Changes in Endogenous Gibberellin Contents during Bulb Development Period in the Cold-type Cultivar of Garlic (Allium sativum L.) of Korea (한지형 마늘의 인경 발육 과정에서 내생 지베렐린류의 함량변화)

  • This study was performed to investigate the role of phytohormones in the bulbing of garlic in order to assess the yield and quality. The effect on endogenous plant hormones such as gibberellin (GA) content was also examined during growth stage i.e. clove differentiation to bulbing in garlic. More than 18 gibberellins in garlic were identified with extensive gas chromatograph-mass spectrometry-selected ion monitoring (GC-MS-SIM) quantitative analysis. The results showed that GAs were biosynthesized by both non C-13 hydroxylation pathway (NCH) and early C-13 hydroxylation pathway (ECH) in garlic plant. It was also revealed that NCH pathway leading to synthesis of bioactive $GA_4$ was the more prominent GA biosynthesis pathway than ECH pathway in which bioactive $GA_1$ was synthesized. Total GAs level was gradually increased from clove differentiation to bulbing and later decreased, which portrays the active role of GA in differentiation. The biosynthesis ratio of bioactive $GA_4$ and $GA_1$ concentration was similar to that of total GAs content, which was closely related with bulb development in garlic.

  • This experiment was carried out to find reasonable semi. hot seedbed system for early transplanting of "Tongil"rice cultivar. The quality of the young rice-plants, yield, and the occurrence of the reddish dry leaves were not significant differences between the seedbed with polyethylene tunnel and that of Rat covering. The per cent of healthy seedling of the soil preparation with the dry soil plowing was increased than that of the water soil plowing. The stability of the seedling cultivation of the thin layer straw mulching seedbed beneath the polyethylene film was higher than that of the common flat seedbed system.ed system.

Identification of Quantitative Trait Loci Associated with Resistance to Bacterial Pustule (Xanthomonas axonopodis pv. glycines) in Soybean (SSR 분자표지이용 콩 불마름병 저항성 관여 양적형질 유전자좌(QTL) 분석)

  • Bacterial pustule (BP), caused by Xanthomonas axonopodis pv. glycines, is prevalent disease in major soybean production areas. BP can reduce seed yield as well as seed quality. To identify the genomic region associated with the resistance to BP, QTL analysis was conducted using $F_{10}$ RIL (recombinant inbred lines) population, Keunolkong${\times}$Shinpaldalkong. Four QTLs for BP disease were identified on the linkage group B2, D2, I and K in field accounts for 36.4% of the phenotypic variation. Especially, QTL at near of Satt135 on LG D2 was identified in green house experiment explaining 20.9% of the phenotypic variation was found to be a major QTL conferring BP. One of these QTLs, Satt135 on the LG D2, was also identified in green house experiment. In both field and green house condition, the position of major QTL for BP was detected between Satt135 and Satt397 on the LG D2. The major QTL for BP may be used for minimizing soybean BP through effective marker-assisted selection (MAS).

Development of an Alcoholic Drink Using Onion Extract. (양파즙을 사용한 알코올 음료의 개발)

  • This study was carried out to develope an alcoholic drink by fermentation of onion extract using Saccharomyces cerevisiae. The optimal conditions for ethanol production were obtained by standing culture at $25^{\circ}C$ for 5 days with 5% inoculum volume. At the results by flask culture, the growth curve of used S. cerevisiae reached to the stantionary phase at 48 hr and the death phase at 90 hr, whereas ethanol production reached maximum at 114 hr. Under the above conditions, a large scale production was carried out. A standing culture in 5 l fermenter showed the similar results to its flask culture, but progressed 24 hr rapidly more than that of the flask culture. A fed-batch culture was performed by addition of the onionic medium supplemented with 10% (v/v) sucrose after 72 hr from the fermenting start. The fed-batch culture could prevent S. cerevisiae from entering into the death phase and maintain constant level of alcohol production. A continuous culture was able to carry out by adding per every 24 hr the onionic medium supplemented with 10% (v/v) sucrose after 72 hr from the fermenting start. Although S. cerevisiae used showed a little decreased growth, alcohol production maintained roughly the constant level at the maximum yield. To enhance the quality of this alcoholic drink, $2-O-{\alpha}-D-glucopyranosyl$ L-ascorbic acid (AA-2G) was supplemented into the onion extract of the substrate for fermentation. As resulted at this study, this alcoholic drink containing AA-2G should be used as a functional fermented alcohol drink strengthened with vitamin C.

  • In previous paper(Lee et al., 1984), preparation formula and processing conditions of the fish meat (mackerel) paste using dielectric heating were described, that included the proper shape and size of product and the conditions of dielectric heating, hot air dehydration, and heating with electric heater to yield the minimum expansion and case hardening during heating and to controll the final rater activity of 0.86 to 0.83 accompanying with a complete reduction of viable cells and good texture. In present study, changes in VBN, pH, total plate count, water activity, texture, the loss of available lysine, color indexes, TBA value, and the content of TI were determined to assess the quality stability and shelf-life of the product during the storage for 35 days at $5^{\circ}C\;and\;25^{\circ}C$, respectively. And the effect of vacuum sealing and hot water treatment before storage on the storage stability of product was also mentioned. As the product was vacuum packed in K-flex film bag, heat treated in boiling water for 6 minutes, and stored, water activity was maintained 0.86 to 0.84 for 35 days regardless of storage temperature, and the increase of total plate count was negligible in case of $5^{\circ}C$ storage while tended to gain slightly after 25 days at $25^{\circ}C$ storage. Changes in VBN was also minimum with an increase of 1.5 mg/100g at $5^{\circ}C$ and 7.0mg/100g at $25^{\circ}C$, but in case of unpacked sample, it was 24.5mg/100g at $5^{\circ}C$ and 42.4 mg/100g at $25^{\circ}C$ even after 7 days. In textural property hardness tended to increase after 28 days and folding test score was down to A or B from AA grade. The loss of available lysine was $7.5\%\;at\;5^{\circ}C$ and $17.0\%\;at\;25^{\circ}C$ but brown color was not deeply developed as the color index score indicated. TBA value was not increased at $5^{\circ}C$ while it tended to increase rapidly after 30 days at $25^{\circ}C$. Changes in TI content was not obvious except that it showed a tendency of increase at the end of storage as well as in the change of lysine and TBA value. It is concluded from the results that the quality of the product, pasteurized and water activity controlled by dielectric heating, and vacuum packed in K-flex film would be stable for more than 35 days at $5^{\circ}C$ and at least 25 days even at room temperature.

A Mid-late Maturing Rice Cultivar with High-Quality and Bacterial Blight Resistance "Jinbaek" (벼 중만생 고품질 흰잎마름병 신균계(K3a) 저항성 품종 "진백")

  • A new rice cultivar "Jinbaek" carrying Xa3 and xa5 was derived from the cross between 'HR15204-38-3' with xa5 gene resistant to bacterial blight K1, K2, K3 and K3a, and F1 plant derived from the cross between Junam and Sindongjin with Xa3 gene. "Jinbaek" has approximately 125 days of growth duration from transplanting to harvesting in the west-southern coastal and Honam plain of Korea. Culm length of "Jinbaek" is 71 cm. In reaction to biotic stresses, it shows moderate resistance to blast, and wide spectrum resistance to bacterial blight pathogen, K1, K2, K3, and K3a but susceptible to rice stripe virus and blast. The milled rice of "Jinbaek" exhibits translucent, relatively clear non-glutinous endosperm and midium short grain. It has lower amylose content (18.8%) and protein content (6.2%) compared with Nampyeong. The milled rice yield of this cultivar was 5.30 MT/ha in local adaptability test of three years from 2006 to 2008. This cultivar would be adaptable to the bacterial blight-prone area in the south-western coast and Honam plain of Korea.

  • 'Dacheong', a new japonica rice variety developed from a cross between Iksan450 having a good eating-quality and multi-disease resistance, and YR21258-GH3 having insect resistance, was developed by the rice breeding team of Department of Rice and Winter Cereal Crop, NICS, RDA in 2008. This variety has about 125 days growth duration from transplanting to harvesting in west-southern coast, Honam and Youngnam plain of Korea. It has 87 cm culm length and tolerance to lodging. In reaction to biotic and abiotic stresses, it shows resistance to blast, bacterial blight pathogen races from $K_1\;to\;K_3$, stripe virus and brown plant hopper. The milled rice of 'Dacheong' exhibits translucent, relatively clear non-glutinous endosperm and medium short grain. It has slightly lower amylose content of 18.8% and lower protein content of 5.7%, and good palatability of cooked rice compared with Nampyeongbyeo. The milled rice yield performance of this variety is about 5.91 MT/ha in local adaptability test for three years. 'Dacheong' would be adaptable to west-southern coast, Honam and Youngnam plain of Korea.

  • 'Honong' is a new japonica rice variety developed and registered by the rice breeding team of Department of Rice and Winter Cereal Crop, NICS, RDA in 2009. This variety was derived from mutagen MNU (N-methyl-N-nitrosourea) treatment on fertilized egg cells of Unbong31. This variety has about 126 days growth duration from transplanting to harvesting in west-southern coast, Honam and Youngnam plain of Korea. It has 79 cm in culm length and is tolerant to lodging. In reaction to biotic and abiotic stresses, it show resistance to blast, bacterial blight pathogen races from $K_1$ to $K_3$ and stripe virus, but susceptible to other major diseases and insect pests. The milled rice of 'Honong' exhibits translucent, relatively clear non-glutinous endosperm and medium short grain. It has higher amylose content (20.3%) and lower protein content (6.3%), and good palatability of cooked rice compared with Nampyeong. The milled rice yield performance of this variety is about 5.44MT/ha in local adaptability test for three years. 'Honong' would be adaptable to west-southern coast, Honam and Youngnam plain of Korea.

Effects of Level of CP and TDN in the Concentrate Supplement on Growth Performances and Carcass Characteristics in Hanwoo Steers during Final Fattening Period (한우거세우 비육후기 배합사료의 적정 조단백질 및 에너지함량 규명에 관한 연구)

  • Seventy two Hanwoo steers in final fattening period ($585.87{\pm}41.02kg$) were randomly assigned to 3 groups, LPLT (relatively low protein and low energy; CP 12%, TDN 73%), LPHT (relatively low protein and high energy; CP 12%, TDN 75%) and HPHT (relatively high protein and high energy; CP 14%, TDN 75%) in concentrate feed for 163 days in order to investigate the effects on growth performance, carcass characteristics, and longissimus dorsi muscle's chemical compositions. Rice straw was also fed as a roughage. Because ADGs were higher in LPLT and HPHT than LPHT, feed efficiencies were improved in LPLT and HPHT group (P<0.05). Feeding concentrates with different CP and TDN levels had affected to improve back fat thickness and rib eye area in HPHT group but had no effect on carcass weight and meat yield index. Carcass weight for LPLT, LPHT and HPHT were $420.75{\pm}30.56$, $417.05{\pm}32.03$ and $418.32{\pm}32.03kg$, respectively. Meat quality grade was improved in HPHT (P<0.001), because the marbling score was highest in HPHT group. Auction prices (carcass/kg) of LPLT, LPHT and HPHT group were 17,904 won, 18,094 won and 18,899 won, respectively. The percentage of animals over grade 1 appeared in LPLT, LPHT and HPHT were 79.2, 72.7 and 90.8%, respectively. The results of chemical analysis of longissimus dorsi muscle showed no difference between groups but crude fat composition tended to be higher in HPHT group (P=0.088) than the other groups. Stearic acid contents in the muscle was significantly increased in HPHT group than LPLT group (P<0.05). Myristoleic acid and oleic acid composition in HPHT group was higher than LPLT and LPHT group. These results supported the hypothesis that supplementation of higher levels of crude protein and energy in concentrates to Hanwoo steers' during final fattening period improved the growth performance and the carcass quality grade.

The Assessment of Cattle Slurry Application on Productivity and Feed Values of Barley and Hairy Vetch Influenced by Sowing Methods (보리와 헤어리 베치의 단, 혼파 재배 시 우분뇨의 시용에 따른 생산성과 사료가치의 평가)

  • This trial was conducted to assess effects of cattle slurry application on productivity and feed values of barley and hairy vetch when they were influenced by single or mixed-sowed cultivation in paddy fields, and to obtain good quality of organic forage resources. The results summarized are as follows. For barley and hairy vetch, single-sowed cultivation was lower in annual dry matter (DM) and total digestible nutrients (TDN) yields than mixed-sowed (P<0.05). Although crude protein (CP) contents for the barley within single-sowed treatments were lowest as 6.5%, those of hairy vetch within the same sowed method were highest as 16.7%. However, mixed-sowed cultivation with barley and hairy vetch, showing 13.8% CP content, was significantly (P<0.05) higher than CP and relative feed value (RFV) of barley alone treatment. For barley alone treatment, cattle slurry application significantly increased annual DM and TDN yields in comparison with treatments of P+K fertilization as chemical fertilizers and no fertilizer as controls. Results from cattle slurry application showed 84% and 82% in contrast with chemical fertilizer for annual DM and TDN yields, respectively. For mixed-sowed cultivation with barley and hairy vetch, cattle slurry application showed 90% and 94% annual DM and TDN yields, respectively as compared with N+P+K fertilization as chemical fertilizers. Crude protein contents ($14.2{\sim}15.9%$) for cattle slurry application treatments were significantly (P<0.05) higher than those of other treatments. Moreover, cattle slurry application treatment had the highest TDN and RFV among treatments, showing $60.7{\sim}61.8%$ and $112.2{\sim}118.1$, respectively. For hairy vetch alone treatments, annual DM and TDN yields of cattle slurry alone application treatment were highest among fertilization treatments. Furthermore, CP, TDN and RFV of cattle slurry alone application treatments were significantly (P<0.05) higher than those of other treatments. The results showed that mixed-sowed cultivation rather than single-sowed for barley or hairy vetch improved their nutritive value and quality, and also within mixed-sowed cultivation, cattle slurry application increased production yield per ha and CP contents. In the application of above system to organic livestock farming, it would be expected that forages produced by cattle slurry application under mixed-sowed method might become a substitute for foreign organic grain as protein sources.
