PikD is a widely known pathway-specific regulator for controlling pikromycin production in Streptomyces venezuelae ATCC 15439, which is a representative of the large ATP-binding regulator of the LuxR family (LAL) in Streptomyces sp. RapH and FkbN also belong to the LAL family of transcriptional regulators, which show greatest homology with the ATP-binding motif and helix-turn-helix DNA-binding motif of PikD. Overexpression of pikD and heterologous expression of rapH and fkbN led to enhanced production of pikromycin by approximately 1.8-, 1.6-, and 1.6-fold in S. venezuelae, respectively. Cross-complementation of rapH and fkbN in the pikD deletion mutant (ΔpikD) restored pikromycin and derived macrolactone production. Overall, these results show that heterologous expression of rapH and fkbN leads to the overproduction of pikromycin and its congeners from the pikromycin biosynthetic pathway in S. venezuelae, and they have the same functionality as the pathwayspecific transcriptional activator for the pikromycin biosynthetic pathway in the ΔpikD strain. These results also show extensive "cross-communication" between pathway-specific regulators of streptomycetes and suggest revision of the current paradigm for pathwayspecific versus global regulation of secondary metabolism in Streptomyces species.

The redundancy allocation problem (RAP) is a famous NP.complete problem that has beenstudied in the system reliability area of ships and airplanes. Recently meta-heuristic techniques have been applied in this topic, for example, genetic algorithms, simulated annealing and tabu search. In particular, tabu search (TS) has emerged as an efficient algorithmic approach for the series-parallel RAP. However, the quality of solutions found by TS depends on the initial solution. As a robust and efficient methodology for the series-parallel RAP, the hybrid metaheuristic (TSA) that is a interactive procedure between the TS and SA (simulated annealing) is developed in this paper. In the proposed algorithm, SA is used to find the diversified promising solutions so that TS can re-intensify search for the solutions obtained by the SA. We test the proposed TSA by the existing problems and compare it with the SA and TS algorithm. Computational results show that the TSA algorithm finds the global optimal solutions for all cases and outperforms the existing TS and SA in cases of 42 and 56 subsystems.
This study was carried to develop the new material of functional food which antioxidant of natural substances of sprouts. We compared antioxdiative activity and antioxidant substances exist in sprouts. Antioxidaive activities of sprouts were measured by total polyphenolic acid contents, electron donating activity(EDA), SOD-liked activity and hydroxyl radical scavenging ability. Total polyphenolic acid content was very higher in Rape sprouts extract(Rap) than other sprout extracts. Also Rape extract was showed the most excellent antioxdiative activity in SOD liked activity at 86.94%. The EDA was ordered Rape sprouts extract(Rap)>Tatsoi sprouts extract(Tat)>Broccoli sprouts extract(Bro)>Alfalfa sprout extract(Alf). Hydroxy radical scavenging ability was the most effective in Rape sprouts extract(Rap). Therefore we could be certain that Rape sprouts extract(Rap) was the most effective in antioxidative activity from sprouts.
PURPOSES : This research was a laboratory study for evaluating the Reclaimed Asphalt Pavement (RAP) mixture added developed rejuvenator for warm mix recycling. Waste asphalt mixtures occupy about 18.2% of construction wastes in Korea. Moreover, most rejuvenators were imported from Europe or the U.S. Therefore, improving usage of RAP with a developed rejuvenator material provides environmental protection at a reduced cost. METHODS : The specimen used for this experiment was performed by only using RAP. A suitable rejuvenator for Target PG was then added. In addition, a conventional rejuvenator was selected to compare performance and specimens introduced with the same procedure as the developed rejuvenator was prepared. In order to evaluate rutting resistance and water susceptibility, we conducted a deformation strength test, a tensile strength ratio test, and a dynamic immersion test with the prepared mixtures. RESULTS :Laboratory test results indicated that both the developed additive and conventional additive improved performance of the recycled asphalt mixtures compared to mixtures without the rejuvenator. In addition, the deformation strength test and TSR test results satisfied standards for domestic recycling asphalt mixtures. The dynamic immersion test showed that the developed rejuvenator has superior scaling resistance than the conventional rejuvenator. CONCLUSIONS : In terms of rutting resistance and moisture susceptibility, the warm mix recycled asphalt mixtures with the developed rejuvenator appeared to effectively recovered performance.
Background: Recurrent abdominal pain (RAP) is one of the frequent complaints in general practice, particularly in pediatrics and is among the common cause of referral to gastroenterology clinics. Purpose: This study is designed to investigate the effects of probiotics for the treatment of RAP and desired therapeutic outcomes. Methods: One hundred twenty-five children with the diagnosis of RAP according to Rome III criteria for irritable bowel syndrome (IBS), functional abdominal pain (FAP), functional dyspepsia (FD), and abdominal migraine (AM), were enrolled in this double-blind randomized controlled trial. Results: Sixty-five subjects received probiotics, and others received placebo treatment for 4 weeks. Lactobacillus reuteri was therapeutically effective in 32 patients compared to 8 patients, responding to the placebo treatment. Compared to baseline, all pain-related variables showed a significant reduction for the IBS and FD at the end of the 4th week. However, it did not respond well in FAP and AM groups. Pain-related outcomes such as, frequency of the pain, severity, and duration of the pain were decreased following the probiotic treatment. No therapeutic response was seen in AM group after the administration of probiotics. L. reuteri significantly led to pain relief in the overall population, and also in FAP, FD, and IBS subgroups. Conclusion: L. reuteri probiotics are likely to lead to RAP relief and can be recommended for the treatment of functional gastrointestinal disorders.
Babesia bovis rap-1 and B equi ema-1 intergenic(IG) nucleotides were analyzed and compared for identifying putative promoter sites using computer programs. The reason to initiate this research was to determine if IG nucleotides of Babesia genes that are predicted to be involved in erythrocyte invasion have functions regulating gene transcription and translation, which can be applied to functional gene knockout. Four IG sequences used included BbIG5(B bovis rap-1 5' IG), BblG3(B bovis rap-1 3' IG), BeIG5(B equi ema-1 5' IG) and BeIG3(B equi ema-1 3' IG). BbIG5 contained a putative promoter at nucleotide 197-246 with a predicted TATA-box and a transcription start site. BbIG3 had a putative promoter at nucleotide 270-320 with two predicted TATA-boxes and a transcription start site. BeIG3 had a putative promoter at nucleotide 155-205 with a predicted TATA-box and a transcription start site. Putative promoter sites in these three sequences mentioned above were identified with score cutoff 0.8, which means detection of about 40% recognized promoters with 0.1-0.4% false positives. In contrast, BeIG5 had a putative promoter at nucleotide 163-213 with score cutoff 0.8, but neither TATA-box nor transcription start site were recognized. However, BeIG5 had a putative promoter at nucleotide 388-438 with a predicted TATA-box and a transcription start site when score cutoff was decreased to 0.18, which means detection of about 70% recognized promoters with 2.2-5.3% false positives. These sequences with putative promoters can be tested if they have functions regulating gene transcription and translation.
PURPOSES : The purpose of this study was to determine the optimum mix design of the content of 100 % reclaimed asphalt pavement (RAP) for spray injection application with different binder types. METHODS : Literature review revealed that spray injection method is the one of the efficient and economical methods for repairing a small defective area on an asphalt pavement. The Rapid-Setting Polymer modified asphalt mixtures using two types of rapid setting polymers-asphalt emulsion and a quick setting polymer asphalt emulsion-were subjected to the following tests to determine optimum mix designs and for performance comparison: 1) Marshall stability test, 2) Retained stability test, 3) Wet track abrasion test, and 4) Dynamic stability test. RESULTS and CONCLUSIONS : Type A, B, and C emulsions were tested with different mix designs using RAP aggregates, to compare the performances and determine the optimum mix design. Performance of mixtures with Type A emulsion exceeded that of mixtures with Type B and C emulsion in all aspects. In particular, Type A binder demonstrated the highest performance for WTAT at low temperature. It demonstrated the practicality of using Type A mixture during the cold season. Furthers studies are to be performed to verify the optimum mix design for machine application. Differences in optimum mix designs for machine application and lab application will be corrected through field tests.
본 연구는 준고온 공법을 이용한 재생 아스팔트 콘크리트의 강도특성을 평가하기 위해 이루어졌다. 굵은 골재 최대치수 13mm의 화강암과 침입도 60-80인 신규 바인더 60-80을 재생 혼합물을 제조하는데 사용하였다. 배합설계는 RAP(굵은 입자 : 잔입자=6 : 4) 첨가비율 20%와 30%를 사용하였고 GPC, 침입도, 절대점도, 동점도를 준고온 첨가제(Evotherm와 Sasobit)의 첨가 함량을 결정하기 위하여 측정하였다. LD(low-density poly ethylene)를 본 연구에서 준고온 재생 아스팔트 혼합물의 개질제로 사용하였다. 본 연구에서는 8개의 준고온 재생 혼합물(2 RAP함량 ${\times}$ 2 준고온 첨가제 ${\times}$ 2 개질제)뿐만 아니라 2개의 일반 재생 혼합물, 1개의 가열혼합 일반혼합물(control)까지 총 11개의 혼합물을 제조하였다. 변형강도 시험, 간접인장강도 시험, 수분민감성 시험, wheel tracking을 통한 소성변형 시험을 준고온 재생혼합물의 기본 특성을 평가하기 위하여 수행하였다.
Purpose: The purpose of this study was to identify the risk factors for recurrent abdominal pain (RAP) in children who presented with nonorganic acute abdominal pain. Methods: A retrospective, single study was conducted on 2-15-year-old children diagnosed with nonorganic acute abdominal pain at the pediatric outpatient department of Vajira Hospital, Nawamindradhiraj University, between January 2015 and December 2019. The potential risk factors were analyzed using univariate and multivariate analyses. Results: Of the 367 patients with nonorganic acute abdominal pain, 94 (25.6%) experienced RAP within three months. In this group with RAP, 76 patients (80.8%) were diagnosed with functional gastrointestinal disorders, including functional dyspepsia, irritable bowel syndrome, functional abdominal pain-not otherwise specified, and functional constipation. History of gastrointestinal infection (p=0.011), mental health problems (p=0.022), abdominal pain lasting ≥7 days (p<0.001), and change in stool frequency (p=0.001) were the independent risk factors associated with RAP in children with nonorganic acute abdominal pain; their odds ratios and 95% confidence intervals were 3.364 (1.314-8.162), 3.052 (1.172-7.949), 3.706 (1.847-7.435), and 2.649 (1.477-4.750), respectively. Conclusion: RAP is a common problem among children who first present with nonorganic acute abdominal pain. The identification of risk factors may provide proper management, especially follow-up plans for this group in the future.
There are several methods determining an appropriate performance grade of virgin binder, which is re(erred as design binder, in RAP contained bituminous mixture design process. However, difficulties have been experienced in utilizing the methods in the field application in Korea, because SHRP binder test, the key tests to determine a design binder, requires well-trained personnel, high price equipment, and time consuming process. Thus, the study investigated the relationship among the binder aging level, RAP contents, and rheologica1 properties of binder. The study results provide mix designer with a simple method in selecting an appropriate grade of virgin binder.
