Increased Catalase Activity by All-trans Retinoic Acid and Its Effect on Radiosensitivity in Rat Glioma Cells (백서 교종 세포에서 레티노인산에 의한 카탈라제의 활성 증가가 방사선감수성에 미치는 효과)

  • Purpose: It has been reported that all-trans retinoic acid (ATRA) can inhibit glioma growing in vitro. However, clinical trials with ATRA alone in gliomas revealed modest results. ATRA has been shown to increase radiosensitivity in other tumor types, so combining radiation and ATRA would be one of alternatives to increase therapeutic efficacy in malignant gliomas. Thus, we intended to know the role of catalase, which is induced by ATRA, for radiosensitivity if radiation-reduced reactive oxygen species (ROS) is removed by catalase, the effect of radiation will be reduced. Materials and Methods: A rat glioma cell line (36B10) was used for this study. The change of catalase activity and radiosensitivity by ATRA, with or without 3-amino-1, 2, 4-triazole (ATZ), a chemical inhibitor of catalase were measured. Catalase activity was measured by the decomposition of $H_2O_2$ spectrophotometrically Radiosensitivity was measured with clonogenic assay. Also ROS was measured using a 2, 7-dichlorofluorescein diacetate spectrophotometrically. Results: When 36B10 cells were exposed to 10, 25 and $50{\mu}M$ of ATRA for 48 h, the expression of catalase activity were increased with increasing concentration and incubation time of ATRA. Catalase activity was decreased with increasing the concentration of AT (1, $10{\mu}M$) dose-dependently. ROS was increased with ATRA and it was augmented with the combination of ATRA and radiation. ATZ decreased ROS production and increased cell survival in combination of ATRA and radiation despite the reduction of catalase. Conclusion: The increase of ROS is one of the reasons for the increased radiosensitivity in combination with ATRA. The catalase that is induced by ATRA doesn't decrease ROS production and radiosensitivity.

Flavonoids of Rosa roxburghii Tratt Act as Radioprotectors

  • Background: To study the radioprotective effects of flavonoids from Rosa roxburghii Tratt (FRT). Materials and Methods: The radioprotective effects of FRT were investigated by examining cell viability, 30-day survival of mice and the number of colony-forming units in spleen (CFU-S) after total-body 60Co irradiation. Results: The survival rates of irradiated cells gradually increased with increasing concentrations of FRT. The survival rate was the highest at 87% with a concentration of $30{\mu}g/mL$. Pretreatment with FRT was needed to realize its radioprotective activity in mice at the dose of 60 mg/kg. With the increasing doses of 30 mg/kg, 60 mg/kg and 120 mg/kg, the numbers of CFU-S increased, and were significantly different compared with the control group. Conclusions: Pretreatment with FRT prior to irradiation resulted in significantly higher cell survival at 24 h after 5 Gy radiation, increased 30-day survival in mice after exposure to a potentially lethal dose of 8 Gy, and resulted in a higher number of CFU-S in mice after exposure to a dose of 6 Gy. These results collectively indicate that FRT is an effective radioprotective agent.

Plasmid DNA damage by neutron and ${\gamma}-$ radiation (중성자 및 ${\gamma}-ray$ 조사에 따른 plasmid DNA 의 손상 관찰)

  • The plasmid was used pBR 322 and ${\varphi}X174$ RF DNA. In neutron experiment, damage of pBR 322 and ${\varphi}X174$ RF DNA were observed according to increasing concentration of BSH and neutron dose. Damage of plasmid DNA appeared obvious by increasing of BSH and neutron irradiation. In ${\gamma}-$ radiation experiment, it was carried out like above neutron experiment but damages of two plasmid appeared no differences from the control unlike neutron result.

  • Since the solar radiation is the main input for sizing any solar system, it will be necessary to understand and evaluate the insolation data. The horizontal global insolation data have been measured since May, 1982 and direct normal solar insolation data since December 1990 at 16 different sites all over the country and considerable effort has been made for constructing a standard value from measured data at each station. In the results, the average global total solar radiation of the nation is $3,055kcal/m^2.day(12.79MJ/m^2.day)$ and the average clear day direct normal solar beam radiation was $4,600kcal/m^2.day(19.26MJ/m^2.day)$, which indicates possible solar energy application of medium and high temperature technologies with high concentration.

Ecological Model Experiments of the Spring Bloom at a Dumping Site in the Yellow Sea (생태계모델을 이용한 황해투기해역에서의 춘계 식물플랑크톤 대증식 연구)

  • To explore limiting factors of spring bloom caused by waste disposal after dumping activity commenced in the Yellow Sea, we used a 1-dimensional temperature-ecological coupled model. The vertical structure of temperature and vertical diffusivity (Kh) are calculated by the temperature model with sea surface temperature using the 2.5 layers turbulence closure scheme. The ecological model applied results at the temperature model consisted of five state variables (DIN, DIP, phytoplankton, zooplankton, and detritus) forced by photosynthetically available radiation. We simulate year-to-year variations of plankton and nutrients using the coupled model from 1998 to 2000 and compare results of the model with observed data. It turned out that temperature is the growth factor of spring bloom in dumping area. During the winter the weak stratification made sufficient supply of the accumulated nutrients from the sea bed into the upper water column and led to the bloom in the coming spring. Radiation also turned out to be another important factor of spring bloom in the study area. Insufficient radiation of March 1999 showed low chlorophyll-a concentration despite sufficient nutrients in the surface.

NO Sensing Characteristics of ZnO Nanorod Prepared by Ultrasound Radiation Method (초음파 처리에 의해 합성된 ZnO 나노로드 센서의 일산화질소 가스에 대한 감응 특성)

  • ZnO nanorod gas sensors were prepared by an ultrasound radiation method and their gas sensing properties were investigated for NO gas. For this procedure, 0.01, 0.005 and 0.001M of zinc nitrate hydrate [$Zn(NO_3)_2\;{\cdot}\;6H_2O$] and hexamethyleneteramine [$C_6H_{12}N_4$] aqueous solutions were prepared and then the solution was irradiated with high intensity ultrasound for 1 h. The lengths of ZnO nanorods ranged from 200 nm to 500 nm with diameters ranging from 40 nm to 80 nm. The size of the ZnO nanorods could be controlled by the concentration of solution. The sensing characteristics of these nanostructures were investigated for three kinds of sensor. The properties of the sensors were influenced by the morphology of the nanorods.

Combined Effect of Radiation and $7{\beta}$-Hydroxycholeslerol on Human Cervical Cancer Cells in vitro

  • [ $7{\beta}$ ]-Hydroxycholesterol (cholest-5-ene-3, 7-diol, $7{\beta}$-OHC) showed the cytotoxicity on human cervical carcinoma cells (HeLa), $10{\mu}M$ of 50% inhibitory concentration. We evaluated $7{\beta}$-OHC as the possibility of radiation sensitizer. The combination effect of $7{\beta}-OHC\;and\;{\gamma}$-irradiation was measured using colony forming assay and flow cytometer with propidium iodide and $DiOC_6$ stained cells, respectively. The combined treatment of $7{\beta}-OHC\;and\;{\gamma}$-irradiation did not show significant enhancing effects on HeLa cells.

  • Tensioned metastable fluids provide a powerful means for low-cost, efficient detection of a wide range of nuclear particles with spectroscopic capabilities. Past work in this field has relied on one-component liquids. Pure liquids may provide very good detection capability in some aspects, such as low thresholds or large radiation interaction cross sections, but it is rare to find a liquid that is a perfect candidate on both counts. It was hypothesized that liquid mixtures could offer optimal benefits and present more options for advancement. However, not much is known about radiation-induced thermal-hydraulics involving destabilization of mixtures of tensioned metastable fluids. This paper presents results of experiments that assess key thermophysical properties of liquid mixtures governing fast neutron radiation-induced cavitation in liquid mixtures. Experiments were conducted by placing liquid mixtures of various proportions in tension metastable states using Purdue's centrifugally-tensioned metastable fluid detector (CTMFD) apparatus. Liquids chosen for this study covered a good representation of both thermal and fast neutron interaction cross sections, a range of cavitation onset thresholds and a range of thermophysical properties. Experiments were devised to measure the effective liquid mixture viscosity and surface tension. Neutron-induced tension metastability thresholds were found to vary non-linearly with mixture concentration; these thresholds varied linearly with surface tension and inversely with mixture vapor pressure (on a semi-log scale), and no visible trend with mixture viscosity nor with latent heat of vaporization.

Investigation of acrylic/boric acid composite gel for neutron attenuation

  • The present work was aimed to show the possibility of using hydrogel (acrylic/boric acid) for evaluation of the neutron radiation shielding. The influence of acrylic acid concentration, different gamma doses and relative contents of boric acid were studied. The physical properties and the thermomechanical stability of the studied samples were investigated. The shielding property of the composite for neutron was tested by Pu-Be neutron source (5 Ci) under room temperature. The neutron fluence rates and gamma fluxes were measured using a stilbene organic scintillator. The macroscopic effective removal cross-section ΣR (cm-1) of fast neutrons and total attenuation coefficient μ (cm-1) of gamma rays has been studied experimentally. The transmission parameters, the relaxation length (??) and the half-value layer (HVL) were obtained. The obtained results indicated that the addition of boric acid to acrylic acid tends to increase the macroscopic effective removal cross-section ΣR (cm-1) to 0.141 compared to 0.094 of ordinary concrete.

  • This paper presents the electrical properties of PTFE nozzle for a electrical apparatus. In the arcing environment in a electrical apparatus, radiation is considered to be the major energy transport mechanism from the arc to the wall. The fraction of the radiation power is emitted out of the arc and reaches the nozzle wall, causing ablation at the surface and in the depth of the wall. The energy concentration in the material leads to the depolymerization and eventually leads to the generation of decomposed gas as well as some isolated carbon particles. Adding some fillers into PTFE is expected to be efficient for improving the endurability against radiation. In this experiment, three kinds of fillers that have endurance in the high temperature environment were added into PTFE. Light reflectance of fillers was investigated. Dielectric constan and dissipation factor of PTFE composites were investigated. Dielectric constant and dissipation factor of the PTFE composites increased with increasing contents of the fillers.
