acetate-administrated rats were lowered significantly in dose dependent manners than those of control groups. 3. The sperm motilities of 1,000, 2,000 or 4,000 ppm/kg of lead acetate-administrated rats decreased in dose-dependent manners compared with those of control groups. 4. The weights of livers increased and the kidney weights decreased and changes were dose-independent manner. 5. Necrosis of hepatocytes around the central veins, infiltrations of neutrophils, accumulations of bile and infiltrations of fine granules-harboring macrophages in psychymal and interstitial tissues were found out in the livers of copper sulfate-administrated rats. The Bowman's capsule, tubular epithelium and includes in nucleus of kidneys were filled with hyaline materials and hematophilic centers appeared in several lymph nodes.

BQ-123, $ET_{A}$ antagonist, decreases clinical sign and inflammatory region on EAE.

  • BQ-123, a selective $ET_{A}$ receptor antagonist, reverses various responses induced by Endothelin-1 and it has been reported that BQ-123 ameliorates the cerebrovascular constriction, hypertension, and decrease of blood flow. Previously, we announced that the level of Endothelin-2 increase in the brain and spinal cord of EAE-induced lewis rat and showed the origin of ET-1 is activated macrophages. Intracisternal injection of $ET_{A}$ receptor antagonist, BQ-123(10nmol) was done for visualizing the role of endothelin-1 on the pathogenesis of EAE. BQ-123 apparently blocked the severity of clinical score of EAE and decreased the histologically observed inflammatory region. The blocking effect on the progression of EAE model following BQ-123, suggests that BQ-123 is a physiological antagonist in terms of development of the sign of multiple sclerosis.

  • Objective: In this study, we investigated the effects of Rhizoma Cimicifugae (RC) on the induction of nitric oxide (NO) in RAW 264.7 murine macrophages treated with lipopolysaccharide (LPS). Methods: We examined whether RC have an antipyretic effect by inhibition of COX and/or NOS in fever that is evoked by an intraperitoneal injection of LPS in rats. Results: 1. RC inhibited NO metabolites produced by activated murine macrophage in a dose-dependent manner. 2. RC had an antipyretic effect in a dose-dependent manner in LPS-induced fever. 3. RC inhibited levels of both COX-2 protein and iNOS protein increased by the treatment of LPS in brain tissues as well as brain blood vessels. 4. There were no changes in proteins of nNOS/COX-1. Conclusions: RC has an antipyretic effect by attenuating the level of iNOS and COX-2 protein.

  • Ginkgetin was previously reported as an inhibitor of group II phospholipase A$_2$. It also inhibited in vitro arachidonate release from the activated macrophages and lymphocyte proliferation. These previous studies suggested an anti-inflammatory nature of ginkgetin, especially on chronic inflammation. In fact, ginkgetin showed potent anti-inflammatory activity against rat adjuvant-induced arthritis, a chronic inflammatory animal model, with comparable analgesic activity. In order to investigate the action mechanisms, tumor necrosis factor and interferone release were studied from human PMMC. It was found that ginkgetin clearly inhibited release of these cytoknes from human PMMC. Ginkgetin was also found to inhibit immunoglobulin M production at 1 - 10 uM. These results may contribute to antiarthritic activity of ginkgetin in vivo.

  • The purpose of this research was to investigate effects of Bangpoongtongsungsan water extract(BTSE) on the immune reaction, anti-allergy action and anti-inflammatory action in BALB/c mice. The administration of BTSE (500mg/kg) enhanced the cell viability of thymocytes and the population of helper T cells in splenic T-lymphocytes. BTSE suppressed the production of nitric oxide, but enhanced the phagocytic activity in peritoneal macrophages. BTSE enhanced hemagglutination titer in mice. BTSE inhibited passive cutaneous anaphylaxis induced by egg albumin in rat, the lethal anaphylaxis induced by platelet activating factor and compound 48/80 in mice, and then inhibited the degranulation of peritoneal mast cells induced by compound 48/80. BTSE did not inhibit Arthus reaction, but inhibited the delayed type hypersensitivity induced by SRBC and contact dermatitis induced by DNFB. BTSE inhibited the acute hind paw edema induced by histamine after 30 minutes, the permeability of evans blue into peritoneal cavity induced by acetic acid and the writhing syndrome induced by acetic acid. These results suggest that BTSE has an immunopotentiative action, anti-allergy action and anti-inflammatory action via the inhibition of histamine release.

  • In the previous study, we reported that 2'-hydroxy-4'-methxoychalcone, synthetic chalcone inhibited PGE2 production in TPA- stimulated rat peritoneal macrophages by inhibiting the induction of COX-2 protein. The present study was carried out to clarify whether 2'-hydroxy-4'-methoxychalcone inhibit angiogenesis by the experimental methods in vitro and in vivo. 2'-Hydroxy-4'-methoxychalcone decreased angiogenesis of both chick embryos in the chorioallantoic membrane assay and basic fibroblast growth factor-induced vessel formation inthe mouse Martigel plug assay.   • There are several subspecies of Bombyx mori, whose silk sericin variants differ. Silk sericin can induce bone morphogenic protein-2 (BMP-2) in macrophages, and silk sericin from different species may have different levels of BMP-2 induction ability. In this study, silk sericin from three B. mori subspecies (Baegokjam, Yeonnokjam, and Goldensilk) was prepared. They were administered to RAW264.7 cells and BMP-2 expression level was studied. Bone regeneration was evaluated using a rat calvarial defect model. BMP-2 expression level was the highest in the Baegokjam group. The bone volume in the Baegokjam group was significantly higher than that in the Yeonnokjam group (P = 0.003). In conclusion, sericin from Baegokjam showed higher levels of BMP-2 expression and bone regeneration than those from Yeonnokjam and Goldensilk.

  • Protein kinase C an enzyme of signal transduction has been known to regulate cell proliferation activation as well as apoptosis in some cancer cell lines. To explore the role of PKC in the course of cell mediated autoimmune disease such as experimental autoimmune encephalomyelitis (EAE) EAE was induced in Lewis rats(6-8 weeks old) with immunization of myelin basic protein supplemented with complete Freund's adjuvants and affected spinal cords were sampled at days 13 postimmunization(PI) as peak stage of EAE and at days 21 PI as recovery stage. The spinal cords with EAE were subjected to Northern blot analysis and insitu hybridization of PKC delta which is one of prominant isotypes of PKC in the haematopoietic cells. Northern blot analysis showed that levels of PKS delta mRNA in the spinal cords of rats withEAE was significantly increased at days 13 PI in which inflammatory cells including T cells and macrophages in the EAE lesions appeared. however the stage. By in situ hybridization signals of PKC delta in EAE lesions was intensely expressed on the delta is also expressed on some brain cells in normal rat central nervous system This finding suggests that PKC plays an important role on either activation of inflammatory cells including encephalitogenic T cells and macrophages or apoptotic elimination of some inflammatory cells depending on the stge of EAE.
