White strains of Hypsizygus marmoreus are more difficult to cultivate than are brown strains; therefore, new white strain breeding strategies are required. Accordingly, we constructed the genetic map of H. marmoreus with 1996 SNP markers on 11 linkage groups (LGs) spanning 1380.49 cM. Prior to analysis, 82 backcrossed strains (HM8 lines) were generated by mating between KMCC03106-31 and the progenies of the F1 hybrid (Hami-18 × KMCC03106-93). Using HM8, the first 23 quantitative trait loci (QTLs) of yield-related traits were detected with high limit of detection (LOD) scores (1.98-9.86). The length, thickness, and hardness of the stipe were colocated on LG 1. Especially, length of stipe and thickness of stipe were highly correlated given that the correlation coefficients were negative (-0.39, p value ≤ .01). And a typical biomodal distribution was observed for lightness of the pileus and the lightness of the pileus trait belonged to the LG 8, as did traits of earliness and mycelial growth in potato dextrose agar (PDA) medium. Therefore, results for color traits can be suggested that color is controlled by a multi-gene of one locus. The yield trait was highly negatively correlated with the traits for thickness of the stipe (-0.45, p value ≤ .01). Based on additive effects, the white strain was confirmed as recessive; however, traits of mycelial growth, lightness, and quality were inherited by backcrossed HM8 lines. This new genetic map, finely mapped QTLs, and the strong selection markers could be used in molecular breeding of H. marmoreus.
The vein-necrosis strain or potato virus Y (PVY-Vff) and black shank (Phytophlhora parasitica roar. nicotianae) causes severe damage on burley tobacco(Wicotiana tabacum L.) in Korea, A new burley tobacco resistance to PVY and black shank, KB 110, was developed by Korea Ginseng and Tobacco Research Institute. It was developed from the cross of Burley 21 with TC 591 in 1990, and was backrossed to Burley 21 in the following season. TC 591 has resistance to PVY and moderate resistance to race 0 of black shank, but it is susceptible to tobacco mosaic vim (TMV). KB 110 was evaluated for its resistance to PVY, TMV and black shank in the greenhouse and at fields for preliminary and performance trials. KB 110 which has secreting glandular trichomes was resistant to PVY-VN, TW and black shank. It had an erect growth habit and two more leaves per plant than that of Burley 21, and matures two to three days later. It yielded approximately 3 percent more cured leaf than the standard cultivar Burley 21, but other plant characteristics were very similar to those of Burley 21. It had acceptable standards for chemical and physical characteristics of lured leaf on regional farm test in 1995-1997. KB 110 produced average yields of good quality tobaccos and was appeared to be resistant to PVY inwhere occurrence of the virus are severe chronic at burley growing area.
Tobacco diseases have not been recorded until 1900s in Korea, where tobacco plants were introduced at early 1700s. Practical researches on the disease have been conducted since mid 1960s. Major ten tobacco diseases were mosaic caused by tobacco mosaic virus·potato virus Y·cucumber mosaic virus, bacterial wilt, hollow stalk, wild fire caused by angular leaf spot strain, black shank, brown spot, powdery mildew and fusarium wilt. But their annual occurrences were varied according to changes of tobacco varieties and their cultivating practices. As no useful chemicals, several biological tactics have been developed to control the viral or bacterial diseases that give significant economic damages on sustainable crop yield, but not practicable to field farming condition yet. Transgenic tobacco plants containing foreign disease resistant genes have been developed by current bio-technology, but not released to farmers yet. Though some disease-resistant tobacco varieties have been developed by the conventional breeding technology and currently used by farmers, their disease controlling efficacy have been diminished by occurrence of the new strain or race. Future research on tobacco diseases has been focused on technical development to produce high quality tobacco with less production cost, which leads Korean tobacco industry to keep its competence against foreign industry and decreasing overall market.
Wolfiporia cocos is an important medicinal fungus that has been used in regions of Northeast Asia including Korea, Japan, and China. W. cocos is classified in Korea into two types (red bokryeong and white bokryeong) based on the internal colors (yellow orange-pale pink and white) of the sclerotium. Generally, the W. cocos type cultivated on farms produces white sclerotium. In this study, we endeavored to select strains that form sclerotium in sawdust medium using 2-way cross-breeding among two cultivated strains and three wild strains. Monospores were isolated from the fruiting bodies of cultivated and wild strains on potato dextrose agar. Thirty-nine strains of 338 hybrid strains isolated formed sclerotia with white or yellow colors upon culture for 3 months in Pinus densiflora sawdust medium. Selection for sclerotium forming strains using sawdust culture follows a very simple and easy procedure that is presented for the first time in this paper. We plan to test selected strains in the field to aid in developing new varieties for the future.
'Sanggang', a new cultivar of Agrocybe aegerita, was bred with mating between monokaryotic strains isolated from 'GMAG45109' and 'GMAG45107' in Mushroom Research Station, Gyonggi Province A.R.E.S. in 2009. The optimum temperature for the mycelial growth of 'Sanggang' was 26 to 28$^{\circ}C$ on PDA(potato dextrose agar) medium. The optimum temperature for the primordia formation and fruiting body development of 'Sanggang', was 18 to 20$^{\circ}C$. In the bottle cultivation of 'Sanggang', the period of spawn running was around 38 days at 22 to 23$^{\circ}C$ and the period from scratching of inoculum to harvest was 12 days. These characteristics of 'Sanggang' were not different from those of 'Mihwang' (control). 'Sanggang' had brown-colored pileus, whereas 'Mihwang' had yellowish brown-colored one. In bottle cultivation, 'Sanggang' hada little smaller but stronger stipe and pilus than 'Mihwang', and 'Sanggang', as well as 'Mihwang', had upright stipe. And 'Sanggang' was shown to have lower veil opening ratio than that of 'Mihwang' at harvest time.The yield of fruiting bodies of 'Sanggang' was 134 g/850ml bottle, which was similar to that of 'Mihwang'. Resistance of'Sanggang' against Trichoderma spp., as well as 'Mihwang', was weak. Freshness of 'Sanggang' was maintained for 13 days under storage temperature 4$^{\circ}C$, while that of 'Mihwang' was maintained for 10 days.

Leaf blight caused by Stemphylium vesicarium is one of the most important fungal diseases of garlic (Allium sativum L.) worldwide, which results in a reduction of quality and yield. The breeding of resistant cultivars is an efficient approach to decrease the use of chemical fungicides and minimize crop losses. In this study, to find the resistant garlic resources against S. vesicarium, we evaluated the resistance degree of 20 garlic germplasms. To do this, garlic seedlings at four-leaf stage were rubbed with nonabsorbent cotton and then inoculated with spore suspension (3.0×105 spores/ml of potato dextrose broth) of S. vesicarium by spray method. Three to seven days after inoculation, the infected leaf area (%) of garlic seedling was measured. 'Daeseo' and 'Namdo' were included as susceptible and resistant control cultivars, respectively. After 3 to 7 days of incubation, the infected leaf area (%) of garlic seedling was measured. Our results showed that IT245512, IT245528, and IT244068 lines exhibited the highest resistance against S. vesicarium, whereas IT257134 and IT253043 lines were more susceptible than the susceptible cultivar 'Daeseo'. Based on the results, the resistant genetic resources selected in this study can be used a basic material for resistant garlic breeding system against leaf blight.

Due to abnormal weather conditions caused by climate change, natural disasters and damages are gradually increasing around the world. Global climate change as accompanied by warming is projected to exert adverse impact on production of potato, which is known as cool season crop. Even though, role of potato as a food security crop is expected to increase in the future, the climate change impacts on potato and adaption strategies are not sufficiently established. Therefore, this study was conducted to analyze the damage pattern of potatoes due to high temperature treatment and to evaluate the response of cultivars. T he high temperature treatment (35~38℃) induced heat stress by sealing the plastic house in midsummer (July), and the quantity and quality characteristics of potatoes were compared with the control group. T otal yield, marketable yield (>80 g) and the number of tubers per plants decreased when heat treatment was performed, and statistical significance was evident. In the heat treatment, 'Jayoung' cultivar suffered a high heat damage with an 84% reduction in yield of >80 g compared to the control group. However, in Jopung cultivar, the decrease was relatively small at 26%. Tuber physiological disturbances (Secondary growth, Tuber cracking, Malformation) tended to increase in the heat stress. Under heat conditions, the tubers were elongated overall, which means that the marketability of potatoes was lowered. T he tuber firmness and dry matter content tended to decrease significantly in the heat-treated group. T herefore, the yield and quality of tubers were damaged when growing potatoes in heat conditions. T he cultivar with high heat adaptability was 'Jopung'. T his result can be used as basic data for potato growers and breeding of heat-resistant cultivars.

Viruses diagnosed on crops including rice plants from farmers or agricultural extension agencies cover the country were 11 species including Broad bean wilt virus 2 (BBWV2) in 2008. Tomato spotted wilt virus (TSWV) was the most important virus having the detection rate of 22.9%. Two viruses of Tomato yellow leaf curl virus (TYLCV) and Tobacco leaf curl virus (TLCV) inducing leaf yellow and curl diseases on tomatoes were occurred newly with the detection rate of 12.2% and 4.0%, respectively, in 2008. Rice stripe virus (RSV) was occurred on 869.5 ha mainly at Jindo and Haenam areas in Jeollanamdo province. At Jindo area, 12 plots were damaged severely with the infected hill rate of 83.8%. At the main production area of oriental melon at Seongju, almost all fruits from whole sale market at Seongju were infected with Cucumber green mottle mosaic virus (CGMMV) as the detection rate of 87%. The areas occurred TSWV in Korea were 25 totally from 2003 including 7 areas newly reported in 2008 including Naju in Jeoallanamdo. TSWV could be reduced as 0.1 % from 5.3% by covering insect proof net in vinyl house after chemical soil sterilization. Tomato yellow leaf curl disease was occurred on April in 2008 at Tongyoung area in Kyeongsangnamdo, and detected continuously at 13 areas, 7 in Kyeongsangnamdo, 4 in Jeollabukdo and 2 in Jejudo. Potato spindle tuber viroid (PSTVd) was occurred abruptly in a confined space of a civil breeding greenhouse and a cultivar evaluation field followed by disuse 17.4 M/T of potato tubers. No PSTVd was detected at 17 fields cultivated the related potatoes to the bred company by RT-PCR.
To obtain basic information for the genetic analysis and breeding of Flammulina velutipes, some factors affecting the release of protoplasts from the fungus were studied. Potato Dextrose peptone Agar medium was suitable for the growth of the mycelium and the protoplast formation of F. velutipes. The culture age for the high yields of protoplast was 5 days on PDPA. Few protoplasts were formed from the mycelium cultured on Mushroom minimum Media. The highest yield of protoplasts was obtained in enzyme solution containing Novozyme 234 plus cellulase CP at 10 mg $ml^{-1}$ concentration, while a half amount of protoplasts was obtained in enzyme solution containing Novozyme 234 only. The optimal reaction time of the mycelium in the Iytic enzyme mixtures was 3 hours. The best osmotic stabilizer for the protoplast formation of the mycelium was 0.6M sucrose without buffer at pH 6.2.
