The ACC deaminase from rhizobateria promoted resistance of salininty stress in seedling and growth of plant

  • Rhizobacteria are a diverse group of free-living soil bacteria that live in plant rhizosphere and colonized the root system. Plant growth-promoting rhizobacteria (PGPR) possessing ACC deaminase (ACCD) can reduce ACC and ethylene in plant tissue and mediated the growth of plants under various stresses including salt stress. ACCD decrease ethylene levels in plant tissue that produce high levels of ethylene in tissue via elevated levels of ACC under salt stress. We selected strains of Pseudomonas sp. possessing ACCD activity for their ability to promote plant growth under salt stress from soil sample collected at Byeonsan, Jeonbuk, South Korea. The Pseudomonas strains possessing ACCD increased the rate of the seedling and growth of chinese cabbage seeds under salt stress. We cloned ACCD gene from P.fluorescens and expressed recombinant protein in Escherichia coli. The active form of recombinant ACCD converted ACC to a-ketobutyrate. The in vivo treatment of recombinant ACCD itself increase the rate of the seedling and growth of Chinese cabbage seeds under salt stress. The polyclonal P.fluorescens anti-ACCD antibody specifically reacted with ACCD originated from Pseudomonas. This indicates that the antibody might act as an important indicator for ACCD driven from Pseudomonas exhibiting plant growth-promoting activity. This study will be useful for identification of newly isolated PGPR containing ACCD and exploioting the ACCD activity from PGPR against various biotic and abiotic stresses.

온도, 일장 및 식물생장조절제에 의한 덴파레의 생육 및 개화 반응 (Flowering and Growth of Dendrobium phalaenopsis as influenced by Photoperiod, Temperature, and Plant Growth Regulators)

  • 덴파레 유묘 및 성묘의 온도, 일장, 생장조절제에 의한 생육 및 개화기 촉진을 목적으로 10시간 및 16시간 일장처리는 3월 24일부터 5월 23일까지 하여 생육 및 개화조사를 하였다. 1년생묘(유묘) 덴파레 'Semi alba'를 온도와 일장처리 결과, $30/25^{\circ}C$로 12주 처리시 생육이 가장 빨랐고, 16시간 일장일 때 신초의 생육과 개화가 촉진되었다. 2년생묘 (성묘) 덴파레 'Candistriape ${\times}$ Tedtakiguz'의 일장처리에서는 16시간 일장에서 소화수, 화폭, 경장 등 개화특성에는 차이가 나지 않았으나, 신초에서부터 개화소요일수는 16시간 일장이 216일, 자연일장 241일, 10시간 일장은 243일로, 16시간 일장이 자연일장이나 10시간일장보다 약 25일 빨랐다. 생장조절제처리는 생장과 개화에 뚜렷한 효과가 없었다.

  • The activities of auxins and cytokinins have been examined in the growth of callus tissue derived from Phaseolus vulgaris L. cv. Damyang. The synthetic auxin, picloram was the most effective in promoting callus growth and the range of effective concentrations (0.1$\mu{M}$ to 32$\mu{M}$) was broad. 2, 4-D also enhanced callus growth at the optimal concentration of 3.2$\mu{M}$. NAA promoted callus growth at relatively higher concentrations than other auxins tested. IAA was less effective in supporting callus growth. Cytokinin bearing saturated side chain ($N^6$-isopentyladenine) was approximately 30 times more active than the corresponding unsaturated compound, $N^6$-($\D^2$-isopentenyl) adenine. The abilities of cytokinin-autonomous growth were also examined. Callus tissues previously grown on concentrations lower and/or higher than optimal concentrations of cytokinins were better habituated in the subsequent passage. It was suggested that the development of cytokinin autonomy may be related to dosage-concentrations of cytokinin in the previous passage.

A Two-Strain Mixture of Rhizobacteria Elicits Induction of Systemic Resistance Against Pseudomonas syringae and Cucumber Mosaic Virus Coupled to Promotion of Plant Growth on Arabidopsis thaliana

  • We evaluated a commercial biopreparation of plant growth-promoting rhizobacteria (PGPR) strains Bacillus subtilis GB03 and B. amyloliquefaciens IN937a formulated with the carrier chitosan (Bio Yield) for its capacity to elicit growth promotion and induced systemic resistance against infection by Cucumber Mosaic Virus (CMV) and Pseudomonas syringae pv. tomato DC3000 in Arabidopsis thaliana. The biopreparation promoted plant growth of Arabidopsis hormonal mutants, which included auxin, gibberellic acid, ethylene, jasmonate, salicylic acid, and brassinosteroid insensitive lines as well as each wild-type. The biopreparation protected plants against CMV based on disease severity in wild-type plants. However, virus titre was not lower in control plants and those treated with biopreparation, suggesting that the biopreparation induced tolerance rather than resistance against CMV. Interestingly, the biopreparation induced resistance against CMV in NahG plants, as evidenced by both reduced disease severity and virus titer. The biopreparation also elicited induced resistance against P. syringae pv. tomato in the wild-type but not in NahG transgenic plants, which degrade endogenous salicylic acid, indicating the involvement of salicylic acid signaling. Our results indicate that some PGPR strains can elicit plant growth promotion by mechanisms that are different from known hormonal signaling pathways. In addition, the mechanism for elicitation of induced resistance by PGPR may be pathogen-dependent. Collectively, the two-Bacilli strain mixture can be utilized as a biological inoculant for both protection of plant against bacterial and viral pathogens and enhancement of plant growth.

Plant Growth Promoting Activities of Some Rhizosphere Bacteria and their Effect on Brassica rapa Growth

  • The necessity to develop economical and eco-friendly technologies is steadily increasing. Plant growth promoting rhizomicrobial strains PGPR are a group of microorganisms that actively colonize plant roots and increase plant growth and yield. Pot experiments were used to investigate the potential of some rhizobacterial strains to enhance the Brassica rapa growth. Microbial strains were successfully isolated from the rhizosphere of Panax ginseng and characterized based on its morphological and plant growth promotion characters. Surface disinfected seeds of Wisconsin Fast B. rapa were inoculated with the selected PGPR microorganisms. The different pots treatments were inoculated by its corresponding PGPR ($10^7cfu\;mL^{-1}$) and incubated in the growth chamber at $25^{\circ}C$ and 65% RH, the light period was adjusted to 24 hours (day). NPK chemical fertilizer and trade product (EMRO, USA) of effective microorganisms as well as un-inoculated control were used for comparison. Plants harvested in 40 days were found to have significant increase in leaf chlorophyll units and plant height and also in dry weight of root and shoot in the inoculated seedlings. Root and shoot length and also leaf surface area significantly were increased by bacterial inoculation in sterile soil. The study suggests that Rhodobacter capsulatus and Azotobacter chroococcum are beneficial for B. rapa growth as they enhance growth and induced IAA production and phosphorus solubilization. This study presents some rhizomicrobial strains that significantly promoted growth of Wisconsin Fast Plant B. rapa in pot experiment under different soil conditions.

Biocontrol Effect of Gliocladium virens G1 and Soil Amendment on Astragal Stem Rot Caused by Rhizoctonia solani

  • In order to find an environment-friendly method to suppress astragal stem rot caused by the isolates of Rhizoctonia solani AG 1 and AG 4, we tested an antagonistic fungus Gliocladium virens G1 was evaluated as a biocontrol agent and estimated inorganic compounds and organic materials were tested for their effect of the disease suppression. G. virens G1 effectively inhibited mycelial growth in a dual culture and caused mycelial lysis in the culture filtrate. No adverse effect was observed when examined for seed germination and seedling growth. Promoted seedling growth was observed with the seed treatment. Seeds of astragal plant were germinated higher in the sterile soil than the natural soil. Of 14 inorganics tested, alum, aluminum sulfate and calcium oxide significantly suppressed the mycelial growth and sclerotial germination. Milled pine bark and oak sawdust also suppressed the mycelial growth. Soil amended with 1% of G. virens G1 composted with pine bark (w/v) significantly controlled astragal stem rot in the glasshouse experiments.

자외선 형광장치현미경으로 본 화분관생장과 석탄 및 aureomycin의 상호작용에 대하여 (On the synergistic action of calcium and antibiotics in pollen growth as observed with ultraviolet fluorescence microscopy)

  • Observations were made on Crinum and Catharanthus pollen growth in artificial media by an ultraviolet transmission fluorescence microscope showing synergistic effect on pollen growth with calcium (Ca) and aureomycin. Bright yellow fluorescence of aureomycin enabling to trace out at tissue or cellular level did reveal that the greater accumulation of fluorescence occurred in the pollen tube wall if Ca was supplemented to the media than when aureomycin alone was present. The promotive pollen growth the media of Ca alone was further enhanced by the addtion of aureomycin. It was assumed that the promoted pollen growth with aureomycin in the Ca media was probably brought about by a supporting role of aureomycin in the Ca action.

Growth and Flowering Responses of Petunia to Various Artificial Light Sources with Different Light Qualities

  • This study was carried out to investigate the effect of artificial light sources with different light qualities on the growth and flowering characteristics of a herbaceous long-day plant, Petunia ${\times}$ hybrida Hort. Seedlings of petunia cultivar 'Madness Rose' were potted, acclimated for one week, and grown in a phytotron equipped with tube- and bulb-type fluorescent lamps (FL tube and bulb), tube-type white light-emitting diodes (LED tube), halogen lamps (HL), metal halide lamps (MH), and high pressure sodium lamps (HPS) for 10 weeks. The temperature, photoperiod, and photosynthetic photon flux density (PPFD) in the phytotron were $22{\pm}2^{\circ}C$, 16 h, and $25{\pm}2{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. Light sources combined with HL promoted stem elongation, and plant height and internode length decreased with increasing red to far-red (R:FR) ratio. FL tube + LED tube, HPS, and FL tube promoted branching, whereas plants grown under light sources combined with HL did not have any branches. Days to flowering (from longest to shortest) occurred as follows: FL tube + HL > FL tube + HL > MH > HPS = FL tube + FL bulb > FL tube + LED tube > LED tube > FL tube, indicating that reducing the R:FR ratio of the light sources promoted flowering. Only 20% of plants grown under an FL tube flowered, whereas under all other treatments, 100% of plants flowered. At 10 weeks after treatment, plants grown under HPS and MH had (cumulatively) 12 open flowers, and those grown under FL tube + FL bulb, LED tube, FL tube + LED tube, and HPS treatment had approximately seven flower buds. These results suggest that light sources with low R:FR ratios promote flowering and stem elongation in petunia, but they reduce its ornamental value due to overgrowth and poor branching.

  • 본 연구는 포자체 배양을 통하여 참쇠고비의 효율적인 번식방법을 구하고자 수행되었다. 배양절편의 기원(근경, 엽병, 엽신)과 절편의 균질화가 신초 계생에 미치는 영향을 조사한 결과, 오직 근경 조직에서만 기관발생능력이 관찰되었고, 식물체 재생은 배양재료를 균질화함으로써 더욱 촉진되었다. MS배지의 무기물 농도를 2배로 첨가한 배지에서 신초의 증식과 생장이 활발히 이루어졌다. 신초 재생에 적합한 배지의 질소함량은 MS배지의 1/2배 수준 이었고, 적정 sucrose농도는 1%였다. 또한 배지에 $NaH_2PO_4$를 첨가함으로써 재생된 식물체의 생육이 전반적으로 촉진되었다. 근경절편의 기관형성능력은 kinetin과 IBA를 혼용 처리함으로써 촉진되었으며, $0.1\sim0.2%$의 활성탄과 생장조절물질을 혼용 처리한 결과, 다수의 싹이 뭉친 듯이 보이는 primordia 형태의 발달이 억제되었고, 포자체의 정상적인 형태형성이 향상되었다.

Antagonistic Potential of Fluorescent Pseudomonads and Control of Crown and Root Rot of Cucumber Caused by Phythophtora drechsleri

  • In this study, 200 isolates of fluorescent pseudomonads were isolated from different fields of East and West Azarbaijan and Ardebil provinces of Iran. These bacterial isolates were screened on the basis of a dual culture assay, the presence of known antibiotic genes, and their ability to successfully colonize roots and to promote plant growth. Twelve isolates exhibited 30% or more inhibition of mycelia growth of $P.$ $drechsleri$. Genes encoding production of the antibiotics 2,4-diacetylphloroglucinol, phenazine-1-carboxylic acid, and pyoluteorin were detected in some strains but none of the strains possessed the coding gene for production of antibiotic pyrrolnitrin. In an $in$ $vitro$ test for root colonization, the population density on roots of plants treated with most of the above strains was more than 6 $\log_{10}$ CFU $g^{-1}$ roots, with a maximum of 7.99 $\log_{10}$ CFU $g^{-1}$ roots for strain 58A. Most of the strains promoted significant plant growth in comparison to non-treated controls. In green house studies, the percentage of healthy plants in pots treated with strains 58A and 8B was 90.8% and 88.7%, respectively. The difference between these treatments and treatment with the fungicide metalaxyl was not significant.
