Effects of human chorionic gonadotropin-producing peripheral blood mononuclear cells on the endometrial receptivity and implantation sites of the mouse uterus

  • Objective: This research investigated the effects of human chorionic gonadotropin (HCG)-producing peripheral blood mononuclear cells (PBMCs) on the implantation rate and embryo attachment in mice. Methods: In this experimental study, a DNA fragment of the HCG gene was cloned into an expression vector, which was transfected into PBMCs. The concentration of the produced HCG was measured using enzyme-linked immunosorbent assay. Embryo attachment was investigated on the co-cultured endometrial cells and PBMCs in vitro. As an in vivo experiment, intrauterine administration of PBMCs was done in plaque-positive female mice. Studied mice were distributed into five groups: control, embryo implantation dysfunction (EID), EID with produced HCG, EID with PBMCs, and EID with HCG-producing PBMCs. Uterine horns were excised to characterize the number of implantation sites and pregnancy rate on day 7.5 post-coitum. During an implantation window, the mRNA expression of genes was evaluated using real-time polymerase chain reaction. Results: DNA fragments were cloned between the BamHI and EcoRI sites in the vector. About 465 pg/mL of HCG was produced in the transfected PBMCs. The attachment rate, pregnancy rate, and the number of implantation sites were substantially higher in the HCG-producing PBMCs group than in the other groups. Significantly elevated expression of the target genes was observed in the EID with HCG-producing PBMCs group. Conclusion: Alterations in gene expression following the intrauterine injection of HCG-producing PBMCs, could be considered a possible cause of increased embryo attachment rate, pregnancy rate, and the number of implantation sites.

  • Objective: The aim of this study was to clone the mRNA sequence of the Acyl-CoA dehydrogenase long chain (ACADL) gene of goats and explore the effect of ACADL on the differentiation of subcutaneous fat cells on this basis. Methods: We obtained the ACADL gene of goats by cloning and used quantitative real-time polymerase chain reaction (qPCR) to detect the ACADL expression patterns of different goat tissues and subcutaneous fat cells at different lipid induction stages. In addition, we transfect intramuscular and subcutaneous adipocytes separately by constructing overexpressed ACADL vectors and synthesizing Si-ACADL; finally, we observed the changes in oil red stained cell levels under the microscope, and qPCR detected changes in mRNA levels. Results: The results showed goat ACADL gene expressed in sebum fat. During adipocyte differentiation, ACADL gradually increased from 0 to 24 h of culture, and decreased. Overexpression of ACADL promoted differentiation of subcutaneous adipocytes in goat and inhibited their differentiation after interference. Conclusion: So, we infer ACADL may have an important role in positive regulating the differentiation process in goat subcutaneous adipocytes. This study will provide basic data for further study of the role of ACADL in goat subcutaneous adipocyte differentiation and lays the foundation for final elucidating of its molecular mechanisms in regulating subcutaneous fat deposition in goats.

Comparative Genomic Analysis and Rapid Molecular Detection of Xanthomonas euvesicatoria Using Unique ATP-Dependent DNA Helicase recQ, hrpB1, and hrpB2 Genes Isolated from Physalis pubescens in China

  • Ground cherry (Physalis pubescens) is the most prominent species in the Solanaceae family due to its nutritional content, and prospective health advantages. It is grown all over the world, but notably in northern China. In 2019 firstly bacterial leaf spot (BLS) disease was identified on P. pubescens in China that caused by both BLS pathogens Xanthomonas euvesicatoria pv. euvesicatoria resulted in substantial monetary losses. Here, we compared whole genome sequences of X. euvesicatoria to other Xanthomonas species that caused BLS diseases for high similarities and dissimilarities in genomic sequences through average nucleotide identity (ANI) and BLAST comparison. Molecular techniques and phylogenetic trees were adopted to detect X. euvesicatoria on P. pubescens using recQ, hrpB1, and hrpB2 genes for efficient and precise identification. For rapid molecular detection of X. euvesicatoria, loop-mediated isothermal amplification, polymerase chain reaction (PCR), and real-time PCR techniques were used. Whole genome comparison results showed that the genome of X. euvesicatoria was more closely relative to X. perforans than X. vesicatoria, and X. gardneri with 98%, 84%, and 86% ANI, respectively. All infected leaves of P. pubescens found positive amplification, and negative controls did not show amplification. The findings of evolutionary history revealed that isolated strains XeC10RQ, XeH9RQ, XeA10RQ, and XeB10RQ that originated from China were closely relative and highly homologous to the X. euvesicatoria. This research provides information to researchers on genomic variation in BLS pathogens, and further molecular evolution and identification of X. euvesicatoria using the unique target recQ gene through advance molecular approaches.

A relationship of healthy eating status and body composition in Korean adults by their constitution type: A cross-sectional study (우리나라 성인의 사상체질에 따른 식생활 상태와 신체구성의 상관성에 관한 단면분석연구)

  • Background Adherence to healthy diet acts as a key role to modify sedentary lifestyle in real life setting. Constitution type of traditional Korean medicine has been prediagnosed to risk factors of cardiometabolic diseases. This study aims to evaluate the associations between body composition and healthy eating status in Korean adults by their constitution type. Methods Of 4046 participants from Korean Medicine Daejeon Citizen Cohort study, Korean Medicine Daejeon Citizen Cohort (KDCC) study were included for analysis. Demographic, health-related behaviors and Korean Medicine (KM) type were surveyed based on a general health-related questionnaire. Anthropometric measurements and dietary factors by using Korean Healthy Eating Index (KHEI) were assessed only in the half of the original participants. Results 50.8% of Taeeum was observed from eligible 1967 participants (66.7%, women). The highest KHEI score was observed in soyang group (52.0±0.3, p=0.006) compared to other two groups. In taeeum group, lower appendicular skeletal muscle mass (ASM)(%)(Taeeum: 38.7±0.1 vs. Soeum/Soyang: 39.2±0.1, p < 0.05), and higher percent of body fat (PBF)(%) (Taeeum: 32.9±0.2 vs. Soeum/Soyang: 29.0±0.2, p < 0.05) by the lowest tertile (T1) of the KHEI score, respectively. When the KHEI score increased by 1 point in taeeum group, a positive relationship was observed, which increased by 0.015% of ASM, however, a negatively related to in which body fat mass (BFM) decreased by 0.022 kg and PBF decreased by 0.024%. Conclusion Customized nutritional management is required that could help maintaining physical health and diet by their constitution type.

Video-conference fatigue of college students (MZ세대의 화상회의 피로감)

  • In an acceleration of 'untact' society during the COVID-19 pandemic, video conferencing platforms have been provided to connect the isolated and to overcome the limit of having interpersonal relationships. While these services brought a great deal of benefits, they also increased the number of people who suffer from additional fatigue from video conferencing. This study, thus, aimed to identify the factors of using video conferencing services and further to examine the relationship between the factors and the users' overall fatigue. Through focus-group interviews and survey, four main factors of fatigue (restriction from untact communication, difficulty of use, background set-ups, discomfort from excessive attention) were identified. This study also found the positive relationships of restriction from untact communication and background set-ups on the users' overall fatigue. The results does not only provide implication on defining new fatigue factors from young generations but also help practitioners and service providers to improve the deficits of services that the real users currently experience.

  • Augmented reality game has differences with traditional games in the way what it includes physical risks happened in the real world. Although accidents related to Pokemon Go are increasing, academic discussions on the issues are rare. Thus, this study explores risk types caused by the augmented reality game and examined how perception of the risks affect usage intention on the game. This study also provides the research implications through comparative analysis between youth group who generally has the tendency toward game addiction and older group. According to the results, teenagers had less usage intention when perceived financial risk increase. In case of over twenty, time risk had negative relationship with usage intention. Physical risk had no significant effect on usage intention for teenagers, but positive relationship was observed in case over twenty. These results imply the necessity of an appropriate regulation for safe game play.

More precisely, we prove that there exists a smooth hypersurface M whose metric is ds2 = dρ2 + ρ2d𝜉2 and whose mean curvature function is ${\frac{1}{n}}(\frac{u^p}{{\rho}^{\beta}})e^{f({\rho}^2)}{\psi}(\xi)$ for any given real constants p, β and functions f and ψ where u and ρ are the support function and radial function of M, respectively. Equivalently, we get the existence of a smooth solution to the following quasilinear equation on the unit sphere 𝕊n, $${\sum_{i,j}}({{\delta}_{ij}-{\frac{{\rho}_i{\rho}_j}{{\rho}^2+|{\nabla}{\rho}|^2}})(-{\rho}ji+{\frac{2}{{\rho}}}{\rho}j{\rho}i+{\rho}{\delta}_{ji})={\psi}{\frac{{\rho}^{2p+2-n-{\beta}}e^{f({\rho}^2)}}{({\rho}^2+|{\nabla}{\rho}|^2)^{\frac{p}{2}}}}$$ under some conditions. Our proof is based on the powerful method of continuity. In particular, if we take $f(t)={\frac{t}{2}}$, this may be prescribed mean curvature problem in Gauss measure space and it can be seen as an embedded result in Gauss measure space which will be needed in our forthcoming papers on the differential geometric analysis in Gauss measure space, such as Gauss-Bonnet-Chern theorem and its application on positive mass theorem and the Steiner-Weyl type formula, the Plateau problem and so on.

  • At the current stage of globalization and European integration of Ukraine, the aspects related to the effective fight against corruption in the system of economic security of our country are receiving more and more attention, as they become a prerequisite for continuing reforms based on international funding. In order to consider this issue and solve this problem, the necessary step is to develop and implement real mechanisms of the system for detecting and preventing corrupt behavior, which are based on international anti-corruption standards. The leading component of this system is the management of corruption risks in the system of economic security in order to identify them and implement measures to reduce them. This study analyzes the corruption perception index in Ukraine in recent years, which showed a positive, albeit somewhat slow dynamics of its growth, indicating a gradual increase in overcoming corruption through the introduction of a number of anti-corruption measures and changes. It is proved that the current stage of socio-economic development of the country contributes to strengthening the processes of combating corruption and preventing corruption risks, creating an effective and efficient anti-corruption system of the state. The concept of "corruption" was studied, it was found that in the field of public administration it is considered from different positions and is closely related to the concept of "corruption risks". The essence and features of corruption risks are studied, the preconditions of their occurrence are formulated, the relationship between the causes of corruption risks and economic security in the field of public authority has been established. The system of corruption risk management is considered and its components are characterized. It is proposed to increase the effectiveness of anticorruption policy through the implementation of measures aimed at investigating the causes of corruption risks, as well as developed effective and effective means of reducing corruption risks within the system of economic security

From Law/Superego to Love: Law, Violence, and the Possibility of Love in Herman Melville's Billy Budd, Sailor (법/초자아에서 사랑으로 -허먼 멜빌의 『빌리 버드』에 나타나는 법, 폭력, 그리고 사랑의 가능성)

  • This essay aims to explore Herman Melville's recognition and resolution of the vicious link between law and violence in his posthumous Billy Budd, Sailor (1924). In order to investigate the issues, this essay refers to Freud, Benjamin, Derrida, Lacan, and Žižek, all perceptive to the uncanny affinity of law and violence. Especially, Žižek's arguments of "superego" as an embodiment of cruel and destructive violence supplementing the official law and of "love" as an ethical possibility beyond the limit of the problematic law are introduced in this study to make Melville's reflection of the inseparableness between law and violence much clearer. John Claggart and Captain Vere embody the legal (superegoic) violence. Claggart even procurs secret enjoyment, in the name of maintaining positive law. Billy Budd discloses another violence defending his justness according to natural law. However, Melville suggests the possibility of suspending the problematic tie of law/violence through "love," as portrayed at the last part of the story. The two final words from Billy and Vere, as a sort of delayed dialogue between them after the event of their secret interview before Billy's hanging, suggest that they finally distance from the obscene nightly law of superego-respectively from outward punitiveness toward Vere and from inward punishment for Vere's excessive enforcement of Billy's hanging-and identify each other's lack as their own. Their love implicated in the last words is for the real other-in Lacan's sense-who discloses the constitutive lack or incompleteness of beings and aporia of the law. This essay's examination of Melville's representations about the superegoic violence as the (im-)possible condition of law and the possibility of withdrawing from it would help us recognize Billy Budd, Sailor as the author's own last word for the possible vision of love cutting the vicious knot of law/violence.

Correlation Extraction from KOSHA to enable the Development of Computer Vision based Risks Recognition System

  • Generally, occupational safety and particularly construction safety is an intricate phenomenon. Industry professionals have devoted vital attention to enforcing Occupational Safety and Health (OHS) from the last three decades to enhance safety management in construction. Despite the efforts of the safety professionals and government agencies, current safety management still relies on manual inspections which are infrequent, time-consuming and prone to error. Extensive research has been carried out to deal with high fatality rates confronting by the construction industry. Sensor systems, visualization-based technologies, and tracking techniques have been deployed by researchers in the last decade. Recently in the construction industry, computer vision has attracted significant attention worldwide. However, the literature revealed the narrow scope of the computer vision technology for safety management, hence, broad scope research for safety monitoring is desired to attain a complete automatic job site monitoring. With this regard, the development of a broader scope computer vision-based risk recognition system for correlation detection between the construction entities is inevitable. For this purpose, a detailed analysis has been conducted and related rules which depict the correlations (positive and negative) between the construction entities were extracted. Deep learning supported Mask R-CNN algorithm is applied to train the model. As proof of concept, a prototype is developed based on real scenarios. The proposed approach is expected to enhance the effectiveness of safety inspection and reduce the encountered burden on safety managers. It is anticipated that this approach may enable a reduction in injuries and fatalities by implementing the exact relevant safety rules and will contribute to enhance the overall safety management and monitoring performance.
