벼 현탁배양을 통하여 분리된 원형질체로부터 식물체 재분화 (Plant Regeneration from Protoplasts Isolated through Embryogenic Cell suspension Culture in Rice)

  • Plant regeneration was accomplished from protoplast culture of rice (Oryza sativa L. cv. Taebaeg). Embryogenic callus was induced from mature seed on MS medium containing 5 mM proline, 2.5 mg/L 2,4-D, 30 g/L sucrose in the dark at 28$^{\circ}C$ and used to establish embryogenic cell suspension culture. Suspension cells were subcultured every one week in N6 medium supplemented with 5 mM proline, 200 mg/L casein hydrolysate, 2.5 mg/L 2,4-D and amino acids of AA medium. Suspension cultures were composed of cells that were densely cytoplasmic, potentially embryogenic and were at least maintained for more than 6 months in liquid medium. Protoplasts were isolated from fast-growing suspension culture cells and cultured in a slightly modified KpR medium by mixed nurse culture. Isolated protoplasts began to divide within 5~7 days and thereafter, protoplast-derived calli were sequentially transferred to callus proliferating medium that soft agar MS medium contained 2 mg/L 2,4-D and produced distinct embryogenic cells. Microcolonies were then transferred to solid medium which consisted of MS medium containing 5 mg/L kinetin, 1 mg/L NAA, 1 mg/L ABA, 30 g/L sucrose and 10 g/L sorbitol under fluorescent light. Mulitple shoots of 4~5 per callus emerged and were transferred to hormone-free MS medium for root initiation. Thereafter, The plantlets were transferred to pots of soil to mature in the culture room.

곶감과 동충하초 첨가에 따른 청주(삼백주)의 발효 특성 변화 (Changes of Quality Characteristics in Brewing of Chungju(Sambaekju) Supplemented with Dried Persimmon and Cordyceps sinensis)

  • 본 연구에서는 곶감과 동충하초의 첨가에 따른 청주(삼백주)의 발효 특성을 조사하였다. 환원당 함량을 측정한 결과 발효가 경과함에 따라 감소하는 경향을 나타내었으며 알코올 함량의 변화 발효가 경과함에 따라 증가하는 경향을 나타내었다. 각 담금 후의 최종 알코올의 함량은 각각 5.8%, 11.4% 및 16.5%를 나타내었다. 총산의 함량과 아미노산도는 발효 과정에 따라 증가 추세를 보였다. 유기산 함량은 젖산의 함량이 가장 높게 나타났으며 기타의 유기산은 초산, 사과산, 호박산등은 검출되었으나 구연산과 주석산은 검출되지 않았다. 유리 아미노산의 함량은 L- proline의 함량이 1151.7 mg%로 가장 높게 나타났으며, 총 유리 아미노산과 필수 아미노산의 함량은 각각 1.19%와 217.70 ppm이었다.

A Proline- and Leucine-rich 19 Amino Acid Oligopeptide from FS1 Functions as a Transcriptional Repression Domain

  • We have used a transient expression assay employing Drosophila S2 cells to study the transcriptional repression activity of a 27 amino acid residue-long repression domain FS1 which was generated by a frame-shift in a pair-rule gene, even-skipped of Drosophila melanogaster. In an attempt to define a minimal requirement for the repression activity, we constructed a series of truncation mutant forms of the FS1, fused to a heterologous GAL4 DNA-binding domain, and measured their activities. All of the mutant forms, including the GAL4-FS1 (5-23) which retains the smallest number (19) of amino acid residues of FS1, were found to repress an initiator, a minimal TATA-lacking promoter, in a GAL4-binding-site-dependent manner. These findings suggest that a 19 amino acid residue-long region, rich in proline and leucine residues, is a transcriptional repression domain and may interact with the general transcription machinery.

Measurement of Metal-Film Removal Rate in a Microemulsion Using QCM

  • A set of Quartz Crystal Microbalances (QCM's) was used to observe the film removal characteristics of three different $CO_2-nitric$ acid microemulsions. QCM's electroplated with nickel or copper were used as specimens. F-AOT, NP-4 and the newly synthesized Proline Surfactant-1 were used as surfactants to create microemulsions. While the F-AOT microemulsion yielded a relatively low removal rate, that of the Proline Surfactant-1 completely removed the Cu metal film within a short period of time. The NP-4 microemulsion removed the metal surface. However, removal rate measurements per QCM were not possible due to the instability of the microemulsion when Cu ions were present in the nitric solution. The reaction kinetics and metal removal capabilities of microemulsions formed by the different surfactants are explained along with the characteristics of reverse micelles.

  • 네품종의 보리(올보리, 영산보리, 사천 6호, 수원 228호)로 부터 Osborne의 방법과 이것을 변형한 전보의 방법으로 단백질을 분획하여 albumin, globulin, hordein 및 glutelin 획분을 얻어 이들 각획분들을 SDS-PAGE로 분석한 결과 품종간 polypeptide 조성 차이를 관찰할 수 있었으며 특히 hordein과 hordein-I을 pH 3에서 PAGE를 수행 하였을 때 각 품종사이의 pattern에 명확한 차이를 볼 수 있었다. 한편, hordein-I의 아미노산조성은 glutamic acid와 proline 함량이 높았으나 품종간의 아미노산조성차이는 거의 볼 수 없었다.

염절임 가지(시키부, Shikibu)의 저장방법에 따른 성분 변화

  • 본 연구는 수출방법의 개선과 절임 가공식품의 개발하고자 일본 수출용 시키부(Shikibu) 염절임 가지의 저장방법과 저장중에 당류, 유기산 및 아미노산의 변화를 분석하였다. 총당 및 환원당 함량은 저장방법에 관계없이 저장기간이 길어질수록 조금씩 감소하였고, 침지 저장한 것보다는 진공포장하여 저장한 것이 변화가 적었다. 가지에서 분리 정량된 유기산은 acetic acid, citric acid, lactic acid, malic acid 그리고 succinic acid 총 5종이었으며 acetic acid와 malic acid의 함량이 가장 높았으며, 저장기간이 길어질수록 저장 초기에 비해 acetic acid의 함량은 증가하였는데 반해 malic acid의 함량은 감소하였다. 가지의 주요한 구성아미노산은 valine, leucine, aspartic acid, glutamic acid, proline, alanine 등이었으며, 필수아미노산 함량은 저장기간이 길어질수록 저장 초기에 비해 진공포장하여 저장한 방법에서는 감소하였다가 증가하였는데 반해 침지하여 저장한 방법에서는 계속 증가하였고 저장 20일째에는 진공포장한 방법보다 l00mg/100g 정도의 차이를 나타내었다. 유리아미노산은 aspartic acid, alanine, cystine과 proline등의 함량이 높았고, 저장 기간이 길어질수록 저장초기에 비해 유리아미노산의 함량이 증가하였는데, 특히 진공 포장하여 저장한 가지에 비해 침지 저장 한 가지에서 그 함량이 높았다. 또한 아미노산 유도체의 함량은 phosphoserine, taurine, ${\gamma}$-aminoisobutyric acid 및 hydroxyproline의 함량이 높았고, 저장중에 phosphoserine의 함량은 감소하였는데 반해 ${\gamma}$-aminoisobutyric acid는 진공포장하여 20일 동안 저장하였더니 생가지 보다 5배 이상의 증가를 보였다. 스키부품종의 절임가지의 저장방법과 저장기간에 따른 성분의 변화를 고려할 때 축양품종과 동일하게 진공포장하는 것이 절임가지의 품질유지할 수 있는 것으로 생각된다.

Ab Initio Conformational Study on Ac-Flp-NHMe: Stereoelectronic Effects on Proline Conformation

  • We report here the results on N-acetyl-N'-methylamide of 4-fluoroproline (Ac-Flp-NHMe) calculated using the ab initio molecular orbital method with the self-consistent reaction field (SCRF) theory at the HF level with the 6-31+G(d) basis set to investigate the stereoelectronic effects on the conformational preference of proline depending on the cis/trans peptide bonds and down/up puckerings along the backbone torsion angle $\square$ in the gas phase, chloroform, and water. In the gas phase, all potential energy surfaces for Ac-Flp-NHMe are quite similar to those of Ac-Pro-NHMe, except that up-puckered conformations are more stabilized than down-puckered ones. In chloroform and water, polyproline structures become dominant, whose populations are larger than those of Ac-Pro-NHMe. In chloroform and water, the populations of polyproline II (i.e., tF conformations) are quite similar to each other, but those of polyproline I (i.e., cF conformations) are larger by 5% in water than in chloroform. In particular, all cis populations for Ac-Flp-NHMe in the gas phase, chloroform, and water are decreased than those of Ac-Pro-NHMe.

진도산(珍島産) 구기자(枸杞子)의 아미노산조성(組成)과 유리당(遊離糖)의 분석(分析) (Quantitative Analysis of Total Aimno Acids and Free Sugars in Lycii fructus)

  • 구기자(枸杞子)의 총(總)아미노산조성(組成) 및 유리(遊離)당을 각각 amino acid autoanalyzer, HPLC로 분석(分析)한 결과(結果)는 다음과 같다. 1. 17종(種)의 총(總)아미노산이 검출(檢出)되었다. 즉 asparatic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystine, valine, methionine, isoleucine, leucine, tyrosine, phenr-lalanine, hisidtine, lysine, arginine인데 이중 threonine을 가장 많이 함유하였으며 aspartic acid는 가장 적었다. 2. 총(總) essential amino acid량은 총(總)아미노산량의53.93%로 양질(良質)의 아미노산을 함유하였다. 3. Glucose, fructose, saccharose 등 3종(種)의 유리(遊離)당이 검출(檢出)되었는데 이중 fructose를 가장 많이 함유하였다.

Differentiation of Roots of Glycyrrhiza Species by 1H Nuclear Magnetic Resonance Spectroscopy and Multivariate Statistical Analysis

  • To classify Glycyrrhiza species, samples of different species were analyzed by $^1H$ NMR-based metabolomics technique. Partial least squares discriminant analysis (PLS-DA) was used as the multivariate statistical analysis of the 1H NMR data sets. There was a clear separation between various Glycyrrhiza species in the PLS-DA derived score plots. The PLS-DA model was validated, and the key metabolites contributing to the separation in the score plots of various Glycyrrhiza species were lactic acid, alanine, arginine, proline, malic acid, asparagine, choline, glycine, glucose, sucrose, 4-hydroxy-phenylacetic acid, and formic acid. The compounds present at relatively high levels were glucose, and 4-hydroxyphenylacetic acid in G. glabra; lactic acid, alanine, and proline in G. inflata; and arginine, malic acid, and sucrose in G. uralensis. This is the first study to perform the global metabolomic profiling and differentiation of Glycyrrhiza species using $^1H$ NMR and multivariate statistical analysis.

Structure and Activity of Angiotensin I Converting Enzyme Inhibitory Peptides Derived from Alaskan Pollack Skin

  • Angiotensin I that converts the enzyme (ACE) inhibitory peptide, Gly-Pro-Leu, previously purified and identified from the Alaskan pollack skin gelatin hydrolysate, were synthesized. In addition, the peptides Gly-Leu-Pro, Leu-Gly-Pro, Leu-Pro-Gly, Pro-Gly-Leu, Pro-Leu-Gly, Gly-Pro, and Pro-Leu, which consisted of glycine, proline, and leucine, were synthesized by the solid-phase method. The $IC_{50}$ values of each tripeptide - namely Leu-Gly-Pro, Gly-Leu-Pro, Gly-Pro-Leu, Pro-Leu-Gly, Leu-Pro-Gly, and Pro-Gly-Leu - were 0.72, 1.62, 2.65, 4.74, 5.73, and $13.93{\mu}M$, respectively. The ACE inhibitory activity of these tripeptides was higher than that of dipeptides, such as Gly-Pro and Pro-Leu with $IC_{50}$ values of 252.6 and $337.3\;{\mu}M$, respectively. Among the tripeptides, Leu-Gly-Pro and Gly-Leu-Pro had higher inhibitory activity than Gly-Pro-Leu that was isolated from the Alaskan pollack skin gelatin hydrolysate. Among the different types of tripeptides that were examined, the highest ACE inhibitory activity was observed for Leu-Gly-Pro. It had the leucine residue at the N-terminal and proline residue at the C-terminal.
