Octapeptide (Alanine Angiotensin) 의 合成 (Synthesis of an Octapeptide (Alanine Angiotensin))

  • We have shown that carboxy-peptidase destroys the biological activity of angiotensin octa-and deca-peptides. Since Proline occurs as the seventh amino acid from the amino end of the chain and since carboxypeptidase does not cleave proline from a peptid chain, it is evident that the heptapeptid H.asp-arg-val-tyr-ileu-his-pro.OH is formed by this hydrolysis. This peptide must then be biologically inactive. In order to determine whether the phenyl group of the C-terminal amino acid was the necessary requirement for biological activity of the octapeptide, $ala^8$ angiotensin octapeptide(amino acids of peptides numbered from amino end) was synthesized. For this synthesis the four dipeptides were prepared: carbobenzoxy-L-prolyl-L-alanine-P-nitrobenzyl-ester, m.p. $134-135^{\circ}C,$ carbobenzoxy-L-isoleucyl-imidazole benzyl-L-histidine methyl ester, m.p. $114-116^{\circ}C,$ carbobenzoxy-L-valyl-L-tyrosine hydrazide and carbobenzoxy B-benzyl-L-aspartyl-nitro-L-arginine. The first three dipeptides were obtained as crystalline compounds. Imidazole-benzyl-L-histidine was used in the hope that it would block the histidine imidazole against side reactions in steps subsequent to the formation of the C-terminal tetrapeptide. Also, it was through that the imidazole benzylated peptides would be easier to crystallize. This, however, was not the case. The tetrapeptide, carbobenzoxy-L-isoleucyl-L-im, benzyl-histidyl, L-prolyl-L-alanine-nitrobenzyl ester was not obtained in a crystalline form. Neither could the mono-or dihydrobromide of the tetrapeptide free base be induced to crystallize. Carbobenzoxy-L-valyl-L-tyrosine azide was condensed with the tetrapeptide free base to yield the protected hexapeptide; carbobenzoxy-L-valyl-L-tyrosyl-L-isoleucyl-L-im, benzyl, histidyl-L-Prolyl-L-alanine-nitrobenzyl ester. Upon removal of the carbobenzoxy group with hydrogen bromide in acetic acid an amorphous free base hexapeptide ester was obtained. This compound gave the correct C, H, N analysis and contained the six amino acids in the correct ratio. The octapeptide was obtained by condensing this hexapeptide with carbobenzoxy-B-benzyl-L-aspartyl-nitro, L-arginine using the mixed anhydride method of condensation. This amorphous product was proven to be homogenous by chromatography in two solvent systems and upon hydrolysis yielded the eight amino acids in correct ratio. The five protecting groups were removed from the octapeptide by hydrogenolysis over palladium black catalyst. Biological assay of the free peptide indicated that it possessed less than 0.1 per cent of both pressor and oxytocic activity of the phenylalanine8 angiotensin. This suggests that the phenyl group is a point of attachment between angiotensin and its biological receptor site.

  • In order to investigate the feasibility of transmucosal delivery of the model peptide, LHRH, metabolism of LHRH and inhibition effect of medium chain fatty acid salts were studied in rabbit mucosal homogenate. LHRH incubated in homogenates of rectal(RE), nasal(NA) and vaginal(VA) mucosa were assayed by HPLC. Five to six degradation products of LHRH were deterted and the degradation of LHRH$(500\;{\mu}g/ml)$ followed the first order kinetics. The main degradation products were found as $LHRH^{1-5}(M-I)$, $LHRH^{1-3}(M-II)$ and $LHRH^{1-6}(M-III)$ by the method of amino acid analysis. The half-lives of LHRH in the mucosal homogenates were found to be less than 20 min at protein concentration of 2.5 mg/ml with the order of VA>NA>RE mucosal homogenate. Medium chain fatty acid salts such as sodium caprylate $(C_8)$, sodium caprate $(C_{10})$ and sodium laurate $(C_{12})$ at the concentration of $0.5%{\sim}1.0%$ inhibit the proteolysis of LHRH significantly. The addition of sodium laurate(0.5%) into the NA and VA mucosal homogenates protected LHRH completely from the degradation.

Determination of Intestinal Disappearance of Lysine and Methionine Derived from Ruminally Protected Lysine and Methionine in Holstein Heifers

  • In order to estimate the availability of ruminally-protected Lys (RPLys) and Met (RPMet), quantitative changes of free Lys and Met in the total abomasal outflow and feces were determined in three Holstein heifers with a T-shaped duodenal cannula. RPLys was prepared by coating Lys with fat and RPMet with a pH-sensitive polymer. RPLys and RPMet containing 30 g of each amino acid were supplied together to heifers, and total abomasal outflow was collected hourly from the T-shaped duodenal cannula for a 48 h period. Collected abomasal outflows were fractionated into liquid and solid phases to measure free Lys and Met concentration. At 2 wk after total abomasal outflow sampling, heifers were supplied RPLys and RPMet together again, and total feces were collected every 12 h for a 3 d period t estimate excreted RPLys and RPMet in feces. The amounts of Lys and Met recovered from the liquid fractions of abomasal outflow were estimated to be $23.9{\pm}8.3%$ and $68.6{\pm}3.6%$ (p=0.008) of RPLys and RPMet ingested, respectively. Final intestinal disappearances of Lys and Met were estimated to be $49.5{\pm}2.6%$ and $78.2%{\pm}6.5%$ (p=0.015) of ingested RPLys and RPMet, respectively.

EFFECTS OF THE SUPPLEMENTAL LEVEL OF PROTECTED LYSINE ON PERFORMANCES OF HOLSTEIN DAIRY COWS

  • The objective of this study was to investigate the optimum level of the rumen protected lysine (RPLys) for early lactating Holstein dairy cow. This experiment was carried out with 16 Holstein dairy cows for 106 days and consisted of 4 treatments : $T_1$ (RPLys 0%), $T_2$ (RPLys 0.1%), $T_3$ (RPLys 0.2%) and $T_4$ (RPLys 0.3%). The results obtained are summarized as follows : 1. The daily intakes of feed were similar among treatments, but the digestibility of crude protein tended to increase 0.5-5.0% with increased level of RPLys and also the crude fiber digestibility increased (p < 0.05). 2. The daily weight gain for cows in $T_1$ was 253 g, which was lower than any other treatments (p < 0.05). The highest was 521 g in $T_3$. Also, the body condition score was changed from 3.22 at initial to 3.45 at final. The lowest increase in body condition score as 0.09 was obtained in control and the highest as 0.60 in $T_3$ (p < 0.01). 3. The total milk production of groups $T_2$, $T_3$ and $T_4$ were higher than $T_1$, as well as total protein, total fat and total solid yield. Especially in $T_4$ treatment group milk yield was higher than other treatments. The content of fat was higher in $T_2$ and $T_4$ compared to other treatments. Other components of milk were not significantly different (p > 0.05). The persistencies of lactation were increased in all RPLys treatments, especially, rate of reduction in milk yield was lowest in $T_4$ (p < 0.05). 4. The total amino acid content in the plasma, as well as plasma lysine content showed no consistent trend with treatments.

새로운 베스타틴 유사체의 입체선택적 합성 (Stereoselective Synthesis of Novel Bestatin Analogs)

  • 두 종류의 새로운 베스타틴(bestatin) 유사체를 $\small{D}$-leucine과 $\small{D}$-valine으로부터 효율적이면서 입체선택적으로 합성하였다. 아미노펩티데이즈 억제제인 베스타틴은 면역조절 효과를 보이며 급성백혈병 치료제로 상품화되어 있다. 주요 중간체인 trans-옥사졸리딘 메틸에스터 2a와 2b는 페닐설포닐나이트로메테인($PhSO_2CH_2NO_2$)과 N-하이드록시 메틸기가 보호기로 도입된 ${\alpha}$-아미노 알데하이드(4a와 4b) 간의 일련의 세 단계 연속반응과 연이은 가오존분해 반응으로부터 20 : 1 이상의 입체선택성으로 합성되었다. 2a와 2b의 가수분해 반응 후에 $\small{L}$-Leu-OMe와의 펩타이드 결합을 통하여 베스타틴의 새로운 유사체인 3a와 3b를 보호기가 도입된 형태로 얻었다. 이소부틸기와 이소프로필기를 갖는 두 종류의 새로운 베스타틴 유사체(1a와 1b)는 해당 ${\alpha}$-아미노알데하이드 4로부터 높은 입체선택성으로 6단계에 걸쳐 각각 51%와 38%의 수율로 합성되었다.

Neuroprotective Compounds Isolated from the Methanolic Extract of Lonicera japonica

  • A $CH_3Cl$ extract from the flower of Lonicera japonica (Lauraceae) significantly protected primary cultures of rat cortical cells injured by the excitotoxic amino acid, L-glutamate. Loganin (1), secoxyloganin (2), caffeic acid (3) rutin (4), hyperoside (5), quercetin-3-O-glucoside (6), lonicerin (7), kaempferol-3-O-rutinoside (8), luteolin-7-O-b-D-glucopyranoside (9), quercetin (10) and luteolin (11) were isolated by bioactivity-guided fractionation from the $CH_3Cl$ fraction and further separated using chromatographic techniques. Caffeic acid, lonicerin, kaempferol-3-O-rutinoside, quercetin and luteolin had significant neuroprotective activities against glutamate-induced neurotoxicity in primary cultures of rat cortical cells at concentrations ranging from $0.1{\mu}M$ to $10.0\;{\mu}M$.

Chemical Modification of 5-Lipoxygenase from the Korean Red Potato

  • The lipoxygenase was purified 35 fold to homogeneity from the Korean red potato by an ammonium sulfate precipitation and DEAE-cellulose column chromatography. The simple purification method is useful for the preparation of pure lipoxygenase. The molecular weight of the enzyme was estimated to be 38,000 by SDS-polyacrylamide gel electrophoreses and Sepharose 6B column chromatography. The purified enzyme with 2 M $(NH_4)_2SO_4$ in a potassium phosphate buffer, pH 7.0, was very stable for 5 months at $-20^{\circ}C$. Because the purified lipoxygenase is very stable, it could be useful for the screening of a lipoxygenase inhibitor. The optimal pH and temperature for lipoxygenase purified from the red potato were found to be pH 9.0. and $30^{\circ}C$, respectively. The Km and Vmax values for linoleic acid of the lipoxygenase purified from the red potato were $48\;{\mu}M$ and $0.03\;{\mu}M$ per minute per milligram of protein, respectively. The enzyme was insensitive to the metal chelating agents tested (2 mM KCN, 1 and 10mM EDTA, and 1 mM $NaN_3$), but was inhibited by several divalent cations, such as $Cu^{++}$, $Co^{++}$ and $Ni^{++}$. The essential amino acids that were involved in the catalytic mechanism of the 5-lipoxygenase from the Korean red potato were determined by chemical modification studies. The catalytic activity of lipoxygenase from the red potato was seriously reduced after treatment with a diethylpyrocarbonate (DEPC) modifying histidine residue and Woodward's reagent (WRK) modifying aspartic/glutamic acid. The inactivation reaction of DEPC (WRK) processed in the form of pseudo-first-order kinetics. The double-logarithmic plot of the observed pseudo-first-order rate constant against the modifier concentration yielded a reaction order 2, indicating that two histidine residues (carboxylic acids) were essential for the lipoxygenase activity from the red potato. The linoleic acid protected the enzyme against inactivation by DEPC(WRK), revealing that histidine and carboxylic amino acids residues were present at the substrate binding site of the enzyme molecules.

Diethylpyrocarbonate Inactivation of Aspartase from Hafnia Alvei

  • An aspartase purified from Hafnia alvei was inactivated by diethylpyrocarbonate (DEP) in a pseudo-first-order inactivation. The first-order plot was biphasic. The inactivation process was not saturable and the second order rate constant was $1.3\;M^{-1}s^{-1}$. The inactivated aspartase was reactivated with NH₂OH. The difference absorption spectrum of DEP-inactivated vs native enzyme preparations revealed a marked peak around 242 nm. The pH dependence of the inactivation rate suggests that an amino acid residue having a pK value of 7.2 was involved in the inactivation. L-aspartate, fumarate (substrates), and chloride ion (inhibitor) protected the enzyme against inactivation, indicating that histidine residues for the enzyme activity are located at the active site of this aspartase. Inspection of the presence and absence of $Cl^-$ ion demonstrated that the number of essential histidine residues is less than two. Thus, one or two histidines are in or near the aspartate binding site and participate in an essential step of the catalytic reaction.

  • A novel O-diethylaminoethyl chitosan (DEAE-chitosan) was synthesized via Schiff's reaction between chitosan and benzaldehyde. $C_2$ amino group was protected via Schiffs base reaction with benzaldehyde to form N-benzylidene chitosan. After reaction with diethylaminoethyl chloride, Schiffs base was removed by reacting O-diethylaminothyl-N-benzylidene chitosan and hydrochloric acid. Tensile strength of DEAE-chitosan was improved due to the incorporation of bulky side group in $C_6$ position of chitosan. DEAE-chitosan showed a pH-dependent swelling characteristics. Release rate of riboflavin was dependent on the water content of DEAE-chitosan that is a function of crosslinking degrees.

Inactivation of Brain myo-Inositol Monophosphate Phosphatase by Pyridoxal-5'-Phosphate

  • Myo-inositol monophosphate phosphatase (IMPP) is a key enzyme in the phosphoinositide cell-signaling system. This study found that incubating the IMPP from a porcine brain with pyridoxal-5'-phosphate (PLP) resulted in a time-dependent enzymatic inactivation. Spectral evidence showed that the inactivation proceeds via the formation of a Schiff's base with the amino groups of the enzyme. After the sodium borohydride reduction of the inactivated enzyme, it was observed that 1.8 mol phosphopyridoxyl residues per mole of the enzyme dimer were incorporated. The substrate, myo-inositol-1-phosphate, protected the enzyme against inactivation by PLP. After tryptic digestion of the enzyme modified with PLP, a radioactive peptide absorbing at 210 nm was isolated by reverse-phase HPLC. Amino acid sequencing of the peptide identified a portion of the PLP-binding site as being the region containing the sequence L-Q-V-S-Q-Q-E-D-I-T-X, where X indicates that phenylthiohydantoin amino acid could not be assigned. However, the result of amino acid composition of the peptide indicated that the missing residue could be designated as a phosphopyridoxyl lysine. This suggests that the catalytic function of IMPP is modulated by the binding of PLP to a specific lysyl residue at or near its substrate-binding site of the protein.
