Purpose: The purpose of this study was to examine effects of decreased locomotor activity on mass, Type I and II fiber cross-sectional areas of ipsilateral and contralateral hindlimb muscles 21 days after establishing the Parkinson's disease rat model. Methods: The rat model was established by direct injection of 6-hydroxydopamine (6-OHDA, 50 ${mu}g$) into the left substantia nigra after stereotaxic surgery. Adult male Sprague-Dawley rats were assigned to one of two groups; the Parkinson's disease group (PD; n=17) and a sham group (S; n=8). Locomotor activity was assessed before and 21 days after the experiment. At 22 days after establishing the rat model, all rats were anesthetized and soleus and plantaris muscles were dissected from both ipsilateral and contralateral sides. The brain was dissected to identify dopaminergic neuronal death of substantia nigra in the PD group. Results: The PD group at 21 days after establishing the Parkinson's disease rat model showed significant decrease in locomotor activity compared with the S group. Weights and Type I and II fiber cross-sectional areas of the contralateral soleus muscle of the PD group were significantly lower than those of the S group. Conclusion: Contralateral soleus muscle atrophy occurs 21 days after establishing the Parkinson's disease rat model.
The electrophoretic patterns of plasma membrane surface proteins of normal rat liver cells and rat hepatomas were compared in 10% non-denaturing and 7-15% gradient non-denaturing gel. Chemical carcinogens, 2-Me DAB (2-methyl-4-dimethylaminoazobenzene) and DENA (diethylnitrosamine), were used to induce hepatoma in rats. One protein which disappeared in hepatoma was identified in normal rat liver by non-denaturing gel electrophoresis. Rabbit antisera were raised against this specific protein, and the protein was purified by Sephacryl S-200 column and immunoaffinity chromatography using the purified antibody. The purified protein showed two bands of molecular weights approximately 50 $kD_{\alpha}$ and 52 $kD_{\alpha}$ by SDS-polyacrylamide gel electrophoresis, which reacted specifically with the antibody. However only one band was observed in non-denaturing gel and also in isoelectric focusing with a pI value of 6.6. This study showed the existence of an unique protein on the plasma membrane surface of normal rat liver cells which disappeared in rat hepatomas induced by chemical carcinogens.
A gene coding for triosephosphate isomerase (TPI) from a rat skeletal muscle cDNA library was cloned and its nucleotide sequence was determined. The 1, 348-bp cDNA clone contains 24 bp $5^I$ noncoding region, the entire 750 bp coding region corresponding to a protein of 249 amino acids, $547bp 3^I$ noncoding region and part of a poly(A) tail. It also contains a polyadenylation signal, AATAAA, starting from 17 bp upstream of the poly(A) tail. The calculated molecular weight of rat TPI is 27.8 kDa and the net charge is +4. The deduced amino acid sequence from rat TPI CDNA sequence has 93% and 94% homology with that of mouse and human clones, respectively. The amino acids at the residue of Asn12, Lys14, His96, Glu 166, His96, His101, Ala177, Tyr165, Glu13O, Tyr2O9, and Ser212 in catalytic site are completely identical, confirming that the functional residues in TPI proteins are highly conserved throughout evolution. The most profound characteristic of rat TPI enzyme, compared with other TPIs, is that there are five cysteine substitutions at the residue of 21, 27, 159, 195 and 204. A Glu123 instead of Gly was found in rabbit, rhesus, mouse and human sequences. Through the method of RT-PCR, the mRNA transcription level of TPI gene was found to be different among various tissues and was highest in muscle.
Chronic liver diseases and hepatic cancer have been reported as 10% of cause of death in Koreans. Regardless of various causes, chronic liver disease accompanies commonly hepatic fibrosis. But still the mechanism of hepatic fibrosis remains poorly understood. Using the dimethylnitrosamine(DMN)-induced hepatic fibrosis rat model, We performed to evaluate the possible therapeutic effect of RIP(extracts of Phellodendron amurense and Patrinia scabiosaefolia) and to investigate the changes in referential connective tissue proteins($TGF-{\beta}_1$, ${\alpha}$-smooth muscle actin, and vimentin) as a marker of fibrogenesis. For these purposes, liver tissues were stained with H & E, and Azan staining for estimation of developing fibrosis. In the DMN-treated rat liver tissue, fibrosis were developed forming incomplete septal fibrosis. Whereas, in the RIP-treated rat liver tissues, the fibrosis were decreased recovering to normal morphology. The expressions of $TGF-{\beta}_1$, ${\alpha}$-smooth muscle actin($\alpha-SMA$), and vimetin were increased in the DMN-treated rat liver tissues, but decreased in the various areas of RIP-treated rat liver tissues. According to these results, RIP could be a possible therapeutic agent to reduce hepatic fibrosis, and the $TGF-{\beta}_1$, ${\alpha}$-SMA, and vimentin could be possible indicative markers of hepatic fibrosis development and recovery.
It was previously reported from our laboratory that the rate of deterioration of contractile force was slower in the heart of the ginseng extract treated rats. It was also found that ginseng may have an ability to sustain the normal function of the heart by sustaining Ca accumulation by sarcoplasmic reticulum. $Ca^{++}-dependent$ ATPase plays the central role in movement of $Ca^{++}$ ion from sarcoplasm into sarcoplasmic reticulum. In this investigation, the fragment of sarcoplasmic reticulum was prepared from rat heart treated with ginseng water extract orally 100mg/kg/day for 7 to 10 days and from normal rat heart. $Ca^{++}-dependent$ APTase activity was estimated by a modified method of Fiske and Subbarow's procedure. Experimental groups were divided into 6 groups, depending on the preincubation time, 5, 30 and 60min. at ${25}^{\circ}C$ and ${37}^{\circ}C$ respectively. In both of the groups of ${25}^{\circ}C$ and ${37}^{\circ}C$, $Ca^{++}-dependent$ ATPase activities of the ginseng treated rat hearts were higher than that of normal hearts. Therefore, it can be concluded that $Ca^{++}-dependent$ ATPase activities in sarcoplasmic reticulum of rat hearts were increased by the treatment with ginseng extract.

The fallowing are the result of the experimental studies of KH-304 on the penile nitric oxide synthase (NOS) activity, SOD and the level of blood testosterone in old rat. KH-304 was tested for the expression and activity of eNOS, nNOS, SOD/Mn, caveol in-1, 3 in penis of old-rat. The penile expression level of the five enzyme were increased significantly after oral administration of the KH-304 100mg/kg for 100ays and the concentration of testosterone in the blood were increased. In vitro, the effect of SOD and free radical scavenging were increased significantly. Conclusively, KH-304 is capable of improving of sexual ability in old-rat.

최근 들어 전 세계적으로 극심한 기후변화와 그에 따른 기상이변으로 국지성 집중호우로 인한 홍수피해가 급증하고 있으며, 그의 피해규모 또한 대형화되고 있는 추세이다. 국내의 경우 1990년 중반부터 매년 국지성 집중호우나 이상호우로 인한 피해가 반복되고 있고, 그밖에 경우 7월과 8월 여름철에 강우가 집중되며 일정한 강우패턴이 사라지고 있다. 이러한 기상재해에 따른 피해를 줄이기 위해 강우-유출 현상에 관한 정교한 해석이 필요하며, 단기간에 발생되는 호우사상에 대한 유출해석은 홍수관리 측면에서 중요한 역할을 한다. 본 연구에서는 국지성 집중호우 및 돌발홍수가 잦은 낙동강수계인 경상남도 양산에 위치한 양산천을 대상유역으로 선정하여 홍수유출모의를 실시하였으며, 홍수유출모의에는 분포형모형인 S-RAT모형과 VfloTM모형을 사용하여 모의하였다. S-RAT모형의 검증을 위해 동일한 사상을 이용하여 보정 및 검증을 하였다. 또한 VfloTM 의 Inundation Tool을 이용하여 홍수범람모의를 하였고, 통계학적 분석을 통하여 두 모형의 오차를 계산하여 S-RAT모형의 국내 유역 적용성을 검토하였다.
The objectives of this study is to find out mechanism of vasodilating effects of ANP in 2K-1C renovascular hypertensive rat aorta and to compare with those of normotensive rat aorta. In 2K-1C renovascular hypertensive rat, average arterial blood pressure and plasma renin activity were higher than in normotensive rat. In 2K-1C renovascular hypertensive rat aorta, NE sensitivity was more increased and maximal contraction of aorta by NE was higher than those of normotensive rat aorta. ANP inhibited NE-induced contraction in both 2K-1C renovascular hypertensive and normotensive rat aorta, concentration-dependently. However, ANP was less effective for relaxing NE-induced contraction in 2K-1C renovascular hypertensive rat aorta than in normotensive rat aorta. ANP inhibited $^{45}Ca^{2+}$ uptake induced by NE in both 2K-1C renovascular hypertensive and normotensive rat aorta. From these results. inhibition of $Ca^{2+}$ influx may be one of the vasodilating mechanism of ANP in 2K-1C renovascular hypertensive rat aorta. Although the potency of ANP in relaxing NE-induced contractions was attenuated, the efficacy of ANP was not changed in 2K-1C renovascular hypertensive rat aorta compared with that of ANP in normotensive rat aorta. 사람에게 행동의 개인차가 있듯이 rat이나 mouse에 있어서도 행동의 차이를 발견할 수 있다. Rat의 행동성향에 따른 (1)학습 및 기억 능력의 차이, (2)기억과 해마의 관계, (3)치매유발단백질의 하나로 알려진 아밀로이드 베타($A{\beta}$ )및 수종의 항 치매 약물효과를 알아보는 것이 본 실험의 목적이다. Rat의 행동관찰을 통해 두 가지 행동패턴을 관찰할 수 있었는데, 이러한 rat의 행동 특성은 심리학자 Jung이 심리유형으로 설명하고 있는 extraversion, introversion의 행동성향과 유사할 것이라는 가정 하에 실험을 계획, 실시하였다. Rat에 water maze test를 실시하여 공간 기억의 단기, 장기 기억을 분석하였는데 그 결과 두 가지 행동 성향을 가진 rat은 서로 다른 학습 및 기억 능력의 특성을 보였다. 즉, extraversion은 단기 기억의 향상을 보인 반면에, introversion은 장기 기억의 향상을 보였다. Rat을 대상으로 water maze test 외에 Y-maze, passive avoidance test를 실시하여 공간 기억(spatial memory), 작동 기억(working memory), passive avoidance memory, 그리고 단기, 장기 기억의 관계를 종합적으로 분석해 보았다. 그 결과 두 가지 행동성향에 따라 서로 영향을 미치는 기억의 종류 및 관계에 차이가 있음을 발견할 수 있었다. 또한 두 가지 행동성향을 가진 rat에 약물을 투여했을 때, 서로 다른 약물 효과를 보였으며, $A{\beta}$ 를 주입했을 때, 기억(memory) 및 해마(hippocampus) 세포 사멸(cell death)에 서로 상반된 결과를 보여주었다. 이러한 연구 결과는 개체의 행동성향에 따라 학습 및 기억의 효과가 다를 수 있음을 보여주는 결과라 할 수 있고, 개인의 적성과 소질의 인식 및 개발의 중요성에 시사하는 바가 크다. 또한 개개인의 행동과 학습 및 기억 능력의 차이를 두뇌과학적으로 이해하여, 두뇌의 장점은 살리고 단점을 보완할 수 있는 이론적 토대를 세우는데 이러한 동물실험이 그 기초를 제공해 줄 수 있을 것이다. 또한 행동성향 및 기억의 종류에 따른 약물효과의 차이는 기억과 관련된 질병인 알츠하이머 환자에 있어 개개인에게 맞는 적절한 특징적인 치료약물이 존재할 것이라는 가능성을 제공해줄 뿐만 아니라 학습과 기억력 증진 효과를 기대해 볼 수 있을 것이라고 생각된다.
Accurate and rapid diagnosis of Pandemic Influenza A/H1N1 2009 virus (H1N1 2009) infection is important for the prevention and control of influenza epidemics and the timely initiation of antiviral treatment. This study was conducted to evaluate the performance of several diagnostic tools for the detection of H1N1 2009. Flocked nasopharyngeal swabs were collected from 254 outpatients of suspected H1N1 2009 during October 2009. This study analyzed the performances of the RealTime Ready Inf A/H1N1 Detection Set (Roche), Influenza A (H1N1) Real-Time Detection Kit (Bionote), Seeplex Influenza A/B OneStep Typing Set [Seeplex Reverse Transcriptase PCR (RT-PCR)], BinaxNow Influenza A & B Test Kit [Binax Rapid Antigen Test (RAT)], and SD BIOLINE Influenza Ag kit (SD RAT). Roche and Bionote real-time RT-PCR showed identical results for the H1N1 2009 hemagglutinin gene. Compared with real-time RT-PCR, the sensitivities and specificities were 83.7% and 100% for Seeplex RT-PCR, 64.5% and 94.7% for Binax RAT, and 69.5% and 100% for SD RAT. The sensitivities of Seeplex RT-PCR, Binax RAT, and SD RAT in patients aged over 21 years were 73.7%, 47.4%, and 57.9%, respectively. The sensitivities of Seeplex RT-PCR, Binax RAT, and SD RAT on the day of initial symptoms were mostly lower (68.8%, 56.3%, and 31.3%, respectively). In conclusion, multiplex RT-PCR and RAT for the detection of H1N1 2009 were significantly less sensitive than real-time RT-PCR. Moreover, a negative RAT may require more sensitive confirmatory assays, because it cannot be ruled out from influenza infection.
