Selection of suitable reference gene for gene expression studies of porcine ovaries under different conditions in quantitative reverse transcription polymerase chain reaction assay

  • The ovary undergoes substantial physiological changes along with estrus phase to mediate negative/positive feedback to the upstream reproductive tissues and to play a role in producing a fertilizable oocyte in the developing follicles. However, the disorder of estrus cycle in female can lead to diseases, such as cystic ovary which is directly associated with decline of overall reproductive performance. In gene expression studies of ovaries, quantitative reverse transcription polymerase chain reaction (qPCR) assay has been widely applied. During this assay, although normalization of target genes against reference genes (RGs) has been indispensably conducted, the expression of RGs is also variable in each experimental condition which can result in false conclusion. Because the understanding for stable RG in porcine ovaries was still limited, we attempted to assess the stability of RGs from the pool of ten commonly used RGs (18S, B2M, PPIA, RPL4, SDHA, ACTB, GAPDH, HPRT1, YWHAZ, and TBP) in the porcine ovaries under different estrus phase (follicular and luteal phase) and cystic condition, using stable RG-finding programs (geNorm, Normfinder, and BestKeeper). The significant (p < 0.01) differences in Ct values of RGs in the porcine ovaries under different conditions were identified. In assessing the stability of RGs, three programs comprehensively agreed that TBP and YWHAZ were suitable RGs to study porcine ovaries under different conditions but ACTB and GAPDH were inappropriate RGs in this experimental condition. We hope that these results contribute to plan the experiment design in the field of reproductive physiology in pigs as reference data.

A survey on the leggings wearing condition of down-aging women in their 30s and 40s (다운에이징 30-40대 여성의 레깅스 착용실태조사)

  • This study aims to investigate the actual condition of wearing leggings and the preferred design of down-aging women in their 30s and 40s who have young sensibility, but are starting to enter a middle-aged body type. By analyzing the problems experienced when wearing leggings in this age group, this study intends to provide basic data for the development of a middle-aged female leggings pattern with an improved fit. Therefore, a survey was conducted on 214 women in their 30s and 40s. The results of the survey are as follows. Among down-aging women who want to look younger, leggings are becoming an essential item, but most leggings brands are producing leggings with patterns suitable for women in their 20s. The results of this study also showed that down-aging women in their 30s and 40s mainly purchase and prefer body-fitting leggings. However, they are experiencing discomfort due to 'Y-zone accentuate', 'undergarment accentuate', 'cloth stucking', and 'waist band curling and tightening'. The results indicate that women in their 30s and 40s, who are starting to change to a middle-aged body type, have fit problems. Women in their 20s generally prefer functions that improve the body line, while those in their 30s and 40s prefer functions that improve movement and cover problems caused by the abdomen and the Y-zone area. Therefore, when designing leggings for down-aging women in their 30s and 40s, based on body-fitting leggings, a material with excellent elasticity must be selected. Also develop patterns that solves 'Y-zone accentuate', 'undergarment accentuate', 'cloth stucking', and 'waist band curling and tightening' problems.

Identification of Alkalophilic Bacillus sp. S-1013 Producing Non-Cariogenicity Sugar Fuc($1{\to}4$)gaINAc($2{\to}6$)NeuAc and Optimization of Culture Condition for Its Production (비우식성 당 Fuc($1{\to}4$)gaINAc($2{\to}6$)NeuAc를 생산하는 호알칼리성 Bacillus sp. S-1013의 동정 및 생산조건의 최적화)

  • The study was performed to identification of producing microbe Non-Cariogenicity Sugar (NCS; Fuc($1{\to}4$) gaINAc($2{\to}6$)NeuAc) with anti-caries activity, and to optimization of production condition. A typical strain which produced the NCS was identified alkalophilic Bacillus sp. S-1013 through the results of morphological, biochemical and chemotaxonomic characteristics and 16S rDNA sequencing. The optimal medium composition for the maximal production of the NCS from alkalophilic Bacillus sp. S-1013 was as follow: soluble starch 30 g, dextrin 15 g, yeast extract 5 g, peptone 10 g, $K_{2}HPO_4$ 2 g in a liter of distilled water. Optimal temperature and pH were 25 and 11.0, respectively. The highest production of NCS was shown 60 hrs cultivation using the optimal medium, and then NCS productivity and dry cell weight of culture broth increased 4.24 and 2.67 time than initial medium, respectively.

Modulation of Escherichia coli RNase E. Action by RraAS2, a Streptomyces coelicolor Ortholog of RraA (Streptomyces coelicolor의 RraA 동족체인 RraAS2에 의한 Escherichia coli RNase E 활성조절)

  • RraA is a recently discovered protein inhibitor that regulates the enzymatic activity of RNase E, which plays a major role in the decay and processing of RNAs in Escherichia coli. It has also been shown to regulate the activity of RNase ES, a functional Streptomyces coelicolor ortholog of RNase E, which has 36% identity to the amino-terminal region of RNase E. There are two open reading frames in S. coelicolor genome that can potentially encode proteins having more than 35.4% similarity to the amino acid sequence of RraA. DNA fragment encoding one of these RraA orthologs, designated as RraAS2 here, was amplified and cloned in to E. coli vector to test whether it has ability to regulate RNase E activity in E. coli cells. Co-expression of RraAS2 partially rescued E. coli cells over-producing RNase E from growth arrest, although not as efficiently as RraA, induced by the increased ribonucleolytic activity in the cells. The copy number of ColEl-type plasmid in these cells was also decreased by 14% compared to that in cells over-producing RNase E only, indicating the ability of RraAS2 to inhibit RNase E action on RNA I. We observed that the expression level of RraAS2 was lower than that of RraA by 4.2 folds under the same culture condition, suggesting that because of inefficient expression of RraAS2 in E. coli cells, co-expression of RraAS2 was not efficiently able to inhibit RNase E activity to the level for proper processing and decay of all RNA species that is required to restore normal cellular growth to the cells over-producing RNase E.

A study on Development Process of Fish Aquaculture in Japan - Case by Seabream Aquaculture - (일본 어류 양식업의 발전과정과 산지교체에 관한 연구 : 참돔양식업을 사례로)

  • When we think of fundamental problems of the aquaculture industry, there are several strict conditions, and consequently the aquaculture industry is forced to change. Fish aquaculture has a structural supply surplus in production, aggravation of fishing grounds, stagnant low price due to recent recession, and drastic change of distribution circumstances. It is requested for us to initiate discussion on such issue as “how fish aquaculture establishes its status in the coastal fishery\ulcorner, will fish aquaculture grow in the future\ulcorner, and if so “how it will be restructured\ulcorner” The above issues can be observed in the mariculture of yellow tail, sea scallop and eel. But there have not been studied concerning seabream even though the production is over 30% of the total production of fish aquaculture in resent and it occupied an important status in the fish aquaculture. The objectives of this study is to forecast the future movement of sea bream aquaculture. The first goal of the study is to contribute to managerial and economic studies on the aquaculture industry. The second goal is to identify the factors influencing the competition between production areas and to identify the mechanisms involved. This study will examine the competitive power in individual producing area, its behavior, and its compulsory factors based on case study. Producing areas will be categorized according to following parameters : distance to market and availability of transportation, natural environment, the time of formation of producing areas (leaderㆍfollower), major production items, scale of business and producing areas, degree of organization in production and sales. As a factor in shaping the production area of sea bream aquaculture, natural conditions especially the water temperature is very important. Sea bream shows more active feeding and faster growth in areas located where the water temperature does not go below 13∼14$^{\circ}C$ during the winter. Also fish aquaculture is constrained by the transporting distance. Aquacultured yellowtail is a mass-produced and a mass-distributed item. It is sold a unit of cage and transported by ship. On the other hand, sea bream is sold in small amount in markets and transported by truck; so, the transportation cost is higher than yellow tail. Aquacultured sea bream has different product characteristics due to transport distance. We need to study live fish and fresh fish markets separately. Live fish was the original product form of aquacultured sea bream. Transportation of live fish has more constraints than the transportation of fresh fish. Death rate and distance are highly correlated. In addition, loading capacity of live fish is less than fresh fish. In the case of a 10 ton truck, live fish can only be loaded up to 1.5 tons. But, fresh fish which can be placed in a box can be loaded up to 5 to 6 tons. Because of this characteristics, live fish requires closer location to consumption area than fresh fish. In the consumption markets, the size of fresh fish is mainly 0.8 to 2kg.Live fish usually goes through auction, and quality is graded. Main purchaser comes from many small-sized restaurants, so a relatively small farmer and distributer can sell it. Aquacultured sea bream has been transacted as a fresh fish in GMS ,since 1993 when the price plummeted. Economies of scale works in case of fresh fish. The characteristics of fresh fish is as follows : As a large scale demander, General Merchandise Stores are the main purchasers of sea bream and the size of the fish is around 1.3kg. It mainly goes through negotiation. Aquacultured sea bream has been established as a representative food in General Merchandise Stores. GMS require stable and mass supply, consistent size, and low price. And Distribution of fresh fish is undertook by the large scale distributers, which can satisfy requirements of GMS. The market share in Tokyo Central Wholesale Market shows Mie Pref. is dominating in live fish. And Ehime Pref. is dominating in fresh fish. Ehime Pref. showed remarkable growth in 1990s. At present, the dealings of live fish is decreasing. However, the dealings of fresh fish is increasing in Tokyo Central Wholesale Market. The price of live fish is decreasing more than one of fresh fish. Even though Ehime Pref. has an ideal natural environment for sea bream aquaculture, its entry into sea bream aquaculture was late, because it was located at a further distance to consumers than the competing producing areas. However, Ehime Pref. became the number one producing areas through the sales of fresh fish in the 1990s. The production volume is almost 3 times the production volume of Mie Pref. which is the number two production area. More conversion from yellow tail aquaculture to sea bream aquaculture is taking place in Ehime Pref., because Kagosima Pref. has a better natural environment for yellow tail aquaculture. Transportation is worse than Mie Pref., but this region as a far-flung producing area makes up by increasing the business scale. Ehime Pref. increases the market share for fresh fish by creating demand from GMS. Ehime Pref. has developed market strategies such as a quick return at a small profit, a stable and mass supply and standardization in size. Ehime Pref. increases the market power by the capital of a large scale commission agent. Secondly Mie Pref. is close to markets and composed of small scale farmers. Mie Pref. switched to sea bream aquaculture early, because of the price decrease in aquacultured yellou tail and natural environmental problems. Mie Pref. had not changed until 1993 when the price of the sea bream plummeted. Because it had better natural environment and transportation. Mie Pref. has a suitable water temperature range required for sea bream aquaculture. However, the price of live sea bream continued to decline due to excessive production and economic recession. As a consequence, small scale farmers are faced with a market price below the average production cost in 1993. In such kind of situation, the small-sized and inefficient manager in Mie Pref. was obliged to withdraw from sea bream aquaculture. Kumamoto Pref. is located further from market sites and has an unsuitable nature environmental condition required for sea bream aquaculture. Although Kumamoto Pref. is trying to convert to the puffer fish aquaculture which requires different rearing techniques, aquaculture technique for puffer fish is not established yet.

Two-step Acid Hydrolysis Method for Producing Fermentable Sugar from Lignocellulosic Biomass (발효당 생산을 위한 목질계 바이오매스의 2단 산당화)

  • For obtain fermentable sugar, we conducted acid hydrolysis with lignocellulosic biomass without enzyme. The lignocellulosic biomass used pinus rigida and Palm residues (EFB; empty fruit bunches). In the acid hydrolysis, we consider the hydrolysis condition to reduce a denatured sugar. So this study was conducted 2-step acid hydrolysis. First-step hydrolysis used high concentration (72 wt%) sulfuric acid at $80^{\circ}C$. At the condition, we obtained 11.49 wt%, 32 wt% glucose conversion for pinus rigida and EFB. After the step, the liquor was dilute until 9~15 wt% acid concentration and conducted second hydrolysis at $50{\sim}120^{\circ}C$. In the second hydrolysis, we obtained maximum glucose conversion (pinus rigida 86.8 wt% (39 g/L) and EFB 95.3 wt% (32.4 g/L)) at 9 wt% acid concentration and $120^{\circ}C$ for 80 min. All samples through the process are analyzed on the basis of mass balance.

Epimerization of L-Arabinose for Producing L-Ribose (L-리보스 생산을 위한 L-아라비노스의 에피머반응)

  • L-ribose has recently attracted interest as a starting material for antiviral drug. It could be obtained from L-arabinose by epimerization reaction. Epimerization reaction was carried out with molybdenium oxide or molybdic acid catalyst and methanol/water solution. Reaction temperature, methanol percentage, and catalyst kind were selected to find an optimum reaction condition. Ion exhange chromatography was used for separating epimerization reaction mixture, and then HPLC chromatogram of L-ribose fraction obtained to calculate the yield of the reaction. Shodex ion exchange HPLC column(Model SC1011) and Phenomenex Luna $NH_2$ HPLC column were compared to employ a convenient HPLC analysis. It was found that the usage of 20% methanol, $60^{\circ}C$, and 40 g/L molybdic acid gives the best reaction condition with a yield of 21%.

Study on the Contrasting Aspects Between Sasang Constitutional Medicine and Traditional Oriental Medicine (사상의학(四象醫學)과 증치의학(證治醫學)의 비교(比較) 연구(硏究))

  • This study was performed to reveal some contrasting aspects between Sasang Constitutional Medicine(四象醫學) and Traditional Oriental Medicine(傳統 韓醫學). This aims ultimately for the unified standardization of Korea-Oriental Medicine. Suggesting the clear contrasting aspects of thse two medical system, this study might give several basic informations for ultimate achievement. As a result, some contrasting aspects were revealed as follows. 1. The interpretation of the man's status is contrasted. In Sasang Constitutional Medicine, man is interpreted as the member of the Society(天下) while in Traditional Oriental Medicine as the part of the Nature(天地). 2. The man's modeling is contrasted. In Sasang Constitutional Medicine, man is in the process of becoming as differentiating itself by the inner emotional producing. While in Traditional Oriental Medicine, man is generalized as one model according to the Nature, one great universe(天地). 3. The pathogenesis is contrasted. In Sasang Constitutional Medicine, emotional inclination is the main factor provoking the primary pathogenesis. While in Traditional Oriental Medicine the external isolated pathogenic factors such as Cold, Heat, Warm etc. are significant for the pathogenesis. 4. The evaluation of the disease is contrasted. In Sasang Constitutional Medicine all the diseases are evaluated according to the inherent condition of patient himself. But in Traditional Oriental Medicine the evaluation of disease isolated from the inheret condition of patient himself is generalized in its own system. 5. The prime method of healing, controlling, and maintaining health of the patient is contrasted. For these, the patient's spontaneous effort for adjusting his emotional inclination is important in Sasang Constitutional Medicine. While in Traditional Oriental Medicine, the external adjustment dependent on the herbal medication is the main method for healing, controlling, and maintaining health of the patient.

Effect of Pre-cooking Conditions on the Quality Characteristics of Ready-To-Eat Samgyetang

  • The aim of this study was to examine the effectiveness of pre-cooking conditions on the quality characteristics of ready-to-eat (RTE) Samgyetang. Raw chickens were steamed under the different conditions of 50℃/30 min (T1), 65℃/30 min (T2), 85℃/30 min (T3), and 90℃/10 min (T4) prior to retorting at 120℃ for 65 min. The results showed that pre-cooking conditions in all treated samples could reduce fat contents in breast and leg meats by 8.5-11.7% and 10.0-11.0% compared to the control, even though there were no significant differences among treatments (p>0.05). The L* and b* values of breast and leg meats treated with the higher temperature and longer time conditions were significantly higher than the control (p<0.05), while a* values tended to decrease despite of not to a significant extent (p>0.05). Moreover, apparent viscosity and water soluble protein showed insignificant differences (p>0.05) among the samples as a result of the retorting process, which might have more negative influences on the quality. T2 samples obtained significantly the highest average Quantitative Descriptive Analysis (QDA) score and transmittance value, representing the most clear broth among the samples, compared to the control. On the other hand, T3 showed the highest cooking loss among the treatments and the lowest QDA scores among the samples. In conclusion, pre-cooking treatment prior to retorting in manufacturing Samgyetang is a plausible way to reduce its fat content. A pre-cooking condition at either 65℃ for 30 min, or 90℃ for 10 min are recommended for producing Samgyetang with optimum quality.

  • The conception of uniform is changing from function and economical efficiency of the past to symbolism and aesthetic sense So, in such a management strategical point of view as enterprises' PR, scouting talent, organization revitalization etc, this is required to settle uniform culture which is proper to each enterprise and design development. The purpose of this thesis is, first, to study to help distribution enterprises' CI or PR by examining the present condition of uniform design in domestic fashion distribution enterprise and planning uniform design on the base of real case and finally making a real thing in person. Second is to suggest to fashion enterprise security of competitive ability of uniform-producing enterprise by developing article design of worth. Third is to present academic area with how to do design education in a wide range not based on trend article development. Following is how and what to study to carry out these purposes. Details about necessity and how and what to study are in an introduction, the conception of uniform in theory through precedential records examination in chapter 2, the present condition of uniform design based on Changwon city in chapter 3, planning uniform design and making a real thing suitable for F enterprise in chapter 4, and the conclusion is in chapter 5. Through this study, I became to know that uniform in distribution enterprise who treats visitors face to face is very important. I supply material and information concluded from this study to uniform industry and hope this will be helpful in developing uniform. It is significant in that this study as being related with enlarged uniform and enterprise, beyond such a uniform area as school uniform and hospital one, expanded the study range of clothing. I already tell you that this study is based on visual expression of uniform and wearer's feeling in clothes is not included.
