Tocotrienols (T3) are minor plant constituents found abundantly in rice bran, which provide a significant source of vitamin E in animal feeds. T3 was reported to have an intrinsic hypocholesterolemic effect by inhibiting HMG-Co A reductase. It has similar antioxidative properties as tocopherols in food and biological system due to their similar chemical structures. However, the antioxidant activity and mechanism of T3 to scavenge free radicals and to inhibit the peroxidation of linoleic acid are less understood. The purpose of this study was to investigate the scavenging effect of T3 on free radicals and its inhibition of peroxide formation. Free radical scavenging activity was monitored by the DPPH (1,1-diphenyl-2-picrylhydrazyl) method whereas inhibition of linoleic acid peroxidation was evaluated using the thiocyanate method. Thiobarbituric acid (TBA) test was used to determine malonaldehyde formation from linoleic acid peroxidation. Free radical scavenging activity increased with increasing concentration levels of T3. T3 exhibited 38.2, 78.6, 92.7 and 96.2% radical scavenging activity at concentrations of 2, 8, 32 and 128 ppm, respectively. At 128 ppm, it was highly effective in inhibiting linoleic acid peroxidation. The activity of T3 evaluated by the thiocyanate method showed low absorbance values indicating a high level of antioxidant activity. All treatments showed similar trends in antioxidant activity when evaluated by both the thiocyanate method and TBA test.
The objectives of this study were to investigate the effect of fermentation medium on the hydroxyl radical scavenging activity of exopolysaccharides from Inonotus obliquus by response surface methodology (RSM). A two-level fractional factorial design was used to evaluate the effect of different components of the medium. Corn flour, peptone, and $KH_2PO_4$ were important factors significantly affecting hydroxyl radical scavenging activity. These selected variables were subsequently optimized using path of steepest ascent (descent), a central composite design, and response surface analysis. The optimal medium composition was (% w/v): corn flour 5.30, peptone 0.32, $KH_2PO_4$ 0.26, $MgSO_4$ 0.02, and $CaCl_2$ 0.01. Under the optimal condition, the hydroxyl radical scavenging rate (49.4%) was much higher than that using either basal fermentation medium (10.2%) and single variable optimization of fermentation medium (35.5%). The main monosaccharides components of the RSM optimized polysaccharides are rhamnose, arabinose, xylose, mannose, glucose, and galactose with molar proportion at 1.45%, 3.63%, 2.17%, 15.94%, 50.00%, and 26.81%.
The antioxidant activity of solvent extracts of leaves, trunk and root Pueraria Radix was determined by measuring the radical scavenging effect on l.l-diphenyl-2-picrylny drazyl(DPPH) radical. Extacts prepared from root showed radical scavenging effect on DPPH radical. But, extracts prepared from leaves and trunk did not show activity. The ethyl acetate ewtract of Pueraria Radix root showed radical scavenging activity at an IC 50 value of 75.7 $\mu\textrm{g}$/$m\ell$.
Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. The ethanol extracts of mixed vegetables (Bioactive Vegetables, BV), mixed fruits (Bioactive Fruits, BF) and their liquid formulation (Chungpae Plus , CP) were evaluated for their antioxidative and antigenotoxic activities. They were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed to show the inhibitory effect of lipid peroxidation as measured by malondialdehyde (MDA) formation although the potencies were not higher than those of vitamin C. They did not possess any pro-oxidant effect on bleomycin-Fe(III) dependent DNA degradation, whereas vitamin C showed strong pro-oxidant effect. Furthermore, oral administration of BV and BF inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. CP also showed significant inhibition under same experimental condition. Therefore, the liquid formulation (CP) containing BV and BF may be a useful natural antioxidative and antigenotoxic agent by scavenging free radicals, inhibition of lipid peroxidation and protecting chromosomal damage.
Quercetin and its glycosides showed a strong free radical scavenging effect to DPPH radical generation. However, there were not big differences between quercetin aglycone and glycosides under experimental condition of this study. On the other hand, quercetin had pro-oxidant effect in bleomycin-dependent DNA assay. Quercetin aglycone and its glycosides, quercitrin inhibited $H_2$$O_2$- induced DNA damage in CHL cells. They also have an anticlastogenicity toward DNA breakage agent by radical generation like bleomycin. These results indicate that quercetin aglycone and its glycosides are capable of protecting the free radical generation induced by reactive oxygen species like $H_2$$O_2$. The mechanism of inhibition in hydrogen peroxide-induced genotoxicity may be due to their free radical scavenging properties. Therefore, quercetin aglycone and its glycosides may be useful chemopreventive agents by protecting of free radical generation which are involved in carcinogenesis and aging. However, quercetin and its glycosides must also carefully examined for pro-oxidant properties before being proposed for use in vivo.
This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.
The optimization and microwave assisted extraction of stem bark of Terminalia arjuna, quantitative estimation of the marker compounds arjunic acid and arjunolic acid using HPTLC and the evaluation of free radical scavenging activity has been performed in this study. The central composite design was used for optimization and the values of parameters for optimized batch of microwave assisted extraction were 1000 W (Power), 3 minutes (Time) and 1/120 (Solid/solvent ratio). The solvent system to carry out the HPTLC was toluene: acetic acid: ethyl acetate (5: 5: 0.5) and quantitative estimation was done using standard equations obtained from the marker compounds. The in-vitro free radical scavenging activity was performed spectrophotometrically using ascorbic acid as standard. The value of estimated percentage yield of arjunic acid and arjunolic acid was 1.42% and 1.52% which upon experimentation was obtained as 1.38% and 1.51% respectively. The DPPH assay of the different batches of microwave assisted extraction and marker compounds taken suggested that the marker compounds arjunic acid and the arjunolic acid were responsible for the free radical scavenging activity as the batch having the maximum percentage yield of the marker compounds showed best free radical scavenging effect as compared to standard ascorbic acid. The $IC_{50}$ value of the optimized batch was found to be 24.72 while that of the standard ascorbic acid was 29.83. Hence, the yield of arjunic acid and arjunolic acid has direct correlation with the free radical scavenging activity of stem bark extract of Terminalia arjuna and have potential to serve as active lead compounds for free radical scavenging activity.

To develop the functional food, methanol extracts of dried plantain (Plantago asiatica L.) was fractionated in hexane, chloroform, ethyl acetate, butanol and aqueous fraction. DPPH radical scavenging effect, reducing power, nitrite scavenging effect and antimicrobial activities of these fractions were investigated. Ethyl acetate fraction among these fractions showed the highest effect of all experiments. Ethyl acetate fraction showed about 60% scavenging effect on the DPPH radicals at 400 $\mu$g/5 mL. Nitrite scavenging effect of ethyl acetate and butanol fraction was more than 70~80% at concentration above of 500 $\mu$g. Among various solvent fractions from methanol extracts of plantain, ethyl acetate fraction showed the highest inhibitory effect on the microorganism such as Staphylococcus aureus, Bacillus cereus and Vibrio parahaemolyticus at 5 mg/disc.
The radical scavenging activity of the methanol extracts was in the order of doenjang〉 meju〉 soybean. The metha-nol extracts were further fractionated with dichloromethane, ethyl acetate, butanol and water in sequence. Among the solvent fractions of the methanol extracts, the ethyl acetate fraction demonstrated high levels of radical scave-nging activity. On the other hands, the nitrite scavenging activity of the methanol extrats was in the order of doen-jang〉soybean〉meju. All fractions from doenjang were exhibited the strong nitrite scavenging effect. Especially, $H_2O$, EtOAc and BuOH fractions with 6gmg/10$m\ell$ concentration exhibited much stronger nitrite scavenging effect than that of L-ascorbic acid. The ethyl acetate fraction showed remarkable antioxidant and nitrite scavenging effects compared with the other fractions obtained from soybean, meju and doenjang. The ethyl acetate fraction of doenjang was fur-ther purified by repeated chromoatograpic methods and isolated three compounds. There are identified as genistin, dai-dzein and genistein on the basis of spectral data.
