Detection and Identification of Bacteriocins Produced by Propionibacteria Isolated from Commercial Swiss Cheese Products

  • Wild propionibacteria isolated from different commercial swiss cheese samples were tested for antimicrobial activities. In initial screening, six of these Propionibacterium isolates showed antagonistic activity against 10 selected indicator organisms by the deferred method. In next, only two Propionibacterium strains JW6 and JW14 showed antibacterial activity in the cell-free supernatants by the modified well diffusion method. Propionibacterium strains JW6 and JW14 were finally identified as bacteriocin producers which exhibited a bactericidal effect against closely related species. The antimicrobial substances were proteins, since their activities were completely destroyed following several degradative enzyme treatments. The bacteriocins showed a narrow inhibitory spectrum of activity against two propionibacteria and two bacilli of strains tested in this study.

  • Perstractive fermentation is a good way to increase the productivity of bioreactors. Us-ing Propionibacteria as the model system, the feasibility of using supported emulsion liquid mem-brane(SELM) fro perstractive fermentation is assessed in this study. Five industrial solvents were considered as the solvent for perparing the SELM. The more polar a solvent, is the higher the par-tition coefficeint However, toxicity of a solvent also increases with its polarity. CO-1055(indus-trial decanol/octanol blend)has the highest partition coefficient toward propionic acid among the solvents that has no molecular toxicity toward Propionibacteria, A preliminary extraction study was conducted using tetradecane as solvent in a hydrophobic hollow fiber contactor. The results confirmed that SELM eliminates the equilibrium limitation of conventional liquid-liquid extrac-tion and allows the use of a non-toxic solvent with low partition coefficient.

Biochemical Properities, Isolation & Identification of the Propionibacterium acnes Picked from Acne Lesion (여드름병변에서 채취한 Propionibacterium acnes의 분리, 동정 및 생화학적 특성 연구)

  • Thirty P. acnes were isolated from the 30 acne patients who visited some skin care shops from Feburary of 2001 to August of 2001. They were identified by morphological features (gram stain), biochemical properties. Only P. acne strains are isolated among Propionibacteria sp. and biochemical properities of P. acnes were not different from P. acnes ATCC 11827 strain significantly. The results of the biotyping was as follow: twenty three strains of biotype I from comedone, 1 strain of biotype II from pauple, 5 strains of biotype III from pauple and pustule, and 1 strain of biotype IV from pauple were isolated respectively.
