  • 유기질비료 시용이 봄 및 가을 작형에서 무 품종의 생육 및 수량에 미치는 영향을 알아보고자 본실험을 실시한 바 다음과 같은 결과를 얻었다. 1) 무의 엽장, 엽중, 근장, 근경, 근중등 생육은 봄, 가을작형 모두에서 3요소, 발효계분 + 1/5 량NK추비, 발효계분, 계분, 무처리구 순으로 촉진하는 경향이었으며, 엽의 chlorophyll 함량과 질소함량도 생육에서와 같은 처리순으로 증가하였다.2) 봄 및 가을작형의 수량에 있어 발효계분 450kg 처리구는 3요소구에 비해 저하되는 경향이었으나 전국평균 수량을 상회하였다.3) 발효계분처리로 4회연속재배한 경우보다 1회재배한 경우보다 3요소처리구와의 무 생육차가 감소하였다.4) 무 재배후 토양의 질소 및 마그네슘함량은 처리간 비슷한 경향이었으며 pH, 유기물함량과 인산 및 칼슘함량은 3요소구에 비해 발효계분에서 높은 경향이었다.

Application of SCAR markers to self-incompatibility genotyping in breeding lines of radish (Raphanus sativus L.)

  • Self-incompatibility (SI) prevents self-fertilization by inhibiting the pollen tube growth of self-pollen. Molecular analysis has revealed that the S locus comprises a number of genes, such as the S-locus glycoprotein (SLG), the S-locus receptor kinase (SRK), and SP11 (SCR). Although molecular markers related to those genes have been developed, a simple S-haplotype detecting method has not been reported due to the highly polymorphic and relatively small coding regions. In this study, the sequence characterized amplified region (SCAR) markers were used to establish an efficient radish genotyping method. We identified the S-haplotypes of 192 radish accessions using 19 different markers, which proved to be highly reliable. The accessions were assigned to 17 types of S-haplotypes, including 8 types of SRKs and 9 types of SLGs. Since the developed SCAR markers are based on their gene sequences, we could easily identify the S-haplotypes by a single specific band, with the highest frequencies detected for SLG 5, SRK 1, and SLG 1, in order. Among the tested markers, the SLG 1, SRK 1, and SRK 5 markers exhibited high reliability, compared to phenotypic results. Furthermore, we identified the seven types of unreported SLGs using SLG Class -I and -II specific markers. Although the developed SCAR markers still need to be improved for the genotyping of all S-haplotypes, these markers could be helpful for monitoring inbred lines, and for developing the MAS in radish breeding programs.

Alternaria brassicicola에 의한 무 검은무늬병에 대한 효율적인 저항성 검정법 개발 (Development of an Effective Method for Testing Resistance to Black Spot of Radish Caused by Alternaria brassicicola)

  • 본 연구는 Alternaria brassicicola에 의해 발생하는 무 검은무늬병에 대한 효율적인 저항성 검정법을 확립하기 위하여 수행되었다. A. brassicicola 7균주의 포자 형성량과 병원성을 검정하였다. 실험한 균주 중, A. brassicicola KACC 40036과 43923 균주는 V-8 juice agar 배지에서 가장 많은 포자를 생산하였다. 그리고 실험한 균주 모두는 무 유묘에 대하여 강한 병원력을 나타냈다. 시판 중인 무 61개 품종의 A. brassicicola KACC 40036 균주에 대한 저항성 스크리닝을 수행하였다. 실험한 품종 중 높은 저항성을 보이는 품종은 없었으나 '금봉'과 '새롬'같이 약한 저항성을 나타내는 품종이 발견되었다. 이들 결과로부터 추후 실험을 위해 저항성 반응에 차이를 보이는 4개 품종을 선발하였다. 이들 4개 품종의 접종하는 무의 생육 시기, 접종원 농도 그리고 접종 후의 재배 온도에 따른 검은무늬병 발생을 조사하였다. 실험한 모든 품종은 어린 유묘일수록 검은무늬병에 대한 감수성이 증가하였으며, 더 많은 A. brassicicola 포자를 접종할수록 검은무늬병이 더 많이 발생하였다. 그리고 접종한 유묘를 $25^{\circ}C$의 습실상에서 배양하였을 때에는 $20^{\circ}C$$30^{\circ}C$에서 배양하였을 때보다 검은무늬병 발생이 증가하였다. 이들 실험의 결과로부터 무 품종의 검은무늬병에 대한 저항성을 검정하기 위해서는 무 종자를 파종하고 온실($25{\pm}5^{\circ}C$)에서 16일 동안 재배하고, 이 유묘에 A. brassicicola 균주의 포자현탁액($2.0{\times}10^5spores{\cdot}mL^{-1}$)을 분무하여 접종하고, 접종한 식물은 $25^{\circ}C$ 습실상에 24시간 동안 배양한 후에 $25^{\circ}C$, 상대습도 80%의 생육상에서 재배하고, 접종 2일 후에 병반면적율을 조사하는 것을 제안한다.

Antioxidant Activity and Anti-inflammatory Effects of Raw and Processed Fruits and Vegetables

  • In this study we investigated antioxidant and anti-inflammatory activities of Malus Domestica (apple), Pyrus Communis L. (pear), Daucus carota L. (carrot), Brassica oleracea var. (broccoli), Brassica oleracea var. capitata (cabbage) and Raphanus sativus L. (radish), that were obtained from local market. As these are common fruits and vegetables that are widely consumed, we aimed to investigate their beneficial properties especially the antioxidant and anti-inflammatory. The samples were processed by an indirect heating method and their properties were compared to their raw forms. Based on DPPH and ABTS assay, processed samples showed better antioxidant activity compared to raw samples, and processed pear sample had the best antioxidant activity. The anti-inflammatory activities of the samples were also investigated in LPS-treated RAW 264.7 cells. The mRNA expressions of pro-inflammatory mediators and cytokines (iNOS, COX-2, $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) were assessed by RT-PCR. Processed samples exhibited better inhibition of iNOS, compared to the raw forms. Processed broccoli and cabbage samples exhibited outstanding anti-inflammatory effects. The samples did not exhibit cytotoxicity against RAW 264.7 cells up to 1mg/ mL as shown in the cell viability assay. Taken together, processed broccoli and cabbage samples exhibited the strongest anti-inflammatory properties.

무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구 (Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. 9월 파종 가을무의 파종시기에 따른 생장 반응 (The Growth of Fall Varieties of Radish (Raphanus sativus L). Sowing in September)

  • 9월 만파시 가을무의 생육과 품종간차를 알아보는 한편 멀칭과 무멀칭재배의 차이를 조사하여 김장무의 파종한계기 구명 및 원활한 수급을 위한 기초 자료를 얻고자 본 실험을 실시하였다. 일반 가을무는 수원지방에서 9월 8일 파종시 무멀칭재배는 550g미만이었고 P.E. 멀칭재배에서는 대형추석이 718g, 백경이 604g으로 나타났다. 부산지방은 9월 16일 파종, 무멀칭재배에서는 11월 23일 수확시 백경이 655g, 대형추석이 813g을 보였고 P.E. 멀칭재배에서는 모두 1kg이상의 근중을 나타냈다. 멀칭의 효과는 대형추석이 백경보다 컸다. 알타리무는 중부지방중 한지에서는 9월 14일경, 난지는 9월 18일 경에도 파종이 가능할 것으로 추정되며 파종기가 늦을수록 근형형성이 빠른 총각알타리가 유리하였다. 부산은 9월 26일까지 파종을 실시한 결과 어느 품종이나 재배가 가능하였다.

Changes in Nutrient Levels of Aqueous Extracts from Radish (Raphanus sativus L.) Root during Liquefaction by Heat and Non-heat Processing

  • The amount of cellular components including soluble sugars, amino acids, organic acids and glucosinolates (GLS) was investigated during radish root processing to develop a radish beverage. The radish root was divided into two parts, white and green tissue, and processed separately by extracting the juice from the fresh tissue and from the boiled tissue to compare differences in the components content among the preparations. The overall palatability of both the fresh and boiled extracts from the green part of the radish was higher than that of the same extracts from the white part. The sweetness of extract by boiling increased and its pungency decreased, thereby the palatability increased by being compared to the fresh radish extract. The sweetness was affected by sucrose not by glucose or fructose of monosaccharides by showing different sucrose contents according to treatment comparing palatability. Malic acid was identified as primary organic acid, and the content was higher in both the fresh and boiled extracts from the white part than in the extracts from the green part of the radish. The fresh extract from the green part of the radish contained more essential amino acids, such as threonine and valine, and more hydrophilic amino acids including glutamic acid, aspartic acid, and arginine than those of the fresh extract from the white part, suggesting the green fresh part is more palatable than the white fresh part. The main sulfur compound was ethylthiocyanate in radish, and others were butyl isothiocyanate, dimethyl-disulfide, and 4-methylthio-3-butylisothiocyanate. The four GLS were detected much more in the fresh green and fresh white parts of the radish because they evaporated during boiling. The contents of the four sulfur compounds were higher in the white fresh part than in the green fresh part, which is likely the reason the pungency was higher and the palatability was lower in the white fresh part than in the green fresh part of the radish. The ascorbic acid content was higher in the fresh extract compared to the boiled extracts from both the green and white parts. Taken together, these findings indicate that fresh radish extract is superior to obtain in terms of retaining desirable nutritional and functional components for health.

유용 Pseudomonas 종의 근면점유와 무우 Fusarium시들음병의 억제에 관한 생물학적 정량 (Root Colonization by Beneficial Pseudomonas spp. and Bioassay of Suppression of Fusarium Wilt of Radish)

  • 무우품종(Raphanus sativus L.) Saxa Nova에 시들음병을 일으키는 Fusarium oxysporum f. sp. raphani (FOR)에 대해 저항성을 증가시켜 방제효과를 얻기 위하여 식물성장을 증진시키는 것으로 알려진 Pseudomonas florescens WCS374 (WCS 374), P. putida RE10 (RE10) 및 Pseudomonas sp. EN41S (EN415)을 병원균처리 토양에 단독 또는 혼합처리하여 4주간 폿트배양한 후 무우에 나타나는 외부 및 내부병징을 조사하여 처리세균에 의한 병억제 효과를 측정하였다. 내부 및 외부병징으로 대조구는 각각 46.5% 및 21.1%를 나타내었고, RE10처리는 내외병징이 각각 12.2%와 7.8%로 발병이 억제됨을 알 수 있었다. 그러나 발병억제력이 높다고 알려진 WCS374는 내외 병징이 각각 45.6%와 27.8%로 나타났다. 한편 RE10균주를 WCS374 또는 EN415 균주와 혼합하여 처리할 경우 내외 병징이 10.0-22.1% 정도이었고 EN415와 혼합처리하면 7.8-20.2%의 병징을 나타낸다. FOR의 뿌리점유율은 뿌리에서 $2.4-5.1{\times}10^3/g$였고 토양내 분포수는 $0.7-1.3{\times}10^3/g$이었다. 대조구는 뿌리에서 $3.8{\times}10^3/g$였으며 RE10의 처리는 $2.9{\times}10^3/g$로 분포수가 적었고, 3종 세균의 혼합처리는 $5.1{\times}10^3/g$으로 많이 관찰되었으나, 처리간에 통계적 차이는 없었다. 토양에서 관찰된 FOR은 부리부분 보다 그 분포 수가 적었다. 처리된 3가지 세균은 뿌리에서 $2.3-4.0{\times}10^7/g$ 범위이고, 토양에서는 $0.9-1.8{\times}10^7/g$으로 뿌리에서 관찰된 수 보다 적게 분포하였다. 뿌리부분에서 대조구나 RE10의 처리토양은 형광성 Pseudomonas spp.의 분포수가 적고 처리간에는 통계적 차이를 나타냈으나, 토양조사에서 이세균은 큰 차이를 나타내지 않았다. 뿌리에 처리된 세균과 FOR의 분포수는 토양에 처리된 것과 대조하였을 때 많았으며, 이 실험 에 사용된 RE10과 EN415은 Fusarium시들음병에 대한 기주의 저항성을 유도하는 것으로 생각된다.

  • 5개 무 품종의 종자를 3시간 물에 침지한 후 cellulose 분말을 코팅하여 UV 광하에서 sinapine 누출정도에 따른 형광색의 분포정도에 따라 무형광(NF)종자, 일부형광(PF)종자, 완전형광(FF)종자로 구분하고, 또 이들 종자와 발아한 유묘의 외부형태를 주사전자현미경(SEM), 광학현미경 및 육안으로 관찰하였다. 종자의 발아율은 무형광(NF)종자>일부형광(PF)종자>완전형광(FF)종자 순으로 낮았으며, 무형광(NF)종자는 종피조직이 치밀하고 둥근 모양이었으나, 일부형광(PF) 및 완전형광(FF)종자는 종피가 주름지거나 상처가 있는 것이 대부분이었다. 또 무형광(NF)종자에서 나온 유묘의 떡잎과 배축은 정상이었으나, 일부형광(PF) 및 완전형광(FF)종자에서 나온 유묘의 떡잎에는 조직이 괴사한 흔적이 많았으며 떡잎과 배축의 크기가 작고 비정상적이었다.

HeLa세포에서 Helicobacter pylori 독소에 의한 공포형성에 미치는 무의 효과 (Effect of Radish on HeLa Cell Vacuolation Induced by Helicobacter pylori Cytotoxin)

  • Helicobacter pylori (H. pyroli) infection it associated with type B gastritis, peptic uler disease, and gastric cancer. The vacuolation of cells induced by H. pylori is thought to be essential for the initiation and maintenance of gastric infection. The roles of H. pylori cytotoxin, urease, and ammonia in the vacuolation of HeLa cells were determined. H. pylori toxin induced vacuolation of HeLa cells. Korean and Japanese radishes significantly prevented the vacuolation of HeLa cells induced by H. pylori toxin. The urease activity in vacuolated cells was also decreased with Korean and Japanese radishes. H. Pylori toxin-induced vacuolation was inhibited by vacuolar type ATPase inhibitors (bafilomycin and N-ethylmaleimide). However, further investigation is required to determine the mechanisms of radish for the inhibition of vacuole formation of eukaryotic cells in response to the H. pylori toxin.
