Changes in SDS-PAGE Pattern of Mung Bean Grain Proteins During Germination (녹두 발아중 분획한 단백질의 전기영동 패턴의 변화)

  • Changes of protein contents and amino acid composition and SDS-PAGE pattern of protein of mung bean which were germinated in dark at $25^{\circ}C$ for 6 day. The total protein contents gradually decreased during germination and the contents of each fractionative soluble proteins were increased shortly after the soaking of mung bean and gradually decreased during the germination afterwards. SDS-PAGE of albumin fraction showed 18 bands, and during the germination the most of bands were diminished or disappeared. But protein bands at 24,000, 40,000, 45,000, 70,000 dalton position remained until 6th day of germination. SDS-PAGE of globulin fraction showed 6 discrete bands, and during the germination the protein band at 45,000 dalton position disappeard. But the protein bands at $14,000{\sim}25,000$ dalton position did not change during the period. SDS-PAGE of glutelin fraction showed 10 discrete bands, and during the germination the bands of $45,000{\sim}70,000$ dalton become diminished or disappeared. But the bands of 30,000, 60,000 dalton did not change throughout the germination period.

  • This study was investigated on the effects of colchicine and EMS. induced mutation, plant on the germination and polypeptide pattern and peroxidase activity monitored by two dimensional gel analysis in Wasabia japonica Matsum (wasabi). Germination rate of Muju was higher than that of Ulrung-Do and optimum concentration for germination was appeared 100 ppm GA3 containing with 10 ppm BAP in these cultivars. Survived plants rate of Muju was higher than that of Ulrung-Do after colchicine and EMS treatment. Peroxidase activity and plant height were decreased by mutagen treatments, while incresed in root and stem thickness. The number of protein spots and pattern showed difference between Muju and Ulrung-Do . The plants treated mutagen increased polypeptide spots, especially EMS treatment showed more different polypeptide pattern compared to control.

  • Background: A human orthologue of mouse S100A6-binding protein (CacyBP), Siah-1-interacting protein (SIP) had been shown to be a component of novel ubiquitinylation pathway regulating $\beta$-catenin degradation. The role of the protein seems to be important in cell proliferation and cancer evolution but the expression pattern of SIP in actively dividing cancer tissues has not been known. For the elucidation of the role of SIP protein in carcinogenesis, it is essential to produce monoclonal antibodies specific to the protein. Methods: cDNA sequence coding for ORF region of human SIP gene was amplified and cloned into an expression vector to produce His-tag fusion protein. Recombinant SIP protein and monoclonal antibody to the protein were produced. The N-terminal specificity of anti-SIP monoclonal antibody was conformed by immunoblot analysis and enzyme linked immunosorbent assay (ELISA). To study the relation between SIP and colon carcinogenesis, the presence of SIP protein in colon carcinoma tissues was visualized by immunostaining using the monoclonal antibody produced in this study. Results: His-tag-SIP (NSIP) recombinant protein was produced and purified. A monoclonal antibody (Korea patent pending; #2003-45296) to the protein was produced and employed to analyze the expression pattern of SIP in colon carcinoma tissues. Conclusion: The data suggested that anti-SIP monoclonal antibody produced here was valuable for the diagnosis of colon carcinoma and elucidation of the mechanism of colon carcinogenesis.

Amino Acid Composition and Scanning Electron Micrographs of Modified Silkworm Larvae Protein Concentrates (변형번데기 농축단백질 아미노산 조성 및 주사전자현미경에 의한 관찰)

  • This study was designed to determine the amino acid composition and to investigate microstructure by scanning electron micrographs of silkworm larvae protein and modified silkworm larvae protein concentrate. The results were as follows: 1, The protein contents of soybean and silkworm larvae protein concentrate were 70.3% and 84.1%, respectively. 2. In general, the essential amino acid content of silkworm larvae protein concentrate were higher than soybean protein concentrate as well as FAO provisional scoring pattern. Silkworm larvae protein concentrate was especially high in lysine and methionine indicating that it could be a good supplemental source for cereals and beans. Succinylation and acetylation resulted in no difference in most amino acid content. 3. The scanning electron microscopic observations revealed that silkworm larvae protein concentrate had smooth surface topography while defatted silkworm larvae flour showed different shapes and sizes with relatively rough surfaces. Acylated silkworm larvae protein concentrate exhibited less cellularity and denser than protein concentrate. However, succinylated silkworm larvae protein concentrate showed especially good texture indicating that it could increase the functional properties of silkworm larvae protein concentrate.

Metabolic Syndrome Risk by Intake Ratio and Intake Pattern of Proteins in Middle-aged Men Based on the 2012-2013 Korean National Health and Nutrition Examination Survey Data (2012년~2013년 국민건강영양조사 자료를 이용한 중년 남성의 단백질섭취비와 섭취패턴에 따른 대사증후군 위험도 비교)

  • Objectives: The purpose of the study was to compare intake of energy nutrients, physical characteristics, and the prevalence of metabolic syndrome according to protein intake group. Methods: Subjects were 827 men aged 40-65 years. The results presented were based on data from the 2012-2013 National Health and Nutrition Examination Survey and analyzed using SPSS. The odds ratio (OR) of metabolic syndrome was assessed according to the protein intake group and intake pattern of protein-rich foods. Results: The mean of protein intake was $73.96{\pm}0.71g$. According to level of protein intake, four groups (deficient, normal, excess 1, excess 2) were created and their percentages were 8.3%, 39.6%, 37.1%, and 15.0% respectively. The mean of daily energy intake was $2,312.33{\pm}24.08kcal$. It was higher in excess group 2 than in the deficiency group (p < 0.001). Moreover, the intake of all energy nutrients increased significantly with protein intake group (p < 0.001). The main contribution to daily protein included mixed grains ($10.96{\pm}0.32g$), milled rice ($7.14{\pm}0.30g$), chicken ($3.50{\pm}0.21g$), and grilled pork belly ($3.04{\pm}0.16g$). With regard to physical characteristics, and blood pressure and blood test results, only body mass index increased significantly according to protein intake groups (p < 0.05). The prevalence of metabolic syndrome in subjects was 38.5%, and there was no significant correlation with protein intake group. The OR of metabolic syndrome increased with protein intake, and was higher 4.452 times in excess group 2 than in the normal group (p < 0.05). Conversely, the OR of metabolic syndrome according to the frequency of protein-rich food intake did not show a significant correlation. Conclusions: The results of this study can be used as significant supporting data to establish guidelines for protein intake in middle-aged men.

  • The information about protein structure gives the clues for the function of protein. It is needed for the improvement for the efficacy and fast development of protein drugs. So, the studies visualizing the structure of protein effectively increase. Most studies of visualization focus on the structural prediction for protein or the improvement on the rendering speed. However, studies of information delivery depending on the form of protein visualization are very limited. The major objective of this study is to analyze the information representation goodness-of-fit for the patterns of the hybrid visualization with primary and secondary structures of protein. Those hybrid visualizations included the patterns which updated current representative visualization services, Chimera, PDB and Cn3D. Information factor to analyze information representation goodness-of-fit is assorted by protein primary structure, secondary protein structure, the location of amino acid and ratio information about protein secondary structure, based on the result of subject-analysis. Subject is the group of experts who are involved in protein drug development over 5 years. The result of this study shows the meaningful difference in the information representation goodness-of-fit by the patterns of hybrid visualization and proves the difference in the information by the pattern of visualization.

A GSK-3/SHAGGY-Related Protein Kinase is Involved in Phytochrome Signal Transduction Pathway

  • Phosphorylation of cellular proteins is a key regulatory mehanism for signal transduction pathway in living cells. Phytochrome, a red/far-red light photoreceptor in plants, is known to employ protein phosphorylation for its light signaling, although its detauked mechanism is still ambiguous. This study is intended to identify the phosphoproteins and protein kinases that are regulated by phytochrome, by employing transgenic rice seedlings that overexpress Arabidopsis phytochrome A. Red light stimulated phsophorylation of a 70 kDa protein and far-red light negated the effect. The red light induced phosphotylation of the 70 kDa protein was strongly activated by heparin and inhibited by poly-L-lysine, suggesting that the 70 kDa protein phosphorylating kinase belongs to GSK-3/SHAGGY protein kinase that has functional roles in establishing cell fate and pattern formation in Drosophila. Taken together with the fact that phytochrome controls plant development, these results may suggest that a GSK-3/SHAGGY-related protein kinase in plant(ASK) is likely to be involved in phytochrome signal transduction.

  • The effect of pH on surface hydrophobicity, sulfhydryl group, infrared spectrum, SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) pattern and enthalpy was investigated in recovered protein from mackerel and frozen blackspotted croaker by alkaline processing. Hydrophobic residue in myofibrillar protein exposed to the surface of protein, and hydrophobic interaction were the highest around 6$0^{\circ}C$. The surface hydrophobicity was different between myofibrillar protein and myofibrillar protein including sarcoplasmic protein (recovered protein). The peak at 1636 c $m^{-l}$ was increased with pH, and the recovered protein was unfolded in alkali pH. Difference of surface and total sulfhydryl group at pH 7.0 and 10 was comparative high, and decrease of surface sulfhydryl group indicated formation of S-S bonds. Mackerel and frozen blackspotted croaker in alkaline pH showed bands of polymerized myosin heavy chain on SDS-PAGE pattern. The transition temperatures of recovered protein were 33.1, 44.3 and 65.5$^{\circ}C$. Gelation of recovered protein from alkali processing was estimated by increase of $\beta$-sheet structure by pH treatment, S-S bonds by oxidation of surface sulfhydryl group in heating, polymerization of myosin heavy chain in order.r.

Studies on Electrophoretic Pattern and Amino Acids of Buckwheat Protein (메밀 단백질의 전기영동유형 및 아미노산 조성에 관한 연구)

  • 1. Amino acid compositions were determined by amino acid analyzer. Through the analysis of these samples, it was found that glutamic acid was the most abundant; glycine, aspartic acid, lysine and threonine were rich; and tryptophan and methionine were the limiting amino acid. 2. Albumins, globulins, gliadins and glutelins were extracted from the Kangwon hull, Kangwon rice buckwheat, and wheat. The relative proportions of protein fractions were 52.45 : 10.14 : 16.61 : 20.80% in Kangwon hull buckwheat, 21.10 : 13.80 : 28.40 : 36.70% in Kangwon rice buckwheat and 6.87 : 1.65 : 42.85 : 48.6% in wheat, in the order of albumins, globulins, gliadins and glutelins. 3. Polyacrylamide gel electrophoresis (PAGE) and SDS-polyacrylamide gel electrophoresis (SDS-PAGE) were performed to identify the subfractions of each protein fraction. The electrophoregrams of PAGE showed that the same fractions of both Kangwon hull buckwheat protein and Kangwon rice buckwheat protein had very similar electrophoretic patterns to each other respectively, but there were significant differences in the patterns between buckwheat proteins and wheat proteins.

  • The purpose of this paper is to present a new algorithm for extracting the consensus pattern, or motif from sequence belonging to the same family. Two methods are considered for feature interval partitioning based on equal probability and equal width interval partitioning. C2H2 zinc finger protein and epidermal growth factor protein sequences are used to demonstrate the effectiveness of the proposed algorithm for motif extraction. For two protein families, the equal width interval partitioning method performs better than the equal probability interval partitioning method.
