  • The putative endochitinase gene, yheB of Escherichia coli K-12 is not expressed under lab culture conditions. The endochitinase gene was amplified by PCR and subcloned into pET28c vector and pQE9 vector, respectively. The endochitinase produced in E. coli harboring pET28c containing yheB or pQE9 vector containing yheE was partly released into the growth medium. The overproduced endochitinase was partially purified by His affinity column chromatography and DE-52 column chromatography. The apparent molecular weight of the endochitinase determined by SDS-polyacrylamide gel electrophoresis was about 97,000. The purified E. coli endochitinase showed maximal chitinolytic activity at pH 6 and $40^{\circ}C$.

  • The aquatic animals of Tan stream were composed of 46 species, 28 families, 11 orders, 6 classes in 4 phyla during the survey period of April, 1996 to December, 1996. They were composed of 31 species in aquatic insecta, 6 species in annelida, 3 species in mollusca, 1 species in crustacea, and 5 species in fishes, respectively. Major dominant species in Tan stream were Chironomidae sp.1, Chironomidae sp.2, Chironomidae sp.3, Tubufucidae sp.1, Physa acuta and hirudo niponica. Dominance indices of benthic macroinvertebrates ranged highly from 95.74 to 100.00% at lower stream(site 4), but ranged 50.00 to 95.85% at site 1 through site 3. The aquatic animals ranged from 25 to 32 species at site 1 through site 3, but they were only 3 species at site 4 for survey period. Tan stream in the light of urban stream ecosystem has a little less riffle areas and hydrophyte areas by cementation of riparian area and channel type of water course. Therefore, the species of aquatic animals in Tan stream decreased because of deterioration of water quality according to reduction of self-purifcation ability and loss of microhabitat according to reduction of hydrophyte areas and riparian areas. The tendency of decreasing species of aquatic animals appeared seriously at lower stream From drive licence test authority at Kangnam-ku, Seoul to conjunction point of the Han river.

  • Distribution of tellurite and tellurate-reducing enzymes in the cell of Nocardia sp, the purifcation and the chemical properties of enzymes were investigated. Tellurite- and tellurate-reducing enzymes were located in the cytoplasm, but T. T. C. reduction part was in the cell membrane. Purification of tellurite- and tellurate-reducing enzymes was possible with the application of ammonium sulfate precipitation method and DEAE-Cellulose or CM-Cellulose column chromatographic method from the crude soluble part of the cell. On investigating the properties of purified enzyme, one of NADP, NADPH and reductive methylene blue(leucomethylene blue) was thought to react as a hydrogen donor. Both NADH and NADPH, or either of them would be physiological hydrogen donor.) In the reaction of this enzyme, either tellurite or tellurate reacts as a hydrogen acceptor, but on the other hand either selenate or selenate also reacts as a hydrogen acceptor.

  • To evaluate the purification capacity of pollutants (COD, $PO_4^{3-}-p$, $NO_3^{-}-N$, Cu, Cd, Pb) in the three tidal flats, Eueunri, Gyewhado, Chunjangdae, the experiments were carried out with the batch reactor equipped with artifical tidal flats. Eueunri tidal flat was $98.8\%$ content of silt and clay and Chungjangdae was $97.84\%$ content of sand. The organic matters (I.L., $COD_{sed}$, POC) in Eueunri tidal flat were 2$\~$8 times higher than other tidal flats. The purification capacity of COD was 0.75 kg/ha/12 hr in Eueunri, 0.60 kg/ha/12 hr in Gyewhado and 0.55 kg/ha/12 hr in Chunjangdae. The mean purification capacity of COD in three tidal flats was 1.27 kg/ha/day. The calculated purification capacity of COD was 25.4 ton/day in the disappeared tidal flat areas (20,000 ha) of the Saemangeum reclamation. The purification capacity of phosphorus was 0.21 kg/ha/12 hr in Gyewhado, 0.39 kg/ha/12 hr in Eueunri and 0.22 kg/ha/12hr in Chunjangdae. The nitrate was 0.53, 0.74 and 0.43kg/ha/12hr, respectively. The purification capacity of heavy metals (Cu, Cd, Pb) were 88.9g/ha/12hr, 11.0g/ha/12hr, 1.7g/ha/12hr in Gyewhado, 89.1g/ha/12hr, 18.0g/ha/12hr, 2.6g/ha/12hr in Eueunri and 55.3g/ha/12hr, 18.0g/ha/12hr, 2.1g/ha/12hr in Chunjangdae, respectively. Accordingly, the purification capacity of pollutants in Eueunri tidal flat with high contents of organic matter were higher than other tidal flats. So, the purification capacity of pollutants were affected by physical and/or chemical characteristics of tidal flats.
