Analysis of nutrient composition of silkworm pupae in Baegokjam, Goldensilk, Juhwangjam, and YeonNokjam varieties

  • Silkworm pupae have been utilized as a food source and have high-quality proteins and fatty acids. However, studies on the nutritional composition of pupae according to their variety, developmental stage, and sex have not been conducted. In this study, the nutritional composition of four Korean silk varieties (Baegokjam, BG; Goldensilk, GS; Juhwangjam, JH; and YeonNokjam, YN) were analyzed according to developmental stage and sex. The main ingredient of the pupa was protein, and the protein levels were higher in females than in males. Fat levels were high in males and increased in YN and GS during the late stage but decreased in BG. Fiber content rapidly increased in the late stages, and the lowest content was observed in BG. The pupae contained all essential amino acids, which were detected at higher ratios in females with a high protein content. Fatty acids had a different predominance depending on the variety. Oleic acid level was high in BG, linoleic acid and linolenic acid levels were high in GS, and palmitic acid and stearic acid levels were high in JH. In the mineral analysis, zinc was dominant in BG, whereas calcium and iron levels were relatively low. Zinc level was low in GS, and calcium and magnesium levels were high in JH. Potassium, sodium, magnesium, and phosphorus levels were low in YN. The silkworm pupae showed differences in components according to their variety, sex, and developmental stage; therefore, selecting a variety suitable for its purpose is necessary.

Studies on the mechanism in the induced to unfertilized eggs(male sterility of Silkworm) by protected environment during pupae period (용기의 보호환경에 따른 불수정란(웅성불임잠)의 유발기구에 관한 조사연구)

  • In order to find out effects of the generative power of silk worm moth which have been brought up in the high temperature accommodation at their pupa stage. For this specific study, 9 different kinds of male silk worms have been selected as specimen. All those specimen were brought up in the normal temperature at their larvae stage and after the pupation period they have been accommodated in the condition of high temperature for a certain length in accordance with the study programme. Afterwards, those mlae specimen were copulated with Suwon jam 103${\times}$104 which were all brought up in normal conditions. This study was carried out to find the copulation function as well as the ratio of unfertilized eggs(male sterility test). Results of study have been revealed as follows: 1) Although some differences were observed, male pura which have been brought up in the condition of high temperature shown the low rate of unfertilized eggs rather than those were brought up in the normal conpition. 2) In this group the eclosion(emergency) has been found to be poor rather, than those specimen brought up in normal conditions. 3) The copulation function of Moran, Daedong, J124 and C108 specimen were found to be poor than those of Suwon jam. 4) Fertility rate of Moran, Daedong, J124 and C108 was found to be around 65%. This figure is rather lower than what we normally expect. 5) Unfertilized egg ratio of Moran, Daedong and C108 were found to be around 20% if they were brought up in the condition of high temperature for one day from the time of pupation: 40% at 2 days, and 70% at 3 days duration. More than 3 days treatment has shown no progress in the unfertilized egg ratio. 6) One day's treatment for the pupa at the later stage has shown the unfertilized egg ratio of about 10%; 20% at 2 day's treatment, 35% at 3 day's treatment, 40-60% at 4 day's treatment, more than 60% at 5 day's treatment, and the 70% of fertilized egg ratio was only observed when the treatment days come to 7 days. It was understood that the unfertilized egg ratio was high at the antepupa stage rather than that of post-pupa stage. 7) According to the result of observation the sperm in copulatory pouch and seminal receptacle out of the normal female silk worm which have been copulated with the male brought up in the condition of high temperature at their moth stage. The reproduction system found in the control zone has been found to be normal and the sperm is amountful and active in motion while the sperm found in the treatment to be limited in amount and slow in motion. The observation was made within 5 hours from the copulation. 8) According to the result of observation of sperm of seminal receptacles of the female silkworm moth, and according to that observation of sperm in the seminal receptacle in female silkworm moth, the amount of sperm and mobility in the female moth brought up in high temperature were poor comparing that were brought up in normal temperature zone. Some of them are even found to be no trace of such. 9) Appearance and mosle of the copulatory organ of the male silkworm moth was found to be no anatomical change. 10) Testis of the later pupa stage which was brought up in the high temperature was found to be almost net developed to anucleate sperm and they are degenerated at stage of between maturation division and sperm abnormal stage. Mean time at control zone, the formation of anucleate sperm was already observed.

Studies on bionomical characteristics of Papilio xuthus (호랑나비(Papilio xuthus)의 생육특성에 관한 연구)

  • This study was investigation for primary bionomical characteristics, emergence rate of wintering type pupa and oviposition preference, of Papilio xuthus. The emergence rate of wintering type pupa stored at cold chamber($4{\pm}1^{\circ}C$) was 85%, after 10 months. The average emergence rate of wintering type pupa during storage was 89.6%, and the emergence periods were $7.9{\pm}1.9days$ in room temperature. Mating ratio in large oviposition room, $6,000{\times}6,000{\times}3,500$ mm($W{\times}L{\times}H$), was $86.7{\pm}5.8%$ higher than $63.3{\pm}15.3%$ in small room, $2,500{\times}3,000{\times}2,000$ mm($W{\times}L{\times}H$). But the number of eggs in the large room were $137.0{\pm}16.5$ fewer than $141.7{\pm}20.4$ in the small room. As a result of oviposition preference experiment on three different host plants, 10 mated female laid $141.7{\pm}27.8$ on the leaves of Phellodendron amurensew more than $67.7{\pm}20.6$ on Zanthoxylum schinifolium and $77.0{\pm}21.8$ on Citrus unshiu. The egg periods were $4.4{\pm}0.8$ days after oviposition, and the hatchability was 92.2% in $25^{\circ}C$ room condition. The larval period were $19.9{\pm}2.1$ days in the high temperature, long day condition($25^{\circ}C$, 14L : 10D). The headwidth of each developmental larval stage were $0.72{\pm}0.02$(1st), $1.19{\pm}0.02$(2nd), $1.65{\pm}0.05$(3rd), $2.43{\pm}0.07$(4th), $3.21{\pm}0.12$(5th). The pupal ratio was 91.6%. The pupal period were $8.8{\pm}0.9$ days, and the emergence rate was 92.2%.

The Studies on the Change of Lipid Phosphorus and Influence of Irradiation with the Origination of Silkworm Eggs (Bombyx mori L.) (가잠난 발생과정에서 Lipid Phosphorus의 변동과 방사선영향에 관한 연구(제일보))

  • The pupa (7th. day after mounting) were irradiated for foully minutes with 1,000${\gamma}$-ray and the copulation was done under the various conditions when the pupa grew to be imagos. The silkworm eggs that the imagos laid were divided into many groups by five, fifteen, foully-five, ninety, seven-hundred, twenty and twenty-four hundred minutes eggs respectively and the lipid phosphorus in the each group of eggs was analyzed as follows. 1. The quantity of 928${\gamma}$/100mg dry powder (D. P.) was shown in the group of the fourt-five minutes eggs whose ovum nucleus and sperm nucleus were not conjugate in the control C (male and female). The number of the greatest quantity of 944${\gamma}$/100mg D.P. after the syngamy of ovum nucleus and sperm nucleus while the smallest quantity was 768${\gamma}$/100mg D.P. in the group of ninety minutes eggs. 2. Not much difference could be seen the copulation group of female irradiation and male control and the copulation group of male irradiation and female control compared to the both male and female control group, but the influence could be seen in the group of irradiated female pupas. 3. The great difference could be seen in the group of irradiated male and female pupa compared to the control group. The five minutes eggs showed the smallest quantity of 536${\gamma}$/100mg D.P. and the quantity of lipid phosphorus rapidly began to increase from the fifteen minutes eggs. The quantity amounted to 1,082${\gamma}$/100 mg D.P. in the case of seven hundred and twenty minutes eggs while the quantity decreased in the twenty-four hundred minutes eggs which showed 912${\gamma}$/100 mg D.P. The general result obtained in the present study was that the irradiated male and female pupa were greatly influenced before and after the syngamy of gamete.

Comparative Chemical Compositions of Four Kinds of Tochukaso (동충하초 품종별 영양성분 비교)

  • Nutritional compositions of three Tochukaso species (Paecilomyces tenuipes hosted by Larva and pupa, Cordyceps militaris, C. sinensis) were compared. Fruiting body and host fractions were separately analyzed. Fruiting body fraction of P. tenuipes (36.6%) hosted by larva was higher than that hosted by pupa (10.2%), an indication that the quality of the former is superior to the latter. Carbohydrate content of C. sinensis (39.6%) was $2.5{\sim}7$ times higher than those of others, probably due to the presence of polysaccharides. Protein and crude lipid contents of C. sinensis and C. militaris were 25.8 and 10.3%, and 75.1 and 3.9%, respectively. C. sinensis showed the lowest Ca content and $30{\sim}75$ times higher Fe content among the samples tested. Vitamin A content of C. militaris was 308.9 IU/100g, two fold higher than those of the other species. Saturated fatty acid content was the highest in P. tenuipes (pupa, 27.7%), whereas unsaturated fatty acid was the highest in P. tenuipes (larva, 83.3%). Aspartic acid, glutamic acid, and glycine were abundant in all species. Cordycepin content of C. militaris was $20{\sim}50$ times higher than those of the other species.

  • The egg, larva, and pupa of Paracycnotrachelus longiceps (Motschulsky) are described and illustrated. The species is a well-known forest pest and lives on such host plants as Quercus acutissima, Q. aliena, Q. mongolica, and Q. serrata. Taxonomic notes and cradle structure are also provided.

  • Author investigated the control measure and the life cycle of the mulberry sucker, Anemoneura mori Schwarz. The results are as follows; 1. It is clear that the life cycle of mulberry sucker is one generation a year, but not obvious about the duration of pupa. 2. Even though various insecticides were used for control, they are not of a practical value during the egg stage. It is, however, believed that the mulberry sucker, when become larvae, can be controlled by spraying about 400 litres per 1 tanbo( DDVP, 1000$\times$). 3. Author recommends the control measures which you may use not only an insecticide, but also cut off branches so as to make leaf get enough sunlight and ventilation.

  • The present paper is a part of the study on the immature stages of genus Tipula in Korea. It described and illustrated egg, each larval stage and pupa of Tipula nova. It is the first to deal the all immature stages of the species in Korea. Their taxonomic remarks and habitats are also given.

Breeding of a New Non-Cocooning Silkworm Variety, Hachojam, Suitable for Autumn Rearing Season

  • A new non-cocooning silkworm variety, Hachojam, suitable for autumn rearing season is single cross Fl hybrid between Japanese race Jam 307 and Chinese race Jam 126. Jam 307, Japanese parent of the Hachojam, which is a source for non-cocooning process showed a high GCA (generation combining ability) in naked pupation rate and Jam 126, Chinese parent, showed a high GCA in pupation rate and single pupal weight. In the local adaptability test performed at 8 local areas in autumn of 1999 to 2000, the naked pupation rate and thin cocoon rate of Hachojam were 64.7% and 35.3% , respectively. The pupal weight calculated from 10,000 of the 3rd molted larvae was 24% heavier in Hachcjam than the cocoon-producing, check variety Daesungjam.

  • Throughout the studies the following experimental results were obtained and so are summarized here. 1) What caused tussah silkworms terrible disease broken ous all over the Korea in 1965, was examined to be “Inclusion body of virus” through microscope. 2) The examined inclusion bodies are easily stained as purple by seller's staining. 3) The proper speed of centrifugation for the purification of inclusion bodies is 1,000 r. p. m for 10 minutes. 4) It is possible, cleanly resulted, to attempt the “Rapid Agglatination Test & Complement Fixation test” with autigen of inclusion bodies. 5) The Anti-polyhedra rabbit serum from antigen of the dilution of 2${\times}$10$\^$6//l$m\ell$ polyhedra made the Ropid Agglutination test possible even with dilution of 1/8 infected pupa blood(antigen).
