고삼투압이 재조합 Erythropoietin의 생산과 당쇄구조에 미치는 효과

  • Effect of hyperosmotic pressure on growth of recombinant Chinese hamster 。 vary cells and Erythropoietin (EPO) production was investigated. Cells were cultivated in batch modes at various osmolalities. When the osmolality increased from 314 to 463mOsm/Kg, specific EPO productivity (qp) was increased up to 1.6-fold but cell growth was inhibited. EPO has a complex oligosaccharide structure that plays an important role in biological activity in vivo. To investigate the influence of hypoerosmotic pressure on the glycosylation, structural analysis of oligosaccharide was calTied out. Recombinant human EPO was produced by CHO cells grown under various osmotic pressure and purified from culture supernatants by heparin-sepharose affinity column and immunoaffinity column. N-linked oligosaccharides were released enzymatically and isolated by paper chromatography. The isolated oligosaccharides were labeled with fluorescent dye, 2-aminobenzamide and analyzed with MonoQ anion exchange chromatography and GlycosepN amide chromatography for the assignment of GU (glucose unit) value. Glycan analysis by HPLC showed that neutral (asialo) oligosaccharide was increased slightly with an increase in osmolality. In portion of sialylated glycan, total relative amount of mono- and di-sialyated glycan was increased but that of tri- and tetra-sialylated glycan decreased as osmolality was increased.

A study on sound radiation from isotropic plates stiffened by unsymmetrical beams (비대칭 보에 의해 보강된 등방성 평판의 음향방상에 관한 연구)

  • The determination of sound pressure radiated from periodic plate structures is fundamental in the estimation of noise level in aircraft fuselages or ship hull structures. As a robust approach to this problem, here a very general and comprehensive analytical model is developed for predicting the sound radiated by a vibrating plate stiffened by periodically spaced orthogonal unsymmetrical beams subjected to a sinusoidally time varying point load. The plate is assumed to be infinite in extent, and the beams are considered to exert both line force and moment reactions on it. Using this theoretical model, the sound pressure levels on axis in a semi-infinited fluid (water) bounded by the plate were calculated using three numerical tools such as the Gauss-Jordan method, the LU decomposition method and the IMSL numberial package. Especially, the variation in the sound pressure levels and their modes were investigated according to the change in frequency, bay spacing and bay distance.

Design of a Dynamic Absorber for the Large-Size Pressure Vessel of the Petrochemical Plant (석유화학 플랜트의 대형 압력용기에 대한 동흡진기의 설계)

  • In this work, two dynamic absorbers are introduced and designed to reduce the vibration of the large-size pressure vessel of a reactor for a petrochemical plant. The vibration modes and harmonic responses of the vessel are firstly analyzed by the finite element method. On the basis of the analyzed results, two dynamic absorbers are designed by a simple design theory. Furthermore, an optimization process is executed and an optimal design of the dynamic absorber is obtained to improve performance and structural safety of the vessel. As a result, the maximum displacement and stress of the vessel is decreased about 85% and 65% respectively, the design criteria being satisfied.

Effect of Bend Angle on the Behavior of pipe Bend under Internal Pressure and In-Plane Bending toads (내압과 내면 굽힘하중 조건에서 곡관의 거동에 미치는 굽힘각의 영향)

  • This study performed finite element analysis on the pipe bend with various bend angles under loading conditions of internal pressure and combined pressure and bending, to investigate the effect of bend angle on the collapse behavior of pipe bend and on the stress state in the bend region. In the analysis, the pipe bends with bend angle of $5\~90^{\circ}$ were considered, and the bending moment was applied as in-plane closing and opening modes. From the results of analysis, it was found that the collapse moment of pipe bend increases with decreasing bend angle. As the bend angle decreases, also, the equivalent stress at intrados region increases regardless of bending mode. Under closing mode bending especially, the increase in stress at intrados is significant so that the maximum stress region moves from crown to intrados with decreasing bend angle.

  • Many research of elastohydrodynamic lubrication (EHL) has been performed under the condition of steady state loading. However, mechanical elements undergo severe high loads that are in the fluctuating modes of frequency and amplitude. Conventional numerical method for the circular contact of EHL study has the difficulty in making the film thickness and pressure of EHL converged in high loads of steady state as well as fluctuating loading conditions. In this work, multigrid multilevel method are used for the stable convergence of film pressure and thickness under the conditions of high as well as varying loads, and very stable solutions of film behaviors with elastic deformation are obtained. Several results of dynamic loading condition are shown and compared with those of steady state condition in the aspects of circular EHL film thickness and pressure.

Buckling characteristics of multiwalled carbon nanotubes under external pressure

  • This article describes recent work on mechanics of carbon nanotubes, one of the most fundamental and amazing man-made nanostructures. The noteworthy point is that "nano"-scale mechanics of carbon nanotubes can be well described by the continuum elastic theories for "macro"-scale thin shells. This provides an efficient means to elucidate mechanical deformation effects of carbon nanotubes on their physical and chemical properties, which is significant to develop new-generation nanomaterials based on nanotubes and their composites. Potential applications of the mechanical deformation of nanotubes in nano-electronics and nano-biology are also commented. In addition, theoretical investigations regarding external pressure buckling is carried out here and we have numerically confirmed that larger N (the number of layers) and a smaller D (the innermost diameter) make "corrugation modes" with a larger mode-index k be energetically favored.

  • The purpose of this research is to study the vibration and acoustic pressure radiation from a thin isotropic flat plate stiffened by a rectangular array of beams, and excited by a time harmonic point force. These constructions on aircraft and ship structures are often subjected to fiequency dependent pressure fluctuations and forces. Forces from the these excitations induce structural vibrations in a wide range of fiequencies, which may cause such things as acoustic fatigue and internal cabin noise in the aircraft. It is thus important that the response characteristics and vibration modes of such periodic structures be horn. From this theoretical model, the sound pressure levels(SPL) in a semi-infinite fluid(water) bounded by the plate with the variation in the locations of an external time harmonic point farce on the plate can be calculated efficiently using three numerical tools such as the Gauss-jordan method the LU decomposition method md the IMSL numerical package.

  • A specific experimental method, the Split Hopkinson Pressure Bar (SHPB) technique has been widely used to determine the dynamic material properties under the impact compressive loading conditions with strain-rate of the order of 103/s∼104/s. This type of test procedure has been used to examine the dynamic response of materials in various modes of testing. In this paper, dynamic deformation behaviors of rubber materials widely used for the isolation of vibration from varying structures under dynamic loading are determined using a Split Hopkinson Pressure Bar technique.

Critical Heat Flux under Forced and Natural Circulations of Water at Low-Pressure, Low-Flow Conditions

  • The CHF phenomenon has been investigated for water flow under forced and natural circulation modes with vertical round tubes at low pressure and low flow condition. Experiments have been performed by using three different test sections for mass fluxes below 400 kg/㎡s under near atmospheric pressure. The experimental data for forced and natural circulation are compared with each other. To predict the flow rate at the two-phase region our test condition has been analyzed by RELAP5/MOD3 because the local two-phase condition inside the stainless steel tube cannot be directly measured. To predict the CHF with accuracy we have to consider the parameters at the single-phase region as well as the flow behavior at the two-phase region.

  • If a wall separates the bounded and unbounded spaces, then the wall's role in transporting the acoustic characteristics of the two spaces is not well defined. In this paper, we attempted to see how the acoustic characteristical of two spaces are really affected by the spatial characteristics of the wall. In order to understand coupling mechanism, we choose a finite space and a semi-infinite space separated by the flexible or rigid wall and an opening. A volume interaction can be occurred in structure boundary and a pressure interaction can be happened in the opening boundary. For its simplicity, without loosing generality, we use rather simplified rectangle model instead of generally shaped model. The source impedance is presented to the various types of boundaries. The distributions of pressure and active intensity are also presented at the cavity and structure-dominated modes. The resulting modification, shifts of mode1 frequencies and changing of standing wave patterns to satisfy both coupled boundary conditions and governing equations, are presented.
