Purification and Characterization of Sulfated Polysaccharide Isolated from Hot Water Extract of Pachymeniopsis elliptica (Pachymeniopsis elliptica의 열수 추출물로부터 분리한 함황 다당류의 정제 및 특성)

  • In the preliminary study, we investigated the anti-complementary activities of 62 extracts from Korean edible seaweeds. Of those, Pachymeniopsis elliptica showed the highest anti-complementary activity. Therefore, it was purified as follows; i) PE-1 by ethanol precipitation, ii) PE-1-C by ultrafiltration, iii) PE-1-CIV by DEAE-Toyopearl 650C, and iv) PE-1-CIV-ii by Sepharose CL-6B. The purified compound, PE-1-CIV-ii, was the complexed homogeneous polysaccharide (molecular mass: 780 kDa) with 82.9% of anti-complementary activity. Also, it contained a significant amount of sulfate group (30.5%), which indicated it as a sulfated algal polysaccharide. Its structural monosaccharides were galactose (44.3%), 3,6-anhydrogalactose (34.0%), glucose (8.2%), fucose (5.4%), xylose (5.2%) and rhamnose (2.9%). After the treatment of periodate on a sample, a significant decrease in anti-complementary activity was found, which was a characteristic of bioactive polysaccharides. And-tumor activity of PE-1-A, B and C was tested in the sarcoma-180 solid tumor model. The PE-1-C with the largest molecular mass (more than 300 kDa) showed 81% of inhibition on the solid tumors, suggesting that the anti-complementary activity was, at least in part, related to anti-tumor activity. Based upon these results, the purified polysacchardes could be an immunopotentiator in vivo.

Legibility Evaluation of Two and Three Syllable Words Used in Pesticides According to Font, Thickness, Gender, and Visual Acuity (시력, 폰트, 굵기, 성별에 따른 2음절 및 3음절 농약 제품 표시글자의 가독성 평가)

  • Safety and health related information for the proper use and handing of pesticides is usually printed on the surface of the pesticide products in the form of texts. But, the guidelines or standards for the appropriate presentation of the texts for the pesticide products are most vague or not practical. Thus, this study aimed to provide the preliminary guidelines for the text sizes based on the legibility experiments. To achieve the objective legibility evaluation experiments were conducted to test the effects of different near vision (0.6, ${\geq}0.8$), gender, font type(thick gothic-type and fine gothic-type), thickness of font(plain and bold), and number of syllables(2 and 3 syllables) in the same age group of 20s. The results showed that legibility was different according to the visual acuity (p<0.05), and no other main effects showed statistically significant effects. The 'maximum illegible size' to read at least one word correctly in all the text conditions was 2 pt when the near vision was ${\geq}0.8$, and 2 pt or 3 pt when the near vision was 0.6. The 'minimum legible size' for 100% correct answer was 9 pt for the near vision of 0.6, and 5.3 pt for ${\geq}0.8$, respectively. Mean character size does not read any discomfort in 0.6 was 15.5 pt in both male and female but male was mean 8.5 pt, female was 10 pt in ${\geq}0.8$. Considering these experimental results, it was recommended that the 16 pt or larger characters should be used the important information such as 'Pesticides' or toxicity, and the minimum character size was 9 pt for the less important information.

Arthroscopic Subscapularis Tendon Repair - Preliminary Report of 8 cases study - (관절경하 견갑하건 봉합술 - 8 례에 대한 예비보고 -)

  • Purpose: To validate and introduce the technique of the arthroscopic repair for the treatment of the Subscapulris tendon tear. Materials and Methods: From April 2003 to April 2004, Arthroscopic repairs were performed in 8 patient with subscapularis tendon tear. The mean follow-up period was 11months. two cases involved only subscaplaris tendon tear. Four cases were associated small sized posterosuperir. rotator cuff tear and two cases were in large size. The type of subscapularis tendon showed upper portion full-thickness tear in 6 cases, partial-thickness tear localized at articular surface in 1 case, complete tear in 1 case. The results were analyzed by using subjective satisfaction, inferior movenent of superior displaced humeral head, Constant-Murley functional scoring Results: Constant-Murley scoring was improved from 55 point preoperatively to 75 point postoperatively following 11 months. Mean score was 71 point except 2 cases of large superoposterior rotator cuff tear, Most humeral heads were distaracted postoperatively. The subjective result for the 8 shoulders were very satisfying in 5 cases, satisfying in 1 case and dissatisfying in 2 cases. Therefore satisfactory results were noted in 6 cases (75%) of this overall treatment group. Conclusion: The arthroscopic repair for the subscapularis tendon tear is thought to be available method, which could reduce severe complications following the weakness of deltoid muscle and postoperative pain. Especially Partial Subscapularis tear not associated with superoposterior rotator cuff tear including PASTA lesion was the sutable indication of the Arthroscopic repair.

  • The habitat typing method of classification based on community has been developed on many forested areas in the ROCKY MOUNTAINS of WESTERN NORTH AMERICA, and is being used in providing an objective principle of guide for classification of forest sites, land classification, vertical distribution and multipurpose management of forest. I actually made a trial of forest habitat type classification - The methodology can be segmented into a number of tasks including reconnaissance, selection of stands to be sampled, location of plots, data gathering, preliminary and computer analysis, preparation of key- and map of habitat type and description for each habitat type in a view of forest management - on UNIVERSITY OF MONTANA FORESTS in order to examine whether its application is fit or not in korean forest circumstances : these are composed mainly of artificial forest and forest in the early stage of succession. The main results obtained from this study were summarized as follows 1. A hierarchical classification-Climax, Habitat type and Phases-of forest sites was developed using the habitat typing method of classification. 2. A diagnostic key is provided for field identification of the types based upon presence and number of indicator species, and forest managers could prepare a map of habitat type and classify forest habirat types by reference to it. 3. The classified habitat types were considered to be effective on providing an objective principle of guide for multipurpose management of forest including potential productivity for timber, feed productivity of forage and wildlife, selection of the right species in the right sites, application of method of treatment and so forth. 4. Forest habitat type classifications based upon extrapolation which used relict species and associated stands is considered to be suitable for korean forest circumstances and objective in studying potential vegetation, vertical and horizontal distribution of forest.

Development and Preliminary Evaluation of a Leukocyte Removal Aptamer Filter (압타머를 이용한 백혈구제거필터의 개발 및 예비평가)

  • Background: Leukocyte reduction filters are widely used to prevent transfusion reactions caused by leukocytes in blood components. Commercial filters are not sufficient for removal of leukocytes for prevention of transfusion associated graft-versus-host disease; therefore, irradiation of blood components was performed using expensive equipment. Techniques using an aptamer substituted for antibody have been developed and are available in clinical areas. The purpose of this study is to develop the aptamer filter system and to evaluate its efficiency and the possibility of its clinical application. Methods: Aptamers targeted to CD45 were selected by the Postech Aptamer Initiative. The aptamer filter in which aptamers attached to beads were bound to leukocytes and removed by magnetic field was developed. Filtration of 14 units of leukoreduction-red blood provided by Korean Red Cross Blood Services was performed using aptamer filters. Leukocyte removal rate and red cell recovery rate were evaluated and bacterial culture was performed. Results: After filtration using the aptamer filters, 45.6% of leukocytes were additionally removed and the red cell recovery rate was 92.8%. No growth in the bacterial culture was observed. Conclusion: In order to apply the cell depletion technique utilizing an aptamer to blood filter system, we developed and evaluated the aptamer filter system. Through improvement of the binding efficiency of the aptamer and the filtering process, and application of the various aptamers for other different cells, we suggest that this technique can be applied in the clinical area, such as a substitution for the irradiation process for TAGVHD prevention.

Application of Potassium Feldspar pIR-IRSL Method to Dating Quaternary Marine and Fluvial Terrace Sediments in Korea: A Case Study on a Fluvial Terrace and Gusan Fault in Uljin, Korea (한반도 해안-하안단구 퇴적층에 대한 K 장석 pIR-IRSL연대측정법 적용가능성 고찰: 울진 하안단구와 구산단층 연대측정)

  • In this paper, in order to test the possibility of applying K feldspar $pIR-IRSL_{290}$ signal(read out at $290^{\circ}C$) to date old terrace sediments(up to ~ 200 ka, MIS 7) in Korea, we investigated luminescence properties of $pIR-IRSL_{290}$ signals in K feldspar extracts from 27 marine and fluvial terrace sediment samples, and these were compared with those of quartz OSL and conventional K feldspar $IRSL_{50}$ (readout at $50^{\circ}C$) signals. The averaged $2D_0$ value of K feldspar $pIR-IRSL_{290}$ growth curves was ~ 700 Gy, which is consistent with that of $IRSL_{50}$ signal, and this is 3 times higher than that for quartz OSL (~ 250 Gy) on average. Where possible, K feldspar $pIR-IRSL_{290}$ ages were compared with quartz OSL and conventional $IRSL_{50}$ ages. Our preliminary K feldspar $pIR-IRSL_{290}$ ages were older than quartz OSL ages by about 200%, while fading rate-corrected conventional $IRSL_{50}$ ages are in good agreement with those based on quartz OSL. This seems to indicate the possibility of K-feldspar $pIR-IRSL_{290}$ age overestimation due to the presence of unbleachable $pIR-IRSL_{290}$ signals, even with a prolonged exposure to sunlight. Both quartz OSL and K-feldspar $pIR-IRSL_{290}$ signals for the samples from Noeum fluvial terrace and Gusan fault site were all in dose saturation level, thus unable to estimate the formation ages of the sediments. However, $2D_0$ values derived from the dose response growth curves strongly indicate that the Noeum fluvial terrace sediments have formed before 109-140 ka, while the fluvial sediments from Gusan fault were desposited before 100-105 ka. Further, this seems to suggest that the previous quartz OSL ages of ~40-50 ka for Gusan fault sediments should be the underestimated ones due to dose saturation problem.

Antioxidant and Anti-inflammatory Effects of Ficus erecta var. sieboldii Leaf Extract in Murine Macrophage RAW 264.7 Cells (좁은잎천선과나무(Ficus erecta var. sieboldii) 잎 추출물이 대식세포 RAW 264.7 세포에서 미치는 항산화 및 항염증 효과)

  • In this study, a preliminary evaluation of the antioxidant and anti-inflammatory activity of the Ficus erecta var. sieboldii (Miq.) King (FES) leaf extract has been performed to assess its potential as a natural resource for food and medicinal materials. FES was extracted using 70% EtOH and then fractionated sequentially using n-hexane, $CH_2Cl_2$, EtOAc, and n-BuOH. To screen for antioxidant and anti-inflammatory agents effectively, the inhibitory effect of the FES extracts on the production of oxidant stresses (DPPH, xanthine oxidase, and superoxide) and pro-inflammatory factors (NO, iNOS, COX-2, $PGE_2$, IL-6, and $IL-1{\beta}$) in the murine macrophage cell line RAW 264.7 activated with lipopolysaccharide (LPS) was examined. Among the sequential solvent fractions of FES, the $CH_2Cl_2$ and EtOAc fractions showed decreased production of oxidant stresses (DPPH, xanthine oxidase and superoxide), and the hexane and $CH_2Cl_2$ fractions of FES inhibited the production of pro-inflammatory factors (NO, iNOS, COX-2, and $PGE_2$). The $CH_2Cl_2$ fraction also inhibited the production of pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$). These results suggest that FES has a significant effects on the production of oxidant stresses and pro-inflammatory factors and may be used a natural resource for antioxidant and anti-inflammatory agents.

A Preliminary research on Sixth-century Wooden Buddha Images from Funan (6세기 푸난 목조불상에 대한 시론(試論))

  • This paper is an introductory study of the wooden Buddha images excavated in the Mekong Delta in present-day southern Vietnam. Approximately thirty examples of wooden Buddha images have been recovered in the Mekong Delta. As they were found among the major historic sites of Funan (the first kingdom in Southeast Asia), including Oc Eo, and likely date to the sixth century, they can be regarded as the works of Funan. Similarly to stone Buddha images, these wooden examples are considered important for their demonstration of aspects of early Buddhist sculpture in Southeast Asia. Most of these Funan wooden Buddha images are presumed to have been produced around the sixth century based on radiocarbon dating and their stylistic characteristics. This paper analyzed the iconography and style of eight examples whose forms are relatively recognizable. The eight images can be divided into four types according to their postures, dress styles, and hand gestures. They all share features of the Sarnath style of the Gupta period in India as seen in their slender bodies and the outer robe that clings to the body. However, some display a South Indian style of Buddha images in their upright posture, style of wearing a monastic robe with the right shoulder exposed, and making the vitarka mudra (preaching gesture) that is often seen in Sri Lankan Buddha images. This suggests that Buddhist sculptors in Funan devised a new style by incorporating the principal styles of Indian Buddhist sculpture. Another notable feature of these Funan images is their material, namely, wood. While none of the contemporaneous Hindu sculptures discovered from the same historic sites are made of wood, numerous Buddhist sculptures are. This paper postulates that the use of wood in Buddhist sculptures was promoted for reasons beyond ease of carving. According to the Buddhist literature, the first-ever Buddha image was the so-called 'Udayana image' made out of sandalwood. This image and its story was well-known in the Southern Dynasty of China around the sixth century. Interestingly, some auspicious sandalwood images of the Buddha was believed to have been brought into the Southern Dynasty from Funan. This suggests the possibility that the legend of the Udayana image might have been known in Funan as well and resulted in the production of wooden sculptures there.

Validation of Korean Diagnostic Scale of Multiple Intelligence (한국형 다중지능 진단도구의 타당화)

  • The purpose of this study is to develop and verify a Korean Diagnostic Scale of Multiple Intelligence(MI), which will be an alternative test to avoid problems with former Shearer's MI test and to adopt H. Gardner's suggestions to develop MI assessment. The test is developed 5 types; kindergartner, elementary lower grader, elementary upper grader, middle schooler, high schooler test. A form of test is diversified with 3 types; multiple-choice items for accomplishment, true or false items for ability, and self-reported items with likert scale for interest and ability. According to H. Gardner's suggestions, we have tried to reanalyze key component of MI, analyze an overlapping or hierarchical relationship between intelligences, develop intelligences-fair items, diversify form of item. We have developed a final standardized test through a primary, secondary preliminary-test analysis, and sampled 5,585 students by age, gender, and regional groups. As a result of this sampling test, we can get a norm score and compare individuals with other's score relatively. To verify this test, we analyzed behavior observation, mean, standard deviation, a percentage of correct answers, reliability of each test type, correlation between intelligence scales, Kruskal-Wallis test of mean rank of career choice by intelligences. As a result of correlation analysis between sub-intelligence scales, we can conclude that this MI test is satisfied with intelligence independent assumption. Besides, as non-parametric statistics test(Kruskal-Wallis) of career choice by intelligences, we can identify that MI is related with domain of career choice. This test is not a linguistic and logical-mathematical biased test but a intelligences-fair test. It makes us compare individual's potential with a norm score. Besides, it could be useful as a means of educational prescription or counsel in comparison with ability, interest, and accomplishment of individual. But this test is limited to do factor or correlation analysis between types of sub-test, because items are minimized for a time-constraint and a heavy burden of test receiver. But if it could be tested with increased items by two sessions, further research could be expected to get over this constraints and do a further validation analysis.

A preliminary numerical analysis on the behaviour of tunnel under construction in fracture zone considering seismic load (지진 하중을 고려한 단층파쇄대에서의 시공 중 터널 거동 분석에 관한 수치해석적 연구)

  • Recently occurred earthquake Gyeongju and Pohang served as a momentum to remind that Korean peninsular is not a safety zone from earthquake anymore. The importance of seismic design, therefore, have been realized and researches regarding design response spectrum have been actively carried out by many researchers and engineers. Current tunnel seismic design method is conducted to check safety of tunnel structure by dynamic numerical analysis with condition of completed lining installation, so, it is impossible to consider safety of tunnel behavior under construction. In this study, therefore, dynamic numerical analysis considering seismic wave propagations has been performed after back analysis using results from field monitoring of tunnel under construction in fractured zone and 1st reinforcement (shotcrete, rockbolt) behaviour are analyzed. Waves are classified by period characteristic (short and long). As a result, the difference depending on period characteristic is minor, and increasements of displacement are obtained at crown displacement due to seismic wave is 28~31%, 14~16% at left side of tunnel in the fractured zone, 13~27% at right side of tunnel in the bed rock, respectively. In case of shotcrete axial force is increased 113~115% at tunnel crown, 102% at left side, 106~110% at right side, respectively. Displacement and axial force of rockbolts which are selected by type of anchored grounds (only fractured zone, fractured zone and bed rock, only bedrock) are analyzed, as a result, rockbolt which is anchored to fractured zone and bed rock at the same time are weaker than any other case.
