Relationship between Poor Immunogenicity of HLA-A2-Restricted Peptide Epitopes and Paucity of Naïve $CD8^+$ T-Cell Precursors in HLA-A2-Transgenic Mice

  • We examined the immunogenicity of H-2 class I-restricted and HLA-A2-restricted epitopes through peptide immunization of HLA-A2-transgenic mice that also express mouse H-2 class I molecules. All four of the tested epitopes restricted by H-2 class I robustly elicited T-cell responses, but four of seven epitopes restricted by HLA-A2 did not induce T-cell responses, showing that HLA-A2-restricted peptide epitopes tend to be poorly immunogenic in HLA-A2-transgenic mice. This finding was confirmed in HLA-A2-transgenic mice infected with a recombinant vaccinia virus expressing hepatitis C virus proteins. We examined the precursor frequency of epitope-specific naïve $CD8^+$ T cells in HLA-A2-transgenic and conventional C57BL/6 mice and found that the poor immunogenicity of HLA-A2-restricted peptide epitopes is related to the paucity of naïve $CD8^+$ T-cell precursors in HLA-A2-transgenic mice. These results provide direction for the improvement of mouse models to study epitope repertoires and the immunodominance of human T-cell responses.

Improved Immobilized Enzyme Systems Using Spherical Micro Silica Sol-Gel Enzyme Beads

  • Spherical micro silica sol-gel immobilized enzyme beads were prepared in an emulsion system using cyclohexanone and Triton-X 114. The beads were used for the in situ immobilization of transaminase, trypsin, and lipase. Immobilization during the sol to gel phase transition was investigated to determine the effect of the emulsifying solvents, surfactants, and mixing process on the formation of spherical micro sol-gel enzyme beads and their catalytic activity. The different combinations of sol-gel precursors affected both activity and the stability of the enzymes, which suggests that each enzyme has a unique preference for the silica gel matrix dependent upon the characteristics of the precursors. The resulting enzyme-entrapped micronsized beads were characterized and utilized for several enzyme reaction cycles. These results indicated improved stability compared to the conventional crushed form silica sol-gel immobilized enzyme systems.

Effects of Precursors and Exogenous Taxanes on Taxane Production by Cell Suspension Cultures (전구체 및 외부공급 Taxane이 세포배양에 의한 Taxane 생산에 미치는 영향)

  • Effects of three kinds of precursors as well as exogenous taxanes on the production of taxol and its derivatives were investigated in Taxus cuspidata and Taxus brevifolia cell suspension cultures. When geraniol was added as a precursor of geranylgeranyl pyrophosphate to enhance diterpenoid metabolism, production of some taxanes including taxol was enhanced. The time of addition and amount of feeding were found to be important factors. Feeding of camphor and menthol resulted in negative effects on taxol production. Influences of exogenous taxanes on taxane production by cell cultures were found to be very complicated. When taxol, baccatin III, cephalomannine were exogenously added into the culture, production of baccatin III, 7-epi-10-deacetyltaxol, 10-deacetyltaxol was increased respectively.

Characterization of Nafion/Pt/Polypyrrole Composite Membrane Prepared by Chemical In-situ Polymerization for DMFC (화학적 합성에 의해 제조된 직접 메탄올 연료전지용 나피온/백금/폴리피롤 복합 막의 특성 분석)

  • Nafion/Pt/Polypyrrole composite membranes were fabricated by chemical in-situ polymerization of pyrrole monomers with Pt precursors in Nafion matrix for DMFC. We demonstrated that positively charged pyrrolinum groups of polypyrrole particles were co-interacted with sulfonic groups of Nafion as verified by FT-IR results. Mutual interaction between $Nafion-SO_3^-$ (or negatively charged Pt precursors) and Polypyrrole$-NH_2^+$ influenced the physical properties of pristine Nafion. Thermal property proton conductivity, methanol permeability, and cell performance of pristine and modified Nafion were analyzed for an application of DMFC membrane. Thermal stabilities of sulfonic groups and side chains in Nafion/Pt/polypyrrole composite membranes were higher than those of Nafion due to mutual interaction between sulfonic groups of Nafion and pyrrolinum groups of polypyrrole. Methanol permeabilities of Nafion/Pt/Polypyrrole composite were reduced more proton conductivities with the increase in the content of Pt particles. As a result of that, the enhancement of cell performance by Nafion/Pt/Polypyrole O2 relative to Nafion was more pronounced under the specific experimental condition such as high temperature and more concentrated methanol solution.

SNS Technology Contribution to B2C Channel Benefits (SNS 기술의 B2C 채널 효용 기여 모형)

  • In this paper, we intend to understand distinguishable SNS technology contributions for B2C commerce. In order to theorize this, we draw upon Technology-to-Performance chain (TPC) theory to develop the model that could explain how the technology value of the SNS could be realizable. For the purpose of capturing SNS technology effect to B2C channel effectiveness, on the basis of TPC theory, personal productivity is selected as a performance variable instead of satisfaction from SNS usage. Accordingly, as the antecedents of personal productivity, two variables of locatability and timeliness are extracted to reflect Task-technology-fit (TTF) dimension of performance precursors. For the utilization dimension of TPC framework, convenience and recreation tendency of the personal traits in shopping are included as the precursors of SNS utilization in the sense of reflecting utilitarian and hedonic value of SNS adoption. Empirical validation is conducted with reference to promotion and sales event on Facebook, which is chosen as the typical SNS based B2C commerce scenario. The data analysis result provide useful insights to further investigate the firm's SNS usage potentials for business applications.

Fabrication of WS2-W-WC Embedded Carbon Nanofiber Composites for Supercapacitors (슈퍼 커패시터를 위한 WS2-W-WC가 내장된 탄소나노섬유 복합체의 제조)

  • $WS_2$-W-WC embedded carbon nanofiber composites were fabricated by using electrospinning method for use in high-performance supercapacitors. In order to obtain optimum electrochemical properties for supercapacitors, $WS_2$ nanoparticles were used as precursors and the amounts of $WS_2$ precursors were controlled to 4 wt% (sample A) and 8 wt% (sample B). The morphological, structural, and chemical properties of all samples were investigated by means of field emission photoelectron spectroscopy, transmission electron microscopy, X-ray diffraction, and X-ray photoelectron spectroscopy. These results demonstrated that the embedded phases of samples A and B were changed from $WS_2$ to $WS_2$-W-WC through carbothermal reaction during carbonization process. In particular, sample B presented high specific capacitance (~119.7 F/g at 5 mV/s), good high-rate capacitance (~60.5%), and superb cycleability. The enhanced electrochemical properties of sample B were explained by the synergistic effect of the using 1-D structure supports, increase of specific surface area, and improved conductivity from formation of W and WC phases.

Metabolic Differentiation of Saccharomyces cerevisiae by Ketoconazole Treatment

  • Azole fungicides are one of the most wide-spread antifungal compounds in agriculture and pharmaceutical applications. Their major mode of action is the inhibition of ergosterol biosynthesis, giving depletion of ergosterol, precursors and abnormal steroids. However, metabolic consequences of such inhibition, other than steroidal metabolitesare not well established. Comprehensive metabolic profiles of Saccharomyces cerevisiae has been presented in this study. Wild type yeast was treated either with glucose as control or azole fungicide (ketoconazole). Both polar metabolites and lipids were analyzed with gas chromatography-mass spectrometry. Approximately over 180 metabolites were characterized, among which 18 of them were accumulated or depleted by fungicide treatment. Steroid profile gives the most prominent differences, including the accumulation of lanosterol and the depletion of zymosterol and ergosterol. However, the polar metabolite profile was also highly different in pesticide treatment. The concentration of proline and its precursors, glutamate and ornithine were markedly reduced by ketoconazole. Lysine and glycine level was also decreased while the concentrations of serine and homoserine were increased. The overall metabolic profile indicates that azole fungicide treatment induces the depletion of many polar metabolites, which are important in stress response.

Characterization of Ozone Distributions in Pohang: Measurement Data during 2002~2006 (포항지역 오존농도의 분포 특성: 2002~2006년 측정자료)

  • Temporal trends and spatial distributions of ozone concentrations in Pohang were investigated using data measured at 4 air quality monitoring stations (i.e., Daedo, Jukdo, Jangheung, and Desong) during 2002-2006. The monthly mean ozone concentrations were highest during April and June and decreased during July and August, which follows the typical trend in the Northeast Asia region. The high springtime ozone concentration might have been strongly influenced by the enhanced photochemical ozone production of accumulated precursors during the winter under increased solar radiations. In July and August, ozone levels were decreased by frequent and severe precipitation that caused lower mean monthly solar radiation and efficient wash-out of ozone precursors. This suggests that precipitation is extremely beneficial in the aspect of ozone pollution control. High ozone concentrations exceeding 80ppb dominantly occurred in May and June during the late afternoon between 16:00~17:00. Ozone concentrations were higher in Jangheung and Daesong relative to Daedo and Jukdo, whereas total oxidants $(O_3+NO_2)$ were higher in Jangheung and Daedo. In the suburban area of Daesong, ozone concentrations seem to be considerably higher than those in urban sites of Daedo and Jukdo due to lower ozone loss by NO titration with lower local NO level.

Effect of Preparation Conditions on the Hydrogenation Activity and Metal Dispersion of Pt/C and Pd/C Catalysts

  • The Pt/C and Pd/C catalysts were prepared from conventional chloride precursors by adsorption or precipitation-deposition methods. Their activities for hydrogenation reactions of cyclohexene and acetophenone were compared with those of commercial catalysts. The Pt/C and Pd/C catalysts obtained from the adsorption procedure reveal higher hydrogenation activity than commercial catalysts and the catalysts prepared by the precipitation-deposition method. Their improved performances are attributed to the decreased metal crystallite sizes of Pt or Pd formed on the active carbon support upon the adsorption of the precursors probably due to the same negative charges of the chloride precursor and the carbon support. Under the preparation conditions studied, the reduction of the supported catalysts using borohydrides in liquid phase is superior to a gas phase reduction by using hydrogen in the viewpoint of particle size, hydrogenation activity and convenience.

  • Compared with organic fluorophores, semiconductor quantum dots (QDs) have the better properties such as photostability, narrow emission spectra coupled to tunable photoluminescent emissions and exceptional resistance to both photo bleaching and chemical degradation. In this work, CdSe/ZnS QDs nanobeads were prepared by the incorporation of CdSe/ZnS QDs with mesoporous silica to use as the optical probe for detecting toxic and bio- materials with high sensitivity, CdSe/ZnS core/shell QDs were synthesized from the precursors such as CdO and zinc stearate with the lower toxicity than pyrotic precursors. The QD-nanobeads were characterized by transmission electron microscopy, FL microscopy, UV-Vis and PL spectroscopy, respectively.
