Changes in Physicochemical Attributes of Potato Slices with Different Cultivars during Cold Storage (품종별 한국산 감자 슬라이스의 냉장 중 이화학적 품질의 변화)

  • Three potato cultivars were prepared as slices and stored for 4 weeks at $5^{\circ}C$. Changes in Vitamin C content and other quality factors, such as color, pH, soluble solid and protein content were determined. Vitamin C content and L value decreased in three potato cultivars. Initial Vitamin C contents of three potato cultivars varied from 49.27 mg% in Sumi to 56.40 mg% in Namjak. Changes in L value showed that the tendency of browning in Daejima was slower than Sumi and Namjak. Changes in pH were small. Soluble solids and protein content increased and varied by cultivar. From the correlation analysis, correlation between browning degree and Vitamin C content was low in three potato cultivars.

Nutrient Components and Physicochemical Properties of Korean Sweet Potato according to Cultivars (국내산 고구마 품종에 따른 영양성분 및 이화학적 특성)

  • This study examined the nutrient components and measured the physicochemical properties of 23 Korean sweet potato cultivars. The proximate composition, free sugars, organic acids, and dietary fiber of sweet potato were analyzed and the water binding capacity, oil absorption and pasting characteristics were measured. The proximate composition was the highest in Happymi and was higher in colored sweet potato than in general sweet potato (p<0.05). The sucrose content was the highest among the free sugars in the general cultivars and yellow/orange colored cultivar. The major organic acids were oxalic acid, citric acid, malic acid, succinic acid, fumaric acid, and acetic acid. Oxalic acid and citric acid were high in the purple colored cultivars and malic acid was high in yellow/orange colored cultivars. Each total dietary fiber content of the general and colored cultivars was 7.51-13.94 and 13.04-16.97%, respectively, but there was no significant difference. The water binding capacity and oil absorption of Juhwangmi was high in all cultivars (p<0.05). The peak viscosity of sweet potato powder showed highest a significant difference of 12.50-1342 cP (p<0.05). The breakdown of Sincheonmi was 512, which was the highest value (p<0.05). The setback of Geonpungmi was significantly low, 6.0 (p<0.05).

  • Genetic improvement in potato can be carried out through several approaches, as sexual crosses, somatic hybridization, mutation and genetic engineering. Although the approach is different, but the goal is the same, to get a superior cultivar. Mutation and genetic engineering are very interesting methods for genetic improvement of potato plants. Mutation by gamma-ray irradiation have been performed to get some new potato cultivars which are more resistant to disease and have higher productivity. We have carried out a mutation of some potato cultivars and obtained some excellent clones to be potentially released as new superior cultivars. By the mutation method, we have released one potato cultivar for the French fries industry, and we registered one cultivar of potato for chips, and two cultivar for vegetable potatoes. Actually we are doing multi-location trial for three clones to be released as new cultivars. Through genetic engineering, several genes have been introduced into the potato plant, and we obtained several clones of transgenic potato plants. Transgenic potato plants containing FBPase gene encoding for fructose bisphosphatase, have a higher rate of photosynthesis and higher tuber productivity than non-transgenic plants. This result suggests that FBPase plays an important role in increasing the rate of photosynthesis and potato tuber productivity. Some transgenic potatoes containing the Hd3a gene are currently being evaluated for their productivity. Over expression of the Hd3a gene is expected to increase tuber productivity and induce flowering in potatoes. Transgenic potato plants containing MmPMA gene encoding for plasma membrane ATPse are more tolerant to low pH than non-transgenic plants, indicating that plasma membrane ATPase plays an important role in the potato plant tolerance to low pH stress. Transgenic potato plants containing c-lysozyme genes, are highly tolerant of bacterial wilt diseases caused by Ralstonia solanacearum and bacterial soft rot disease caused by Pectobacterium carotovorum. Expression of c-lyzozyme gene plays an important role in increasing the resistance of potato plants to bacterial diseases.

Phylogenetic analysis and association of markers and traits related to starch contents in Korean potato cultivars using SSRs

  • Diversity of 30 Korean potato cultivars was evaluated using 14 microsatellite markers. Twelve microsatellite markers representing 12 loci in the potato genome detected 84 polymorphisms among 30 cultivars and revealed alleles with a mean of 7.00 alleles per primer. The polymorphism information content (PIC) value ranged from 0.57 to 0.93 with average of 0.82. Based on polymorphism, cluster analysis was conducted by the unweighted pair-group method with arithmetic average (UPGMA) methods. Thirty potato varieties were distinctly separated into 2 groups and similarity coefficient of cluster ranged from 0.58 to 0.95. Thirty tetraploid cultivars were evaluated for six important agronomic traits. One-way analysis of variance was done to look for the degree of relationships between individual markers and traits. K1 and K2 markers showed a significant association with amylose contents, starch contents, and specific gravity.

Physicochemical Properties of Starches Isolated from New Potato Cultivars in Korea

  • Physicochemical properties of starches isolated from 3 new potato cultivars developed by Potato Valley Ltd. were investigated and compared to those of starch isolated from 'Superior' being distributed prominently in Korea. Significant differences were observed in physicochemical properties such as granule size, amylose content, phosphorus content, water binding capacity, swelling power, solubility, and in vitro digestibility of starches from different potato cultivars. Thermal properties were evaluated using differential scanning calorimetry (DSC), and onset gelatinization temperatire ($T_o$), peak temperature ($T_p$), and enthalpies of gelatinization (${\triangle}H$) of different potato cultivars ranged as $58.0{\pm}0.3-61.7{\pm}0.4^{\circ}C,\;63.7{\pm}0.2-$ $66.5{\pm}0.0^{\circ}C$, and $15.6{\pm}0.5-17.0{\pm}0.3$ J/g, respectively. Pasting properties were evaluated using a rapid visco analyzer (RVA), and pasting temperature, peak viscosity, and final viscosity of different potato cultivars ranged as $65.0{\pm}0.1-68.9{\pm}0.1^{\circ}C$, $8,163.7{\pm}196.3-9,035.7{\pm}6.4$ cp, and $4,397.7{\pm}166.7-7,025.0{\pm}271.3$ cp, respectively. Especially, 'Gogu valley' starch showed the highest values in the amylose and phosphorus content among tested potato cultivars and higher water binding capacity, swelling power, and solubility than those of other tested starches. And it also showed high pasting temperature, peak viscosity, trough viscosity, and final viscosity as compared to other tested starches.

Biocontrol of Potato White Mold Using Coniothyrium minitans and Resistance of Potato Cultivars to Sclerotinia sclerotiorum

  • This study was conducted in Bahar and Lalehjin, Hamadan, Iran to assess the biocontrol efficacy of Coniothyrium minitans Campbell against potato white mold caused by Sclerotinia sclerotiorum (Lib.) de Bary under field and greenhouse conditions. In addition, the resistance of common potato cultivars against S. sclerotiorum was determined in a greenhouse experiment. After straw inoculation of six potato cultivars (Pashandi, Istambouli, Agria, Marfauna, Alpha and Spartaan) with S. sclerotiorum, the least disease severity was observed in Spartaan and Marfauna. Agria showed the most susceptibility to S. sclerotiorum. Compared with the healthy control, different concentrations of C. minitans conidia ($10^7$, $10^8$ and $10^9$ conidia/mL) reduced disease severity under greenhouse condition, and a concentration $10^9$ was the most effective treatment. During 2008 and 2009, four field trials were conducted to evaluate the efficacy of C. minitans in different soil and aerial applications on disease incidence of potato white mold. In 2008, soil application of $Contans^{(R)}$ WG (a commercial product of C. minitans) showed the greatest biocontrol capacity whereas soil application of solid-substrate C. minitans was found inferior when compared with other treatments in both Bahar and Lalehjin field sites. In 2009, benomyl application was the most effective treatment in reducing disease incidence in both tested field sites.

Food Composition of Raw and Boiled Potatoes (생감자와 삶은 감자의 식품성분 비교)

  • Purpose: This study examined the food composition of four raw and boiled domestic potato cultivars: Superior, Dejima, Bora Valley and Rose. Methods: Proximate composition (moisture, protein, ash, dietary fiber and fat), minerals (calcium, phosphorus, iron, potassium, sodium and magnesium) and vitamins (thiamine, riboflavin and niacin) were analyzed in the study. Results: In this study, it was observed that the moisture contents of raw and boiled Bora Valley potato were 83.74 g/100 g and 81.57 g/100 g, respectively, which was highest among the studied cultivars. The protein content of the raw and boiled Rose potato (12.23 g/100 g and 11.02 g/100 g) was higher than that of the other cultivars. Protein contents of boiled Bora Valley and Rose potatoes were significantly lower than those of their respective raw samples. The potassium, phosphorus and sodium contents of Bora Valley potato were 2,397.69 mg/100 g, 389.13 mg/100 g and 14.12 mg/100 g, respectively. In case of raw potato, the calcium, iron and magnesium contents of Dejima potato were 51.52 mg/100 g, 3.21 mg/100 g and 137.62 mg/100 g, respectively, which were the highest values among the tested potato cultivars. Total dietary fiber content of Dejima potato was 14.78 g/100 g, which was the highest level. The dietary fiber contents of four cultivars of potatoes were lower after boiling. Thiamine content was highest in the Bora Valley potato (0.43 mg/100 g). Riboflavin contents of the Rose potato was 0.35 mg/100 g, which was higher than that of the others. Conclusion: Overall, four cultivars of raw and boiled potatoes had enhanced food composition. Therefore, these potato cultivars are expected to be highly valuable food items for the development and application of functional foods.

Changes in Polyphenol Oxidase Activity, Phenol Concentration and Browning Degree of Potato Slices with Different Cultivars during Cold Storage (품종별 한국산 감자 슬라이스의 냉장 중 Polyphenol oxidase, 페놀함량 및 갈변정도의 변화)

  • Three potato cultivars, Sumi,Daejima and Namjak, were prepared as slices. They were dipped in distilled water for 20 seconds. The potato slices were packed in polyethylene bags and stored at $5^{\circ}C$. Browning degree, total phenol and chlorogenic acid contents and polyphenol oxidase (PPO) activity were measured. And the correlation analysis of browning parameters were conducted. The results were as follows. There were increase in browning degree, phenolic content and PPO activity during cold storage of potato slices with different cultivars. Among three cultivars, Sumi showed the highest browning degree, phenolic content and PPO activity and also showed the highest % increse of browning and PPO activity during cold storage. On the contrary, Daejima was the lowest. But Daejima showed the highest % increase in phenolic contents during cold storage. With Sumi, browning degree was significantly correlated with PPO activity and phenolic contents (p<0.05). With Daejima and Namjak, a significant correlation was found between browning degree and PPO activity (p<0.05). From the above results, enzymatic browning reactions of potato slices and factors affecting them were dependent on cultivar. Among the tested three cultivars, Daejima showed the lowest browning degree during cold storage and thus seems to be desirable for minimal processing.

On Selection of Resistant Potato Cultivars to Common Scab(S. scabies) (감자 더뎅이병 저항성 품종 선발)

  • It was conducted to select the resistant potato cultivars to common scab, to replace the existing susceptible cv Dejima in Jeju island conditions both 1997 and 2001. Among cultivars, cvs Jemchip, Alpa, Atlantic and Jopung were more resistant than others in the field with both low and high incidence of the disease. Also among the existing cultivars grown in Korea, cvs Superior, Jopung and Chubaek were resistant, expecially in terms of production of marketable potatoes.

Seasonal Incidence of Potato virus f Infection on Potato Cultivars for the Double Crops in Korea (2기작 감자 품종의 재배 시기별 PVY 감염 정도 조사)

  • One of major potato viruses is Potato virus Y (PVY) in Korea. In the southern part of Korea, potatoes have been grown as double crops in a year by using cv. 'Dejima' and 'Chubak' due to very short dormancy. However, they have caused a serious problem such as a rapid degeneration. It has been thought that the degeneration is affected by the high incidence of PVY in neighboring potato fields. Therefore, the investigation of factors causing the degeneration is very important in the production of healthy seed potato. In this study, the PVY reinfection rates of several potato varieties and the different seed sources of cv. 'Chubak' have been investigated. Results show that the lowest infection rate of PVY among four potato cultivars derived from minitubers is cv. 'Superior'. The others are in order of 'Dejima', 'Atlantic' and 'Chubak'. Also, the incidences of PVY differ significantly when several seed sources are examined. When the seed potatoes (G2, the progeny of microtuber) as spring potato crops are planted in area without potato field nearby, the infection rate of PVY is as low as that of microtubers. However, PVY incidence in the progenies of minitubers as fall potato crops largely increases. Therefore, the best way of potato production under double cropping system is to use the healthy seed potato produced in area without potato field and plant relatively resistant cultivar such as Dejima.
