B-6 Vitamers and $\beta$-Glucoside Conjugates in Milk of American and Egyptian Women during the first Six Months of Lactation

  • Levels and distribution of five B-6 vitamers(PMP, PM, PLP, PL, and PN) and pyridoxine $\beta$-glucoside conjugates(PN-glucoside) were examined in milk of American women who received supplements of 2.5 or 10mg PN HCl/d and of unsupplemented Egyptian women during the first six months of lactation. B-6 vitamer and PN-glucoside levels in human milk were determined by reverse-phase HPLC. Pyridoxal(PL), which has been reported to be the most rapidly absorbed form of vitamin B-6 and may facilitate bioavailability, was the predominant vitamer in human milk of all three groups. Pyridoxal made up 72% of total vitamin B-6 for the 2.5mg supplemented group, 76% for the 10mg group, and 59% for the Egyptian group. Level and Percent PL were significantly lower for Egyptian women. Mean growth of the two American groups was similar to each other and within the normal range of the NCHS reference, however, Egyptian infants showed growth faltering at 6 months. The Percent of PN-glucoside, a less bioavailable form of vitamin B-6 in humans was 1% in milk of American women and was 11% in Egyptian women and these values were significantly different. for Egyptian women, total vitamin B-6 levels in breast milk correlated Positively with animal protein intake(r=0.91) and percent PN-glucosides(r=0.53) and negatively with plant protein intake(r=-0.55). These findings showed that high plant protein intake was associated with low concentrations of PL and total vitamin B-6 in human milk.

Molecular Size and Distribution of Zinc-binding Ligands in Rat Pancreatic Tissue

  • The pancreas is an important organ in the maintenance of zine homeostasis. The pancreatic tissue used in this study was obtained from rats fed varying levels of dietary Ca nd phytate followed by intraperitoneal {TEX}${65}^Zn${/TEX} injection. THe objective of this study was to determine the molecular size and distribution of compounds that may represent zinc-binding complexes in pancreatic tissue homogenates. The supernatant of the homogenized pancreatic tissue was separated using a Sephadex G-75 column with Tris buffer at pH 8.1. All subfractions were assayed for zinc, protein and {TEX}${65}^Zn${/TEX} activity. The elution of subfractions from pancreatic tissue homogenates showed a prominent peak corresponding to the high molecular weight protein standard (>66kd). A sall molecular weigth protein (<6.5kd), that was absorbed at 280nm, was also present: prominently in low Ca group, however not much as in high Ca group. These small compounds may combine weakly with zinc in pancreatic tissue an serve as zinc-binding ligands in pancreatic/biliary fluid. In the duodenum, these ligands dissociate zinc into an ionic form which becomes vulnerable to phytate complexation.

  • Cationic liposomes for cancer treatment have been developed in the field of chemotharpy. It was well combined on the surface of anionic tumor cell membrane by electrostatic interaction. Thus, the object of this study was to prepare the cationic liposomes capable of forming an ionic complex with the anionic cell membrane. To prepare the cationic liposomes, 1,2-distearoyl-sn-glycero-3-phosphoethanolamine (DSPE) as a cationic lipid material and polyethylenimine (PEI) as a cationic polymer were synthesized. Ionic property on the surface of liposomes was determined by the zeta potential. The adsorption characteristics of plasma protein for liposome in bovine serum were determined by the particle size and turbidity change. To estimate the stability of liposome in buffered solution, the change of particle size was measured at room temperature for seven days. The cationic liposomes were absorbed a large amount of plasma protein in bovine serum because plasma protein having anionic charge was fixed on the surface of cationic liposomes. This result indicate that the modification on the surface of liposomes using cationic polyethylenimine enhances the protein adsorption in bovine serum. Additionaly, cationic liposomes showed good stability in buffered solution for seven days.

Evaluation of Diet for Buffalo Dairy Cows Using the Cornell Net Carbohydrate and Protein System

  • The aim of this paper was to use the Cornell Net Carbohydrate and Protein System (CNCPS), that reports diet energy and protein value and animal requirements, as net energy for lactation ($NE_1$) and metabolizable protein (MP) respectively, to evaluate some rations for lactating Italian Mediterranean buffaloes. The investigation was carried out on six farms in the province of Caserta (southern Italy), where the milk production was controlled four times monthly on 10 animals (changing every time) chosen at different lactation days (5 categories): <2 months (A), 2-4 months (B), 4-6 months (C), 6-8 months (D), >8 months (E). Milk fat and protein were determined. Diet $NE_1$ and MP were estimated with the CPM-Dairy program (1998) using diet component chemical characteristics; then energy and protein intakes were estimated. $NE_1$ and MP requirements were estimated with two methods: 1) using CPM-Dairy that considers produced milk, fat and protein content, lactation phase and body condition score as main factors; 2) by applying the theory that to produce 1 kg of energy corrected milk, the buffalo needs 3.56 MJ of $NE_1$ and the efficiency to convert the absorbed aminoacids into milk protein is lower than cow (CNCPS). As regards energy, with method 1 the requirements were satisfactory starting from category A (4 out of 6 farms) and category B (5/6 farms); however, a surplus resulted for category E (5/6 farms). With method 2 a deficit in category A (5/6 farms) and B (3/6 farms) was observed, while the energy requirements were satisfied for all categories except E, where on only one buffalo farm had a surplus of energy intake. As regards protein, with method 1 the requirements were substantially satisfied for all the categories except E (3/6 farms); with method 2 the MP trend was much less favourable than with method 1. Indeed, a protein deficit was observed for all animals in categories A and B (5/6 farms). Moreover, on one farm the protein intake never satisfied animal requirements. In our experimental conditions, the use of the CNCPS to characterise diets for lactating buffalo and to calculate their requirements led to satisfactory results. By contrast, we cannot say the same for method 2, which applies a lower use efficiency of NE and MP for lactation in buffalo compared to cow.

Biopharmaceutical studies on copper(II) chelates of sulfanilamide derivatives (Sulfanilamide유도체의 동(II)착화합물에 대한 생물약제학적 연구)

  • Cu(II) chelates of several sulfanilamide derivatives (Sulfa-Cu) were prepared and their effects on solubility, absorptivity in intestinal lumen, biding tendency with serum protein and erythrocytes, concentration in rabbit blood, and acetylation rate were studied in comparison with their free ligand forms. For solubility concerned, the partition coefficients of Sulfa-Cu are decreased as following order: Sulfadimethoxine Copper chelate (SDM-Cu), Sulfamethoxypyridazine Copper chelate (SD-Cu), Sulfamerazine Copper chelate (SM-Cu), Sulfaisoxazole Copper chelate (SIX-Cu). The partition coefficients of SDM-Cu and ST-Cu were much greater than those of ligands. this phenomenone acounts for the rapid absorption of SDM-Cu and ST-Cu in the rat small intestine (in situ). The Sulfa-Cu were absorbed at the intestinal lumen of a rat in the rate of first order and there was no difference between long acting sulfa drugs and their Cu0chelates in biological half lives. In binding experiments, sulfa-Cu binded with serum protein in lower ratio than their ligands except SIX-Cu. On other hand, acetylation rates of sulfa-Cu were higher than those of free sulfa drugs and the acetylation rate were higher than those of free sulfa drugs and powder. In a experiment on Sulfa-Cu concentration in rabbit blood, the half lives of SD-Cu, SIX-cu, ST-Cu, and SM-Cu were longer than those of their ligands. Above all, the half life of SD-Cu appeared to be approximately 3.5 times logner than that of corresponding ligand, SD. When absorption of sulfa drugs or sulfa-Cu at the small intestinal lumen of a rat and the concentration in rabbit blood after absorption were compared, it was found that there was not always conrrelated.

Lactation Performance of German Fawn Goat in Relation to Feeding Level and Dietary Protein Protection

  • Effects of high and low levels of feeding with or without protected protein on the performance of lactating goats were studied. Twenty four German Fawn goats either from 1st ($43.37{\pm}3.937$ kg and 2 year old) or 3rd $62.64{\pm}6.783$ kg and 4-5 year old) parity were used for the trial. Feeding levels were 7.2 (I) and 5.2 (II) MJ ME/litre of milk of 3.5% fat in addition to that of the maintenance allowance. At each feeding level, diet had either unprotected (U) or formaldehyde protected (P) soya-meal. Thus, four diets were IU, IP, IIU and IIP, having six animals in each. The diets were composed of hay and pellet (10:4:1 of beet pulp : barley : soya-meal). Effect of feeding level, protein protection, parity, health status and kid number on intake, milk yield, milk composition, growth rate of goats were recorded across the 21 weeks of study. High feeding level resulted increase (p<0.001) in estimated metabolizable energy (ME) and metabolizable protein (MP) availability. Dietary inclusion of protected soya-meal increased (p<0.001) the estimated MP but not the ME availability. Animals in 1st parity ate more (p<0.001) DM (111 vs. 102 g/kg $W^{0.75}$/d) than those in 3rd parity. Animals with twin kids (110 g/kg $W^{0.75}$/d) had higher (p<0.001) DM intake than those with single kid (102 g/kg $W^{0.75}$/d). Fat (4%) corrected milk (FCM) yield was not effected by high (1,924 g/d) or low (1,927 g/d) feeding level but increased (p<0.001) with protected (2,166 g/d) compared with unprotected (1,703 g/d) soya-meal. FCM yield for four dietary combinations were 1,806, 2,078, 1,600 and 2,254 g/d for diets IU, IP, IIU and IIP, respectively. For unit increase (g) in estimated MP availability relative to ME (MJ) intake, FCM yield increased ($1,418{\pm}275.6$) g daily ($r^2$=0.58; p<0.001). Milk fat (3.14 vs. 3.54%; p<0.001) and protein (2.94 vs. 3.04% p<0.05) contents were lower at high than the low feeding level. Protected protein increased (p<0.001) the fat, lactose and net energy (NE) content of milk. Milk urea concentration of 175, 183, 192 and 204 mg/l for diets IU, IP, IIU and IIP, respectively indicated lower RDP content of these diets. The RDP contents were 6.97, 6.70, 7.30 and 6.83 g/MJ of ME for diets IU, IP, IIU and IIP, respectively. Live weight change over the experimental period were 41, 6, 17 and 19 g/d. Absence of any positive response of high feeding was probably due to inefficient rumen fermentation resulting from inadequate RDP supply. Protected protein improved production performance apparently by increasing MP:ME ratio in the absorbed nutrient.

  • Soil humic substances are defined as a humified part of the soil organic matters and regarded to play beneficial roles for colloid chemical properties and the fertility of the soils. This paper is referred to review the present trend of the studies on the forming processes of humic substances and on the effect on plant metabolism by some organic compounds that are directly absorbed by plants. It is generally considered that the humic substances are formed organic matters in soil or plant materials through numerous organic or biochemical processes. However, the nature of the constituting "core" and of attachment of carbohydrate, nitrogen containing compounds like protein, phenolic compounds and metals to the core are unclear though various models are suggested. It is reviewed that some organic compounds, phenclic acids, derived from humic substances are effective on plant metablism in many cases, although the mechanisms are remained to be clarified.

  • In order to observe the absorption pattern of mineral elements and the nutritional components of brown rice grown in reclaimed saline areas in Korea, the four commercial varieties of rice were cultured at the average salt concentration of 6.05 mmhos/cm, $25^{\circ}C$, (0.3%) in saline soils and non-saline silty loam soils. In the saline areas the absorptin of Na and Si was relatively high while the absorption of P, K and Ca was relatively low. N was absorbed actively before the emergence of the rice grains but showed relative decrease thereafter. There was no difference in Mg content. Though brown rice in the saline areas contained sugar abundantly with small contents of protein, fat and crude fiber. There was little difference in total carbollydrate and ash contonts.

The Orientation of CO in Heme Proteins Determined by Time-Resolved Mid-IR Spectroscopy: Anisotropy Correction for Finite Photolysis of an Optically Thick Sample

  • A systematic way of determining the equilibrium orientation of carbon monoxide (CO) in heme proteins using time-resolved polarized mid-IR spectroscopy is presented. The polarization anisotropy at pump-probe delay time of zero in the limit of zero photolysis and the angular distrbution function of CO are required to obtain the equilibrium orientation of CO. An approach is developed for determining the polarization anisotropy in the zero-photolysis limit from the anisotropy measured under finite photolysis conditions in an optically thick sample where the fraction of molecules photolyzed decreased as the pump pulse passes through and is absorbed by the sample. This approach is verified by measuring the polarization anisotropy of CO of carbonmonoxy myoglobin at various levels of photolysis. This method can be readily applied to other photoselection experiments determining precise angle between transition dipoles.

  • The foam concrete was fabricated by adding the foaming agent which composite ordinary Portland cement with plant and animal protein into cement paste, and the electromagnetic wave absorption properties were studied for the first time as well. The studies showed that the electromagnetic waves can be absorbed by multiple reflections and scattering within the porous material. Thickness and filling ratio have a great influence on the electromagnetic wave absorbing properties in 2-18 GHz of the foam concrete, the greater the thickness, the better the performance of absorption; filling ratio was about 52 vol.%, the absorbing properties achieved the best.
