Inhibitory activity of Euonymus alatus against alpha-glucosidase in vitro and in vivo

  • The major goal in the treatment of diabetes mellitus is to achieve near-normal glycemic control. To optimize both fasting blood glucose and postprandial glucose levels is important in keeping blood glucose levels as close to normal as possible. ${\alpha}-Glucosidase$ is the enzyme that digests dietary carbohydrate, and inhibition of this enzyme could suppress postprandial hyperglycemia. The purpose of this study was to test the inhibitory activity of methanol extract of Euonymus alatus on ${\alpha}-glucosidase$ in vitro and in vivo to evaluate its possible use as an anti-diabetic agent. Yeast ${\alpha}-glucosidase$ inhibitory activities of methanol extract of E. alatus were measured at concentrations of 0.50, 0.25, 0.10, and 0.05 mg/ml. The ability of E. alatus to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without E. alatus extract (500 mg/kg) was administered to diabetic rats by gastric intubation after an overnight fast. Plasma glucose levels were measured at 30, 60, 90, 120, 180, and 240 min. Plasma glucose levels were expressed in increments from baseline, and incremental areas under the response curve were calculated. Extract of E. alatus, which had an $IC_{50}$ value of 0.272 mg/ml, inhibited yeast ${\alpha}-glucosidase$ activity in a concentration-dependent manner. A single oral dose of E. alatus extract significantly inhibited increases in blood glucose levels at 60 and 90 min (p<0.05) and significantly decreased incremental response areas under the glycemic response curve (p<0.05). These results suggest that E. alatus has an antihyperglycemic effect by inhibiting ${\alpha}-glucosidase$ activity in this animal model of diabetes mellitus.

자일로올리고당을 함유한 설탕이 건강한 성인의 Glycemic Index (GI)와 혈당에 미치는 효과 (Effects of xylooligosaccharide-sugar mixture on glycemic index (GI) and blood glucose response in healthy adults)

  • 본 연구는 자일로올리고당을 설탕에 적용하여 임상시험을 통해 자일로올리고당 첨가량에 따른 설탕의 GI 저감효과와 혈당저감효과를 확인하였다. 건강한 성인 남녀 11명를 대상으로 표준식품인 포도당과 비교하여 분말 자일로올리고당을 14% 함유한 Xylo 14과 분말 자일로올리고당을 20% 함유한 Xylo 20을 섭취 후 표준식품과 비교식품의 IAUC를 통해 GI를 산출한 결과 Xylo 14는 $60.0{\pm}23.5$로 Medium GI 식품으로 나타났으며, Xylo 20은 $54.3{\pm}17.7$로 Low GI 식품으로 나타났다. 순수한 설탕의 GI 68과 비교하면 각각 11.8%와 20.1% 혈당상승 저감효과를 나타낼 수 있었다. AUC는 15~90분까지 모든 구간에서 유의적인 차이 (p < 0.05)를 나타내었다. 따라서 Xylo 14와 Xylo 20은 혈당 저감효과를 나타냄과 동시에 7~10 g 이상 섭취하면 자일로올리고당의 1일 유효섭취량도 만족시킬 수 있으므로 기능성 설탕으로서의 활용이 기대된다.

Autumn olive (Elaeagnus umbellata Thunb.) berry reduces fasting and postprandial glucose levels in mice

  • BACKGROUND/OBJECTIVES: Fasting and postprandial hyperglycemia should be controlled to avoid complications of diabetes mellitus. This study investigated the effects of autumn olive (Elaeagnus umbellata Thunb.) berry (AOB) on fasting and postprandial hyperglycemia in mice. MATERIALS/METHODS: In vitro ${\alpha}$-glucosidase inhibitory effect of AOB was determined. Maltose solution (2 g/kg) with and without AOB extract at 500 mg/kg or acarbose at 50 mg/kg was orally administered to normal mice after overnight fasting and glucose levels were measured. To study the effects of chronic consumption of AOB, db/db mice received the basal diet or a diet containing AOB extract at 0.4% or 0.8%, or acarbose at 0.04% for 7 weeks. Blood glycated hemoglobin and serum glucose and insulin levels were measured. Expression of adiponectin protein in epididymal white adipose tissue was determined by Western blotting. RESULTS: In vitro inhibitory effect of AOB extract on ${\alpha}$-glucosidase was 92% as strong as that of acarbose. The AOB extract (500 mg/kg) or acarbose (50 mg/kg) significantly suppressed the postprandial rise of blood glucose after maltose challenge and the area under the glycemic response curve in normal mice. The AOB extract at 0.4% or 0.8% of diet or acarbose at 0.04% of diet significantly lowered levels of serum glucose and blood glycated hemoglobin and homeostasis model assessment for insulin resistance values in db/db mice. The expression of adiponectin protein in adipose tissue was significantly elevated by the consumption of AOB at 0.8% of diet. CONCLUSIONS: Autumn olive (E. umbellata Thunb.) berry may reduce postprandial hyperglycemia by inhibiting ${\alpha}$-glucosidase in normal mice. Chronic consumption of AOB may alleviate fasting hyperglycemia in db/db mice partly by inhibiting ${\alpha}$-glucosidase and upregulating adiponectin expression.

  • To keep blood glucose levels as close to normal as possible is the major goal of diabetes mellitus treatment $\alpha$-Glucosidase is the enzyme that digests die1my carbohydrate and inhibition of this enzyme may suppress postprandial hyperglycemia. The methanol extract of Polygonum multiflorum Thunberg was tested for inhibitoty activity against $\alpha$-glucosidase in vitro and in vivo. Polygonum multiflorum Thunberg extract inhibited yeast $\alpha$-glucosidase activity in a concentration-dependent manner. Polygonum multiflorum Thunberg showed an $IC_{50}$ value of 0.48 mg/mL. The ability of Polygonum multiflorum Thunberg extract to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without the methanol extract of Polygonum multiflorum Thunberg extract (500 mg/kg) was administered to diabetic rats by gastric intubation after an overnight fast A single oral dose of Polygonum multiflorum Thunberg extract significantly inhibited increases in blood glucose levels at 60 and 90 min (p<0.05) and significantly decreased incremental response areas under the glycemic response curve (p<0.05). These results suggest that Polygonum multiflorum Thunberg may have an antihyperglycemic effect by inhibiting $\alpha$-glucosidase activity in the animal model of diabetes mellitus.

Hypoglycemic and antioxidant effects of Daraesoon (Actinidia arguta shoot) in animal models of diabetes mellitus

  • BACKGROUND/OBJECTIVES: The primary objective of the treatment of diabetes mellitus is the attainment of glycemic control. Hyperglycemia increases oxidative stress which contributes to the progression of diabetic complications. Thus, the purpose of this study was to investigate the hypoglycemic and antioxidant effects of Daraesoon (Actinidia arguta shoot) in animal models of diabetes mellitus. MATERIALS/METHODS: Rats with streptozotocin-induced diabetes received an oral administration of a starch solution (1 g/kg) either with or without a 70% ethanol extract of Daraesoon (400 mg/kg) or acarbose (40 mg/kg) after an overnight fast and their postprandial blood glucose levels were measured. Five-week-old C57BL/6J mice were fed either a basal or high-fat/high-sucrose (HFHS) diet with or without Daraesoon extract (0.4%) or acarbose (0.04%) for 12 weeks after 1 week of adaptation to determine the effects of the chronic consumption of Daraesoon on fasting hyperglycemia and antioxidant status. RESULTS: Compared to the control group, rats that received Daraesoon extract (400 mg/kg) or acarbose (40 mg/kg) exhibited a significant reduction in the area under the postprandial glucose response curve after the oral ingestion of starch. Additionally, the long-term consumption of Daraesoon extract or acarbose significantly decreased serum glucose and insulin levels as well as small intestinal maltase activity in HFHS-fed mice. Furthermore, the consumption of Daraesoon extract significantly reduced thiobarbituric acid reactive substances and increased glutathione levels in the livers of HFHS-fed mice compared to HFHS-fed mice that did not ingest Daraesoon. CONCLUSIONS: Daraesoon effectively suppressed postprandial hyperglycemia via the inhibition of ${\alpha}$-glucosidase in STZ-induced diabetic rats. Chronic consumption of Daraesoon alleviated fasting hyperglycemia and oxidative stress in mice fed a HFHS diet.

성인을 대상으로 D-리보오스 함유 비율을 달리한 설탕 섭취에 따른 Glycemic Index 및 혈당 반응 연구 (Comparing the effects of intake of sugar containing different levels of D-ribose in sugar on glycemic index and blood glucose response in healthy adults)

  • 본 연구는 10 주 동안 건강한 20대 성인 남녀 14명을 대상으로 D-리보오스 함유 비율이 다른 설탕 (D-리보오스 5%, 10%, 14% 함유) 섭취가 혈당 반응과 GI에 미치는 영향을 분석하여 혈당 저감화 효과를 확인하고자 하였다. 설탕과 D-리보오스함유 설탕의 혈당반응값을 비교하였을 때 D-리보오스 함유 설탕인 RB5, RB10, RB14 순으로 낮아졌으며 RB10, RB14는 설탕에 비해 유의적으로 낮은 혈당반응값을 보였다. 백설탕, RB5, RB10, RB14의 GI는 각각 67.39, 67.07, 47.57, 45.62로 설탕과 RB5는 중 GI 식품으로, RB10과 RB14는 저 GI 식품으로 분류되었으며, RB10, RB14의 GI는 설탕과 RB5의 GI에 비해 유의적으로 낮았다. 혈당 변화에 따른 Cmax를 보면 포도당 61.73 mg/dL에 비해 RB5, RB10, RB14가 각각 49.43 mg/dL, 37.25 mg/dL, 35.15 mg/dL로 유의적으로낮았으나, 설탕의 Cmax 58.18 mg/dL과 비교 시 RB10, RB14의 Cmax는 유의적으로 낮았으나, RB5와는 유의적인 차이를 보이지 않았다. 따라서 D-리보오스 함유 설탕의 혈당 상승 억제 효과는 D-리보오스를 5% 함유 시 보다는 D-리보오스를 10% 이상 함유할 때 더 효과적인 것으로 나타났다. 그러나 D-리보오스를 10%, 14% 함유한 설탕의 GI값과 혈당 변화율이 유사하게 나타나고 있으므로 D-리보오스 14% 함유보다는 D-리보오스 10% 함유하는 것이 혈당 저감화 효과 및 비용적인 측면을 고려할 때 기능성 설탕의 비율로 더 적절할 것으로 사료된다.

Streptozotocin 유발 당뇨쥐에 있어서 콩나물 메탄올 추출물의 헐당강하효과 (Hypoglycemic Effect of the Methanol Extract of Soybean Sprout in Streptozotocin-Induced Diabetic Rats)

  • 콩나물 메탄을 추출물의 yeast $\alpha$-glucosidase 저해활성을 in vitro에서 측정한 결과 5 mg/mL 농도에서 24.5%로 나타났다. 콩나물 용매분획 중 ethyl acetate층에서 $\alpha$-glucosidase 저해활성이 36.3%로 가장 높게 나타났다. STZ로 당뇨를 유발한 흰쥐에 있어서 콩나물 추출물의 탄수화물 소화효소 저해활성을 측정하였다. 공복상태의 동물에게 전분(1 g/kg)과 함께 콩나물 메탄올 추출물 (500 mg/kg)을 경구 투여한 경우, 전분(1 g/kg)만 투여 경우에 비해 투여 후 60, 90, 120, 180분에 혈당 증가가 유의적으로 낮았으며 (p<0.05), 식후 혈당 증가곡선의 면적(AUC)도 유의적으로 작게 나타났다(p<0.05). 이상의 결과에서 콩나물 메탄올 추출물은 $\alpha$-glucosidase 저해 활성에 의해 식후 혈당 조절 효과를 나타내어, 콩나물의 섭취는 당뇨병 치료 및 예방에 도움을 줄 수 있는 것으로 사료된다.

메밀추출물 첨가가 In Vitro 전분가수분해율 및 정상성인의 혈당반응에 미치는 영향 (Effects of Added Buckwheat(Fagopyrum esculentum) Extract on Starch Hydrolysis In Vitro and Glucose Responses in Healthy Subjects)

  • This study determined the effects of added buckwheat extract on the rate of corn starch hydrolysis in vitro as well as blood glucose responses through its supplementation in healthy subjects. The rate of corn starch hydrolysis in the presence or absence of various buckwheat extracts was determined in an in vitro enzyme/dialysis system for 2 hr. The buckwheat was extracted by water, ethanol(40%, 70%, 100%) and methanol(40%, 70%, 100%), respectively. Twenty percent(w/w) additions of the ethanol, methanol and water buckwheat extract to corn starch solution significantly reduced the starch hydrolysis at every minute for 2 hr(p<0.05). The calculated hydrolysis indices of the buckwheat extracts were in the order of 100% ethanol extract(50), 100% methanol(54), 40% ethanol(58), 40% methanol(62), 70% methanol(64), 70% ethanol(68), water (82). For the blood glucose response study, groups of 12 volunteers were given 50 g of boiled rice with or without buckwheat extract(10% and 20% of starch weight) using the 100%, 70%, and 40% ethanol extracts, respectively. The addition of each buckwheat ethanol extract significantly reduced blood glucose concentrations at three or more points during 2 hr and also reduced the mean peak rise and area under the blood glucose curve(p<0.05). The calculated glycemic index(GI) values for all ethanol buckwheat extract groups were significantly decreased compared to the control(rice). At the concentrations of 20%, the buckwheat 100% ethanol extracts lowered the GI by 68%. The 100% ethanol extract was more effective than the 70% and 40% extracts for reducing GI. Therefore, the 100% ethanol buckwheat extract would be the most therapeutically useful in modifying postprandial hyperglycemia.

치커리추출물 첨가가 in vitro 전분가수분해율 및 정상성인의 혈당반응에 미치는 영향 (Effects of Addition of Chicory Extract on Starch Hydrolysis in vitro and Glucose Response in Healthy Subjects)

  • 본 연구는 치커리추출물 첨가가 in vitro에서 전분가수분해율 및 정상성인에서의 혈당반응에 미치는 영향을 실험하였다. In vitro 실험에서 고형분량으로 계산된 5% (w/w) 건조치커리 및 볶은치커리추출물을 전분용액에 각각 첨가하고 2시간 동안 가수분해시킨 후 15, 30, 60, 90, 120분에 각각 유리된 당을 측정하여 전분가수분해율 및 전분가수분해지수를 구하였다. 정상 성인을 대상으로한 실험에서는 고형분량으로 계산된 2.5, 5, 10및 20% (w/w) 건조치커리 및 볶은치커리추출물을 각각 포도당용액(60 g/300 mL)에 첨가하여 섭취시킨 다음 공복시 혈당 및 실험용액 섭취후 15, 30, 60, 90, 120, 150분에 각각 혈당을 측정하고 혈당반응 면적 및 혈당지수를 구하였다. 본 실험결과, 전분에 5% 건조치커리 및 볶은치커리추출물 첨가는 2시간 동안의 전분가수분해율, 가수분해면적 및 가수분해지수를 유의하게 감소시켰으나(p<0.05), 건조치커리군과 볶은치커리군사이의 유의적인 차이는 없었다. 포도당 용액에 2.5, 5, 10, 20% 건조치커리 및 볶은치커리추출물을 첨가했을때 모든 실험군에서 혈당곡선면적 및 최고혈당값은 낮아졌으며 최고혈당값을 나타내는 시간이 지연되는 경향을 보였다. 포도당 섭취시와 비교하여 모든 건조치커리군과 5, 10 및 20% 볶은치커리군의 혈당지수는 유의하게 감소되었으며(p<0.05), 특히 5 및 10% 건조치커리군의 혈당지수는 36%씩 감소되었고 20% 볶은치커리군의 경우 39% 감소되었다. In vitro 및 in vivo 실험결과로 볼때 치커리추출물첨가는 식후 혈당증가를 낮출 것으로 사료된다.

구기자 추출물의 항비만 및 혈당강하 효과 (Study on Anti-obesity and Hypoglycemic Effects of Lycium chinense Mill Extracts)

  • 구기자는 식용가능하고 제2형 당뇨병 합병증인 지방간분해, 고혈압 및 고혈당에 효능이 있다고 알려져 있다. 하지만 당 대사 및 지질 대사에 작용하는 정확한 기작에 대한 연구는 아직 미비하기에 탄수화물 소화효소인 ${\alpha}$-amylase와 ${\alpha}$-glucosidase의 저해활성 및 정상생쥐를 이용한 혈당강하 효과와 3T3-L1 adipocyte를 통하여 비만축적 억제효과를 보았다. 구기자 추출은 80% MeOH, 70% EtOH 및 열수추출하였으며 이들 중 ${\alpha}$-amylase 50% 저해 활성($IC_{50}$)은 saliva ${\alpha}$-amylase에서 acarbose, 80% MeOH, 70% EtOH, aqueous(7.04, 3.19, 3.20, 3.66 mg/mL), pacreas ${\alpha}$-amylase acarbose, 80% MeOH, 70% EtOH, aqueous(10.97, 4.16, 3.07, 3.55 mg/mL)로 saliva ${\alpha}$-amylase에선 acarbose보다 80% MeOH, 70% EtOH 추출물이 가장 낮은 농도에서 $IC_{50}$값을 나타냈고 pacreas ${\alpha}$-amylase에서는 70% EtOH 추출물이 가장 낮은 농도에서 $IC_{50}$값을 나타냈다. 또한 10 ${\mu}g/mL$ 농도에서 yeast ${\alpha}$-glucosidase 활성저해 $IC_{50}$값은 80% MeOH, 70% EtOH, aqueous(5.01, 4.98, 4.80 ${\mu}g/mL$)로 구기자 추출물 모두 acarbose(5.05)보다 우수한 $IC_{50}$값을 나타냈다. 정상생쥐에게 탄수화물 소화효소의 저해활성이 높은 70% EtOH 추출물을 전분과 함께 경구투여 한 후 내당능을 측정한 결과, 전분과 함께 100, 200 mg/kg body weight를 경구투여 한 경우, 대조군에 비해 투여 후 30, 60, 90, 120분에 혈당 증가가 낮았다. 또한 추출물을 100 mg보다 200 mg 투여하였을 경우 대조군에 비해 각각 19% 및 40%씩 저하되었으며, 120분에는 대조군 수준이 되었다. 그리고 3T3-L1 adipocyte에서 비만축적 억제효과를 측정한 결과 70% EtOH 추출물 농도가 증가함에 따라 leptin, 중성지방 농도가 감소하였고 유리지방산 농도는 증가하였다. 이상의 연구 결과 구기자 추출물 중 70% EtOH는 acarbose보다 높은 ${\alpha}$-glucosidase, ${\alpha}$-amylase 저해활성을 나타냈으며, 정상생쥐를 통해 식후 혈당증가를 저하시키는 효과를 확인하였고 GLUT4 발현 역시 증가함을 확인했다. 이를 통해 구기자 추출물이 지방축적 억제와 인슐린 저항성 억제 및 혈당강하 기전을 확인하여 제2형 당뇨병 개선에 효과적인 것으로 판단, 추후 더 연구하여 보다 더 정확한 성분을 조사할 필요성이 있다고 사료된다.
