National Cultural Dimensions and their Impact on Quality Management Maturity and Project Quality Performance: Focusing on ITER Project (국가의 문화차원이 품질경영 성숙도 수준과 프로젝트 품질에 미치는 영향: ITER 프로젝트를 중심으로)

  • Purpose: The study is aimed to identify the national cultural dimensions that are affecting the quality management (QM) maturity level and the project quality performance, and analyze their relationships. Methods: This study collected the data of QM Maturity level based on Crosby's QM maturity model and the project quality performance using the Iron Triangle (Quality, Time/Schedule and Cost) from the employees who are participating in the ITER Project across the major 8 countries (China, France, Italy, Japan, Korea, Russia, U.K. and U.S.A.). Three research hypotheses are proposed concerning the national cultural dimensions in this study and Hofstede's five cultural dimensions framework are used for the statistical test. Results: The results are two folds in the study: First, there is a significant positive correlation between the QM maturity level and the project quality performance. Second, three cultural dimensions (Collectivism, Large Power Distance and Strong Uncertainty Avoidance) and five cultural dimensions (Collectivism, Large Power Distance, Strong Uncertainty Avoidance, Feminity and Long Term Orientation) have a positive impact on the QM maturity level and the project quality performance respectively. Conclusion: From the results, the understanding and consideration of the culture difference among the countries participating International Collaboration R&D project are recommended.

Quality Characteristics of Wheat Flours from New Released Iksan370 with Long Spike and Domestic Wheat Cultivars (신육성 다수확 밀 익산370호의 원맥과 밀가루의 품질 특성)

  • Iksan370 is a long-spike wheat developed by the Rural Development Administration yielding excellent features components such as cold resistance, disease resistance, and viviparous germination. The physicochemical and material properties of the raw wheat and milled flour of Iksan370 were analyzed to derive its appropriate uses. The raw wheat of Iksan370 showed high contents of ash and proteins at 1.71% and 13.7%, respectively. Its test weight of 763.0 g/L was similar to those of other varieties and its 1,000 kernel weight was high at 45.38 g. The milled flour of Iksan370 had an ash content of 0.45%, which corresponds with a class 1 flour, and its protein content is 12.18%, corresponding with strong flour. The damaged starch was 5.41%, which was lower than that of other varieties. The average grain size was $70.67{\mu}m$ and the grain distribution was at the level of a typical hard wheat. In the farinogram, the water absorption was 58.63%, which corresponded to the level of medium flour. The development time was 7.00 minutes, which was significantly lower than those of Jokyung and Keumkang. The degree of softening was 67.00 BU, similar to those of Yunbaek and Baekjoong. Among the physico-chemical characteristics, the high protein content and typical hard wheat grain distribution of Iksan370 were similar to those of strong wheat, usually used for bread making. However, in the farinogram, the dough development time was short and the degree of softening was high. As a result, Iksan370 was expected to have poor breadmaking properties and a small volume of the final bread product due to insufficient dough durability. On the other hand, Iksan370 showed the highest maximum gelatinization viscosity at 864.00 BU. Therefore, Iksan370 is expected to show glutinous texture when used for noodles and its flour appears to be appropriate for frying powders as well.

Neurokinin B-related Peptide Suppresses the Expression of GnRH I, Kiss2 and tac3 in the Brain of Mature Female Nile tilapia Oreochromis niloticus

  • Neurokinin B (NKB) and neurokinin B related peptide (NKBRP) belong to tachykinin peptide family. They act as a neurotransmitter and/or neuromodulator. Mutation of NKB and/or its cognate receptor, NK3R resulted in hypogonadotropic hypogonadism in mammals, implying a strong involvement of NKB/NK3R system in controlling mammalian reproduction. Teleosts possess NKBRP as well as NKB, but their roles in fish reproduction need to be clarified. In this study, NKB and NKBRP coding gene (tac3) was cloned from Nile tilapia and sequenced. Based on the sequence, Nile tilapia NKB and NKBRP peptide were synthesized and their biological potencies were tested in vitro pituitary culture. The synthetic NKBRP showed direct inhibitory effect on the expression of GTH subunits at the pituitary level. This inhibitory effect was confirmed in vivo by means of intraperitoneal (ip) injection of synthetic NKB and NKBRP to mature female tilapia (20 pmol/g body weight [BW]). Both NKB and NKBRP had no effect on the plasma level of sex steroids, E2 and 11-KT. However, NKBRP caused declines of expression level of GnRH I, Kiss2 and tac3 mRNAs in the brain while NKB seemed to have no distinct effect. These results indicate some inhibitory roles of NKBRP in reproduction of mature female Nile tilapia, although their exact functions are not clear at the moment.

Comparative Study on Characterization of Recombinant B Subunit of E. coli Heat-Labile Enterotoxin (rLTB) Prepared from E. coli and P. pastoris

  • Escherichia coli (E. coli) heat-labile enterotoxin B subunit (LTB) was regarded as one of the most powerful mucosal immunoadjuvants eliciting strong immunoresponse to coadministered antigens. In the research, the high-level secretory expression of functional LTB was achieved in P. pastoris through high-density fermentation in a 5-1 fermentor. Meanwhile, the protein was expressed in E. coli by the way of inclusion body, although the gene was cloned from E. coli. Some positive yeast and E. coli transformants were obtained respectively by a series of screenings and identifications. Fusion proteins LTB-6$\times$His could be secreted into the supernatant of the medium after the recombinant P. pastoris was induced by 0.5% (v/v) methanol at $30^{\circ}C$, whereas E. coli transformants expressed target protein in inclusion body after being induced by 1 mM IPTG at $37^{\circ}C$. The expression level increased dramatically to 250-300 mg/l supernatant of fermentation in the former and 80-100 mg/l in the latter. The LTB-6$\times$His were purified to 95% purity by affinity chromatography and characterized by SDS-PAGE and Western blot. Adjuvant activity of target protein was analyzed by binding ability with GMI gangliosides. The MW of LTB-6$\times$His expressed in P. pastoris was greater than that in E. coli, which was equal to the expected 11 kDa, possibly resulted from glycosylation by P. pastoris that would enhance the immunogenicity of co-administered antigens. These data demonstrated that P. pastoris producing heterologous LTB has significant advantages in higher expression level and in adjuvant activity compared with the homologous E. coli system.

The effect of plant extracts on the activity and the expression of MMPs (matrix metalloprotease) induced by UVA

  • UV irradiation on a skin brings about the qualitative and quantitative alterations of the extracellular matrix. Repeated-UV irradiation suppressed the synthesis of collagen and activated the expression of the matrix metalloprotease (MMP). In this paper, on the purpose of development of novel anti-aging agents from natural sources, effects of several natural products on in vitro MMP-1 activity and UVA induced MMP-1 synthesis in human dermal fibroblast (HDF) culture were studied. We measured MMP-1 activities by fluorescence assay using gelatin as substrates. As a result, the extract of Dicentra spectabilis, and flower buds of Tussilago farfara showed strong inhibitory effect. Among them, the extract of flower buds of Tussilago fartara and Dicentra spectabilis inhibited MMP-1 activity by 92% and 87% at 0.05% (w/v). And UVA induced MMP-1 expression were analyzed by enzyme-linked immunosorbent assay (ELISA) and gelatin-based zymography in HDF culture. The extract of flower buds of Tussilago farfara and Dicentra spectabilis suppressed the UVA induced expression of MMP-1 by similar level of Vitamin C 200$\mu$M at 0.1% (w/v). These results suggest that the extract of Dicentra spectabilis, and flower buds of Tussilago farfara effectively prevent skin from the UV-induced photoaging. So the extracts are thought to have potential as effective raw materials for anti-aging cosmetics.

Analysis of Dominant Microorganisms of Bulking Sludge at Low Dissolved Oxygen Concentration using 16S rRNA Sequences (16S rRNA 염기서열을 이용한 낮은 용존산소농도에서 발생한 벌킹슬러지의 우점종 분석)

  • Maintaining dissolved oxygen (DO) at sufficiently low concentration in the aeration tank at a wastewater treatment plant (WWTP) is essential for reduction of the costs of operation and maintenance. On the other hand, the low DO level may result in adverse effect on the integrity of the activated sludge, A typical and disastrous outcome frequently experienced is the outgrowth of filamentous microorganisms, which is called as filamentous bulking, In addition to the traditional methods such as sludge settleability and microscopic observation of the culture, molecular techniques including polymerase chain reaction (PCR) amplification followed by 16S rRNA sequencing were applied to identify filamentous bacteria present in bulking sludge under a condition of low DO concentration, Two morphologically distinct groups, presumably consisting of Sphaerofilus nafans, and Eikelboom Type 1701 or Type 1851, were identified through microscopic observation. They were further confirmed by subsequent 16S rRNA sequencing. Dominant filamentous bacteria identified by the molecular techniques were consisted of three major groups. Sequences of partial 16S rRNA cloned showed that the filamentous bulking organisms were closely related to Eikelboom Type 021N and Eikelboom Type 1701, and Sphaerotilus natans, respectively. Molecular methods were found to possess a strong potential of direct examination of the microbial community of an activated sludge system.

  • The acinar cells and secretory granules of the parotid salivary gland were examined in the lesser white toothed shrew, Crocidura suaveolens and the big white-toothed shrew, C. lasiura. The parotid gland of both species were a serous gland having only one kind of serous acinar cells, and had conventional arrangement of acini and intercalated, granular and striated ducts. In case of C. suaveolens, serous acinar cells had well developed rER, prominent Golgi complex, several large mitochondria and abundant moderate dense secretory granules with various stages of the maturing or fusing process. Immature acinar secretory granules were only or mainly filled with fine strong dense specks and had an indistinct limiting membrane, and mature granules were filled with homogeneous pale large round center and had fine strong dense specks at the periphery of the homogeneous pale center and a distinct limiting membrane. In case of C. lasiula, serous acinar cells had well developed rER, prominent Golgi complex, several large mitochondria and abundant dense secretory granules with maturing or fusing process. Immature acinar secretory granules were only filled with pale rough specks and had an indistinct limiting membrane, and mature granules were only filled with rough dense specks and had a distinct limiting membrane. Eventually The acinar secretory granules of C. suaveolens were seen moderate at the light and ultrastuctural level, those of C. lasiura were strong dense at the light microscopic level and dense at the ultrastructural level.

Formulation and Skin Penetration Characteristics of Aceclofenac Plaster for Transdermal Delivery (아세클로페낙의 경피 제제설계 및 피부투과 특성)

  • Aceclofenac is an non-steroidal antiinflammatory drug which has been used in the treatment of rheumatoidal rthritis and osteo-arthritis. In order to decrease the gastric ulcerogenic effects and contol the plasma level of aceclofenac, we have developed the transdermal delivery system of aceclofenac plaster, which were formulated employing matrix polymers of acrylates and penetration-enhancers such as $Lauroglycol^{\circledR}$, $Transcutol^{\circledR}$, oleic acid and linoleic acid. Using Franz diffusion cells mounted with a rat skin, transdermal penetration characteristics of the formulations were evaluated by the HPLC assay of aceclofenac and diclofenac, an active metabolite, in the receptor compartment of pH 7.2 phosphate buffered solution. Skin penetration was increased when the content of aceclofenac increased, showing the flux $(J,\;{\mu}g/cm^2/hr)$ of 0.37 and 2.50 for 2% and 6.75% of the content, respectively. The flux$(J,\;{\mu}g/cm^2/hr)$ from plasters made of $Durotak^{\circledR}$ 87-2074, $Durotak^{\circledR}$ 87-2510 and $Durotak^{\circledR}$ 87-2097 were 2.50, 2.77 and 4.39, respectively. $Durotak^{\circledR}$ 87-2074 showed the lowest penetration due to the carboxylic acid group in the polymer, which might form a strong hydrogen bonding with a secondary amine of aceclofenac. Although both $Durotak^{\circledR}$ 87-2510 and $Durotak^{\circledR}$ 87-2097 are amine-resistant adhesives, $Durotak^{\circledR}$ 872510 showed lower penetration than $Durotak^{\circledR}$ 87-2097 because of the hydroxyl group in $Durotak^{\circledR}$ 87-2510, which might form a weak hydrogen bonding with aceclofenac. These results reveal that the functional group in acrylic polymers would greatly affect the release of aceclofenac from the matrix, which is the rate-limiting step in the penetration of aceclofenac through rat skins. The penetration of aceclofenac from plasters using different penetration-enhancers increased in the following order: Transcutol < linoleic acid < oleic acid. And the flux from the plasters containing oleic acid as a penetrationenhancer was 2.22 times greater than that of creams, which suggest that a newly deveolped aceclofenac plaster could be used in the treatment of rheumatoidal arthritis and osteo-arthritis as an advanced transdermal delivery system.

A Study on Psychoticism in College Students (정신증적 척도에 의한 대학생의 정신건강 평가)

  • The authors studied Psychoticism, using SCL-90, in the subjects of 3,893 male college students and 1,976 female college students of Yeung Nam University, collecting data during the periods from October to November, 1984. The results could be summarized as follows: 1. There was significant difference in the mean averages of total psychoticism scores between male and female students; male students scored $6.81{\pm}5.13$ female students scored $8.14{\pm}5.05$. 2. Eighty eight male students (2.4%) showed seriously high psychoticism scores of 20 or higher, while 63 female students (3.2%) showed the same scores. 3. In a comparison between male students in different school grades, freshmen showed the highest level of psychoticism scores, and as the grades of students became higher, the levels of psychoticism scores were lower. 4. Among the psychosocial factors, there were strong tendencies toward higher psychoticism scores in the students who were dissatisfied with their college, and hsd pessimistic views of self image in the past, present or future, in both groups. 5. The male students whose educational fees were paid by their parents, and whose spending money per month was under 10,000won, showed higher level of psychoticism scores. 6. The female students whose maturation locations were city, who were dissatisfied with their departments, who resided in dormitory of other residencies, and whose educational fees were paid by their brothers or sisters, showed higher level of psychoticism scores. 7. The items relating to 5(Feeling lonely with people) 9 (Never feeling close to another) 10 (Idea something with mind) in psychoticism and the item of depressed affect in SDS were significantly correlated over 0.40 of correlation coefficient.

A Study on the Application Method of Artificial Injection Test according to the Hydraulic Conductivity of Aquifer (대수층 수리지질특성에 따른 인공함양시험 적용 방법에 관한 연구)

  • Artificial recharge technology is a method for solving problems such as groundwater level drop and ground subsidence caused by groundwater withdrawal. This study investigated the applicability of using the hydraulic conductivity of an aquifer to predict injection test results for aquifer restoration. Pumping and injection tests were performed under the same conditions as those for the artificial injection facility located in Icheon, Gyeonggi-do. The hydraulic conductivity of the aquifer, which plays a decisive role in restoring the groundwater level, was derived from the pumping test. A numerical model of a simplified on-site aquifer was constructed, and a transient analysis was applied with the same conditions as the pumping test. The correlation between the measured and the resulting model values is strong (R2 = 0.78). The injection test was performed in a sedimentary layer composed of silt sand and clay sand. From the results of the injection test, an empirical formula was derived using Theim's formula, which is a common well analysis solution to determine the parameters of the aquifer from time-level data. The model values from the empirical formula have a high degree of correlation (R2 = 0.99) with measured values. Under specific conditions, for areas where it is difficult to conduct an injection test, the formula from this study, which relies on the hydraulic conductivity of the aquifer determined through the pumping test, may be used to predict reliable injection rates for groundwater restoration.
