Detection of Clavibacter michiganensis subsp. michiganensis Assisted by Micro-Raman Spectroscopy under Laboratory Conditions

  • Clavibacter michiganensis subsp. michiganesis (Cmm) is a quarantine-worthy pest in $M{\acute{e}}xico$. The implementation and validation of new technologies is necessary to reduce the time for bacterial detection in laboratory conditions and Raman spectroscopy is an ambitious technology that has all of the features needed to characterize and identify bacteria. Under controlled conditions a contagion process was induced with Cmm, the disease epidemiology was monitored. Micro-Raman spectroscopy ($532nm\;{\lambda}$ laser) technique was evaluated its performance at assisting on Cmm detection through its characteristic Raman spectrum fingerprint. Our experiment was conducted with tomato plants in a completely randomized block experimental design (13 plants ${\times}$ 4 rows). The Cmm infection was confirmed by 16S rDNA and plants showed symptoms from 48 to 72 h after inoculation, the evolution of the incidence and severity on plant population varied over time and it kept an aggregated spatial pattern. The contagion process reached 79% just 24 days after the epidemic was induced. Micro-Raman spectroscopy proved its speed, efficiency and usefulness as a non-destructive method for the preliminary detection of Cmm. Carotenoid specific bands with wavelengths at 1146 and $1510cm^{-1}$ were the distinguishable markers. Chemometric analyses showed the best performance by the implementation of PCA-LDA supervised classification algorithms applied over Raman spectrum data with 100% of performance in metrics of classifiers (sensitivity, specificity, accuracy, negative and positive predictive value) that allowed us to differentiate Cmm from other endophytic bacteria (Bacillus and Pantoea). The unsupervised KMeans algorithm showed good performance (100, 96, 98, 91 y 100%, respectively).

Standoff Raman Spectroscopic Detection of Explosive Molecules

  • We developed a standoff Raman detection system for explosive molecules (EMs). Our system was composed of reflective telescope with 310 mm diameter lens, 532 nm pulse laser, and Intensified Charge-Coupled Device (ICCD) camera. In order to remove huge background noise coming from ambient light, laser pulses with nanosecond time width were fired to target sample and ICCD was gated to open only during the time when the scattered Raman signal from the sample arrived at ICCD camera. We performed standoff experiments with military EMs by putting the detector at 10, 20 and 30 m away from the source. The standoff results were compared with the confocal Raman results. Based on our standoff experiments, we were able to observe the peaks in the range of 1200 and $1600cm^{-1}$, where vibrational modes of nitro groups were appeared. The wave numbers and shapes of these peaks may serve as good references in detecting and identifying various EMs.

  • FT-IR and FT-Raman spectra were measured for 15Li$_{2}$O.3Al$_{2}$O$_{3}$.78SiO$_{2}$. 4K$_{2}$O glass system after UV irradiations. Optimum UV irradiation time of Li$_{2}$O.SiO$_{2}$ crystalline phase was 60 seconds and crystalline phase of Li$_{2}$O.SiO$_{2}$ was leached out on 5% HF. 977 cm$^{1}$ band of FT-Raman spectra can be attributed to two-non bridging oxygen in unit cell for 1 hour and optimum crystallization was confirmed for 3 hrs, 630.deg. C.

Label-Free Molecular Imaging of Living Cells

  • Optical signals based on Raman scattering, coherent anti-Stokes Raman scattering (CARS), and harmonic generation can be used to image biological molecules in living cells without labeling. Both Raman scattering and CARS signals can be used to detect frequencies of molecular vibrations and to obtain the molecular distributions in samples. Second-harmonic optical signals can also be generated in structured arrays of noncentrosymmetric molecules and can be used to detect structured aggregates of proteins, such as, collagen, myosin and tubulin. Since labeling techniques using chemical and biological reactions may cause undesirable changes in the sample, label-free molecular imaging techniques are essential for observation of living samples.

  • In this study, PLZT stock solutions were prepared by sol-gel processing to fabricate PLZT thin films. The stock solutions were spin-coated on ITO-glass and the film were annealed by rapid thermal annealing(RTA). The variation of tile crystallographic structure of the thin films and the phase transition with respect to it were observed using Raman spectra. Raman result showed that the band of spectra are broad as the amount of Zr substitution increased and specially, abrupt change occurs in the raman spectra upon crossing the tetragonal-rhombohedral phase boundry at 2/55/45 PLZT thin film. So, the fact that the crystallographic structure was transitted from tetragonal to rhombohedral structure was certified.

The pH Dependence of Metal Tetrakis (4-sulfonato-phenyl) porphine Structure Probed by Raman Spectroscopy

  • The pH dependence studies of Raman spectra are reported for water-soluble free-base, Zn, Co and Cu tetrakis (4-sulfonatophenyl) porphine in pH 4, pH 7 and pH 13.9 aqueous solution. For free base porphine, the substantial differences are found in absorption and Raman spectra between pH 4 and pH 7 or pH 10 aqueous solutions due to the protonation at low pH. For Zn and Co porphyrins, the hydrolysis equilibrium constants are obtained by spectrophotometric titration experiments. The consistent shifts in Raman frequencies are found at high pH due to the hydrolysis. For Cu porphyrins, instead of hydrolysis the aggregation effect is detected at high pH through the absorption and Raman studies.

  • The compounds associated with corrosion, in metallic archaeological samples of carbon steel of insular origin were evaluated to establish their degree of deterioration and structural damage against air pollution. The iron phases present in samples of archaeological artifacts were detected by Raman spectroscopy and confocal Raman microcopy. These samples mainly exhibited ing mainly ${\beta}$-FeO(OH) type goethite oxyhydroxides and small amounts of akaganeite ${\alpha}$-FeO(OH) lepidocrocite ${\gamma}$-FeO(OH) due to dominant chloride in a marine environment and non-stoichiometric oxyhydroxides Fe (II + / III +) as indicators of early corrosion. Some parts showed the presence of magnetic maghemite indicating high corrosion. ${\gamma}$-FeO(OH) is a precursor of phases associated with advanced marine corrosion. By studying its decomposition by Raman spectroscopy, it was synthesized with the following sequence: ${\gamma}-FeO(OH){\rightarrow}{\alpha}-FeO(OH)+{\gamma}-FeO(OH)$, ${\rightarrow}{\gamma}-Fe_2O_3+Fe_3O_4$. Ferric compounds provided evidence for the effect of intensity of laser on them, constituting a very useful input for the characterization of oxidation of iron in this type of artifacts. Thus, destructive analysis techniques should be avoided in addition to the use of small amounts of specimen.

  • 용융법으로 제작된 인산염 유리시편의 조성변화에 따른 밀도와 물에 대한 안정성 및 각각의 구조변화를 조사하기 위해 Raman과 적외선 흡수 스펙트럼이 측정되었다. 그 결과 $P_2O_5$가 60mol% 부근에서 밀도와 물에 대한 안정성이 급격히 증가하는 구조적인 이상성을 나타내었다. 이러한 변화는 $Mg^{2+}$ 이온을 중심으로 DBO(Double-Bonded Oxygen)이 배위하면서 유리구조의 강한 수축을 유발시켜 나타난 구조변화에 기인하는 것으로, $Mg^{2+}$ 이온을 배위하고 있는 DBO은 이중결합의 성격을 잃어버리고, 다른 하나의 NBO(Non-Bonding Oxygen)과 함께 공명구조를 이룬다고 생각된다.

  • (111) 및 (100) 단결정 실리콘 웨이퍼에 대하여 #140 mesh와 #600 mesh의 연삭숫돌을 사용하여 연삭가공을 행하고 가공에 의한 표면품위의 변화를 관찰하였다. 이를 위하여 Atomic Force Microscope(Am)을 사용하여 미세거칠기를 분석하고 라만 분광법(Raman spectroscopy)을 통하여 결정구조, 상변화 및 잔류응력을 분석하였다. 그 결과 연삭가공 후에 (111) 면에서 미세거칠기가 더 크게 나타났으며 실리콘이 다이아몬드구조의 Si-I에서 Si-III(body centered tetragonal) 및 Si-XII(rhombohedral)로 전이하는 현상을 라만스펙트럼 분석을 통해 확인하였다. 또한 연삭가공에 의하여 패인 구덩이에서는 일반적인 가공표면과 다른 라만스펙트럼이 관찰되어, 표면에 인가된 잔류응력이 새로운 표면생성으로 해소되었음을 알수 있었다. 또한 열처리에 의한 재료의 표면품위개선효과를 분석하기 위하여 대기 중에서 열처리를 시행한 결과 열처리는 잔류응력의 해소와 상전이의 회복에 효과적인 것으로 나타났다.

Raman Spectroscopy of Irradiated Normoxic Polymethacrylic Acid Gel Dosimeter

  • A quantitative analysis of the decreasing rate of the monomer and increasing rate of the polymerization was made by monitoring radiation level increments using Raman spectroscopy within the therapeutic radiation range for a normoxic polymethacrylic acid gel dosimeter. The gel dosimeter was synthesized by stirring materials such as gelatin, distilled water, methacrylic acid, hydroquinone and tetrakis phosphonium chloride at $50^{\circ}C$, and the synthesized gel was contained in a 10- mm diameter and 32-mm high vial to conduct measurement. 24 hours after gel synthesis, it was irradiated from 0 Gy to 20 Gy by 2 Gy using a Co-60 radiotherapy unit. With use of the Cryo FE-SEM, structural changes in the 0 Gy and 10 Gy gel dosimeters were investigated. The Raman spectra were acquired using 532-nm laser as the excitation source. In accordance with fitting the changes in C-COOH stretching (801 $cm^{-1}$), C=C stretching (1639 $cm^{-1}$) and vinyl $CH_2$ stretching (3114 $cm^{-1}$) vibrational modes for monomer and $CH_2$ bending vibrational mode (1451 $cm^{-1}$) for polymer, sensitive parameter S for each mode was calculated. The values of S for monomer bands and polymer band were ranged in $6.0{\pm}2.6$ Gy and $7.2{\pm}2.3$ Gy, respectively, which shows a relatively good conformity of the decreasing rate of monomer and the increasing rate of polymerization within the range of error.
