Effects of a variety of treatments affecting Chinese cabbage protoplast culture, and plant regeneration from protoplast-derived callus (배추 원형질체 배양에 미치는 다양한 처리의 효과와 원형질체 유래 캘러스로부터 신초 재분화)

  • Here we describe a procedure for Chinese cabbage protoplast culture and effects of various treatments. Chinese cabbage protoplasts were isolated from different parts of young seedlings as using an enzyme mixture, of which yield was maximized in seven hours around after digestion. The highest rate of initial cell division followed by micro-callus formation was obtained in the medium with 1.0 mg/L 2,4-D, 0.5 mg/L NAA, and 1.0 mg/L BA when the cotyledon-derived protoplasts were cultured. Initiation of cell division and micro-callus proliferation significantly depended upon Chinese cabbage genotype under a same culture circumstances. The micro-calli developed from cotyledon tissue of Norang-Bom cultivar successfully grew toward callus colonies on the solidified medium with 0.2 mg/L zeatin and 0.1 mM spermidine. The callus colonies generated de novo shoots at the maximum frequency of 4.3% on the medium with 5.0 mg/L BA and 1.0 mg/L NM. Our results might be helpful for further studies to enhance the regeneration efficiency in Chinese cabbage protoplast culture.

  • A method isolating Populus euramericana cv. I-214 mesophyll protoplasts was developed to facilitate application of genetic engineering techniques to this species. The suitable medium for shoot multiplication in vitro was MS basal medium with $0.1mg/{\ell}$ BAP. The effects of several factors influencing protoplast isolation could be evaluated quickly by using leaf in vitro and known volumes of maceration and washing media. The best yields of mesophyll protoplasts were obtained using leaves in vitro in 2.0% Cellulase R-10, 0.8% Macerozyme R-10, 1.2% Hemicellulase, 2.0% Driselase, 0.05% Pectolyase Y-23, and O.6M Mannitol in addition to DTT and MES buffer adjusted to pH 5.6. Over $2.4{\times}10^6$ protoplasts per gram of leaf were produced using these conditions. For protoplast purification, the most favorable sucrose concentration of floating solution was 0.6M after washing them with CPW solution. This method of screening factors affecting protoplast isolation could be applicable to other species.

  • The protoplast formation of Rhizoctonia solani in the fast growing anastomosis groups (AGs) 1 and 4, the intermediate AG-2 and AG-5, and the slow AG-3 yielded the most, moderate and the least in that order, respectively. Sclerotia formation varied with AGs. A high yield of protoplasts from AGs was obtained with a combined lytic enzyme system containing cellulase 'Onozuka' R-10, macerozyme R-10 and ${\beta}-glucuronidase$. When 3g (fresh weight) of 30 hr old mycelia was incubated for 3 hr at $32^{\circ}C$ with the enzyme mixture in 0.6 M mannitol, maximum protoplasts were obtained in the five AGs. A protoplast fusion between sclerotia forming AG-1 inactivated with heat and non-forming AG-5 was induced by polyethylene glycol and ${Ca}^{2+}$. Seven fusants obtained were based on characteristics of colony and sclerotium formation on culture plates. The fusants were confirmed by isozyme patterns of esterase and killing reaction between AG-1 and a fusant F1501.

Conditions for Transformation of Alkalophilic Bacillus sp. K-17 (호알칼리성 Bacillus속 B-17의 형질전환조건)

  • To investigate the possibility of using alkalophilic Bacillus sp. K-11 as a host for molecular cloning, plasmid pUB110 and pBD64 were introduced into alkalophilic Bacillus sp. K-17 by protoplast transformation system. Protoplasts of Bacillus sp. K-11 were prepared by treatment with 200 $\mu\textrm{g}$/$m\ell$ Iysozyme in SMM buffer containing 0.4M sucrose. Optimal temperature, pH and culture time for protoplast formation were 4$0^{\circ}C$, 7.0 and 4hrs, respectively. Cell wall was regenerated efficiently on DM3 medium containing 0.8% agar and 0.5M sodium succinate. Under these conditions for protoplast formation and regeneration, the highest transformation efficiency was obtained with cells incubated for 4hrs, and using 30%(V/V) of 40%(W/V) PEG6,000, In characteristics of transfer-mants, plasmid pUB110 was more stable than plasmid pBD64 in Bacillus sp. K-17. Maximum xylanase production of both transformants carrying pUB110 and pBD64, respectively was similar, but under the same conditions, enzyme secretion by transformant carrying pUB110 was earlier than that of transformant carrying pBD64.

  • This study was conducted to investigate an effective method of protoplast isolation, the plating efficiency for cell division, and fusion of plant protoplasts by polyethylene glycol for somatic hybridzation. The effectiveness of protoplast isolation was different with the various enzyme concentrations, but, in the protoplast isolation from tobacco mesophyll, the enzyme solution with 0.5% macerozyme and 2.0% cellulase was very effective. The protoplast isolation from callus cultured in vitro for a long period was not obtained in any of the enzyme solution used. Protoplasts divided actively at cell densities above $10^44/ml and at $25^{\circ}C$ under 12hr illumination by inflorecient light (l50 Lux), regardless of presence of agar. The highest frequency of protoplast fusion was obtained after treatment with a solution of 0.33 M polyethylene glycol 1500.

  • Isolation and culture of leaf protoplasts from two chilli cultivars (Capsicum annuum var. accumnatum and Bird chilli) were developed to enhance selection process in the somatic hybridization programmes. In order to isolate the protoplasts from leaves of these two chilli cultivars different incubation periods (3, 5 and 10 hours) were tested with combinations of enzyme mixtures containing cellulase and macerozyme. Leaves were incubated on three enzyme mixtures (2% cellulase +0.4% macerozyme, 1% cellulase +0.2% macerozyme and 0.5% cellulase +0.1% macerozyme in 13% mannitol) at 251oC in the dark. Three hours of incubation using 2% cellulase and 0.4% macerozyme was the best for the protoplast isolation of both chilli cultivars tested. The yield was 5${\times}$108protoplasts/m1/g leaf tissue in both chilli varieties. It was found that in the mixed nurse method using Nagata and Takebe (NT) medium supplemented with 1.0mg/12,4-D, NAA and BAP with 0.5M mannitol and 1.2% Sea Plaque agarose is the best medium for protoplast culture. Protoplasts of Capsicum annum var. accumnatum were alive for 14 days forming cell walls and initiating cell division.

Identification of Regenerable Cells in MesophyII Protoplast Cultures (엽조직에서 나출된 원형질체의 재생 가능 세포판별)

  • This study was rimed out to examine the difference in the cell vitality between mesophyII protoplast (MP) and paraveinal mesophyII protoplast (PVMP) of Nicotiana tabaccum 'Xanti', Petunia hybrida 'Blue Star' and Chrysanthemum morifolium 'Baeckwang' by using urea permeability technique. The effects of various enzyme solutions and incubation time, NAA and thidiazron on plant regeneration from isolated protoplasts were also investigated. The vibratome technique was used for protoplast isolation and urea permeability test because the fresh living, thin tissue stripes (50 ${\mu}{\textrm}{m}$ of thickness) could be obtained with minimal damage with the vibratome. For the three plants examined, the urea permeability on the tested tissue stripes was relatively higher in PVMP than in MP by about Ks = 2.0 $\times$ 10$^{-5}$ cm/sec. The treatment of an enzyme mixture of 1.5% cellulase R-10, 1% Driselase, 0.5% Macerozyme R-10, and 0.5% Pectinase for 4 to 8 h was effective on the isolation of PVMP. The highest frequency of callus formation and plant regeneration from the isolated protoplasts was obtained with NAA 2 mg/L and thidiazuron 0.01 mg/L. Furthermore, the results demonstrated that cell devision and plantlet regeneration was more frequent in the PVMP than in the MP of the same leaf or plant We, therefore, conclude that UM is an excellent experimental material for the callus formation and regeneration from isolated protoplasts.

An Electron Microscopic Study on the Cell Wall Regeneration of Culture Panax ginseng Callus Protoplast (인삼 캘러스 원형질체의 배양에 따른 세포벽 재생의 전자현미경적 연구)

  • Ultrastructural changes of the isolated and cultured protoplasts from ginseng (Panax ginseng C. A. Meyer) callus were studies with electron microscopy. In the 3-day-cultured protoplasts, the cell organelles such as rough endoplasmic reticulum, ribosome, Golgi complex, mitochondria, proplastid increased in number and observed microtubules. Many vesicles derived from the Golgi complex were evenly distributed in the cytoplasm. Some of such vesicles protruded the outer surface of the plasmalemma, and formed the protuberances. Vacuole derived from endoplasmic reticulum included Golgi vesicles by the invagination of vacuoles. These vacuoles migrated toward the plasmalemma by a fusion process (exocytosis), after fusing the plasmalemma the cell wall materials released from the outer surface of the plasmalemma, and lastly deposited on the plasmalemma. Proplastids containing many starch grains, and microtubules parallel to the plasmalemma were observed near the plasmalemma. Connected fibrils which were observed on the outer surface of the 3-day-cultured protoplast were interpreted as the component of cellulose.

Plant Regeneration from Mesophyll Protoplasts of Dianthus superbus (술패랭이꽃(Dianthus superbus)의 엽육원형질체로부터 식물체 재분화)

  • Leaf mesophyll protoplasts of Dianthus superbus were cultured in MSP1 liquid medium supplemented with 0.5 mg/L BAP, 2.0 mg/L NAA and 9% mannitol. Protoplast-derived colonies were formed after 3 to 4 weeks of culture in the dark at 27$^{\circ}C$. These colonies were kept under continuous illumination (21.5 $\mu$E. m-2 sec-1) for 2 weeks and finally most of the colonies became green microcalli, about 3 mm in diameter. When green microcalli were transferred to MS solidified medium with 2.0 mg/L 2,4-D, they formed embryogenic calli after 4 week of culture. These calli were then transferred onto $N_{6}$ medium containing 0.1mg/L 2,4-D, 0.1 mg/L NAA, 2.0 mg/L kinetin and 2.0 g/L casein hydrolysate and cultured under illumination. After 5 weeks of culture the calli gave rise to multiple shoots of 10 to 15 per callus. Upon transfer onto MS medium containing 2.0 mg/L NAA, they were noted. The regenerates were successfully transplanted into potting soil.

Isolation and Culture of Protoplasts from Embryonic Cotyledons of Pinus densiflora S. et Z. (소나무 자엽(子葉)을 이용(利用)한 원형질체(原形質體) 나출(裸出) 및 배양(培養))

  • Protoplasts were isolated from cotyledons of germinating Pinus densiflora S. et Z. seeds. The seeds were germinated for nine days, and excised embryonic cotyledons about 3-4mm long were incubated with Cellulase Onozuka R-10 and Macerozyme. After 8 hours of incubation, large number of viable protoplasts were isolated. Isolated protoplasts were cultured in a medium containing basal salts of $B_5$ medium, vitamines, amino acids, organic acid, sugars, and growth hormones. The first evidence of protoplast budding was observed after twelve hours in culture, and it suggested that high potential of the embryonic cotyledons for rapid cell division affected the early budding, rather than effect of culture medium was shown in twelve hours. The three- to four-cell stage was reached after three to four days of culture. Most cell divisions were achieved by additional buddings rather than equal binary cell division. No further cell division was observed beyond the four-cell stage. Protoplasts isolated from fully expanded cotyledons (germinated for 17 to 24 days) seldom initiated or failed to initiate cell division.
