Quantitative and Classification Analyses of Lupenone and ${\beta}$-Sitosterol by GC-FID in Adenophora triphylla var. japonica Hara and Codonopsis lanceolata

  • A simple GC method with a FID detector was developed in order to determine two main compounds (${\beta}$-sitosterol and lupenone) for Adenophorae Radix. ${\beta}$-Sitosterol and lupenone were analyzed by the gradient thermal ramping method. Nitrogen was used as the carrier gas at 108 kPa. The flow rate of gas was 2.0 mL/min; $2{\mu}L$ of filtered sample was injected at a split ratio of 1 : 80. This method was fully validated with respect to linearity, precision, accuracy and robustness. Further, this GC-FID method was applied successfully in order to quantify two compounds in an Adenophorae Radix extract. The GC analytical method for classification analysis was performed by repeated analysis of 59 reference samples in order to differentiate between Adenophora triphylla var. japonica Hara and 14 Codonopsis lanceolata. The results indicate that the GC-FID method is suitable and reliable for the quality evaluation of Adenophorae Radix.

Quantitative Analysis of the Seven Marker Components in Asarum sieboldii using the LC-MS/MS and GC-MS (LC-MS/MS와 GC-MS를 이용한 세신 추출물 중 7종 성분의 함량분석)

  • Asarum sieboldii has been used for treatment of fever, pain, common cold, and chronic sinusitis in Korea. In this study, we performed quantification analysis of seven major constituents including aristolochic acid I, aristolochic acid II, ${\alpha}$-asarone, ${\beta}$-asarone, elemicin, methyl eugenol, and safrole in the 70% ethanol extract of Asarum sieboldii and its solvent fractions, n-hexane, ethylacetate, n-butanol, and water ones using a ultra-performance liquid chromatography-electrospray ionization-mass spectrometer(UPLC-ESI-MS) and gas chromatography-mass spectrometer(GC-MS). Regression equations of seven components were acquired with $r^2$ values >0.99. The values of limit of detection(LOD) and quantification(LOQ) were 0.1-3.9 ng/mL and 0.3-11.7 mg/mL, respectively. The amount of the seven compounds in Asarum sieboldii were not detected -143.66 mg/g. The established LC-MS/MS and GC-MS methods will be helpful to improve quality control of Asarum sieboldii.

  • In Food revolution of Korea, colorimetries or a titration methods are introduced for the analysis of alcoholic liquors. But, these wet analyses have disadvantages such as individual errors, long process time, and sometimes tedious pretreatments. In the process of making alcoholic liquors, fusel oils are produced as by products. Five main fusel components that could be produced are 2-propanol, n-propanol, iso-butanol, n-butanol, and isoamyl alcohol. Also acetaldehyde and methanol could be produced as by-products of ethanol. With using capillary FFAP column in GC or GC/MSD, we analysed these five fusel components as well as internal standard (acetonitrile) including methanol, acetaldehyde and ethanol simultaneously. We obtained excellent mass spectra as qualitative data of all species. We also took excellent quantitative data with GC by using the internal standard method.

Knockdown of Circ_0000144 Suppresses Cell Proliferation, Migration and Invasion in Gastric Cancer Via Sponging MiR-217

  • Previous studies have uncovered the role of circ_0000144 in various tumors. Here, we investigated the function and mechanism of circ_0000144 in gastric cancer (GC) progression. The expression of circ_0000144 in GC tissues and cells was detected through quantitative real-time polymerase chain reaction (qRT-PCR) method. Gain- and loss-of-function experiments including colony formation, wound healing and transwell assays were performed to examine the role of circ_0000144 in GC cells. Furthermore, western blot was conducted to determine the expressions of epithelial mesenchymal transition (EMT)-related proteins. The interaction between circ_0000144 and miR-217 was analyzed by bioinformatic analysis and luciferase reporter assays. The circ_0000144 expression was obviously upregulated in GC tissues and cells. Silencing of circ_0000144 inhibited cell proliferation, migration and invasion of GC cells, but ectopic expression of circ_0000144 showed the opposite results. Moreover, circ_0000144 sponged miR-217, and rescue assays revealed that silencing miR-217 expression reversed the inhibitory effect of circ_0000144 knockdown on the progress of GC. Our findings reveal that circ_0000144 inhibition suppresses GC cell proliferation, migration and invasion via absorbing miR-217, providing a new biomarker and potential therapeutic target for treatment of GC.

  • A gas chromatographic procedure is described for the quantitative analysis of ephedrine hydrochloride, pseudoephediiiie hydrochloride and norephedrine hydrochloride in pharmaceutical preparations. The procedure was based upon the determination of thioureas formed from amines and phenylisothiocyanate. The recoveries of ephedrine, pseudoephedrine and norephedrine from pharmaceutical preparations were 100.7(S.E. 0.9), 99.5(0.8), 99.1(0.8) and 99.1%(O.9), respectively. The high accuracy and precision of this assay were inherent in the flame photometric detectors normally used in gas chromatography.

Absence of Tocotrienol Form of Vitamin E in Purple Perilla (Perilla frutescens var. acuta Kudo) Seeds Confirmed by Comparative Analysis Using HPLC and GC (GC 및 HPLC 비교분석에 기초한 차조기 종실내 tocotrienol 부재의 평가)

  • Lipid soluble vitamin E consists of tocopherols and tocotrienols depending upon double bonds in phytyl side chains attached to chromanol ring. Recent reports on antioxidative, anticancer, and cholesterol-lowering effects of tocotrienols have increased researches and commercialization of tocotrienols. Purple perilla (Perilla frutescens var. acuta Kudo) has been reported as a plant containing tocotrienols along with tocopherol forms of vitamin E based upon normal phase HPLC analysis. To confirm the existence or absence of tocotrienol form of vitamin E in purple perilla, comparative analysis using HPLC, GC/FID, and GC/MSD has been conducted for 14 purple perilla genetic accessions collected from Korea and Japan. Normal phase HPLC analysis showed ${\alpha}-$, ${\beta}-$, ${\gamma}-$, and ${\delta}-tocopherols$ along with peaks with retention times quite similar to ${\beta}-$ and ${\gamma}-tocotrienols$. Same purple perilla samples, analysed by GC exhibited ${\alpha}-$, ${\beta}-$, ${\gamma}-$, and ${\delta}-tocopherols$ quantitatively equivalent to HPLC results. However, no peaks for ${\beta}-$ and ${\gamma}-tocotrienols$ could be observed and unknown two peaks of similar retention times with ${\beta}-$ and ${\gamma}-tocotrienols$ were identified not corresponding tocotrienols by GC/MSD. These results suggest the absence of tocotrienol form of vitamin E in purple perilla as well as the necessity of using GC-based qualitative and quantitative vitamin E analysis to avoid misinterpretation of peaks with similar retention times as tocotrienol isomers when analysed by an HPLC.

Optical and Near-IR Photometry of the NGC 4874 Globular Cluster System with the Hubble Space Telescope

  • We present our study of analyzing the photometric properties of the globular cluster (GC) system which resides in the extended halo of the central bright Coma cluster galaxy NGC 4874. The core of the Coma cluster of galaxies (Abell 1656) was observed with both the HST Advanced Camera for Surveys (ACS) in the F475W (g475) and F814W (I814) and Wide Field Camera 3 IR Channel (WFC3/IR) in the F160W (H160) filters. The data analysis procedure and GC candidate selection criteria are briefly described. We investigate the interesting "tilt" features in color-magnitude diagrams for this GC system and their link to the nonlinear color-metallicity relation for GCs. The NGC 4874's GC system exhibits a bimodal distribution in the optical g475-I814 color and much more than half the GCs fall in the red side at g475-I814 ~ 1.1. This bimodality is weakened in the optical-IR I814-H160 color; the quantitative analysis on the features of both color distributions using the Gaussian Mixture Modeling code proves the bimodalities are different. Both colors, thus, cannot linearly reflect the bimodality of an underlying metallicity, supporting the suggestion that observed bimodalities in extragalactic GC colors are the metallicity-to-color projection effect.

  • The prescription of Gal Gun Tang, which has been used for treatment of cold, fever, and muscular pain in Chinese herb medicine, is produced in the form of decoction However, the storage problem for this dosage form remains unsettled. Using HPLC and GC, we examined quantitative change of major constituents caused by time-progress and temperature-change. Nine major constituents, such as cinnamaldehyde, cinnamic acid, ephedrine, puerarin, paeoniflorin, daidzin, benzoic acid, glycyrrhizin and liquiritin, were selected as references. The content of cinnamaldehyde significantle decreased by the increase of temperature. While that of cinnamic acid increased. Benzoic acid showed the most significant change of the content in three months at $40^{\circ}C$. It is suggested that most of constituents are considerably stable when kept frozen.

  • The famous prescription of So Cheong Ryong Tang (小靑龍湯)in Chinese herb medicine, which has been used for the treatment of common cold, influenza, asthmatic bronchitis, and bronchial asthma and, is being commercially produced in the form of decoction. However, the storage problem for this dosage form remains unsettled. The quantitative changes of the major constituents, at different temperature and time course, were examined employing HPLC and GC. Cinnamaldehyde, cinnamic acid, ephedrine, albiflorine, paeoniflorin, benzoic acid, glycyrrhizin, and liquiritin were choosen as phytochemical markers. The content of cinnamaidehyde significantly decreased when stored at increased temperature, while that of benzoic acid has increased. By overall consideration of the present experimental data, it was suggested that most constituents are considerably stable when preserved below freezing temperature.

Quantitative Analysis of Ethyl Carbamate in Korean Alcoholic Beverages by Chromatography with Mass Selective Detection (GC/MS 를 이용한 한국 주류 중의 Ethyl Carbamate 정량)

  • In order to determine the contents of ethyl carbamate in Korean traditional alcoholic beverages and general beverages, GC/MS-SIM method was used after extraction of beverages with dichloromethane. The contents of ethyl carbamate in Korean traditional alcoholic beverages, non-distilled alcohol, and whisky were detected in the range of $4.6-50.2{\mu}g/L$, $27.8-45.4{\mu}g/L$, and $24.8-55.1{\mu}g/L$, respectively. The recoveries were ranged from 83.3 to 104.8 %. The values of relative standard deviation were ranged from 1.8 to 14.8 % and the detection limit was $0.3{\mu}g/L$.
