Waksman과 Heinrich(1943)에 의해 Streptomyces속이 기술되고 actinomycin, streptomycin등의 많은 항생물질이 발견되면서 방선균(actinomycetes)은 산업적으로 중요한 미생물이 되어 왔으며, 근래에는 항생물질 뿐만 아니라 효소, 효소저해제, 면역조절물질 등 소위 생리활성물질(bioactive compound)의 생산균으로서 주목을 받고 있다. 그러나 지금까지 대부분의 스크리닝이나 연구의 대상은 주로 중성 pH에서 잘 자라는 호중성 방선균(neutrophilic actionmycetes)이었지만 산성이나 알칼리성 토양, 고온, 고열, 고압 등과 같은 특수환경에서 분리된 균주나 희귀 방선균(rare actinomyces)에 대한 관심도 점점 높아지고 있다. 산성 토양에서 분리된 방선균에 관한 연구는 Jensen(1928)의 연구로부터 시작하였지만 Lonsdale(1985)이 침엽수림, 산성탄광 등에서 분리된 방선균을 수리분류할 때까지 활발하게 이루어지지 못하였다. 본 고에서는 호산성 방선균위 분포, 생리적 특성, 수리분류학적 연구, 화학적 분류(chemotaxonomy), 산업적 응용가능성 등을 살펴보고자 한다.
A cyclic undecapeptide-family natural product, cyclosporin A (CyA), which is one of the most valuable immunosuppressive drugs, is produced nonribosomally by a multifunctional cyclosporin synthetase enzyme complex in a filamentous fungal strain named Tolypocladium niveum. Previously, structural modifications of cyclosporins such as a regionspecific hydroxylation at the $4^{th}$ N-methyl leucine in a rare actinomycetes called Sebekia benihana were reported to lead to dramatic changes in their bioactive spectra. However, the reason behind this change could not be determined since a system to genetically manipulate S. benihana has not yet been developed. To address this limitation, in this study, we utilized the most commonly practiced gene manipulation techniques including conjugation-based foreign gene transfer-and-expression as well as targeted gene disruption to genetically manipulate S. benihana. Using these optimized genetic manipulation systems, a putative cytochrome P450 hydroxylase (CYP) gene named CYP506, which is involved in CyA hydroxylation in S. benihana, was specifically disrupted and genetically complemented. The S. benihana${\Delta}$CYP506 exhibited a significantly reduced CyA hydroxylation yield as well as considerable yield restoration by functional complementation of the S. benihana CYP506 gene, suggesting that the genetically manipulated S. benihana CYP mutant strains may serve as a more efficient bioconversion host for various valuable metabolites including CyA.
Actinomycosis is a chronic infectious disease, which is produced by Gram-positive anaerobic organisms, actinomycetes, normally inhabit in the mouth, bowel and female genital tract. Primary endobronchial actinomycosis is relatively rare infection and can be misdiagnosed as endobronchial tuberculosis or malignancies. We experienced a case of primary endobronchial actinomycosis in a 49-year-old man presented with fifteen-days history of hemoptysis and cough. He had a past history of extraction of teeth because of dental caries six months ago. Chest X-ray showed irregular consolidation ad bronchoscopic findings revealed nearly obstruction by tumor mass with active bleeding in RLL. Pathologic finding of the bronchial mass showed sulfur granule with granulation tissue formation. Intravenous administration of penicillin G followed by oral ampicillin therapy for 6 months resulted in marked improvement in symptoms and chest X-ray findings. We report this case with review of literature.
노카디아병 (nocardiosis)은 actinomycetales 목, nocardia 과에 속하는 호기성 양성 간균에 의한 감염으로 노카디아종은 사람과 동물에서 국소적, 전신적인 화농성 질환을 일으킬 수 있다. 노카디아병은 일반적으로 기회감염으로 생기지만 감염자의 1/3은 면역기능이 정상이다. 저자들은 고령의 크론병 (Crohn's disease) 여자 환자에서 노카디아종에 의해 발생한 폐 노카디아병과 뇌농양의 증례를 보고하고자 한다. 이 병변은 영상 검사상 주위 부종을 동반하며 조영 증강을 보여 수술 전에는 종양으로 생각되었고, 전이성 뇌암처럼 공격적인 병의 진행을 보였다. 노카디아병의 조기 진단, 기저 질환이 없는 경우, 적절한 항생제 치료가 이루어 졌을 때 예후가 좋다. 저자들은 뇌에 생긴 노카디아병을 보고하여 자기공명영상과 컴퓨터 단층촬영 소견을 알리고 악성 병변과의 감별에 도움이 되고자 한다.

Members of the genus Streptomyces produce more than 70% of antibiotics. The rise in antibiotic resistance globally enhanced the search for novel species with the ability to produce new bioactive compounds. This study was initiated to investigate different regions in Jordan for previously uncultured and rare Streptomyces species capable of producing novel antimicrobial compounds especially active against bacteria resistant to antibiotics. A total of 191 Streptomyces strains were isolated from 26 soil samples collected from different geographic regions in Jordan. Isolates were characterized based on colony and cellular morphology as well as using 16S rRNA gene sequencing. These isolates were screened for their ability to produce antibiotics by the perpendicular-cross streak method, and then tested by well diffusion method against tested pathogens. Fifty-four isolates showed potential to produce antimicrobial products especially active against resistant bacteria, 20.1% of the isolates showed inhibitory effect against Staphylococcus aureus, 16.9% against clinical MSSA strains, and 18.0% against MRSA: whereas only 4.2% against Esherichia coli, 3.2% against Klebsiella pneumonia, 2.7% against Pseudomonas aeruginosa, and 10.0% against clinical Candida albicans. Three isolates were selected for further identification due to their antibacterial activity against S. aureus, MRSA, and MSSA. These isolates were identified as follows; Streptomyces aburaviensis DSa3, Streptomyces alboniger SAb7 and Streptomyces misionensis ZAb2, based on cultural, biochemical characteristics and molecular analysis of the 16S rRNA.
Nocardia farcinia, an aerobic, gram-positive bacilli actinomycetes of the genus Nocardia, is an uncommon pathogen found in humans. The most common Nocardia infection sites are the lung, central nervous system, and skin. Even though hematogenous dissemination can occur, isolation of the organism from blood cultures is very rare. We report a case of Nocardia infection that was isolated on blood cultures. A 59-year-old male with a medical history that includes a liver transplantation 6-years prior due to hepatocellular carcinoma secondary to chronic hepatitis B, developed pneumonia and was transferred to Severance Hospital. At the time of admission, the patient's initial exam showed hypothermia, tachypnea, and hypotension. His chest radiograph showed severe pneumonia and a large abscess on left upper lobe. Under the presumptive diagnosis of bacterial pneumonia or other opportunistic infection, we started broad spectrum antibiotics. However, he developed Nocardia sepsis, rapidly deteriorated, and subsequently died.
