Chemical Characterization of Industrial Hemp (Cannabis sativa) Biomass as Biorefinery Feedstock

  • Chemical composition and enzymatic saccharification characteristics of hemp woody core were investigated by their chemical composition analysis and enzymatic saccharification with commercially available cellulases (Celluclast 1.5L and Novozym 342). Hemp woody core have higher xylan and lower lignin contents than its bast fiber. Based on hemicelluloses and lignin composition, hemp woody core is similar with hardwood biomass. However, cellulose was more easily converted to glucose than xylan to xylose and this trend was confirmed both hemp woody core and yellow poplar. Hemp woody core biomass shows higher saccharification than yellow poplar (hardwood biomass) based on cellulose and xylan hydrolysis. With easier enzymatic saccharification in cellulose and xylan, and similar chemical composition, hemp woody core have better biorefinery feedstock characteristics than hardwood biomass.

실내 사용 목재의 연소 특성 분석 (II) (Combustion Properties of Woods for Indoor Use (II))

  • 본 연구에서는 목재의 화재 안정성 평가에 대한 기초 자료를 구축하기 위하여 연소 성능 및 열적 특성을 분석하였다. 열특성 분석은 콘 칼로리미터 시험 방법과 열중량 분석(TGA)으로 열방출률, 총 방출열량, 연소 가스 발생, 그리고 중량 감소를 분석하였다. TGA 분석에서 시료의 열분해 온도는 스트로브 잣나무 $359.83^{\circ}C$, 서어나무 $359.80^{\circ}C$, 이태리 포플러 $363.14^{\circ}C$, 졸참나무 $358.59^{\circ}C$, 산벚나무 $362.11^{\circ}C$로 나타났다. 열방출률의 최댓값은 서어나무로 나타났으며, 최솟값은 스트로브 잣나무로 나타났다. 총방출열량을 분석한 결과, 서어나무가 가장 높은 값을 나타내었고, 이태리 포플러가 가장 낮은 값을 나타내었다. 가스분석 결과에서는 산벚나무가 $CO/CO_2$ 비율이 최소치로 0.021로 확인되었고, 졸참나무가 0.031로 최대치를 나타내었다. 중량감소율의 최솟값은 산벚나무가 87.57%로 나타났으며, 졸참나무가 95.03%로 CO와 $CO_2$의 발생과 목재 연소의 거동과의 상관관계를 나타내었다. 본 연구는 목재의 실내 적용 시 화재 안정성 향상 등의 기초 자료로써 매우 유용할 것으로 판단된다.

  • Effects of aqueous ammonia soaking treatments to yellow poplar (Liriodendron tulipifera L.) were investigated to focus on chemical compositional changes and enzymatic hydrolysis characteristics changes by this treatment. Treatment temperature and time were main variables. At 3 different levels of aqueous ammonia soaking temperature and time ($145^{\circ}C$ -1 h, $90^{\circ}C$ -16 h and $45^{\circ}C$ - 6 days), lower temperature and longer soaking time led to more xylan removal based on carbohydrate compositional analysis. However, at higher temperature treatment led to more enzymatic saccharification of cellulose to glucose by commercial cellulose mixtures (Celluclast 1.5L and Novozym 342 from Novozyme, Denmark). Cellulose hydrolysis was gradually increased with increasing enzymatic hydrolysis time but xylan hydrolysis was leveled out at early stage (less than 10 h) of enzymatic hydrolysis.

Optimization of The Organosolv Pretreatment of Yellow Poplar for Bioethanol Production by Response Surface Methodology

  • We investigated the optimization of the organosolv pretreatment of yellow poplar for bioethanol production. Response surface methodology was used to determine the optimal conditions of three independent variables (reaction temperature, reaction time, and sulfuric acid (SA) concentration). Reaction temperature is the most significant variable in the degradation of xylan and lignin in the presence of an acid catalyst, and ethanol production increased with a decrease in the lignin content. The highest ethanol concentration ($42.80g/{\ell}$) and theoretical ethanol yield (98.76%) were obtained at $152^{\circ}C$ (2.5 bar) with 1.6% SA for 16 min. However, because of excessive degradation of the raw material, the overall ethanol yield was less than under other pretreatment conditions which has approximately 50% of WIS recovery rate after pretreatment. The optimal conditions for the maximum overall ethanol yield ($146^{\circ}C$ with 1.22% SA for 15.9 min) were determined with a predicted yield of 17.11%, and the experimental values were very close (17.15%). Therefore, the quadratic model is reliable.

Characterization of by-products from organosolv pretreatments of yellow poplar wood (Liriodendron tulipifera) in the presence of acid and alkali catalysts

  • Organic by-products derived from cellulose and lignin during organosolv pretreatments of yellow poplar wood (Liriodendron tulipifera) in the presence of $H_2SO_4$ and NaOH as catalysts, respectively, were subjected to various analyses to elucidate their effects on further performance of biological ethanol fermentation and provide preliminary data for the structure and utilization of organosolv lignin. Monomeric sugars amounted to ca. 2.2-7.7% in the organosoluble fraction of the organosolv pretreatment with $H_2SO_4$, while significantly low amount of sugars (0.2-0.3%) were determined in that of the organosolv pretreatment with NaOH. In case of addition of $H_2SO_4$ during organosolv pretreatment of biomass, a fermentation of the organosoluble fraction could be considered as an essential process to increase an efficiency of biomass utilization as well as yield of bioethanol. Precipitates, insoluble by-products in the solvent mixture, were also cficiency oed by diverse analytical methods and revealed that these were typically composed of a lignin moiety regardless of catalyst. According to the results of nuclear magnetic resonance (NMR), Fourier Tcinsform Infrared Spectroscopy (FT-IR) and Gel permeation chromatograp r (GPC), the main components of precipitates seem to be lignin polymers. However, their structures could be slightly modified during pretreatment and mixed with some carbohydrates by chemical bonds and/or physical associations.

라왕 푸라타누스 및 포푸라 단판(單板)을 구성(構成)한 합판(合板)의 성질(性質)에 관(關)한 연구(硏究) (A study on the properties of plywoods constructed by sycamore. poplar and lauan veneers)

  • This study was carried out to exploit and utilize American sycamore (Platanus occidentalis) and poplar grown in Korea as veneer species for plywood manufacture. At this study to save the imported lauan logs and dollars, the effects affecting to the properties of plywood constructed by sycamore, poplar and lauan veneers were studied. Important items dealt with this study were dry and wet shear strength, moisture content, and specific gravities By the results and discussion it may be summarized as followings. 1) Sycamore core lauan plywood (A-II type) was shown best dry shear strength, and the next were all lauan veneer plywood (A-I type), poplar core lauan plywood (B-I type), lauan core sycamore plywood (A-III type) in order. There are no differences between dry shear strength of A-II type and A-I type, A-II type and B-I type plywood. 2) Wet shear strength showed the same order with dry shear strength. A-II type plywood showed best wet shear strength and it showed more difference than other plwoods. A-I type, B-I type and A-III type plywood were all very good without significant difference in wet shear strength. 3) B-I type plywood showed highest moisture content of all type plywood and the next were all sycamore(A-IV type) all poplar plywood (B-III type), A-III type, B-II type, A-II type pllywood in order. Generally high moisture content showed when two or three veneers were same species in the three layer plywood. 4) A-III type plywood showed highest specific gravity of all and the next was A-IV type plywood without a difference to A-I type plywood on the whole. plywoods constructed by lauan and sycamore were shown high specific gravities and the next was lauan, sycamore and poplar, lauan and poplar. It seemed to be that connected wi th the specific gravity of veneer itself in different species. 5) The plywood manufactured by extension of potato flour is not only superior but also profitable in the price aspect than the plywood manufactured by extension of wheat flour.

Utilization of Robinia pseudoacacia as Sawdust Medium for Cultivation of Edible and Medicinal Mushrooms

  • This study was undertaken to examine the feasibility of black locust (Robinia pseudoacacia) as substrates for several edible mushrooms. For the cultivation of several edible and/or medicinal fungi on black locust, optimum bulk densities, synthetic or semisynthetic additives, natural additives and pretreatment methods were investigated. Fruit body yields of the fungi on various sawdust media composed of different wood species were also analyzed for testing the capability of black locust as a substrate for mushroom production. Mycelial growths decreased proportional when the bulk density increased. The most suitable carbon and nitrogen sources as additives to promote the mycelial growth were sucrose (2%, w/w) and ammonium phosphate (0.2%, w/w) respectively. When corn-powder and beer-waste as natural additives were added to sawdust of black locust showed the significant growth of mycelia. And the optimum mixing ratio was 10:2:1 (sawdust: corn-powder: beer-waste, w/w). Black locust after cold water treatment showed the outstanding mycelial growths. Any significant changes of pH, moisture content (%) and dry-weight losses (%) could not be found among culture substrates (sawdust of black locust, oak and poplar wood) examined before and after harvesting of fruit bodies. Yield of fruit bodies on black locust culture media were comparable with those culture media composed with oak and poplar wood. The present work indicated strongly the potentiality of black locust as raw materials for edible and medicinal mushrooms.

Tension Wood as a Model System to Explore the Carbon Partitioning between Lignin and Cellulose Biosynthesis in Woody Plants

  • Tension wood, a specialized tissue developed in the upper side of the leaning stem and drooping branches of angiosperm, is an attractive experimental system attractive for exploring the development and the biochemical pathways of the secondary cell wall formation, as well as the control mechanism of the carbon flux into lignin, cellulose, and hemicellulose. However, the mechanism underlying the induction and the development of the tension wood is largely unknown. Recently, several researchers suggested the possible roles of the plant growth hormones including auxin, gibberellin, and ethylene mainly based on the expression pattern of the genes in this specialized tissue. In addition, expressed sequence tag of Poplar and Eucalyptus provide global view of the genetic control underlying the tension wood formation. However, the roles of the majority of the identified genes have not yet been clearly elucidated. The present review summarized current knowledge on the biosynthesis of tension wood to provide a brief synopsis of the molecular mechanism underlying the development of the tension wood.

반탄화 온도와 승온속도에 의한 바이오매스 열분해 거동 (Thermal Degradation Behavior of Biomass Depending on Torrefaction Temperatures and Heating Rates)

  • 본 연구에서는 반탄화 과정에서 바이오매스 열분해 거동을 열중량 분석과 발생되는 기체분석으로 확인하였다. 반응온도는 $220{\sim}300^{\circ}C$, 승온속도 $10{\sim}30^{\circ}C/min$, 반응시간은 110분으로 고정하여 분석하였다. 백합나무 중량감소율은 $220^{\circ}C$에서 8.01~8.81%, $300^{\circ}C$에서 71.86~77.38%로 나타났으며, $240^{\circ}C$ 이상에서 급격하게 증가하였다. 반면 낙엽송 중량감소율은 $300^{\circ}C$에서 49.58~54.15%로 백합나무와 비교하여 낮았다. 백합나무에 대한 활성화 에너지는 87.32~91.24 kJ/mol으로 승온속도에 따라 큰 차이를 나타내지 않았으며, 낙엽송에 대한 활성화 에너지는 83.85~91.60 kJ/mol으로 나타났다. 반탄화 과정에서 발생하는 가스성분은 대부분 헤미셀룰로오스 유래 성분이었으며 반응온도가 증가하면서 성분의 종류 및 농도도 증가하였다. 백합나무에서는 furfural, acetic acid가 고농도로 검출되었다.

Modification of cell wall structural carbohydrate in the hybrid poplar expressing Medicago R2R3-MYB transcription factor MtMYB70

  • The isolation, cloning, and characterization of an R2R3-MYB transcription factor gene (MtMYB70) from the model legume Medicago truncatula is reported. MtMYB70 consists of a 768-bp coding sequence corresponding to 255 amino acids. Sequence alignment revealed that MtMYB70 cDNA contains conserved R2R3-type MYB domains with highly divergent C terminal regions. MtMYB70 was found to have relatively low sequence homology with known R2R3-MYB genes. Phylogenetic analysis placed the R2R3-MYB domain of MtMYB70 closest to PtMYB1, a known activator of lignin biosynthesis. Overexpression of MtMYB70 under the control of the 35S promoter in transgenic poplar did not cause a significant difference in total lignin content relative to the control, but glucan content was significantly increased in transgenic poplar. Therefore, MtMYB70 might have regulatory role in the biosynthesis of cell wall structural carbohydrates.
