Expression of Placenta-Related Genes (Cdx2 and GATA6) in Cloned Porcine Development

  • Abnormal development and fetal loss during the post-implantation period are key concerns in the production of cloned animals by somatic cell nuclear transfer (SCNT). We hypothesized that the problems in cloned porcine offspring derived from SCNT are related to interactions between the conceptus and the endometrial environment. In the present study, we investigated expression patterns in the formation of placenta-related genes (Cdx2 and GATA6) in whole in vivo normal porcine embryos (from single cell to blastocyst) and each tissue of a normal fetus at Days 25, 35 and 55 by quantitative mRNA expression analysis using real-time PCR. The expression of Cdx2 and GATA6 mRNA increased to around the blastocyst stage. These genes were gradually decreased from the peri-implantation to post-implantation stage. Moreover, we examined the expression patterns of Cdx2 and GATA6 in Day 35 normal and SCNT cloned fetuses by the same methods. And, the level of Cdx2 and GATA6 gene expression in the extraembryonic tissue of SCNT was significantly higher than that of control tissues. From the present results, it can be postulated that the aberrant expression of Cdx2 and GATA6 genes in the endometrial and extraembryonic tissues at pre- and peri-implantation stages may be closely related to the lower efficiency of animal cloning.

  • Abiotic stresses such as high/low temperature, drought, salinity, and submergence directly or indirectly influence the physiological status and molecular mechanisms of rice which badly affect yield. Especially, the low temperature causes harmful influences in the overall process of rice growth such as uneven germination and the establishment of seedlings, which has become one of the main limiting factors affecting rice production in the world. It is of great significance to find the candidate genes controlling low-temperature tolerance during seed germination and study their functions for breeding new rice cultivars with immense low-temperature tolerance during seed germination. In this study, 120 lines of Cheongcheong/Nagdong double haploid population were used for quantitative trait locus analysis of low-temperature germinability. The results showed significant difference in germination under low different temperature conditions. In total, 4 QTLs were detected on chromosome 3, 6, and 8. A total of 41 genes were identified from all the 4 QTLs, among them, 25 genes were selected by gene function annotation and further screened through quantitative real time polymerase chain reaction. Based on gene function annotation and level of expression under low-temperature, our study suggested OsGPq3 gene as a candidate gene controlling viviparous germination, ABA and GA signaling under low-temperature. This study will provide a theoretical basis for marker-assisted breeding.

배추 유래 저온 저항성 관련 유전자, BrCSR의 특성 분석 (Characterization of a Cold Tolerance-related Gene, BrCSR, Derived from Brassica rapa)

  • 본 연구는 배추에서의 저온 저항성 유전자를 개발하는데 목적이 있으며 이를 위해 먼저 저온($4^{\circ}C$) 스트레스가 처리된 내혼계배추를 대상으로 한 KBGP-24K oligo chip의 결과 [BrEMD(Brassica rapa EST and Microarray Database)]를 분석하였다. 그 결과 23,929개의 배추 unigene 중 저온 처리시 대조군 대비 5배 이상 발현이 증가하는 417개(1.7%)의 저온 반응 유전자를 1차 선발하고, 이들 중 기능이 정확히 알려지지 않았으나 완전장을 갖추고 있는 BrCSR로 명명한 유전자를 선발하였다. 이 유전자의 저온 저항성을 분석하기 위하여 형질전환용 과발현 vector인 pSL101 binary vector를 제작하여 담배에 형질전환시켰다. BrCSR이 과발현된 $T_1$ 세대 담배 형질전환체들은 PCR과 Southern hybridization 분석에 의해 선발하였고, BrCSR의 기능은 저온 처리 시 유전자의 발현 수준 분석과 표현형 검정을 통해 확인하였다. Quantitative real-time RT-PCR과 Northern blot hybridization 분석 결과, 형질전환 담배에서 BrCSR의 발현이 대조군보다 약 2배 정도 높게 발현되었으며 실제로 $4^{\circ}C$ 처리 후 표현형 분석에서 BrCSR이 과발현된 형질전환체들이 대조군보다 우수한 저온 저항성을 보여 주었다. 위 결과들에 근거하여 BrCSR 유전자가 저온 환경 하에서 식물의 생장과 저항성 향상에 중요한 역할을 담당하고 있음을 확인할 수 있었다.

초등학교 6학년 학생들의 교과서 비례 문제 해결과 비례 추론에 관한 연구 (A Study on the Solving Proportion Problems of Mathematics Textbooks and Proportional Reasoning in 6th Graders)

  • 본 연구에서는 7차 교육과정에서 6학년 때 도입되는 교과서의 비례 문제들이 학생들의 비례 추론 능력과 어떠한 관련이 있는지를 알아보기 위해 교과서의 비례 문제 해결 실태를 파악하고, 수준별 비례 추론 능력을 하위영역으로 나누어 교과서 비례 문제 해결 능력에 따라 하위영역별로 비례추론 능력이 어떠한지 분석하였다. 연구결과 교과서의 비례 문제에서 정답률이 높은 문제들은 설명에서 비례식을 이용해서 풀 수 있도록 제시되어 있었으며, 비례식을 세웠을 때 두 비 사이의 관계가 정수비로 계산이 간단하였다. 비례 추론 하위 영역 중 비감각 영역의 문제 해결을 잘하였고, 양과 변화 영역에 대한 부분의 능력은 가장 뒤떨어졌다. 교과서의 비례 문제 해결 능력과 비례 추론의 관계에 대해서는 교과서의 비례 문제 해결이 우수한 학생일수록 비례 추론 능력이 우수였다. 교과서 비례문제의 해결 결과가 비례추론 능력을 예언할 수 있다고 볼 수 있다. 교과서 비례문제 수준에 따라서 비례 추론 문제 해결의 수준차를 알아본 결과, 차이가 많이 나지 않는 문제는 꼭 비와 비율 관련 단원이 아니라도 수학 교과서에서 다양하게 접할 수 있는 문제였고, 수준별 차이가 많이 나는 문제는 그동안 교과서에서 쉽게 접해보지 못한 유형으로 단순히 비례식을 이용해서는 해결할 수 없는 문제들이었다. 따라서, 비례추론 하위 영역별로 모든 영역에 대하여 능력을 향상시키기 위해서는 교과서에 비례식 외에 다양한 상황과 내용의 비례문제를 포함하여 지도하여야 할 것이다.

스텝 애니메이션과 감성 표현 사이의 정량적 상호관계에 관한 연구 (A study of quantitative correlation between step animation and emotional expressions)

  • 본 논문은 스텝 애니메이션에서 표현되어지는 감성을 정의하고, 스텝 관련 감성 자극 요소들을 추출 측정, 분석하여 스텝의 직관적인 감성 표현을 정량화시키는 것을 목적으로 하고 있다. 실험 방법으로는 스텝 관련 27개의 감성 표현 어휘와 36개의 샘플 스텝 동영상의 상관관계를 설문 조사하였다. 통합된 데이터를 수량화 이론 III류를 이용하여 스텝의 감성적 멘털구조를 2차원 평면화 하였다. 이를 분석한 결과, 감성 스텝은 가로의 자신감 축과 세로의 안정감 축으로 형성되어 있음을 밝혀내었다. 감성 스텝의 2차원 분포도를 분석한 결과, <자신감, 불안감 쪽> 2사분면과 <자신감, 안정감 쪽> 3사분면은 데이터가 풍부한 반면 <소심함, 불안감 쪽> 1사분면과 <소심함, 안정감 쪽> 4사분면의 데이터 분포가 희박함을 보여주었다. 이러한 그래프 분석을 통해 불안감이 느껴지면서 소심한 감성과 안정감이 느껴지는 소심한 캐릭터의 감성은 스텝만으로 표현하기에는 난해한 감성 표현임을 파악할 수 있었다. 감성에 영향을 미치는 스텝의 신체 물리적 요소로는 <스텝의 간격> <스텝의 속도> <골반(Y축)의 움직임 각도> <팔 스윙 폭> <척추(x축)의 움직임 각도> <신체의 기울기> 6가지로 선정하여 분석하였다. 그 결과 <스텝의 속도>와 <신체의 기울기>가 스텝의 감성 자극에 가장 큰 영향을 미치는 요소임을 밝혀낼 수 있었다. 본 연구를 통해 캐릭터의 미묘한 감성 상태를 스텝 애니메이션에 자연스럽게 표현해 내는 과정에 있어 애니메이터들의 주관적인 가성 표현들을 객관화, 정량화시키고자 하였으며 향후 이 데이터들을 활용한 멀티컨텐츠의 적용을 목표로 하고 있다.

Molecular Characterization and Expression Analysis of Creatine Kinase Muscle (CK-M) Gene in Horse

  • Since ancient days, domestic horses have been closely associated with human civilization. Today, horse racing is an important industry. Various genes involved in energy production and muscle contraction are differentially regulated during a race. Among them, creatine kinase (CK) is well known for its regulation of energy preservation in animal cells. CK is an iso-enzyme, encoded by different genes and expressed in skeletal muscle, heart, brain and leucocytes. We confirmed that the expression of CK-M significantly increased in the blood after a 30 minute exercise period, while no considerable change was observed in skeletal muscle. Analysis of various tissues showed an ubiquitous expression of the CK-M gene in the horse; CK-M mRNA expression was predominant in the skeletal muscle and the cardiac muscle compared to other tissues. An evolutionary study by synonymous and non-synonymous single nucleotide polymorphism ratio of CK-M gene revealed a positive selection that was conserved in the horse. More studies are warranted in order to develop the expression of CK-M gene as a biomarker in blood of thoroughbred horses.

구강편평상피암종에서 stromal cell-derived factor-1의 발현 (Stromal cell-derived factor-1 (SDF-1) expression in the oral squamous cell carcinoma)

  • Purpose: Chemokines are structurally related, small polypeptide signaling molecules that bind to and activate a family of transmembrane G protein-coupled receptors, the chemokine receptors. Recently, interaction between the chemokine receptor CXCR4 and its ligand, stromal cell-derived factor 1 (SDF-1 or CXCL12), has been found to play an important role in tumorigenicity, proliferation, metastasis and angiogenesis in many cancers such as lung cancer, breast cancer, melanoma, glioblastoma, pancreatic cancer and cholangiocarcinoma. Hence, the goal of this study is to identify the correlation of clinicopathological factors and the up-regulation of SDF-1 expression in oral squamous cell carcinoma. Material and methods: We studied the immunohistochemical staining of SDF-1, quantitative RT-PCR (qRT-PCR) of SDF-1 gene in 20 specimens of 20 patients with oral squamous cell carcinoma. Results: 1. In the immunohistochemical study of poor differentiated and invasive oral squamous cell carcinoma, the high level staining of SDF-1 was observed. And the correlation between immunohistochemical SDF-1 expression and tumor nodes metastases (TNM) classification of specimens was significant.($x^2$ test, P < 0.05) 2. In the SDF-1 gene qRT-PCR analysis, SDF-1 expression was more in tumor tissue than in carcinoma in situ tissue. Paired-samples analysis determined the difference of SDF-1 mRNA expression level between the cancer tissue and the carcinoma in situ tissue.(Student's t-test, P < 0.05) Conclusion: These findings suggest that up-regulation of the SDF-1 may play a role in progression and invasion of oral squamous cell carcinoma.

황칠나무 줄기 추출물의 MCF-7과 MDA-MB-231 유방암 세포주에 대한 세포증식억제 효과 (Anti-cancer Effects of Dendropanax Morbifera Extract in MCF-7 and MDA-MB-231 Cells)

  • Objectives : Dendropanax morbifera is known as a tree that has been used in traditional medicine for various diseases. However, its biological activities in cancer have not yet been clearly elucidated. In this study, we investigated the anti-cancer effects of water extract of Dendropanax morbifera (DP) on 2 human breast cancer cell lines (estrogen dependent MCF-7 and estrogen independent MDA-MB-231). Methods : The MTT assay and flow cytometry were used to assess cell proliferation, along with cell cycle analysis. Nitric oxide production was detected by Griess assay. The expression of apoptosis related gene was assessed by quantitative real-time PCR. Results : Our data revealed that DP inhibits the cell growth in a dose dependent manner (0, 50, 100, 250, and 500 μg/ml) of both estrogen independent MDA-MB-231 and estrogen dependent MCF-7 breast cancer cells. Also, LPS induced nitric oxide production was significantly reduced by DP. Cell cycle analysis showed an increased G1 phase in the MCF-7 cell and G2/M phase in the MDA-MB-231 cell. DP decreased mRNA expression of apoptotic suppressor gene Bcl-xL, and increased mRNA expression of pro-apoptotic genes. DP increased mRNA expression of p21, and Rip1 in both cell. And DP decreased mRNA expression of survivin in the MCF-7 cell. Conclusions : Taken together, these results indicate that DP extract are source of anti-cancer potential and could be developed botanical drug.

The High Expressed Serum Soluble Neural Cell Adhesion Molecule, a High Risk Factor Indicating Hepatic Encephalopathy in Hepatocelular Carcinoma Patients

  • Objective: To investigate whether the expression of serum soluble neural cell adhesion molecule (sNCAM) is associated with hepatic encephalopathy (HE) in hepatocelular carcinoma (HCC) patients. Materials and Methods: The Oncomine Cancer Microarray database was used to determine the clinical relevance of NCAM expression in different kinds of human cancers. Sera from 75 HCC cases enrolled in this study were assessed for expression of sNCAM by enzyme linked immunosorbent assay (ELISA). Results: Dependent on the Oncomine Cancer Microarray database analysis, NCAM was down regulated in 10 different kinds of cancer, like bladder cancer, brain and central nervous system cancer, while up-regulated in lung cancer, uterine corpus leiomyoma and sarcoma, compared to normal groups. Puzzlingly, NCAM expression demonstrated no significant difference between normal and HCC groups. However, we found by quantitative ELISA that the level of sNCAM in sera from HCC patients with HE ($347.4{\pm}151.9ng/ml$) was significantly more up-regulated than that in HCC patients without HE ($260.3{\pm}104.2ng/ml$), the p-value being 0.008. sNCAM may be an important risk factor of HE in HCC patients, the correlation coefficients was 0.278 (P<0.05) on rank correlation analysis. Conclusions: This study highlights that up-regulated level of serum sNCAM is associated with HE in HCC patients and suggests that the high expression can be used as an indicator.

MicroRNA-21 Regulates the Invasion and Metastasis in Cholangiocarcinoma and May Be a Potential Biomarker for Cancer Prognosis

  • Background: MicroRNAs are noncoding RNA molecules that posttranscriptionally regulate gene expression. The aim of this study was to determine the role of microRNA-21 in cholangiocarcinomas and its relationship to cholangiocarcinoma RBE cell capacity for invasion and metastasis. Methods: MicroRNA-21 expression was investigated in 41 cases of cholangiocarcinoma samples by in situ hybridization and real-time PCR. Influence on cholangiocarcinoma cell line invasion and metastasis was analyzed with microRNA-21 transfected cells. In addition, regulation of reversion-inducing-cysteine-rich protein with kazal motifs (RECK) by microRNA-21 was elucidated to identify mechanisms. Results: In situ hybridization and real-time quantitative PCR results for patients with lymph node metastasis or perineural invasion showed significantly high expression of microRNA-21 (P<0.05). There was a dramatic decrease in cholangiocarcinoma cell line invasion and metastasis ability after microRNA-21 knockdown (P<0.05). However, overexpression significantly increased invasion and metastasis (P<0.05). Real-time PCR and Western-blot analysis showed that microRNA-21 could potentially inhibit RECK expression in RBE cells. Survival analysis showed that patients with higher expression levels of microRNA-21 more often had a poor prognosis (P<0.05). Conclusions: MicroRNA-21 may play an important role in cholangiocarcinoma invasion and metastasis, suggesting that MicroRNA-21 should be further evaluated as a biomarker for predicting cholangiocarcinoma prognosis.
