An Experimental Study on Growth Pattern and Ultrastructure of Human Gingival Fibroblasts Treated by Low Level Laser (저출력레이저가 성인의 치은섬유아세포의 성장양상과 미세구조에 미치는 영향에 관한 실험적 연구)

  • In order to verify the acceleration effect of low level laser (LLL) on oral mucosal wound healing process at cell biological level, the author studied growth pattern and ultrastructure in human gingival fibroblasts flashed by GaAlAs LLL-830 nm, 15mW for 10 minutes/flash one to three times at interval of 3-4 days through the evaluation of cell growth rate, protein conent/cell, DNA content/cell and ultrastructural changes for 14 days. The results were as follows : 1. The growth rate in gingival fibloblasts treated by LLL showed 4 orderly stages-decreasing stage after LLL treatment, acute increasing stage 3 days after LLL treatment, restring stage and recovering stage. 2. The effect of multiple flashes on LLL at interval of 3 days more or less was not proportional to times of flash on acceleration of growth in gingival fibroblasts. 3. The total protein content per gingival fibroblast was not significantly changed by LLL treatment in comparison with control group. But some kinds of protein which might be cell growth promoting factors were decreased immediately after LLL treatment, thereafter were acutely increased in cellular protein profile. 4. In ultrastructural changes of gingival fibroblasts treated by LLL, more prominent rough endoplasmic reticulum, mitochondrial hyperplasia/hypertrophy and increased extracellular fibrillar matrix were observed in comparison with control group under same experimental period.

  • a- and c-raf protein kinase in the brain of rat, the protein pattern of cerebellum during postnatal development of rat by polyacryamide gel electrophoresis, and the existence of c-raf protein kinase by using Western blotting method. The results were as follows: The cytoplasm of Purkinje cells was, in general, strongly labeled with the antibodies of a- and c-raf protein kinases in the cortex regions such as Pyramis cerebelli, Unula, Nodulus, Paraflocculus, and Flocculus. C-raf protein kinase appeared stronger immunoreactivity than a-raf protein kinase. In peripheral of cytoplasm of Nucleus emboliformis, A-raf Protein kinase was labeled markedly. During postnatal development, the protein of 38,000 dalton increased gradually in the cytosolic fraction of cerebellum, and the protein of 260,600 dalton appeared in the membrane fraction of cerebellum. By immunoblotting method, the protein band of 74,000 dalton was detected in crude and cytosolic fractions, but it was not exhibited in membrane fraction, In this fact, it was identified that a - and c-raf proteins were distributed throughout neuronal cells, especially in the Purkinje cells, in normal cerebellum cortex of rat. Also, this phenomenon was assumed that raf protein kinase in cytoplasm of neuronal cell had to do with a certain functional mechanism and signal transduction of neurotransmitter as Protein kinase C.

Dietary Self-selection and Nutrient Feeding Systems for Egg-type Growing Pullets and Layers (난용계의 선택채식과 영양소 공급체계)

  • From the observations of dietary self-selection by growing pullets, step-up protein or reverse protein and single-stage low protein pullet feeding systems were developed. They offered another pullet feeding concept that appears to control the body weight effectively and to reduce the consumption of feed and nutrients without impairment of subsequent laying performance. It is obvious from the feed and nutrient consumption pattern of layers fed diets for self-selection of energy, protein and calcium that they have a daily cyclic requirement rather than a constant requirement for nutrients. It seems that a practical self-selective feeding system is needed to meet the daily cyclic requirement for nutrients without consuming an excess of energy and protein at certain times of the day as compared to the complete or single diet where layers have to consume extra energy and protein in the afternoon when they have a specific appetite mainly for calcium.

  • In this study, a simple procedure is described for patterning biotin on a glass substrate and then selectively immobilizing proteins of interest onto the biotin-patterned surface. Microcontact printing (CP) was used to generate the micropattern of biotin and to demonstrate the selective immobilization of proteins by using enhanced green fluorescent protein (EGFP) as a model protein, of which the C-terminus was fused to a core streptavidin (cSA) gene of Streptomyces avidinii. Confocal fluorescence microscopy was used to visualize the pattern of the immobilized protein (EGFP-cSA), and surface plasmon resonance was used to characterize biological activity of the immobilized EGFP-cSA. The results suggest that this strategy, which consists of a combination of $\mu$CP and cSA-fused proteins. is an effective way for fabricating biologically active substrates that are suitable for a wide variety of applications. one such being the use in protein-protein assays.

Blood Biochemical Profile and Rumen Fermentation Pattern of Goats Fed Leaf Meal Mixture or Conventional Cakes as Dietary Protein Supplements

  • The expediency of replacing cost prohibitive and often inaccessible traditional protein supplements prompted the monitoring of hematological parameters was carried out in female goats at 0, 30, 60 and 90 days post feeding. Rumen environment was (3), respectively fed supplements containing either a leaf meal mixture (LMTM) of Leucaena leucocephala-Morus alba-Tectona grandis (2:1:1) or traditional protein supplements groundnut cake (GNC) or soybean meal (SBM) and wheat straw as basal diet. The periodic monitoring of hematological parameters was carried out in female goats at 0, 30, 60 and 90 days post feeding. Rumen environment was studied in bucks in a $3{\times}3$ switch over design. Rumen liquor was collected at 0, 2, 4, 6 and 8 h post feeding after 4 weeks of feeding. The goats fed on LMTM or GNC had similar dry matter intake (g/kg $W^{0.75}$), which was significantly (p<0.05) higher than SBM. Except for packed cell volume (PCV), none of the blood biochemical constituents (Hemoglobin, serum glucose, total protein, serum albumin (A) and globulin(G), A:G ratio, alkaline phosphatase, transaminases) varied significantly due to replacement of 50% dietary protein by LMTM throughout the experiment. GNC group had significantly higher level of PCV than other treatments. However, the level of serum total protein (p<0.01) tended to increase from 60th day onwards irrespective of dietary treatments. The average rumen pH was significantly higher (p<0.001) on SBM followed by LMTM and GNC, respectively. Total volatile fatty acid (TVFA) production was comparable in goats given LMTM or GNC supplements, the corresponding values were significantly different (p<0.001) when compared with SBM. The ammonical-N, total-N and TCA-precipitable-N (mg/100 ml SRL) did not differ significantly among dietary treatments. It may be concluded that supplementing wheat straw with LMTM based concentrate had no adverse effect on voluntary intake, blood biochemical profile and rumen fermentation pattern of the goats.

  • Proteomics is a useful approach to know protein expression, post-translational modification and protein function. We investigated the protein expression pattern and identity in chickens fed with the tissue culture medium waste after harvest of Korean wild ginseng (TCM-KWG) (Panax ginseng). Two groups (n=60/group) of day old broiler chickens were administered with 0 (control) and 0.8% (treatment) TCM-KWG through drinking water. After 5 weeks, we examined the protein expression pattern of fibularis longus and superficial pectoral muscle by Two-dimensional electrophoresis analysis. Interestingly, TCM-KWG treatment significantly increased five spot's density, and markedly reduced five spot's density in the muscles. We identified 10 proteins (desmin, myosin light chain 1, heat shock 25 kDa protein, collapsin response mediator protein-2A, alpha enolase, vimentin, actin alpha 1, my023 protein, pyruvate kinase and troponin T) by the matrix-assisted laser desorption ionization time of flight (MALDI-TOF).

  • In this paper, we firstly reported a C-type lectin cDNA clone of 1029 bps from the larvae of A. Pernyi (Ap-CTL) using PCR and RACE techniques. The full-length cDNA contains an open reading frame encoding 308 amino acid residues which has two different carbohydrate-recognition domains (CRDs) arranged in tandem. To investigate the biological activities in the innate immunity, recombinant Ap-CTL was expressed in E. coli with a 6-histidine at the amino-terminus (Ap-rCTL). Besides acted as a broad-spectrum recognition protein binding to a wide range of PAMPs and microorganisms, Ap-rCTL also had the ability to recognize and trigger the agglutination of bacteria and fungi. In the proPO activation assay, Ap-rCTL specifically restored the PO activity of hemolymph blocked by anti-Ap-rCTL antibody in the presence of different PAMPs or microorganisms. In summary, Ap-rCTL plays an important role in insect innate immunity as an pattern recognition protein.

An Efficient Method to Find Accurate Spot-matching Patterns in Protein 2-DE Image Analysis (단백질 2-DE 이미지 분석에서 정확한 스팟 매칭 패턴 검색을 위한 효과적인 방법)

  • In protein 2-DE image analysis, the accuracy of spot-matching operation which identifies the spot of the same protein in each 2-DE gel image is intensively influenced by the errors caused by the various experimental conditions. This paper proposes an efficient method to find more accurate spot-matching patterns based on multiple reference gel images in spot-matching pattern analysis in protein 2-DE image analysis. Additionally, in order to improve the reduce the execution time which is increased exponentially along with the increasing number of gel images, a "partition then extension" framework is used to find spot-matching pattern of long length and of higher accuracy. In the experiments on real 2-DE images of human liver tissue are used to confirm the accuracy and the efficiency of the proposed algorithm.

Effects of the Pattern of Energy Supply on the Efficiency of Nitrogen Utilization for Microbial Protein Synthesis in the Non-Lactating Cows Consuming Grass Silage

  • Effects of the pattern of energy supply on the efficiency of nitrogen utilization for microbial protein synthesis (MPS) were examined in cows consuming grass silage (7.1 kg DM/d) and supplement of 1 kg sucrose per day given as an intraruminal infusion. Three non-lactating cows received three experimental treatments in a $3{\times}3$ Latin square design with each period lasting 14 days. The treatments were (1) the basal diets of silage alone given in one meal each day at 09:30 h (BASAL), supplemented with (2) 1.0 kg sucrose given a 4-h infusion starting at 09:30 h (SYNC), (3) 1.0 kg sucrose given a continuous infusion for 24 h (CONT). Compared with BASAL, sucrose infusions altered (p<0.05) the pattern of variation in ruminal pH and the concentration of ammonia at 4 h after feeding but none of the sucrose treatments resulted in any changes in the ruminal concentration of VFA. All sucrose treatments increased (p<0.05) MPS relative to BASAL by 14% and 33% for SYNC and CONT, respectively, and that for CONT was greater (p<0.05) than for SYNC. It is concluded that synchronization of energy and nitrogen over the shorter term has no further advantage of the efficiency for MPS relative to CONT.

Different Level of Plasma Free Hemoglobin between Qi-deficiency and Fire Heat among Korean Stoke Subjects (한국인 중풍환자의 기허군 화열군의 plasma free hemoglobin의 비교)

  • The purpose of this study was to fine proteins, which have significantly different level in plasma between Qi-deficiency and Fire-heat group of Korean Oriental Stroke pattern identification (PI) among Korean stroke patients. Eighteen stroke patients with Qi-deficiency and forty nine patients with Fire-heat, which had critical syndrome of each PI, were participated in this study. Plasma protein pattern were analyzed by SELDI-TOF MS using Q10 strong anion exchange chip and Mass spectral data (m/z) statistically determined. The expression level of proteins, which were different between Qi-deficiency and Fire-heat in the results by SELDI-TOF MS, were confirmed by western blot. As a result of analyzing plasma protein by SELDI-TOF MS, six protein peaks were significantly higher in Fire-heat group than Qi-deficiency group. Two peaks among of them, M15003 and M15745, were respectively identified as hemoglobin alpha and beta in previous study. Expression level of plasma free hemoglobin of Fire-heat group was also confirmed higher in Fire-heat group than in Qi-deficiency group. These findings suggest that plasma free hemoglobin is a candidate for discriminating Qi-deficiency and Fire-heat group according to pattern identification (PI) of stroke.
