Proteomic Changes by Acupuncture Stimulation at HT7 in the Hippocampus of Rat Pups (신문혈 자침이 어린 백서 해마의 단백질 발현에 미치는 영향)

  • Objectives : Hippocampus, a region of temporal lobe, plays an important role in the pathogenic mechanisms of brain diseases such as Alzheimer's disease, depression and temporal lobe epilepsy. This research is designed to investigate hippocampal changes after acupuncture stimulation at Shinmun(HT7) using 2-dimensional gel electrophoresis(2-DE). Methods : On postnatal-day 15, rat pups were randomly devided into Normal(NOR) or HT7 group. All of Pups kept with their mothers for 7 days, but pups in HT7 group received acupuncture stimulation at HT7 daily. On postnatal-day 21, hippocampus of each rat pup was dissceted 30 minutes after last acupuncture stimulation and the protein expressions were investigated using 2-DE. Results : After acupuncture stimulation at HT7, expression of 20 proteins were significantly increased. Succinate semialdehyde dehydrogenase, glyceraldehyde-3-phosphate dehydrogenase-like, transketolase, aconitate hydratase and phosphoglucomutase-1 were related to glucose methabolism. Eukaryotic initiation factor(eIF) 4A-II, eIF 4A-III, mitochondrial Tu translation elongation factor and chain A of crystal structure of the 70-Kda heat shock cognate protein involve in the protein synthesis in ribosome. Tubulin ${\beta}$-4 chain, tubulin T ${\beta}$-15 and tubulin ${\alpha}$-1B chain comprise cytoskeleton. Glutathione S-transferase(GST) ${\omega}$-1, GST P and GST Yb-3 can reduce oxidative stress. ${\beta}$-soluble N-ethylmaleimide-sensitive fusion protein attachment protein is required for vesicular transport between the endoplasmic reticulum and the Golgi apparatus, glycerol-3-phosphate dehydrogenase plays a major role in lipid biosynthesis, creatine kinase U-type catalyses the conversion of creatine and consumes adenosine triphosphate to create phosphocreatine and adenosine diphosphate. Platelet-activating factor acetylhydrolase IB subunit alpha and voltage depedent anion-selective channel protein 2 were also increased. Conclusions : The results suggest that acupuncture stimulation at HT7 may enhance glucose and lipid metabolism, protein synthesis, cytoskeletal substance and anti-oxidative stress in hippocampus.

Influence of Prenatal Noise and Music on the Expressions of c-Fos and Nitric Oxide Synthase in the Hippocampus of Rat Pups

  • The expressions of c-Fos and nitric oxide synthase (NOS) represent neuronal activity and play' a crucial role in the shaping of the development of brain. During the late pregnancy, stresses may influence neuronal activity of prenatal rats. In the present study, the effects of prenatal noise and music on the expressions of c-Fos and NOS in the hippocampus of rat pups were investigated. Exposure to the noise during pregnancy decreased c-Fos and NOS expressions in the hippocampus of rat pups, whereas exposure to music during pregnancy increased c-Fos and NOS expressions in the hippocampus of rat pups. The present results show that prenatal music stimulation may increase neuronal activity of rat offspring, whereas exposure to noise during pregnancy may reduce the neuronal activity of offspring. The present study suggests that prenatal stimuli including noise and music could affect the fetal brain development.

The Effect of Early Intervention and Rehabilitation in the Expression of Aquaporin-4; and Ultrastructure Changes on Rat's Offspring's Damaged Brain Caused by Intrauterine Infection

  • Objective : To study the effect of early intervention and rehabilitation in the expression of aquaporin-4 and ultrastructure changes on cerebral palsy pups model induced by intrauterine infection. Methods : 20 pregnant Wistar rats were consecutively injected with lipopolysaccharide intraperitoneally. 60 Pups born from lipopolysaccharide group were randomly divided into intervention group (n=30) and non-intervention group (n=30); intervention group further divided into early intervention and rehabilitation group (n=10), acupuncture group (n=10) and consolidate group (n=10). Another 5 pregnant rats were injected with normal saline intraperitoneally; 30 pups born from the normal saline group were taken as control group. The intervention group received early intervention, rehabilitation and acupuncture treatment. The motor functions of all pups were assessed via suspension test and modified BBB locomotor score. Aquaporin-4 expression in brain tissue was studied through immunohistochemical and western-blot analysis. Ultrastructure changes in damaged brain and control group were studied electron-microscopically. Results : The scores of suspension test and modified BBB locomotor test were significantly higher in the control group than the intervention and non intervention group (p<0.01); higher in the intervention group than the non-intervention group (p<0.01). The expression of Aquaporin-4 was lower in intervention and non intervention group than in the control group (p<0.01); also lower in non-intervention group than the intervention group (p<0.01). Marked changes were observed in ultrastructure of cortex and hippocampus CAI in brain damaged group. Conclusion : Early intervention and rehabilitation training can improve the motor function in offspring with brain injury and reduce the expression of aquaporin-4 in damaged brain.

The Effects of Multi-minerals on Susceptibility to Lead Toxicity in Rats

  • Female Wistar rats were randomly divided into 5 groups: Control, received distilled water; Low lead, received 0.5 g/ιlead (as acetate) in drinking water; High lead, received 2.0 g/ιlead; Low lead + Minerals, received 0.5 g/ιlead in drinking water and received minerals (Ca$^{2+}$, 25 mg/kg/day; Fe$^{3+}$, 0.47 mg/ kg/day; Zn$^{2+}$, 0.33 mg/kg/day; Se, 0.83 $\mu\textrm{g}$/kg/day) by gavage; High lead + Minerals, received 2.0 g/ιlead and received the same minerals. Animals exposure to lead was from 10 days before mating till postnatal day 21; and the minerals was administered from the first day of pregnancy and during lactation. No statistical difference was found either in body weights or in blood lead levels between the pups received minerals and those only exposed to lead at the same dose. The developmental and behavioral teratological effects of lead on pups, such as time-lag of eye opening, pinna detachment, fur developing, incisor eruption, ear unfolding, and surface righting were observed in this study; and the minerals decreased the toxicity of lead either in low or in high lead exposure pups. The numbers of step-down were significantly increased in lead exposed animals, and the effect of intervention by the minerals was appeared only in the pups exposed to low lead. The ChAT activity and levels of glutamate and aspartate in hippocampus decreased in treated animals compared to control animals, no effect of intervention by the minerals was found. The results of this study indicate that the applied multi-minerals can alter the outcome of develop-mental lead poisoning in rats.s.s.s.

Elimination of Lead by TTFD and TPD from Central Nervous System of Postnatally Lead-exposed Rats (출생후 납중독흰쥐에서 TTFD 및 TPD에 의한 중추신경계 납의 제거 작용에 관한 연구)

  • Amount of lead burden in a tissue reflects poisoning of lead in that tissue, so is the removal of lead directly connected to curement of lead poisoning. The purpose of present study was to investigate the relative effects of penicillamine and thiamine tetrahydrofurfuryl disulfide (TTFD) or thiamine propyl disulfide (TPD) in the removal of lead from rat brain tissue treated with excessive lead. Wistar rat pups of both sexes were used in this experiment. Within 1 day of parturition, experimental mothers nursing their pups as well as rat pups were given drinking water containing 0.2% lead acetate, TTFD 20mg/1.2 L (2 mg/kg/day), TPD 20 mg/1.2 L (2mg/kg/day), penicillamine 40 mg/1.2 L (40 mg/kg/day), 0.2% lead acetate+TTFD 20mg/1.2 L (2 mg/kg/day), 0.2% lead acetate+ TPD 20 mg/1.2 L (2 mg/kg/day) or 0.2% lead acetate+ penicillamine 40 mg/1.2 L (40 mg/kg/day) ad libitum, throughout the entire period of experiment. Rat pups in the control group received normal tap water. The animals were sacrificed by decapitation on the day when they become 2 or 8 weeks of age. Brains were dissected into five regions: telencephalon, diencephalon, midbrain, pons/medulla and cerebellum. The dissected brain tissues were lyophillized and then solubilized by acid mixture (nitric acid + sulfuric acid). Lead levels in the solubilized brain tissues were measured by the inductively coupled plasma. In lead-exposed rats, lead levels were significantly higher than those of control group in all brain legions, lead levels in brain regions of TTFD or TPD group were generally lower than those of control group. The simultaneous administration of lead with TTFD or TPD to animals caused significant decrement of lead from all brain regions. In the elimination of lead from brain regions, effectiveness of TTFD or TPD was equivalant to penicillamine.

  • Objectives : Alteration of hippocampus was demonstrated in the maternal social separation(MSS) pups, separated from dams on postnatal day(pnd) 14 and placed alone. Therefore, to understand the molecular events involved in the MSS, we have initiated a search for gene profiles that are up or down-regulated in the hippocampus of MSS pups. Methods : Analysis of cDNA microarray was performed by using total RNA extracted from the hippocampus of control and MSS pups on pnd 17. Also, passive-avoidance test was demonstrated on pnd 35. Results : Up-regulation of Nedd4a was observed in the hippocampus of MSS pups. Also, MSS rats showed less elongation of latency in passive avoidance test. Conclusion : We suggest that environmental effects of MSS may be altered the neural and/or glial differentiation and synapse formation-related genes which may lead cognitive alterations in MSS rats.

  • The objective of this study was to assess that the effects of DEHP administration on reproductive characteristics and blood hematological and chemical values in pups born after DEHP administration in pregnant mice. DEHP was administrated to pregnant mice by intraperitoneally injection with 0, 0.5, 1.0 and 10.0mg/kg B.W, 5 times at 3 days interval from Day 1 to Day 16 in the gestation period. The body weight and reproductive organ weight(testis, epididymis and coagulating gland) in male pups on 45 day after birth was not affected in all experimental groups, but vesicular gland in DEHP groups was significantly lower than that of control group(P<0.05). The semen characteristics of male pups were not affected in DEHP treatment groups. The WBC, HB, HT, MCH and albumin values in male pups were not affected in all experimental groups, but RBC MCV, MCHC, PLT and total protein values were significantly different among the experimental groups(P<0.05). In female pups, the effects of DEHP administration were not affected the body and uterus weight, but the left ovary in 10.0mg DEHP group was significantly heavier than in control and 0.5mg DEHP group(P<0.05). The WBC, MCV, MCH, MCHC, PLT, albumin, BUN and total protein values in female were not different in all experimental groups. The RBC, HB and HT values were significantly different among the experimental gruop(P<0.05). The historical evaluation of testis in male pups that were grown to 45 days after birth was not different in all experimental groups. The ovary in female pups had many corpus luteum in 10.0mg DEHP group. The endometriosisi of uterus was significantly decreased in DEHP group. There results suggest that low concentration of DEHP administration in pup born after DEHP administration in pregnant mice was not affered on reproductive characteristic, but was affected on blood hematological and chemical values.

INFLUENCE OF ENDOGENOUS SOMATOSTATIN ON LACTATION IN RATS

  • The role of endogenous somatostatin on lactation in rats was examined by passive immuno-neutralization of Wistar rats. In one study, the rats were given either immunoglobulin raised in sheep against somatostatin, or non-specific sheep immunoglobulins by daily s.c. injection from parturition through the first two weeks of lactation. The growth of the pups was recorded by weighting every second day, and the milk yield calculated from the pup weight and weight gain. Immunoneutralization of maternal somatostatin during pregnancy had a slight effect (p < 0.05) on the mean birth weight of the pups but no subsequent effect on postnatal growth rate of the pups or milk yield ($25.32{\pm}0.88g/day$) compared with young control rats given normal sheep serum ($25.55{\pm}1.04g/day$). Similarly, passive immunization against somatostatin during lactation ($21.96{\pm}1.57g/day$) also did not affect milk yields compared with controls ($24.85{\pm}1.03g/day$). These data do not support a significant role for endogenous somatostatin in regulating milk production in lactating rats.

Effect of dietary n-3 Polyunsaturated Fatty Acids on Bony Remodeling during Eruptive Tooth Movement

  • The aim of this study was to investigate the effect of n-3 polyunsaturated fatty acids (PUFAs) on eruptive movement during tooth development. Sprague-Dawley (SD) rat pups were randomly divided into two groups; control group and experimental group. The experimental group was administered daily with n-3 PUFA by intraperitoneal (IP) injection. After 10 days postpartum, rat pups were sacrificed to evaluate the effect of n-3 PUFA on eruptive tooth movement. Histological analyses were by hematoxylin-eosin (H&E) staining. Tartrate-resistant acid phosphatase (TRAP) assay was performed to compare the osteoclast distribution in the bone matrix above the developing molar teeth. Incisor teeth eruptions were noticeably observed in IP group, as compared to control group. Rat pups in IP group showed faster tooth eruption on day 8 after birth. Through histological analyses, IP group showed thinner bone matrix and more osteoclasts above the $1^{st}$ molar teeth, as compared to control group. TRAP assay showed significantly stronger stained pattern that the osteoclast above the $1^{st}$ molar teeth in IP group, as compared to control group. The results suggested that n-3 PUFA could affect osteoclastic activity involved in bony remodeling during eruptive tooth movement.

A Transfer of Silver Nanoparticles from Pregnant Rat to Offspring

  • Silver nanoparticles (size: $7.9{\pm}0.95$ nm, dosage: 250 mg/kg) were orally administered to pregnant rats. At 4 days after parturition, four pups were randomly selected (one pup from one dam) and silver level in liver, kidney, lung and brain was determined by ICP-MS and electron microscope. As results, silver nanoparticles highly accumulated in the tissues of the pups. Silver level in the treated group was $132.4{\pm}43.9$ ng/g in the kidney (12.3 fold compared to control group), $37.3{\pm}11.3$ ng/g in the liver (7.9 fold), $42.0{\pm}8.6$ ng/g in the lung (5.9 fold), and $31.1{\pm}4.3$ ng/g in the brain (5.4 fold). This result suggested that the possible transfer of silver nanoparticles from pregnant dams to the fetus through mainly placenta.
