청국장에서 분리된 Bacillus licheniformis의 Protease 생산을 위한 배지 최적화 (Medium Optimization for the Protease Production by Bacillus licheniformis Isolated from Cheongkookjang)

  • 청국장의 기호도를 개선하는 발효균으로 분리된 protease 생산균은 Bacillus licheniformis로 확인되었다. Protease 생산을 위한 배지를 최적화하기 위한 탄소원, 질소원, 인, 금속이온의 성분을 변화시키면서 균의 성장과 효소 생산성을 비교하였다. Glucose를 탄소원으로 사용하였을 때는 균의 성장은 정상적으로 일어나지만, protease 생산이 심하게 억제되는 것으로 나타났으며, Potato starch를 탄소원으로 사용하였을 때 효소 생산성이 가장 높았다. 질소원으로는 yeast extact가 효소 생산에 가장 적합하였다. 한편 2가 금속이온중 $Zn^{2+}$, $Cu^{2+}$, $Co^{2+}$을 배지에 첨가하였을 때는 균의 성장이 심하게 저해되었으며, 효소 생산도 되지 않았다. $CaCl_2$를 첨가한 배지에서는 균이 성장과 효소 생산성이 증가되었다. Potato starch(1.5%), yeast extract(1.5%), $CaCl_2$(0.7%), $K_2HPO_4$(0.03%)와 $KH_2PO_4$(0.03%)를 포함하는 것으로 구성된 최적화 배지에서 최대효소 생산성은 800 U/mL로 나타났으며 28시간 이후에는 배양액내 효소활성이 서서히 감소하였다.

Effects of potato byproduct on growth performance, blood metabolites, and carcass characteristics of Hanwoo steers

  • This study was conducted to investigate effects of a potato byproduct on growth performance, blood metabolites, and carcass characteristics of Hanwoo steers. The palm oil coated potato byproduct was supplemented to the diet of Hanwoo steers to estimate the effect on growth performance, blood metabolites, and carcass characteristics during the late fattening period. Thirteen steers with initial body weight of control ($676.8{\pm}31.7kg$), treatment 1 ($671.8{\pm}46.2kg$) and treatment 2 ($672.8{\pm}31.1kg$) were used for 60 days, respectively. Average daily gain of steers in treatment 2 and control was greater than that in treastment 1 (p > 0.05). All steers in treatment 2 had a grade quality grading system than B in meat quantity and had a meat quality higher than the $1^{st}$ grade. According to the physicochemical analysis of longissimus muscle, treatment 2 had high brightness resulting from high meat quality (p > 0.05), and a decrease in redness and yellowness is seen as a dilution effect due to muscle hypertrophy (p > 0.05). The fatty acid composition showed low levels of linoleic acid (p = 0.039) and arachidonic acid (p = 0.008) in treatment 2. This resulted in lower polyunsaturated fatty acid (PUFA) levels (p = 0.034). On the other hand, high levels of oleic acid resulted in high levels of MUFA (p > 0.05). These results indicate that potato byproduct had similar effects with ${\beta}_2-adrenergic$ agonist (${\beta}_2-AA$). And there was no negative effect on the intramuscular fat. In conclusion, palm oil coated potato byproduct could be potentially used as an alternative growth enhancer.

  • 밀가루 대체원료(代替原料)로써 쌀보리가루 또는 고구마가루를 활용한 복합분(複合粉)으로 국수를 만들고 그들의 품질(品質)을 조사하여 다음과 같은 결과를 얻었다. 1) 면대(麵帶) 및 건면(乾麵)생산에 있어서 밀가루에 대한 보리가루 대체율(代替率)은 100%까지, 고구마가루 대체율(代替率)은 70%까지 가능하였으며 품질개량제(品質改良劑)로서 glyceryl monostearate 및 sodium polycrylate의 첨가는 이들 제품의 texture 특성(特性)을 향상시켰다. 2) 면대(麵帶)와 건면(乾麵)의 texture 특성치(特性値)인 견고성(堅固性), 응집성(凝集性), gumminess는 보리가루나 고구마가루의 배합에 의하여 떨어졌으나 첨가제의 효과가 뚜렷하였고 면대(麵帶)는 견고성(堅固性) 6.2 이상, gumminess 430 이상, 건면(乾麵)은 견고성(堅固性) 6.8 이상이 필요하였다. 3) 조리면(調理麵)에 있어서 밀가루 대체(代替)와 첨가제의 사용은 texture 특성(特性)을 떨어뜨리는 경향이 있었으며 조리특성(調理特性)으로 보아 보리가루는 60%까지, 고구마가루는 40%까지 대체(代替)할 수 있었다. 4) 조리면(調理麵)의 색깔, 맛, texture에 관한 관능(官能)시험에서 20% 대체복합분(代替複合粉)은 밀가루와 5% 수준에서 유의차(有意差)가 없었으며 품질저하(品質低下)는 주로 착색(着色)에 기인(起因)하였다.

A Simple Detection of Sweetpotato Feathery Mottle Virus by Reverse Transcription Polymerase Chain Reaction

  • A reverse transcription polymerase chain reaction (RT-PCR) protocol was developed using two specific 22-mer primers located in coat protein gene of SPFMV. A 411 bp PCR-product was detected in virus infected plants as well as tissue culture raised sweet potato but not in healthy plants. For optimization of RT-PCR protocol, the optimum crude nucleic acid concentration, annealing temperature, primer concentration and numbers of PCR-cycle for maximum sensitivity and specificity were determined. The optimum condition for RT-PCR was as follows: RT-PCR reaction mixture was one-step mixture, containing 50 pmol of primer, 30 units of reverse transcriptase, 5 units of RNasin, and the crude nucleic acid extracts (200 ng). In RT-PCR, cDNA was synthesized at $42^{\circ}C$ for 45 min before a quick incubation on ice after pre-denaturation at $95^{\circ}C$ for 5 min. The PCR reaction was carried out for 40 cycles at $96^{\circ}C$ for 30 see, $63^{\circ}C$ for 30 sec, $72^{\circ}C$ for 1 min, and finally at $72^{\circ}C$ for 10 min. The viral origin of the amplified product was confirmed by sequencing, with the sequence obtained having $95-98\%$ homology with published sequence data for SPFMV. The benefits of this RT-PCR based detection of SPFMV would be simple, rapid and specific.

중소형 CIM을 위한 ROPP 정보 모델러 개발 (A Development of ROPP Information Modeler for Mid-Sized CIM)

  • Currently a hot potato in industries is to reduce the lifecycle of product and maximize the time-to-market benefits. A lot of methodologies and the concrete systems are in use for performing this function. One of these systems is that process are reconstructed by BPR and systems are implemented by PDM. First of all, th analyses of existing product, process and resource data must be preceded. New process is established on the basis of the analyzed information of a company and PDM implements the new process in the company. The consecutive activities like this provide the basic environment for CIM. However, the current system have been developed on the favor of a big company. There are many problems in applying it too a mid-sized company. A lot of function in a PDM tool is not necessary for the mid-sized CIM characteristic. The purpose of this study is to define the ROPP information model that is an appropriate information integration for a design process. The model shows an associated complicated information of a company effectively. Also the modeler makes it possible to manage consistently the constraints of design process on the basis of the analysis of dependency among each resource.

Efficacy of Sweet Potato Powder and Added Water as Fat Replacer on the Quality Attributes of Low-fat Pork Patties

  • The present study was conducted to investigate the efficacy of sweet potato powder (SPP) and water as a fat replacer in low-fat pork patties. Low-fat pork patties were developed by replacing the added fat with combinations of SPP and chilled water. Three different levels of SPP/chilled water viz. 0.5/9.5% (T-1), 1.0/9.0% (T-2), and 1.5/8.5% (T-3) were compared with a control containing 10% animal fat. The quality of low-fat pork patties was evaluated for physico-chemical (pH, emulsion stability, cooking yield, $a_w$), proximate, instrumental colour and textural profile, and sensory attributes. The cooking yield and emulsion stability improved (p<0.05) in all treatments over the control and were highest in T-2. Instrumental texture profile attributes and hardness decreased, whereas cohesiveness increased compared with control, irrespective of SPP level. Dimensional parameters (% gain in height and % decrease in diameter) were better maintained during cooking in the low-fat product than control. The sensory quality attributes juiciness, texture and overall acceptability of T-2 and T-3 were (p<0.05) higher than control. Results concluded that low-fat pork patties with acceptable sensory attributes, improved cooking yield and textural attributes can be successfully developed with the incorporation of a combination of 1.0% SPP and 9.0% chilled water.

Sclerotium rolfsii에 의한 약모밀 흰비단병 (Occurrence of Stem Rot of Houttuynia cordata Thunb. Caused by Sclerotium rolfsii)

  • 2005년도 6월에서 8월 사이에 경상북도농업기술원 신물질연구소의 약용식물시험재배포장에서 재배하는 약모밀에서 줄기와 지제부가 수침상으로 물러지고 썩으면서 암갈색으로 변하는 증상이 발생하였고, 병반부와 지제부 주위의 토양에 흰색의 곰팡이가 생기고 시간이 경과함에 따라 갈색의 둥글고 부정형의 균핵이 형성되었다. 병원균을 순수 분리하여 감자한천 배지(PDA)에서 배양학적 형태적 특성을 조사한 결과 병원균의 생육적온은 $28{\sim}30^{\circ}C$ 이었으며, PDA에서 생육중인 균사의 폭은 $3.0{\sim}9.0{\mu}m$이었고, 형성된 균핵의 크기는 $1.0{\sim}2.5mm$이었으며, 균사 특유의 clamp connection 형태가 현미경하에서 관찰되었다. 병원성 실험결과 병원성이 인정되었다. 이상과 같이 병징과 균학적 특징 및 병원성을 검정한 결과, 이 병을 Sclerotium rolfsii에 의한 약모밀 흰비단병으로 명명하고자 한다.

고구마 소주 주박의 항균 및 항혈전 활성 (Anti-microbial and Anti-thrombosis Activities of Lees of Sweet Potato Soju)

  • 고구마 소주는 전분과 섬유질이 많은 고구마를 발효시킨 후 상압증류하여 제조되어 특유의 부드러운 맛과 향미를 가진다. 본 연구에서는 별도의 용도가 없어 폐기되고 있는 고구마 소주 주박의 효율적인 이용을 위해, 주박의 ethanol 및 열수 추출물을 조제하고 이들을 순차적 유기용매 분획하여 각각의 hexane, ethylacetate (EA), butanol 분획물과 물 잔류물을 조제하여 각 시료의 항균 및 항혈전 활성을 평가하였다. 그 결과, 주박의 ethanol 추출 효율이 열수 추출 효율보다 1.36배 높았으며, 분획물 중에서는 EA 분획물이 가장 높은 total polyphenol 함량을 나타냄을 확인하였다. 고구마 소주 주박의 추출물 및 분획물(0.5 mg/ml)은 인간 적혈구에 대한 용혈활성을 나타내지 않았으며, 열수 및 ethanol 추출물의 EA 분획물에서만 광범위 항세균 활성과 강력한 혈액응고 저해 활성이 나타났다. 또한 혈소판 응집저해 활성의 경우 열수 추출물의 EA 분획에서만 아스피린과 유사한 응집저해를 나타내었다. 본 연구결과는 폐기되고 있는 고구마 소주 주박의 열수 추출물의 EA 분획이 신규의 항세균 및 항혈전제로 개발될 수 있음을 제시하고 있다.

Potential Water Retention Capacity as a Factor in Silage Effluent Control: Experiments with High Moisture By-product Feedstuffs

  • The role of moisture absorptive capacity of pre-silage material and its relationship with silage effluent in high moisture by-product feedstuffs (HMBF) is assessed. The term water retention capacity which is sometimes used in explaining the rate of effluent control in ensilage may be inadequate, since it accounts exclusively for the capacity of an absorbent incorporated into a pre-silage material prior to ensiling, without consideration to how much the pre-silage material can release. A new terminology, 'potential water retention capacity' (PWRC), which attempts to address this shortcoming, is proposed. Data were pooled from a series of experiments conducted separately over a period of five years using laboratory silos with four categories of agro by-products (n = 27) with differing moisture contents (highest 96.9%, lowest 78.1% in fresh matter, respectively), and their silages (n = 81). These were from a vegetable source (Daikon, Raphanus sativus), a root tuber source (potato pulp), a fruit source (apple pomace) and a cereal source (brewer's grain), respectively. The pre-silage materials were adjusted with dry in-silo absorbents consisting wheat straw, wheat or rice bran, beet pulp and bean stalks. The pooled mean for the moisture contents of all pre-silage materials was 78.3% (${\pm}10.3$). Silage effluent decreased (p<0.01), with increase in PWRC of pre-silage material. The theoretical moisture content and PWRC of pre-silage material necessary to stem effluent flow completely in HMBF silage was 69.1% and 82.9 g/100 g in fresh matter, respectively. The high correlation (r = 0.76) between PWRC of ensiled material and silage effluent indicated that the latter is an important factor in silage-effluent relationship.

Effects of Drying Condition and Binding Agent on the Quality Characteristics of Ground Dried-Pork Meat Products

  • The purpose of this study was to investigate the influence of processing conditions (temperature and time) and binding agent types (glutinous rice flour, potato starch, bean flour, and acorn flour) on the physicochemical and sensory characteristics of ground dried-pork meat product. For this purpose, ground dried-pork meat product was produced by adding several binding agents at different drying temperatures and times. The drying time affected moisture content and water activity in all drying temperature. However, under the similar drying conditions, the extent of drying varied depending on the type of binding agents. The results of sensory evaluation for texture degree and overall acceptability indicated the following: overall, higher drying temperatures and longer drying time heightened the degree of texture, and the overall acceptability varied depending on binding agent type. Physicochemical and sensory characteristics were analyzed to determine any possible correlation. The results revealed a high correlation between moisture content, water activity, shear forces, and sensory evaluation (p<0.01). However, there was no correlation with respect to overall acceptability.
