Difference of Catechins Extracted Level when Fermented Sun-dried Salt and Green Tea (천일염과 녹차를 발효시켰을 때 Catechin류의 추출량 변화)

  • In an experiment in which fermentation was done by adding fungal species that have antibiosis but do not have cellulase, the extraction amount of EGC, EC, EGCG, and ECG increased in all samples that fermented by adding sun-dried salt compared to those that fermented only with green tea after fermenting green tea by mixing it with sun-dried salt. In the analysis conducted according to the days of fermentation, the high extraction amounts of EGC(epigallocatechin), ECG(epicatechin gallate), EC(epicatechin), and EGCG(epigallocatechin gallate) were detected on the second and third day. Furthermore, when fermentation was done by adding ferment bacillus, all types of catechin(EGC, EC, EGCG, ECG) extraction increased in Paenibacillus spp but in Bacillus amyloliquefaciens, EGC and EC decreased while EGCG and ECG increased; whereas in Bacillus pumilus and Bacillus subtilis all types of catechin(EGC, EC, EGCG, ECG) decreased. The results of the above experiment reveal that the largest amount of catechin was extracted from the result which conducted fermentation for three days together with sun-dried salt and Paenibacillus spp in the green tea.

  • Sixteen species of macrochelid mites were collected on the surface of scarab dung beetles in Mount Gede-Pangrango National Park, West Java, Indonesia. Of these, one species, Macrocheles gedeensis sp. nov. is described here as new to science. The other fifteen species were Holostaspella katakurai Hartini and Takaku, 2003, H. oblonga Hartini and Takaku, 2010, H. villosa Hartini and Takaku, 2010, Neopodocinum halimunense Hartini and Takaku, 2003, N. subjaspersi Hartini and Takaku, 2003, Neopodocinum sp., Macrocheles dispar (Berlese, 1910), M. entetiensis Hartini and Takaku, 2005, M. hallidayi Walter and Krantz, 1986, M. jabarensis Hartini and Takaku, 2003, M. kraepelini (Berlese, 1905), M. pumilus Hartini, Dwibadra and Takaku, 2009, M. turgoensis Hartini, Dwibadra and Takaku, 2009, M. sukabumiensis Hartini and Takaku, 2003, and M. sukaramiensis Takaku, 2001.

Isolation and Identification of Microorganisms in Korean Traditional Soybean Paste and Soybean Sauce (전통 된장 및 간장의 숙성기간별 생육 미생물의 분리 및 동정)

  • As a basic study for quality improvement of Korean soybean paste and soybean sauce, we investigated on microflora of soybean paste and soybean sauce fermentation. Major Microorganisms were isolated from the sample pastes and sauces, and identified systematically. Selected Microorganisms were identified by MIS whole cell fatty acid analysis by gas chromatography. Identification results showed that Bacillus licheniformis, bacillus pumilis and Bacillus subtilis were dominant in soybean paste and Staphylococcus vitulus, Bacillus subtilis, Bacillus pumilus, and Lactobacillus fermentum were dominant in soybean sauce. It seemed that these Microorganisms played an important role in soybean paste and soybean sauce fermentation and could be used for the further studies such as protease and amylase activities.

Aggregation Property of Chitosan and Chitooligosaccharides (Chitosan과 Chitooligosaccharides의 응집성)

  • Chitosan을 Bacillus pumilus BN-262 유래의 chito-sanase로 처리한 경우에는 trimet, tetramer, pentamer 가 전체 올리고당 중 64,3%에 달하는 비교적 저분자의 chitooligosaccharides로 구성괸 chitooligosacch-aride I을 얻을수 있었다 그러나 Trichoderma viride 유래의 cellulase로 처리한 경우에는 중합도 7이상의 것이 전체 올리고당 중 49.3% 에 달하는 상대적으로 분자량이 큰 chitooligosaccharides로 구성된 chitoolig-osaccharide II을 얻을수 있었다. 따라서 생리적 기능성이 높은 hexamer 이상의 chitooligosaccharides를 얻기 위해서는 chitosanase의 처리조건을 달리하여 분해가 덜 일어나도록 하거나 cellulase와 같은 효소를 처리함으로써 chitosan의 부분적인 분해를 유도하는 것이 필요하고 판단되었다. Chitin과 chitooligosac-charides의 응집성에 대하여 조사하였다 고분자의 chitosan은 2가 음이온을 함유하는 무기화합물 및 고분자의 유기화합물과 반응하여 응집이 일어났다. 분해가 많이 일어난 chitooligosaccharide I 은 무기물질과는 침전하지 않았으나 고분자화학불을 함유하는 유기화합물과는 반응하여 침전이 일어났다 분해가 적게 일어난 chitooligosaccharide II 는 2가 음이온을 함유하는 무기화합물 및 고분자의 유기화합물과 반응하여 침전이 일어났다.

Adhesive Membrane of Oocyte in Korean Cobitid Species (Pisces, Cobitidae) (한국산 기름종개속 어류의 난모세포의 부착막)

  • Through the histolosical observation of ovary in the nine species of the genus Cobitis, we found that specific adhesive membranes were attached to the outer zone radials during the vitellosenesis. These adhesive membranes could be classified into four forms as follows: 1) granular form of Colitis lutheri, C. striate, and C. sinensis, 2) villous form of C. longicorpus, C. konensis koreensis, C. k. pumilus, and C. granoei, 3) saw-like form of C. rotundicaudata, and 4) filamentous form of C. chou. The various forms of adhesive membrane of egg in the cobitid fishes are showed a species specificitv with reference to their habitats and spawning property.

Toxigenic Bacilli Associated with Food Poisoning

  • The genus Bacillus includes a variety of diverse bacterial species, which are widespread throughout the environment due to their ubiquitous nature. A well-known member of the genus, Bacillus cereus, is a food poisoning bacterium causing both emetic and diarrhoeal disease. Other Bacillus species, particularly B. subtilis, B. licheniformis, B. pumilus, and B. thuringiensis, have also recently been recognized as causative agents of food poisoning. However, reviews and research pertaining to bacilli have focused on B. cereus. Here, we review the literature regarding the potentially toxigenic Bacillus species and the toxins produced that are associated with food poisoning.

  • This studies were carried out to purification and seperation of chitooligosaccharides which containing excellent biological active substance. After deacetylation of chitosan (DAC%), DAC-45%, DAC-70%, DAC-95% and DAC-99% were used substrates and hydrolyzed by chitosanase (Bacillus pumilus BN-262) DAC-99% has excellent hydrolyzate which contained several chitooligosaccharides. Therefore, chitosan was hydrolyzed DAC-90 as substrate by chitosanase, and then purified and seperated of chitooligosaccharides Gel filteration and HPLC. This oligosaccharides composed with GlcN0, GlcN2, GlcN3, Glc5 and GlcN6.

Effect of Korean Fermented Food Extracts and Bacteria Isolated from the Extracts for the Control of Rice Seed-borne Fungal Diseases (국산 발효식품 추출물과 발효식품유래 미생물을 활용한 벼 종자전염성 진균병 방제)

  • When we investigated seed infestation by fungal pathogens from 51 varieties in 9 crops, the contamination rate of rice and sesame seeds was high. Therefore, to control seed-borne diseases, we obtained extracts from commercial products of Kimchi, Gochujang, Doenjang, Ganjang, Makgeolli and Tohajut and their suppressive effects against seed-borne diseases were studied. In addition, bacterial strains were screened to control rice seed-borne diseases in vitro and in vivo. Among forty food extracts, eleven food-extracts suppressed incidence of seedling rots in vitro and five food extracts increased 8-33% of healthy seedling in the greenhouse. Among 218 isolates from 40 fermented foods, 43 isolates showed high antifungal activity against seven fungal pathogens. When we tested 43 isolates for the reduction of rice seed borne disease, 32 isolates were able to reduce the rice seed borne disease. Among 32 isolates, 17 isolates reduced significantly seedling rot and increased healthy seedlings, the other isolates except for Kc4-2 and Mkl 2-2 increased shoot emergence and the percentage of healthy plants. Thirty isolates with high antifungal activity and suppressive effect against rice seedling rots were identified by 16S rRNA sequencing. Twenty one of thirty isolates were identified as Bacillus spp. Three isolates from Makgeolli were identified as Saccharomyces cerevisiae. B. amyloliquifaciens were isolated from six Korean traditional fermented foods except for Ganjang. B. amyloliquifaciens were majority in the effective bacterial population of Gochujang and Jutgal. Relatively diverse Bacillus species including B. subtilis, B. pumilus, and B. amyloliquificiens were isolated from Kimchi. The selected effective microorganisms from Korean fermented foods founded to be effective for controlling seed-borne diseases of rice in vitro and in the greenhouse. We think that Korean fermented foods and their useful microorganisms can be used as biocontrol agents for suppressing rice seed-borne diseases based on above described results.

Characterization of Water Quality and the Aerobic Bacterial Population in Leachate Derived from Animal Carcass Disposal (가축 매몰지 침출수에 대한 수질 특성 및 호기성 미생물 분포에 관한 연구)

  • Leakage of leachate from animal carcass disposal is a significant issue because disease can easily spread to humans and other livestock. In this study, we analyzed the physicochemical properties of leachate and tested for the presence of aerobic bacteria in leachate using molecular biology methods, for 16 animal carcass disposals in the first stage (after burial for 5 months). Leachate physicochemical analysis revealed higher total coliforms, TOC, $NH^{4+}$, and $NO^{3-}$ concentrations compared with previously published data. In most leachate samples, the concentrations of $NH^{4+}$ and $NO^{3-}$ exceeded the Korean guideline values for drinking water. In 16S rRNA sequence analysis of the distribution of leachate under aerobic conditions, Bacillus pumilus, Lysinibacillus sphaericus, and B. sphaericus were observed with high frequency, whereas no food-poisoning-related bacteria such as B. cereus or Salmonella were detected. The present findings improve our knowledge of the transport of leachate from animal carcass disposal sites through geologic media, and are useful in risk analysis and for subsequent studies.

  • In this study, the physicochemical properties of leachate and the bacteria existence in leachate using molecular biology methods for 4 animal carcass disposals on the disposal lapse time was analyzed. The result of leachate physicochemical analysis in the middle stage (been buried 20 months) showed higher EC, DO, $HCO_3{^-}$, TOC, T-N and $SO_4{^{2-}}$ concentration compared to the first stage data (been buried 5 months). For identification of leachate using 16S rRNA method, Lysinibacillus sphaericus, Bacillus pumilus, Pseudoclavibacter helvolus, Pseudochrobactrum saccharolyticum and Corynebacterium callunae in the first stage, Bacillus cereus, Lysinibacillus sphaericus, Bacillus circulans and Corynebacterium glutamicum in the middle stage was observed, while there were detections of pathogenicity bacteria such as B. cereus and L. sphaericus. This study improves our knowledge of the fate and transport in geologic media, treatment, risk analysis on the leachate from animal carcass disposal sites.
