Salt Tolerance in Transgenic Pea (Pisum sativum L.) Plants by P5CS Gene Transfer

  • Slices of embryonic axis of mature pea (Pisum sativum L. cv. Green Arrow) seeds were used as explant. Transformation of explants was done via Agrobacterium tumefaciens bearing vector pBI-P5CS construct. The best results for inoculation of explants were obtained when they were immersed for 90 s at a concentration of $6{\times}10^8$ cell $ml^(-1)$ of bacterial suspension. Transformed pea plants were selected on $50\;mg\;l^(-1)$ kanamycin and successful transformants were confirmed by PCR and blotting. Transgenic plants were further analyzed with RT-PCR to confirm the expression of P5CS. Transgenic plants and non-transgenic plants were treated with different concentrations of NaCl 0 (control), 100, 150 and 200 mM in culture medium. Measurement of proline content indicated that transgenic plants produced more amino acid proline in response to salt in comparison with non-transgenic plants. Photosynthetic efficiency in transgenic plants under salt-stress was more than that of non-transgenic plants.

  • Phenylurea, phenylthiourea, benzylcarbamate and toluenesulfonate of L-alanyl-L-proline(comp. $3{\sim}6$) were synthesized and their effects against angiotensin-converting enzyme (ACE) were tested. Comp. 3 showed only mild inhibitory activities against ACE while comp. $4{\sim}6$ were inert indicating that those functional groups were not suitable for interactions with ACE. Ortho-hydroxy- or ortho-carboxy-benzamide of proline (comp. 7) and phenylalanine (comp. 8 and 9) were also tested. Of the benzamides, ortho-hydroxy function was unsuitable to exert inhibitory activities against ACE. Ortho-carboxy group of 9 seemed to have mild interactions with active site of ACE possibly because of the shorter distance between the amide and ortho-carboxyl group of the compound than the corresponding two active sites of the enzyme.

AN ELECTRON MICROSCOPIC RADIOAUTOGRAPHIC STUDY OF PROTEIN SYNTHESIS IN VITRO IN THE PALATAL MUCOSA OF THE RAT

  • The incorporation of ³H-proline by epithelial and connective tissue elements of rat palatal mucosae was studied in order to investigate the relative levels of protein synthesis by the epithelium and underlying connective tissue cells. Following a sixty minutes incorporation of the radioactive tracer in vitro, it was found that the suprabasal cells had most grains per unit area. Furthermore, the grains were more concentrated over the cytoplasm than the nucleus. This was in contrast with the labeling of basal cells which had twice as many grains over the nucleoplasm than that over the cytoplasm. In intermediate cells; i.e., the spinous layer, the number of silver grains per unit area was decreased from that of the suprabasal cells. In areas where desmosomes were more prominent, many grains were in touch with such desmosomes. However, the labeling appeared to be reduced as soon as the cells became flattened. Moreover, the epidermal keratohyalin granules were relatively free of grains. Except for certain intercellular surfaces the keratinized cells were generally free of grains. On the connective tissue side, silver grains were primarily localized over the fibroblasts with occasional grains being found over palatal muscle cells, neural elements and so on. Most grains over collagenous fibers were found in relation to mature collagen fibrils. Thus, protein synthesis in isolated mucosae of the rat palate appeared to take place both in epithelial and connective elements. There were no apparent tissue alterations caused by the in vitro incorporation procedure utilized under conditions of this study.

  • 녹차의 맛을 내는데 주요 성분인 유리아미노산과 유리당을 HPLC로 분석한 결과를 요약하면 다음과 같다. 1. 16종의 유리아미노산이 검출되었다. 즉, aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, histidine, lysine, arginine이 그것이었다. 2. 증제차가 볶음차보다 유리아미노산의 함량이 많았고, 녹차중에는 특히 threonine과 aspartic acid, lysine의 함량이 많았다. 3. Fructose, glucose, sucrose, raffinose등 4종의 유리당이 검출되었다. 4. 유리당은 가열시간 증가에 따라 각 성분의 함량이 현저히 감소하였다.

  • (1) 쌀에 있어서 보다 녹두(綠豆)에 있어서 전질소량(全窒素量), 가용성질소양(可溶性窒素量), 단백태질소양(蛋白態窒素量) 및 Alcohal 가용성(可溶性) 질소양(窒素量)이 더욱 많았다. (2) 쌀혼합녹두(混合綠豆)를 지진 경우가 지지기 전(前)보다 전질소양(全窒素量)과 단백태질소양(蛋白態窒素量)이 더 많았고 가용성질소(可溶性窒素), Alcohol 가용성(可溶性) 질소(窒素)에서는 별 차이(差異)가 없었으나 Alcohol 가용성질소(可溶性窒素)에서는 약간의 감소(減少)를 보였다. (3) 녹두(綠豆)를 지짐으로 해서 Valine, ${\gamma}-aminobutyrate$, Glutamine, Arginine, Methionine, Unknown 1,2 가 소실(消失)되었고 Proline, Lysine, Homoserine, Tyrosine은 검출(檢出)되었다. (4) 쌀혼합녹두(混合綠豆)리 지진 경우 지짐으로 해서${\gamma}-aminobutyrate$, Lysine, Histidine, Tyrosine, Cysteine 이 소실(消失)되었고 Phenylalanine, Leucine, ${\beta}-alanine$이 검출(檢出)되었다.

  • 화학 발암제인 dimetylnitrosamine(DMN)을 Hamster에 구강투여하였을 때 간의 아미노산 조성에 미치는 영향을 규명하기 위하여 reversed phase liquid chromatography법으로 아미노산을 정량하였다. DMN을 투여한 실험군의 간에서의 아미노산의 농도는 대조군에 비해 aspartic acid, glycine, glutamine, histidine, proline, tyrosine, leucine 등은 약 2배, valine, tryptophan은 약 3배, phenylalanine은 약 10배 증가하였고, threonine은 23%로 감소하였으며, serine, alanine, arginine, methionine, isoleucine, lysine은 큰 변화가 없었다.

중회귀분석을 이용한 보리간장 맛의 평가 (Evaluation of Taste in Kanjang Made with Barley Bran Using Multiple Regression Analysis)

  • 본 연구에서는 맛성분과 관능검사자료를 중회귀 분석하여 보리간장 맛을 평가하였다. 단순상관에서는 proline, alanine, methionine, lysine, histidine, lavulinic acid, ${\alpha}$-ketogutaric acid 및 citric acid와 관능검사 점수사이의 상관계수가 유의성이 높은 것으로 나타났으며, threonine, serine, cystein, phenylalanine, succinic acid, arabinose, xylose 및 sucrose 둥의 성분은 관능검사 점수에 거의 상관이 없는 것으로 나타났다. 하지만 단순상관분석으로는 보리간장 맛의 품질을 측정하는 것이 불가능하였다. 중회귀 분석을 실시한 결과 결정계수가 0.93 이상을 나타내어 맛성분 함량과 중회귀식을 이용하여 보리간장 맛의 93%를 추정하는 것이 가능하였다.

  • 환원탈지유배지에서 자란 Lactobacillus helveticus YM-1의 cell-free filtrate로부터 Streptococcus thermophilus CH-1의 생육촉진을 확인한 결과 이 물질은 분자량이 5,000정도의 peptide로 나타난다. 이 peptide 중에는 aspartic acid, Iysine, proline, leucine, valine, alanine, glutamic acid등의 amino산이 비교적 풍부하였으며, 그중에서 glutamic acid 함량이 가장 높았다. 또 이 Peptide가 생육 촉진효과 나타내는데 가장 중요한 역할을 담당하는 것은 glutamic acid와 Phenylalanine으로 판단되었다.

Metabolic Differentiation of Saccharomyces cerevisiae by Ketoconazole Treatment

  • Azole fungicides are one of the most wide-spread antifungal compounds in agriculture and pharmaceutical applications. Their major mode of action is the inhibition of ergosterol biosynthesis, giving depletion of ergosterol, precursors and abnormal steroids. However, metabolic consequences of such inhibition, other than steroidal metabolitesare not well established. Comprehensive metabolic profiles of Saccharomyces cerevisiae has been presented in this study. Wild type yeast was treated either with glucose as control or azole fungicide (ketoconazole). Both polar metabolites and lipids were analyzed with gas chromatography-mass spectrometry. Approximately over 180 metabolites were characterized, among which 18 of them were accumulated or depleted by fungicide treatment. Steroid profile gives the most prominent differences, including the accumulation of lanosterol and the depletion of zymosterol and ergosterol. However, the polar metabolite profile was also highly different in pesticide treatment. The concentration of proline and its precursors, glutamate and ornithine were markedly reduced by ketoconazole. Lysine and glycine level was also decreased while the concentrations of serine and homoserine were increased. The overall metabolic profile indicates that azole fungicide treatment induces the depletion of many polar metabolites, which are important in stress response.

Dipeptides Inhibit Melanin Synthesis in Mel-Ab Cells through Down-Regulation of Tyrosinase

  • This study investigated the effects of proline-serine (PS) and valine-serine (VS) dipeptides on melanogenesis in Mel-Ab cells. Proline-serine and VS significantly inhibited melanin synthesis in a concentration-dependent manner, though neither dipeptide directly inhibited tyrosinase activity in a cell-free system. Both PS and VS down-regulated the expression of microphthalmia-associated transcription factor (MITF) and tyrosinase. In a follow-up study also described here, the effects of these dipeptides on melanogenesis-related signal transduction were quantified. Specifically, PS and VS induced ERK phosphorylation, though they had no effect on phosphorylation of the cAMP response element binding protein (CREB). These data suggest that PS and VS inhibit melanogenesis through ERK phosphorylation and subsequent down-regulation of MITF and tyrosinase. Properties of these dipeptides are compatible with application as skin-whitening agents.
