Three flavonoids (1 - 3) and three phenolic compounds (4 - 6) were isolated from the whole plant of Geranium thunbergii Sieb. et Zucc (Geraniaceae). Their structures were determined by chemical and spectral analysis. These compounds were examined for the inhibitory activity of IL-6 production in $TNF-{\alpha}$ stimulated MG-63 cell. Among the isolated compounds, gallic acid (4) and gallic acid methyl ester (6) showed potent inhibitory activity.
The effect of carbon sources on tacrolimus production by a mutant strain of Streptomyces clavuligerus CKD 1119, an isolate from soil, was examined. Among the carbohydrates and oils tested in this work, a mixed carbon source of soluble starch and com oil was the best. An analysis of the culture kinetics also showed that, in contrast to the carbohydrates, the com oil was consumed later in the antibiotic production phase, implying that the oil substrate was the principal carbon source for the biosynthesis of tacrolimus, and this was directly proven by experiments using $^{14}C$-glucose and $^{14}C$-oleate substrates. Furthermore, com oil induced the formation of lipase by the mutant strain, whereas the addition of glucose significantly repressed lipase activity. The lipase activity exhibited by the FK-506-overproducing mutants was also observed to be directly proportional to their tacrolimus yield, indicating that a high lipase activity is itself a crucial factor for tacrolimus production. A feasibility study with a 200-1 pilot-scale fermentor and the best strain (Tc-XII-15322) identified in this work revealed a high volumetric and specific productivity of about 495 mg/l and 0.34 mg/mg dry mycelium, respectively.
For biotechnological production of high-valued ${\beta}-{\text\tiny{D}}$-hexyl glucoside, the catalytic properties of Hanseniaspora thailandica BC9 ${\beta}$-glucosidase purified from the periplasmic fraction were studied, and the transglycosylation activity for the production of ${\beta}-{\text\tiny{D}}$-hexyl glucoside was optimized. The constitutive BC9 ${\beta}$-glucosidase exhibited maximum specific activity at pH 6.0 and $40^{\circ}C$, and the activity of BC9 ${\beta}$-glucosidase was not significantly inhibited by various metal ions. BC9 ${\beta}$-glucosidase did not show a significant activity of cellobiose hydrolysis, but the activity was rather enhanced in the presence of sucrose and medium-chain alcohols. BC9 ${\beta}$-glucosidase exhibited enhanced production of ${\beta}-{\text\tiny{D}}$-hexyl glucoside in the presence of DMSO, and 62% of ${\beta}-{\text\tiny{D}}$-hexyl glucoside conversion was recorded in 4 h in the presence of 5% 1-hexanol and 15% DMSO.
Ochnaflavone, a naturally occurring biflavonoid composed of two units of apigenin (5,7,4'-trihydroxyflavone) joined via a C-O-C linkage, was first synthesized and evaluated its inhibitory activity on $PGE_2$ production. Total synthesis was accomplished through modified Ullmann diaryl ether formation as a key step. Coupling reactions of 4'-halogenoflavones and 3'-hydroxy-5,7,4'-trimethoxyflavone were explored in diverse reaction conditions. The reaction of 4'-fluoro-5,7-dimethoxyflavone (2c) and 3'-hydroxy-5,7,4'-trimethoxyflavone (2d) in N,N-dimethylacetamide gave the coupled compound 3 in 58% yield. Synthetic ochnaflavone strongly inhibited PGE2 production ($IC_{50}=1.08{\mu}M$) from LPS-activated RAW 264.7 cells, which was due to reduced expression of COX-2. On the contrary, the inhibition mechanism of wogonin was somewhat different from that of ochnaflavone although wogonin, a natural occurring anti-inflammatory flavonoid, showed strong inhibitory activity of $PGE_2$ production ($IC_{50}=0.52{\mu}M$), and seems to be COX-2 enzyme inhibition. Our concise total synthesis of ochnaflavone enable us to provide sufficient quantities of material for advanced biological studies as well as to efficiently prepare derivatives for structure-activity relationship study.
Cirsium japonicum var. ussuriense is known to have a variety of biological activities, including anti-inflammatory, analgesic activity and antipyretic activity. In this study we investigated the role of polyacetylene compound, 1-Heptadecene-11, 13-diyne-8, 9, 10-triol (PA) from the root of Cirsium japonicum var. ussuriense as an immune-modulator. PA was evaluated as inhibitors of some macrophage functions involved in the inflammatory process. We tested the effect of PA on the production of pro-inflammatory cytokines, interleukin-1beta (IL-$1{\beta}$) and tumor necrosis factor-alpha (TNF-$\alpha$), and nitric oxide (NO) in murine macrophage cell line, RAW264.7. There was no effect on cytokine production of macrophages by PA itself. However, PA inhibited lipopolysaccharide (LPS)-induced IL-$1{\beta}$ and TNF-$\alpha$ production by macrophages at a dose dependent manner. PA also suppressed the NO production of macrophages by LPS. LPS-induced NF-${\kappa}B$ activity was decreased by treatment of PA. Therefore, these results suggest that PA has anti-inflammatory effect by inhibiting the NF-${\kappa}B$ activation.
This study was conducted to investigate the tyrosinase inhibitory and melanin production inhibitory activity of the distilled water extract of Dendropanax morbifera leaf (DMW) and the fermented extract by Lactobacillus plantarum (DMF). DMF was prepared by inoculation of L. plantarum after the extraction procedure with distilled water. Fermentation for 48 hours at $37^{\circ}C$ is the most effective condition in this study. In DPPH radical scavenging ability, $SC_{50}$ values of the fermented DMF was $37.9{\mu}g/ml$ as a result of more effective than DMW extract ($52.6{\mu}g/ml$). Moreover, tyrosinase inhibitory activity of DMF showed higher activity than DMW. In nontoxic concentration range, DMF showed strong melanin production inhibitory effect in ${\alpha}$-melanocyte stimulating hormone-stimulated B16F10 cell. As a result, the fermentation of the distilled water extract of D. morbifera leaf by L. plantarum could be applicable to functional materials production for skin-whitening agents.
To investigate the possibility of development as a whitening agent using arctigenin, we measured DPPH assay, NBT/XO assay, intracellular ROS scavenging assay, tyrosinase assay and MSH-induced melanin production in B16 melanoma cells. Arctigenin dose-dependently had anti-oxidant activity in DPPH, NBT/XO and intracellular ROS assay. Although arctigenin did not inhibit purified tyrosinase activity, it dose-dependently inhibited tyrosinase activity and melanin production in B16 melanoma cells stimulated by $1{\mu}M$${\alpha}$-MSH. In particular, arctigenin at a concentration $100{\mu}M$ inhibited ${\alpha}$-MSH-stimulated tyrosinase activity and melanin production by $50.9{\pm}2.9%$ and $69.0{\pm}6.5%$ respectively. And typical tyrosinase inhibitor, arbutin, inhibited $57.7{\pm}2.9%$ and $65.1{\pm}5.0%$ respectively. Such an similar inhibitory effect of arctigenin and arbutin in B16 melanoma cells may be due to the inhibition of MSH signal pathway rather than the direct inhibition of tyrosinase. Therefore, these results suggest that arctigenin may be useful for the development as whitening agents.

Acne vulgaris, the most common skin disease. can be formed as only a few comedons or severe inflammatory lesions. The pathogenesis of acne involves various factors; excessive androgen, excessive sebum production, abnormal alteration of follicular epithelium, proliferation of Propionibacterium acnes, and inflammation. We investigated acne therapy using oriental herbs described in the Korean traditional medical book(Dong-ui-bo-gam). Oriental herbs(Angelica daurica, Arctium lappa, Coptidis rhizoma, and Glycyrrhiza glabra) were chosen based on their respective property of sebum control, anti-inflammatory activity, and anti-bacterial activity. We examined the effect of acne treatment, in terms of chemotactic inhibition, lipogenesis inhibition, and anti-bacterial activity for P. acnes. 1. Neutrophil chemotaxis assay; P. acnes secrete chemotactic factors and other pro-inflammatory extracellular products. Neutrophil chemotactic activity of P. acnes was measured by 48-well chemotaxis method. Angelica daurica clearly suppressed chemotactic activity of P. acnes. 2. Using sebaceous gland of hamster ear lipogenesis assay; Sebaceous lipogenesis was measured using ear biopsies by incubation or $C^{14}$-acetate in culture media. The $C^{14}$-labeled lipids were extracted and determined by liquid scintilation counting. Coptidis rhizoma markedly inhibited sebum production. 3. Anti-bacterial assay for P. acnes(MIC test); Glycyrrhiza glabra showed anti-bacterial activity. P. acnes did not develop resistance against Glycyrrhiza glabra. Retinoids are effectively to inhibit sebum production and regulate follicular keratinization process, with little anti-inflammatory activity. Angelica daurica suppressed neutrophil chemotaxis, Coptidis rhizoma inhibited sebum production, and Glycyrrhiza glabra showed anti-bacterial activity against P. acnes. A combined formulation of Angelica daurica, Coptidis rhizoma. and Glycyrrhiza glabra is expected to provide effective acne treatment.

Chlamydomonas reinhardtii is a green algae that can use light energy and water to produce hydrogen under anaerobic condition. This work reports the effect of limiting factors on hydrogen production in sulfur deprived anaerobic C. reinhardtii culture. In order to confirm the relationship between hydrogen production and limiting factors such as residual PSII activity and endogenic substrate degradation, the increase in chlorophyll concentration and the decrease in starch concentration was investigated during sulfur deprivation. The overall hydrogen production increased depending on cell density in range of $0.4{\sim}0.96\;g$ DCW/l. At this time, the increase in chlorophyll concentration during 24 h after sulfur deprivation increased in proportion to hydrogen production, however, the decrease in starch concentration was not proportional to that. Therefore, hydrogen production under sulfur deprivation using green alga was closely associated with the residual PSII activity than the endogenic substrate degradation.
