  • We attempted to purify radiolabeled RNA using Sephadex G-15 and G-50 chromatography instead of commercial RNA purification kit. In the Sephadex G-15 chromatography the major portion of RNA was eluted in the fractions ranging from 3rd to 5th whereas broad elution profile of RNA was obtained from the Sephadex G-50 chromatography. The elution profile and purity of RNA obtained from Sephadex G-15 chromatography was very similar to that by commercial RNA purification kit. Furthermore, operating time required for purification of RNA by Sephadex G-15 was rather smaller than that by commercial kit. Overall results suggest that the purification of radiolabeled RNA using Sephadex G-15 is more money and time saying than using commercial RNA purification kit.

  • The thermal properties of ramie leaf ${\beta}$-amylase (RBA) were examined to develop a novel process for enzyme purification. The thermostability of RBA extract prepared from ramie leaf powder was examined at various temperatures. RBA activity decreased slightly, whereas other carbohydrate-active enzymes, such as $\small{D}$-enzyme, were rapidly inactivated during 30 min incubation at $60^{\circ}C$. When the heat-treated extract was incubated with various substrates, maltose was produced exclusively as the major product, whereas the untreated crude extract produced maltose and other maltooligosaccharides. In sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis, fewer protein bands were observed for the heat-treated extract than the untreated extract, indicating that the thermostable RBA was partially purified and other thermolabile enzymes were eliminated. Thus, the treatment of the RBA extract at $60^{\circ}C$ for 30 min resulted in 5.4-fold purification with a recovery yield of 90%.

  • Objective is to design a high performance purification filter system of reactor coolant and seal injection system at nuclear power station. The purification filter systems play an important role in the stability of the nuclear and volume control system which consist the primary network systems of the nuclear power station. But the users of the purification filter systems frequently suffer from high maintenance cost which comes from lack of understanding of the system technology and domestic suppliers. It is time to establish a high performance domestic filter system manufacturing technology and optimum design for wide use in industrial applications.

  • This study investigated the purification process of groundwater contaminated with zinc and arsenic using a permeable reactive barrier with a zero-valent iron/pumice mixture. We determined the removal rates of the contaminants for 30 days. In this study, column reactor filled with the zero-valent iron/pumice reactive mixture was used. Experimental results showed that the mixture exhibited an almost complete removal of the zinc and arsenic ions. Arsenic was removed via co-precipitation and adsorption processes while zinc ions were asorbed in active sites. The purification process of water from the metal ionscontinued for 30 days with constant hydraulic conductivity because of the enhanced porosity of the pumice and interparticle distance between the zero-valent iron and pumice. Contaminants removal rates and the remediation mechanism for each reactive system are described in this paper.

  • Membrane filtration is a promising technology for efficient solid/liquid separation in water purification. In FY 1991, the Ministry of Health and Welfare, Japanese Government launched a comprehensive research project "MAC 21" for development of membrane technology and its application to public water supply. The project was conducted by the Water Purification Process Association (WPPA), under the supervision of the Institute of Public Health. By the research project from FY 1991 to FY 1993, we confirmed that microfiltration (MF)/ultrafiltration (UF) technology was applicable to water purification and MF/UF was a effective method for the removal of such contaminants as particulate matter and coliforms. The Guideline Committee organized under the Technical Committee prepared a the guidelines on application of membrane system to small-scale public water supplies, based on the results as written above. The guidelines has been published in Dec., 1994 by WPPA.4 by WPPA.

  • As crude oil with heavier and/or highly oxidized components prevails, purification technologies such as desulfurization, denitrilization and demetalization have become important issues to control contents of sulfur and other impurities affecting the quality of petroleum. Also, the importance of catalyst technologies related with crude oil purification has been emphasized to control the production and yield of products. In this paper, technology trends of impurity removal such as sulfur, nitrogen and metal components from crude oil and catalysts related with purification of crude oil were studied through patent analysis. The patents published or registered in Korea, U. S. A., Japan, and Europe from mid 1970's to 2009 had been analyzed based on the application tendency, the distribution of major applicants, and their active indices, etc. The technology flow was figured out to see the technology trends.

  • Water purification facilities utilizing three technologies - a detention pond, an artificial wetland and an ecological revetment - were installed in waterfront spaces, including river embankments and watersides that have so far been left neglected, and then their water purification levels were analyzed in this study. The water purification facilities were found to show average removal efficiencies of 48.6% in suspended solid (SS), 30.5% in biochemical oxygen demand (BOD), 18.4% in total nitrogen (T-N) and 27.3% in total phosphorus (T-P) during rainfall. The removal efficiencies during non-rainfall were 33.2% in SS, 28.6% in BOD, 13.7% in T-N and 17.3% in T-P. These results showed that the water purification facilities using a detention pond, an artificial wetland and an ecological revetment can be used as a useful natural water purification technology in in waterfront spaces.

  • The concern over the use of melittin in honey bee venom due to its adverse reaction caused by allergens such as phospholipase A2 ($PLA_2$) and hyaluronidase (HYA) has been an obstacle towards its usage. We developed a novel single-step method for melittin purification and the removal of $PLA_2$ and HYA. This study explores the influence of pH, buffer compositions, salt concentration, and types of cation-exchange chromatography resins on the recovery of melittin and the removal of both HYA and $PLA_2$. Melittin was readily purified with a strong cation-exchange resin at pH 6.0 with sodium phosphate buffer. It resulted in a recovery yield of melittin up to 93% (5.87 mg from a total of 6.32 mg of initial melittin in crude bee venom), which is higher than any previously reported studies on melittin purification. $PLA_2$ (99%) and HYA (96%) were also successfully removed. Our study generates a single-step purification method for melittin with a high removal rate of $PLA_2$ and HYA, enabling melittin to be fully utilized for its therapeutic purposes.

  • HF purification performance of an ion exchange membrane(IEM) was evaluated with 0.5% HF spiked with 10ppb of Fe, Ni and Cu nitrates. The result show that after less than five turnovers through an IEM, the metallic impurity concentration drops below 1ppb. The decrease rate can be fitted to a model assuming the experimental tanks to be continuously stirred tank reaction and that the metallic impurity concentration after the IEM is a function of the single-pass purification efficiency of the membrane, the concentration before purification and the metals desorbed form the IEM. The Concentration after purification was investigated up to a cumulative Fe loading of 300ppb in the 23 liter recirculated loop. It increases linearly vs. cumulative loading and can be explained by the Langmuir theory resulting in a purification efficiency at the equilibrium of close to 99.5% in this loading regime.
