  • Plasticizers are materials added to give softness and elasticity to plastics having rigid properties to give soft properties as products, and they are mainly added to high molecular materials to give flexibility to improve workability and to improve cold resistance, resistance to volatility and electrical properties. It is used for the purpose. Most plasticizers are inert liquids, similar in function to solvents but with high molecular weight and no volatility. In addition, when dissolved in petrochemical products, only the plasticizer is separated by the matrix effect with other compounds, and qualitative and quantitative analysis. In this study, qualitative and quantitative analysis of DOA and DOP, which are representative components of petrochemical products, were conducted using MD-GC/MS and developed an optimal plasticizer analysis method.

CircCOL1A2 Sponges MiR-1286 to Promote Cell Invasion and Migration of Gastric Cancer by Elevating Expression of USP10 to Downregulate RFC2 Ubiquitination Level

  • Gastric cancers (GC) are generally malignant tumors, occurring with high incidence and threatening public health around the world. Circular RNAs (circRNAs) play crucial roles in modulating various cancers, including GC. However, the functions of circRNAs and their regulatory mechanism in colorectal cancer (CRC) remain largely unknown. This study focuses on both the role of circCOL1A2 in CRC progression as well as its downstream molecular mechanism. Quantitative polymerase chain reaction (qPCR) and western blot were adopted for gene expression analysis. Functional experiments were performed to study the biological functions. Fluorescence in situ hybridization (FISH) and subcellular fraction assays were employed to detect the subcellular distribution. Luciferase reporter, RNA-binding protein immunoprecipitation (RIP), co-immunoprecipitation (Co-IP), RNA pull-down, and immunofluorescence (IF) and immunoprecipitation (IP) assays were used to explore the underlying mechanisms. Our results found circCOL1A2 to be not only upregulated in GC cells, but that it also propels the migration and invasion of GC cells. CircCOL1A2 functions as a competing endogenous RNA (ceRNA) by sequestering microRNA-1286 (miR-1286) to modulate ubiquitin-specific peptidase 10 (USP10), which in turn spurs the migration and invasion of GC cells by regulating RFC2. In sum, CircCOL1A2 sponges miR-1286 to promote cell invasion and migration of GC by elevating the expression of USP10 to downregulate the level of RFC2 ubiquitination. Our study offers a potential novel target for the early diagnosis and treatment of GC.

Analysis of Quantitative Trait Loci (QTL) Associated with the Gel Consistency in Rice (쌀의 호응집성에 대한 QTLs 분석)

  • The purpose of this study was to identify the quantitative trait loci (QTL) associated with gel consistency (GC), and to evaluate the relationships between GC and physico-chemical properties related to eating quality in rice. A total of 120 doubled haploid (DH) lines derived from a cross between 'Samgang' and 'Nagdong' and 26 varieties were used for this study. The 120 DH lines were divided into four groups, soft (61~100 mm), medium (41~60 mm), hard (26~40 mm) and whole population, according to the gel length. A negative correlation between GC and amylose content was observed only in the soft group. The GC negatively correlated with lipid content in the medium, and the whole population, but positively correlated with lipid content in the soft group. The positive correlation between GC and protein content was observed in the soft group, while negative correlations were observed in the medium group and the whole population. Two significant QTLs regarding GC were detected on chromosomes 4 and 11. They collectively explained 23% of phenotypic variation with LOD score of 3.1 and 2.9, respectively. The DNA markers of S4026 and RM287 were tightly linked to GC in the DH population and 26 varieties, respectively.

Classification and identification of organic aerosols in the atmosphere over Seoul using two dimensional gas chromatography-time of flight mass spectrometry (GC×GC/TOF-MS) data (GC×GC/TOF-MS를 이용한 서울 대기 중 유기 에어로졸의 분류 및 동정)

  • To identify a variety of organic compounds in the ambient aerosols, the two-dimensional gas chromatography-time of flight mass spectrometry (GCxGC) system (2DGC) has been applied. While 2DGC provides more peaks, the amount of the generated data becomes huge. A two-step approach has been proposed to efficiently interpret the organic aerosol analysis data. The two-dimensional 2DGC data were divided into 6 chemical groups depending on their volatility and polarity. Using these classification standards, all the peaks were subject to both qualitative and quantitative analyses and then classified into 8 classes. The aerosol samples collected in Seoul in summer 2013 and winter 2014 were used as the test case. It was found that some chemical classes such as furanone showed seasonal variation in the high polarity-volatile organic compounds (HP-VOC) group. Also, for some chemical classes, qualitative and quantitative analyses showed different trends. Limitations of the proposed method are discussed.

Study on the new approaching method to determine limit of detection by gas chromatography (GC에서 검출한계 결정을 위한 새로운 접근 방법에 대한 연구)

  • The purity methods to determine LOD/LOQ using standard deviation of the residual, intercept and blank by IUPAC and ACS describe many of the pitfalls and pose significant challenges to analytical chemists. Therefore, the aim of this study is the development of the simple, easy, convenient and statistically significant method to determine LOD in quantitative analysis of organic solvents by GC. The new approaching method by linearization in the given concentration range used coefficient of variation ; ${\sigma}_{n-1}$/S(standard deviation, ${\sigma}_{n-1}$ and average, S) of sensitivity(Response/concentration). The comparison of results among the purity methods(IUPAC and ACS) and the linearization have been fulfilled the F-test for standard deviations and t-test for LOD range values. The results of F-test and t-test are satisfied within 95 % confidence level, respectably. The LOD values determined by the new procedure are n-Hexane 0.0116 mg/$m^3$, Toluene 0.0807 mg/$m^3$, and o-Xylene 0.0494 mg/$m^3$. Because the standard deviation of the residual, intercept and blank and the slope of calibration curve are not calculated and the new approaching method use the coefficient of variation of sensitivity by linearization, this new method is simple, easy, convenient and statistically significant. In future, many chemical analysts will expect to applicate and routinely use this method in the all quantitative analysis.

Experimental Study on the Deformation of MLCC Compressed Bar by Quantitative Analysis for Outgas (Outgas 분석을 통한 MLCC Bar 변형률 측정)

  • MLCC (multi-layer ceramic capacitor) is usually fabricated by lamination of predetermined number of single layers. Often, the state of MLCC before sintering is called the green state, whose strength comes from the adhesion between the dielectric material and the polymer binder. Therefore the lamination of a single layer before sintering can be easily deformed by environment due to the relatively lower strength. After the compression process, which helps single sheets cohereto with adjacent sheets, the MLCC green bar is preheated to resolve the probable internal stress. Unfortunately, unexpected deformation after preheating resulted in problems during cutting of the MLCC green bar. In this study, one of 2 primary hypotheses which were proposed to resolve the unexpected deformation after preheating was examined by quantitative experiment with GC/MS (gas chromatograpy/mass spectrometer). The proportion of deformation caused by DOP evaporation, which was primarily evaporated componet during preheating, to the total deformation of the MLCC green bar was found to be 53%.

Installation and Test Run of Comprehensive Analysis System for SF6 in Power Equipment

  • After $SF_6$, which is being used in power equipment as an insulating material, is classified as one of the 6 major greenhouse gases, the maintenance and the refinement of used $SF_6$ started to get attention. In regard to this, KEPCO Research Institute (KEPRI) is developing $SF_6$ recovery and refinement technology starting with establishing a comprehensive $SF_6$ analysis system. With the analysis system, qualitative and quantitative analyses of the purity and the impurities of $SF_6$ before and after recovery, and before and after refinement have been carried out. The analysis system is comprised of GC-DID (Gas Chromatograph -Discharge Ionization Detector) for trace impurities analysis, GC-TCD (Thermal Conductivity Detector) for analyses of $SF_6$ purity and major impurities concentration from several hundred ppm up to percent range, GC-MSD (Mass Selective Detector) for analyses of impurities not included in standard gas, FT-IR (Fourier Transform-Infrared) Spectrometer for analysis of HF and $SO_2$, and moisture analyzer for analysis of moisture below 100 ppm. With this analysis system, complete analysis method of $SF_6$ has been established. This analysis system is being used in the maintenance of power equipment and the development of $SF_6$ recovery and refinement technologies. In this paper, the analysis results of four samples - gas and liquid phase $SF_6$ samples from a $SF_6$ refinement system before and after refinement are presented.

Determination of Volatile Flavor Compounds during Storage of Cereal Added Yogurt using HS-SPME (곡류 첨가 발효유의 저장 중 HS-SPME를 이용한 휘발성 향기성분 정량분석)

  • In this study, quantitative analysis of major volatile flavor compounds from yogurt was conducted using headspace-solid phase microextraction (HS-SPME) GC-FID analysis technique, and the changes of volatile aroma compounds during the storage period were evaluated. The yogurt was prepared with the addition of 2% cereals, such as, white rice (WR), brown rice (BR), germinated brown rice (GBR) and saccharified germinated brown rice (SGBR). After fermentation, the products were stored at $5^{\circ}C$for 15 d. The major volatile aroma compounds in yogurt, such as acetaldehyde, acetone, diacetyl and acetoin were able to be extracted using HS-SPME technique efficiently. The regression ($r^2$) value of standard curve prepared with various concentrations of individual flavor chemicals was analyzed over 0.9975, and reproducibility was acceptable to apply quantitative analysis. The analysis of volatile components of control sample during storage showed that the acetaldehyde on 0 d was 10.83 ppm, and that contents were increased to 15.67 ppm after 15 d of storage. However, addition of BR, GBR and SGBR decreased the acetaldehyde contents during storage periods. The acetone content of all treatments during storage was not significantly different. The diacetyl content of all treatments were increased during storage and the addition of SGBR showed the highest amount of diacetyl (0.84 ppm) among treatments on 15 d of storage. The acetoin content of yogurt added with grains was higher than that of control during storage. As a result, the content of volatile aroma compounds in yoghurt during storage period could be analyzed HS-SPME extraction technique effectively, and HS-SPME/GC analysis can be considered for quality control of fermented milk products.

Development and Characteristic Study of a Portable Gas Chromatography (소형 GC 모듈의 개발 및 특성)

  • In the present study, we developed a portable GC module for real-time, quantitative determinations of gas mixtures in air sample. Capillary or packed column was coiled together with a heater wire and thermocouple in a small case. Together with the small and light weight sensors and valves as well as the rechargeable carrier gas canister, which permits collection and separation of samples, this system can determine the components of complex mixtures of air contaminants at low concentrations with a duty cycle of 10 min. When measured the various samples with a FID and TCD, the system showed, for a capillary column, a good resolution (R=8.3), high sensitivity, reproducibility, and linear dynamic range greater than three orders of magnitude. These results indicate that the portable GC module is expected to be used for a wide range of applications, particularly for in situ environmental monitoring, chemical processes, and regulation of contaminant emission.

Quantitative Analysis of Alkylpyrazines in Snow Crab Cooker Effluents

  • Alkylpyrazines in snow crab cooker effluent(SCCE) and effluent concentrate(EC) were quantitatively analyzed and compared by simultaneous steam distillation-solvent extraction/gas chromatography/mass spectrometry(SDE/GC/MS). A total of 11 pyrazines were identified in both SCCE and EC. Amounts of tetramethylpyrazine, 2,5-dimethylpyrazine, trimethylpyrazine, and 2,6-dimethylpyrazine were 23.0, 21.1, 13.8, and 13.3 times higher, respectively, in EC than those in SCCE. The total amount of pyrzines in EC (1664.0${\pm}$171.1ng/g) was 8.1 times higher than that in SCCE(204.5${\pm}$32.2). The compounds, ethylpyrazine and 2-ethyl-3,6-dimethylpyrazine, were only detected in EC.
