Establishment of Optimal Rearing Conditions for the Production of Tenebrio molitor Larvae (갈색거저리 유충 생산을 위한 효율적인 사육조건 확립)

  • Tenebrio molitor larvae contain large amounts of proteins, lipids and other functional materials, enabling this insect to be used as an edible food source in animal feeds and for industrialization. Although many efforts have been made to set up mass rearing systems, few studies have been conducted to establish optimal rearing conditions for the production of high quality T. molitor larvae. Herein we investigated 1) the effects of additional diets on the survival and fecundity of the insect, 2) the relationship between oviposition period and the uniformity of larval size, 3) the effects of rearing density and temperature on insect development, and 4) the storage stability of eggs and pupae at low temperatures given possible temporary production discontinuation. The addition of carrot and zucchini to the traditional wheat bran diet significantly increased the survival and fecundity rate of adult T. molitor. Of the three different oviposition sampling periods (3, 7, and 14 days) used to investigate the uniformity of the hatched larvae in each treatment, the period of 3 and 7 days provided higher uniformity than the 14 days oviposition period. Larval development was faster at $30^{\circ}C$ than at 20, 25, and $35^{\circ}C$. Interestingly, oviposition rates were highest at $20^{\circ}C$ but showed much slower larval development and lower uniformity at $30^{\circ}C$. Regarding the effect of larval rearing densities (1, 10, 20, 30, 40, and 50 larvae per 90 mm diam. dish), larval weight was significantly reduced at higher rearing densities, but larval longevity and length were not influenced by rearing density. The 30 larvae/dish is suggested to be a reasonable density to be applied to mass production systems. When kept at $4^{\circ}C$, T. molitor eggs showed a significant reduction in hatching rate; however, when stored under the same conditions, pupae emergence rates remained high until 10 weeks, suggesting that storage at low temperatures is more suitable for the pupal stage than the egg stage. Our findings suggest that an increase in T. molitor adult survival and fecundity rates and a uniformity of hatched larval development can be achieved with the following recommendations: a combination diet (including wheat bran), a 7-day oviposition period; a larvae-rearing temperature of $30^{\circ}C$, a rearing density of 30 larvae/dish, and the storage of pupal stages at low temperatures in the case of rearing discontinuation. This study serves as a strong foundation for the successful mass production of high quality T. molitor larvae.

Biological Characteristics and Mass Rearing System for Cadra cautella (Walker) as a Substitute Diet for Natural Enemies (천적의 대체먹이로서 줄알락명나방(Cadra cauteiia)의 생물적 특성과 대량사육)

  • Biological characteristics of the almond moth, Cadra cautella, of which eggs will be substitute diets for Orius spp. and Trichogramma spp., were investigated and a mass-rearing system for the moth was developed. At 25$^{\circ}C$, egg, larval, and pupal period was 4.2, 29.8 and 8.3 days, respectively, and adult longevity was 5.8 days for female and 4.8 days for male. Total number of eggs at 20, 25 and 30$^{\circ}C$ was 128.9,207.9 and 139.9, respectively. The moth could be successfully reared with all food substrates tested, of which rice bran (50%)+chick feed (50%) assumed to be proper for massrearing in view of cost. Eggs could be stocked at 9 C for 7 days, representing 82% hatchability. In the rearing cage (16${\times}$24${\times}$9cm) used, 1,000 eggs was better for initial level of inoculation, showing relatively high emergence rate and adult weight. Mass-rearing procedures were explained in detail.

A Possible Mechanism Related with Non-spinning Syndrome of Bombyx mori that Intimidates the Sericultural Industry in Northern Kyungbuk (경북 북부지역의 양잠산업에 피해를 주고 있는 누에(Bombyx mori) 미화용 기작에 관한 연구)

  • Non-spinning syndrome of Bombyx mori has been serious issue in sericulture industry near Kyungbuk area. This study was focused on the analysis of the mechanism and on screening candidate chemicals inducing the anti-metamorphosis of the silkworms. Rearing temperatures or initial body weight of the final instar larvae did not affect a normal larval to pupal metamorphosis of B. mori. However, pyriproxyfen (a juvenile hormone (JH) agonist) induced follicle patency significantly even at its 10$\^$-8/ M concentration and inhibited metamorphosis of B. mori in both developmental time and dose dependent manners. Pyriproxyfen induced JH esterase (JHE) activity and downregulated expression of JH binding protein of 5. mori. These results suggests that pyriproxyfen induced JHE activity as a JH agonist and that the elevated JHE activity degraded endogenous JH and resulted in JHBP gene expression. Based on the fact that the JH agonist induced follicle patency and inhibited metamorphosis of B. mori, follicle patency bioassay suggested that three commercial pesticides including simazine, molinate or alachlor were proved to give potent JH agonistic effect on B. mori. Further direct exposure experiments to these candidates are required to determine the chemicals responsible for the non-spinning syndrome of 8. mori.

Breeding of Daepoongjam, a Sex-limited Larval Marking and High Silk Yielding Silkworm Variety for Spring Rearing Season (다수성 편친한성(片親限性) 봄누에 품종 "대풍잠" 육성)

  • A new silkworm variety "Daepoongjam" for spring rearing season is $F_1$ hybrid between Jam151, a Japanese race bred from introduction breeding and Jam152, a Chinese race from introduction breeding. In the local adaptatability test performed at 8local areas in spring of 2005, the heavy cocoon yield of "Daepoongjam" was recorded 6% higher than "Kumokjam". The concentration of Deokynojirimycin (DNJ) was measured 4.22 mg in "Daepoongjam", similar to "Kumokjam" (4.26 mg). In the Peacilomyces tenuipes production ability test, the pupal weight of "Daepoongjam" was 1.61 g/individual 13% heavier than "Kumokjam" (1.41 g/individual)

Inhibitory Effect of Cotesia plutellae Bracovirus (CpBV) on Development of a Non-natural Host, Spodoptera exigua (프루텔고치벌(Cotesia plutellae) 유래 폴리드나바이러스의 비자연 기주 파밤나방(Spodoptera exigua)에 대한 발육 억제 효과)

  • Polydnavirus is a symbiotic virus of some endoparasitic wasps and plays crucial roles in inhibiting immune responses and retarding development of the parasitized hosts. Cotesia plutellae bracovirus (CpBV) is a polydnavirus suggesting a major causative to change developmental physiology of the parasitized host. Here, we investigated whether CpBV can interrupt development of non-natural host. Beet armyworm, Spodoptera exigua, is used as a non-permissible host for parasitization of C. plutellae. Extract from the calyx region of C. plutellae contained CpBV, which was confirmed by immunoblotting with a polyclonal antibody raised against CpBV. One female equivalent of CpBV extract was injected into hemocoel of late 4th instar larvae of S. exigua. The injected larvae showed delayed larval period, decrease of body weight gain, and inability of pupal metamorphosis. These inhibitory effect of the CpBV extract was rescued by injection along with CpBV antibody, though the antibody itself did not give any effect on development of the larvae. This result clearly shows that CpBV can interrupt developmental physiology of a non-natural host for its symbiotic wasp.

Investigation of Biological Characteristics of Rice Stem Borer (Chilo suppressalis Walker) and Variation in its Occurrence Patterns Among Crops (작물별 이화명나방의 발생양상 변동과 생물적 특성 연구)

  • We analyzed the variation in occurrence time and intensity of attacks by the rice stem borer (Chilo suppressalis Walker) on several crops. This study aimed to understand how C. suppressalis damages crops and to utilize this basic data for the establishment of environment-friendly control methods like pheromone traps. This study surveyed the changes in occurrence patterns of C. suppressalis, biological characteristics of overwintering larvae, and the efficacy of different types of pheromone traps the years. We found similar occurrence patterns of C. suppressalis in different crops. In addition, occurrence time of the first generation was advanced. Overwintering larvae showed no difference in pupal period and weight compared to the non-overwintering ones. However, the larval period was reduced to 19.1 days in Miscanthus field, in contrast to that in the paddy fields. It was confirmed that larvae of C. suppressalis generally prefer the lower part of the stems of Miscanthus. Efficiency of the emergence trap was confirmed to be greater than that of the delta trap for capturing C. suppressalis adults. However, it is necessary to adjust the control period because of the advancement in occurring time of C. suppressalis in recent years. The larvae of C. suppressalis experience favorable environmental conditions for overwintering in Miscanthus fields. The major Miscanthus fields are generally located in the areas protected for sources of drinking water, owing to which spraying of chemical pesticides is very limited. The results of this study provide important inputs for the development of environment-friendly control methods.

Physiological Effects of Herbicide-resistant Genetically Modified Rice (Milyang 204 and Iksan 483) Developed in Korea on Non-target Insects and a Spider (국내에서 개발된 제초제저항성 벼(밀양 204호, 익산 483호)의 비표적 곤충과 거미에 미치는 생리적 영향)

  • In the present study, we investigated the effects of two herbicide-resistant genetically modified rice (GM rice) varieties, Milyang 204 and Iksan 483, recently developed in Korea on non-target insects and a spider. No difference in host preferences of the English grain aphid Sitobion avenae and the brown planthopper Nilaparvata lugens were observed between GM rice and non-GM rice. Wolf spider Pirata subpiraticus, feeding on N. lugens reared on GM rice or non-GM rice, revealed no significant difference in body weight. P. subpiraticus, fed with N. lugens reared on Milyang 204, showed survival rates similar to that in P. subpiraticus fed with N. lugens reared on non-GM rice. However, P. subpiraticus feeding on N. lugens reared on Iksan 483 demonstrated significantly lower survival rates than that in P. subpiraticus feeding on N. lugens reared on Milyang 204 or non-GM rice. In addition, when larvae of the western honeybee Apis mellifera were supplied with Iksan 483 pollen, a significantly longer pupal period occurred, as compared with that of A. mellifera supplied with pollen of Milyang 204 or non-GM rice. As GM rice has negative effects on P. subpiraticus, which is an important predator in agricultural ecosystems, and on A. mellifera, which plays important roles in pollination and honey production, additional studies on risk assessment of GM rice should be conducted before releasing newly developed herbicide-resistant GM rice to the agricultural environment.

Developmental Characteristics of Zophobas atratus (Coleoptera: Tenebrionidae) at Varing Temperatures (온도에 따른 아메리카왕거저리(Zophobas atratus)의 발육 특성)

  • We investigated the developmental characteristics of super mealworm (Coleoptera: Tenebrionidae) at four different temperatures (25℃, 27℃, 30℃, and 33℃). The rearing conditions were 9L/15D, 65% RH, and 1,330 to 1,800 lux with wheat bran. The length of developmental period of 1 to 18 instars at each temperature showed that 30℃ was the shortest at 120.0±5.8 days, compared to that at 27℃ (132.6±10.7 days), 33℃ (136.5±9.2 days), and 25℃ (156.7±7.5 days). The larval developmental period was statistically significantly longer at 25℃, 27℃, and 30℃ compared to the length at 33℃. However, the death rate of larvae at 33℃ was 2.7-3.3 times higher than the rate at other temperatures. Body weight was heaviest at 30℃ followed by 27℃, 33℃, and 25℃. The patterns of head capsule, body capsule, and body length were similar to that of body weight. Regression analyses of developmental period, larval body weight, and length according to temperature revealed 29-30℃ as the most suitable temperature. The prepupa rate was 43.1% in 17 instars, 30.3% in 18 instars, 15.4% in 16 instars, 7.1% in 19 instars, 2.2% in 15 instars, and 1.9% in 20 instars, accounting for 88.8% in 16-18 instars. Prepupal period was longer at lower temperatures. For the average prepupal period of 15-18 instars, prepupa time was 18.8±1.9 days at 27℃, 18.8±2.3 days at 30℃, 23.0±2.4 days at 33℃, and 23.1±2.9 days at 25℃. The average pupal period of females and males was 11.1±2.2 and 11.6±2.4 days, respectively. The data indicate that the most suitable rearing temperature of super mealworm was 30℃.

Breeding of New Silkworm Variety Golden silk, a Yellow Cocoon Color for Spring Rearing Season (황색실크 생산용 특수누에 품종 '골든실크' 육성)

  • A new silkworm variety Golden silk for spring rearing season is $F_1$ hybrid between Jam311, a Japanese race bred from introduction breeding and Jam312, a Chinese race from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2006, "Golden silk" was showed 7 percent higher in hatchability than authorized silk-worm races laval. "Golden silk" was receded higher 1.7 percent, 2.2 percent in pupation rate, best cocoon rate respectively than Chunsujam. The larval period shortened for 1.12 day even though low record to the several items including productivity than Chunsujam. "Golden silk" was showed 1,031 m, 2.97 denier in length of cocoon filament, weight of cocoon filament respectively. The concentration of Deoxynojirimycin(DNJ) was measured 4.34 mg in "Golden silk", high then to "Chunsujam"(3.81 mg). In the Paecilomyces tenuipes production ability test, the pupal weight of "Golden silk" was 1.20g.

Effect of Juvenile Hormone Analog Manina on Silkworm, Bombyx Mori L. II. Varietal Differences in Cocoon Productivity of the Leading Silkworm Varieties by Topical Application of Juvenile Hormone Analog "Manina". (유약 홀몬에 관한 연구 II. 장려 잠품종에 대한 합성유약홀몬 "마니나"의 증견효과)

  • Since 1966, practical use of active heavy analogs of the hormones has been also worked out as an insecticide and brought the features of it to the light as cocoon producer. On the other hand, it is expected to afford the increase of silk productivity resulted from control of the fifth larval period by delaying normal development. With these regards, some of analogs have been tried to apply practically to the silkworm for an increase of cocoon productivity. One of the synthesized juvenile hormone available is "Manina". And it is presently used for the increase of silk productivity in Korea. For the practical use, it is very essential the varietal differences in the increase of silk productivity by topical application was tested and the obtained results are summarized as follows. 1. It was evident that the fifty larval period was extended by topical application after 48 hrs. of the last ecdysis, ranging from 8 hrs. to one day, as compared to the control. 2. In pupal rates, there is no significance between control and treatments. It proved that there was no toxicity to silkworm by topical application in general, except jam 120. With regards to an increase of cocoon yield in Japanese, it was resulted from 17∼24% of cocoon yield from 10,000 larvae, as compared to that of control. In case of Chinese, the incrasing rates were varied from 15∼26% of cocoon yield, 17.8kg of it with 26% increase for Jam 122 and 16.7kg of it with 25% increase for Jam 118. In case of all hybrids, an increase of the cocoon yield took places from 20% to 31% and the weight of cocoon layer for the Japanese increased by 6 to 14%, those for the Chinese by 4 to 7% and those for the hybrids ranged from 21 to 29% increase. 3. It was recognized that the hybrid vigor rate took places with the hybrids between high responsing parents to juvenile hormones.
