Characterization of the v-cath Gene of Bombyx mori Nuclear Polyhedrosis Virus K1

  • A cathepsin L-like cysteine protease, v-cath, encoded by the baculovirus has been shown to playa role in host liquefaction. We have identified a v-cath gene in the silkworm virus, Bombyx mori nuclear polyhedrosis virus (BmNPV) K1 strain. The 969 bp v-cath has an open reading frame of 323 amino acids. A putative cleavage site and catalytic sites were conserved in BmNPV-K1 v-cath. The predicted three-dimensional structure of BmNPV-K1 v-cath revealed that the overall fold of BmNPV-K1 v-cath is similar to that of other proteases of the papain family. The deduced amino acid sequence of BmNPV-K1 v-cath showed 98% and 97% protein sequence identity to BmNPV T3 strain and to Autographa californica nuclear polyhedrosis virus, respectively. The BmNPV-K1 v-cath differed at 4 amino acid positions from BmNPV T3. The v-cath gene in BmNPV-K1 genome is located on the EcoRV 6 kb and XhoI 9 kb fragments. Northern hybridization analysis of BmNPV K1 v-cath gene revealed that it is expressed late in infection.

CFD Analysis of Trap Effect of Groove in Lubricating Systems: Part I - Variation in Cross-Sectional Shape of Groove (그루브의 Trap 효과에 대한 CFD 해석: 제 1부 − 그루브 단면 형상의 변화)

  • Trap effect of groove is evaluated in a lubricating system using computational fluid dynamics (CFD) analysis. The simulation is based on the standard k-ε turbulence model and the discrete phase model (DPM) using a commercial CFD code FLUENT. The simulation results are also capable of showing the particle trajectories in flow field. Computational domain is meshed using the GAMBIT pre-processor. The various grooves are applied in order to improve lubrication characteristics such as reduction of friction loss, increase in load carrying capacity, and trapping of the wear particles. Trap effect of groove is investigated with variations in cross-sectional shape and Reynolds number in this research. Various cross-sectional shapes of groove (rectangular, triangle, U shaped, trapezoid, elliptical shapes) are considered to evaluate the trap effect in simplified two-dimensional sliding bearing. The particles are assumed to steel, and defined a single particle injection condition in various positions. The “reflect” boundary condition for discrete phase is applied to the wall boundary, and the “escape” boundary condition to “pressure inlet” and “pressure outlet” conditions. The streamlines are compared with particles trajectories in the groove. From the results of numerical analysis in the study, it is found that the cross-sectional shapes favorable to the creation of vortex and small eddy current are effective in terms of particle trapping effect. Moreover, it is found that the Reynolds number has a strong influence on the pattern of vortex or small eddy current in the groove, and that the pattern of the vortex or small eddy current affects the trap effect of the groove.

  • In this study is performed to investigate the pumping characteristics of three-stage disk-type molecular drag pump (DTDP). The experiments are measured using five vacuum pressure gauges in the positions for rotors of DTDP. The experimented DTDP is consisted of three rotors and four stator. In the DTDP, spiral channels of three rotors are cut on the both upper surface and lower surface of a rotating disk, and corresponding stator is a planar disk. The experiments are performed in the outlet pressure range of $0.2{\sim}533\;Pa$. The pressure of each rotors are measured under the various condition of outlet pressure and throughputs, and nitrogen gas is used for test gas. In the numerical study, the pumping characteristics of each rotor are studied for the variation of throughputs in the all rotating channel. Pressure contour and velocity are obtained by the numerical simulation.

  • [ $A_{3-2x/3}Al_{1-z}In_{z}O_4F:Eu_x^{3+}$ ](A = Ca, Sr, Ba, x = -0.15, z = 0, 0.1) oxyfluoride phosphors were simply prepared by the solid-state method at $1050^{\circ}C$ in air. The phosphors had the bright red photoluminescence (PL) spectra of an $A_{3-2x/3}Al_{1-z}In_{z}O_4F$ for $Eu^{3+}$ activator. X-ray diffraction (XRD) patterns of the obtained red phosphors were exhibited for indexing peak positions and calculating unit-cell parameters. Dynamic excitation and emission spectra of $Eu^{3+}$ activated red oxyfluoride phosphors were clearly monitored. Red and blue shifts gradually occurred in the emission spectra of $Eu^{3+}$ activated $A_3AlO_4F$ oxyfluoride phosphors when $Sr^{2+}$ by $Ca^{2+}$ and $Ba^{2+}$ ions were substituted, respectively. The concentration quenching as a function of $Eu^{3+}$ contents in $A_{3-2x/3}AlO_4F:Eu^{3+}$ (A = Ca, Sr, Ba) was measured. The interesting behaviors of defect-induced $A_{3-2x/3}Al_{1-z}In_{z}O_{4-{\alpha}}F_{1-{\delta}}$ phosphors with $Eu^{3+}$ activator are discussed based on PL spectra and CIE coordinates. Substituting $In^{3+}$ into the $Al^{3+}$ position in the $A_{3-2x/3}AlO_4F:Eu^{3+}$ oxyfluorides resulted in the relative intensity of the red emitted phosphors noticeably increasing by seven times.

  • Failure aspects of cut-slope, which induce the sequential collapses during the excavation stage, have been analyzed. Slope rock structures are investigated by examining the orientations and positions of discontinuity planes calculated based on the BIPS image inside the boreholes. Drilled core log has been also used to identify the structural defects. Clay minerals of swelling potentials are detected through XRD analysis. Numerical analysis for slope stability has been performed by utilizing the joint shear strength acquired from the direct joint shear test. Cut-slope collapse characteristics have been studied by investigating the posture of failure-prawn joint planes and the stability of tetrahedral blocks of different sizes. Cross-section analysis has been also performed to analyze the cut-slope behavior and to estimate the amount of reinforcement required to secure the stability of cut-slope. Behavior of reinforced cut-slope is also investigated by analyzing the slope monitoring data.

The Comparison of Trunk Muscles Activity During Bridging Stabilization Exercises on Swiss Ball According to Change of Position (스위스 볼을 적용한 교각 자세 변화에 따른 체간의 근 활성도 비교)

  • Purpose : The purpose of this study was to assess the effect of bridging stabilization exercises of trunk muscles activity on a Swiss ball according to change of position. Methods:30 healthy university students volunteered to participate in this study. Subjects were required to complete following three exercise positions. Exercise position 1; Supine bridge with Swiss ball, Exercise position 2; Side bridge with Swiss ball, Exercise position 3; Prone bridge with Swiss ball. Surface electromyography from selected trunk muscles was normalized to maximum voluntary isometric contraction. Results : A repeated measures of ANOVA with Duncan's correction was used to determine the influence of exercise type on muscle activity for rectus abdominis, external oblique, erector spinae. The erector spinae of exercise position 1 showed significantly higher muscle activity than exercise position 2, 3(p<.05). The external oblique of exercise position 2, 3 showed significantly higher muscle activity than exercise position 1(p<.05). The rectus abdominis of exercise position 3 showed significantly higher muscle activity than exercise position 1, 2(p<.05) Conclusion: These results indicate that muscle activity can be influenced by addition of a Swiss ball in bridging exercises. It is recommend to use a Swiss ball for trunk stabilization exercise.

Analysis of the Phylogenetic Relationships in the Genus Spiraea Based on the Nuclear Ribosomal DNA ITS Region (핵 리보솜 DNA ITS 부위에 의한 조팝나무속 식물종의 계통 관계 분석)

  • Genus Spiraea is composed of many long-lived woody species that are primarily distributed throughout Asia and Europe. In this study, we evaluated a representative sample of the 38 taxa in the world, including 14 in Korea, with nuclear ribosomal DNA internal transcribed spacer sequences (ITS) to estimate genetic relationships within the genus. The molecular data allowed us to resolve well-supported clades in the taxa. In 47 world accessions (38 taxa: 14 Korean taxa, 33 world taxa, and 9 overlapping taxa), total alignment length was 689 positions, of which 452 were parsimony informative, 527 variable, 75 singleton, and 159 constant characters. Although the phylogenic tree showed that many taxa of genus Spiraea were well separated from each other, many branches were not congruent with the morphological characteristics and geographical distributions of the genus. There were 430 segregating sites and the nucleotide diversity (${\pi}$) value was 0.281. Under the neutral mutation hypothesis, the probability that the Tajima test statistic (D) is positive (2.325) is more than 0.5. Therefore, there may be a site at which natural selection, which increases genetic variation, is operating.

A study on the heat transfer characteristics of swirling flow in a circular sectioned $180^{\circ}C$bend with uniform heat flux (균일 열플럭스가 있는 $180^{\circ}C$ 원형단면 곡관의 선회유동 열전달특성 연구)

  • An experiment was performed to local heat transfer coefficient and Nusselt number in the circular duct of 180.deg. bend for Re=6*10$^{4}$, 8*10$^{4}$ and 1*10$^{5}$ at swirling flow and non-swirling flow conditions. The test tube with circular section was made by stainless which has curvature ratio 9.4. The wall of test tube was heated directly by electrical power to 3.51 kw and swirling motion of air was produced by a tangential inlet to the pipe axis at the 180 degree. Measurements of local wall temperatures and bulk mean temperature of air are made at four circumferential positions in the 16 stations. The wall temperatures show particularly reduced distribution curve at bend for non-swirling flow but this effect does not appear for swirling flow. Nusselt number distributions for swirling flow which was calculated from the measured wall and bulk temperatures were higher than that of non-swirling flow. Average Nusselt number of swirling flow increased about 90 ~ 100% than that of non-swirling flow whole through the test tube. The Nu/N $u_{DB}$ values at the station of 90.deg. for non-swirling flow and swirling flow are respectively about 2.5 and 4.8 at Re=6*10$^{4}$. Also that is good agreement with Said's result for non-swirling flow. flow.

On-Site Measurement of the Inlet Air Evaporative Cooling Performance for a Centrifugal Turbo Compressor (CDA 인입공기 증발냉각을 통한 압축기 성능 개선장치 개발 연구)

  • In the present study, water vapor is injected at various positions in a Clean Dry Air (CDA) system such as a system inlet duct, compressor inlet, and compressor outlet by humidified air turbines. The application of evaporative cooling reduces the compression work and enhances the Energy Consumption Index (ECI) per unit volume. The main purpose of this study is to investigate the compressor power performance with different inlet air temperatures and humidity conditions. It is found that the actual power consumption and discharge flow are significantly influenced by the inlet air temperature as well as relative humidity. The results obtained during this study are compared both numerically and experimentally and are found to be in very good agreement.

  • This study investigates a parallel manipulator that can move over two parallel sliders and in which the end-effector of the manipulator can be adjusted arbitrarily. Through the direct and inverse kinematics of the manipulator, position equations are derived. These equations represent the relationship between the positions of the sliders and the position of the end-effector. The Jacobian matrices of the direct and inverse kinematics are obtained by these equations. By using the condition number defined from these matrices, the local performance index of the manipulator is proposed. By using the simulation results of the performance index, we find that the manipulator can smoothen movements in only one quadrant and that the distribution of the maximal performance index is affected by the ratio of the length of links and the orientation of the end-effector.
