Polymorphonuclear (PMN) leucocytes are fundamental importance to the body's defense mechanism and play a major role in the local and systemic reactions to infectious disease. Investigation of the physiological and pathological role of the various leucocyte subtypes in host defence mechanisms is dependent upon the isolation of adequate numbers of viable, pure leucocyte fractions. This report describes the separate frequency of PMN leucocytes both from buffy coat layer and from packed RBC layer when bovine peripheral blood was treated with various anti-coagulants such as acid-citrate-dextrose(ACD), ethyldiaminetetraacetic acid(EDTA), sodium citrate and heparin. The separate frequencies of PMN leucocytes from buffy coat layer was 60.4$\pm$9.6%(heparin), 56.8$\pm$11.8%(sodium citrate), 30.6$\pm$14.1%(ACD) and 6.2$\pm$3.7%(EDTA), in order. Those from packed RBC layer monitored with EDTA, ACD, sodium citrate and heparin was 85.0$\pm$4.7%, 84.3$\pm$5.5%, 83.8$\pm$6.5% and 76.3$\pm$7.7%, respectively. The Ficoll-hypaque(FH) density gradient method was used to remove a small part of lymphocytes and/or monocytes from leucocytes in packed RBC layer. With the result that it increased separate frequency of PMN leucocytes from EDTA(89.9$\pm$2.4%), ACD(89.5$\pm$3.6%), and sodium citrate(83.6$\pm$10.3%) than heparin(68.4$\pm$13.9%). These results indicate that the use of EDTA and ACD as anticoagulant Is suitable for the separation of PMN leucocytes from bovine peripheral blood, and that the FH density gradient method is able to increase the separate frequency of PMN leucocytes from packed RBC layer.

To determine contents of inorganic elements of Ganoderma lucidum, the horn-shaped carpophores and the pileus of Ganoderma lucidum were incinerated and analyzed by inductively coupled plasma atomic emission spectrophotometry. The ash contents of the pileus and the horn-shaped carpophore were 1.48% and 1.40%, respectively. The pileus contained calcium, magnesium, sodium, manganese, iron, zinc and germanium in that order. The horn-shaped carpophore contained magnesium, calcium, zinc, manganese, iron, copper and germanium in that order. To examine the protein-bound polysaccharide from Ganoderma lucidum for immunopotentiating activity, its fruit bodies were extracted with hot water. Purification of the extract was carried out by acetone precipitation and dialysis. The fraction obtained during the purification procedure consisted of a polysaccharide moiety (51%) and a protein moiety (5%). When the compound was administered intraperitoneally to the mice at a dose of 50mg/kg, it enhanced the accumulation of the peritoneal exudate cells, macrophage and polymorphonuclear leucocytes, thereby indicating immunopotentiation.

Polysaccharide fractions were prepared from Ginseng root, Mori Radicis Cortex (M. R. C. ), Phellodendri Cortex (Ph. C. ), Sappan Wood (S. W. ) and Tigli Semen (T. S.). Water extract was also prepared from the mixture of ph. C., S. W. and T. S. Ginseng polysaccharide and water extract of the mixture showed marked antitumor activity against sarcoma 180. Ginseng polysaccharide showed a mild increasing effect on the number of circulating leucocytes and a marked increasing effect on the number leucocytes and a marked increasing effect on the number of plaque forming cells (PEC). Polysaccharides from ginsing root, S. W., Ph. C. + T. S. and water extract of the mixture showed dramatic inducing activities of periotoneal exudate cells (PEC), polymorphonuclear leucocytes (PMN) and macrophages. These results suggest the possibility that water extract of the mixture may have the lentinan like effect and ginseng polysaccharide may have stimulating effects on the general immune system.

The present study was designed in order to clarify ultrastructural characteristies of the spermatogenetic process and to examine survival state of the spermatozoa introduced into the female reproductive tract after autumnal coitus in the Korean greater horseshoe bats(Rhinolophus ferrumeguinum korai). The general morphological characteristics of spermatogenesis were principally similar to those of the other mammalian species; acrosomal formation, flagellar formation, middle piece formation and concentration of the spermatozoan nucleus. The spermatozoa introduced into the vagina were found to be dead forming a vaginal plug, the opaque central core of which consisted of trapped dead spermatozoa. Some spermatozoa introduced into the uterus were observed to be phagocytized by the polymorphonuclear leucocytes infiltrated into the uterine glandular lumen. The oviductal epithelium, consisted of ciliated and secretory cells; the luminal surface of secretory cells were covered by a number of microvilli with well developed glycocalyx, suggesting a close relationship to nutrient (e.g. glycogen) supply for the spermatozoa during hibernation.

The effect of Roentgen rays on carcinoma of the cervix has long been of great interest to both radiologists and gynecologists. Since most cervical carcinomas are treated by irradiation, any additional knowledge either concerning the radiosensitivity of cervical tumors or their ultimate prognosis would be of value. The vaginal smear is considered to be one of convenient and rapid methods to study the effects of radiation on cervical malignancy. We observed morphologic changes in 297 cytologic preparations obtained from 60 patients who had underwent irradiation for cancer of the cervix. With the morphologic parameters such as cytoplasmic vacuolization, cytoplasmic basophilia, multinucleated giant cell formation, polymorphonuclear leucocytes (PMNL) sticking and postradiation dysplasia, we analyzed the findings in relation to the follow up time interval. The most common effect was the cytoplasmic vacuolization with basophilia of basaloid cells, which were noted in more than 90% of followed patients. The multinucleated giant ceil formation and PMNL stickering were noted in 38 cases(63%) and 48 cases(80% ) respectively. The differential diagnosis of postradiation dysplasia from recurrent or persistent carcinoma, reparative atypical cells, and regressing tumor cells was difficult and further study seems to be needed to clarify the more accurate morphologic features and biologic behavior.

The anti-inflammatory effect of Spirulina fusiformis on monosodium urate crystal-induced inflammation in mice has been investigated and compared with the non-steroidal anti-inflammatory drug Indomethacin. The paw volume, lysosomal enzyme activities, lipid peroxidation, anti-oxidant status and inflammatory mediator tumour necrosis factor-$\alpha$ were studied in control and monosodium urate crystal-induced mice after oral administration of Spirulina platensis in an experimental model for gouty arthritis. In the induced mice, the levels of lysosomal enzymes, inflammatory mediator tumour necrosis factor-$\alpha$, lipid peroxidation and the paw volume increased significantly, whereas the antioxidant status decreased when compared to control mice. $\beta$-glucuronidase and lactate dehydrogenase level were also found to be increased in untreated monosodium urate crystal-incubated polymorphonuclear leucocytes. After the oral administration of Spirulina fusiformis, the physical and biochemical changes observed in monosodium urate crystal-induced animals were significantly restored to near normal levels. The results clearly indicated the anti-inflammatory role of Spirulina fusiformis, a promising drug for gouty arthritis.

동면기 (11월부터 3월까지) 동안의 한국산 관박쥐(Rhinolophus ferrumequinum korai) 자성 생식도관 내의 정자저장, 정자 생존 여부 및 정자이동을 알아보기 위하여 전자현미경으로 관찰한 결과는 다음과 같았다. (1) 자궁내강, 자궁선내의 정자들은 다수의 백혈구들에 의해 포식되고 소멸되었다. (2)정자들은 수란관 미측 협부에서만 저장되었고, 정자들의 두부는 상피세포쪽을 향하고 있었다. 이는 교미기(10월 초-중순경)에 사출된 정자들이 긴 동면기 동안에 수란관 미측 협부에서만 생존 가능함을 의미하며, 수란관의 미측 협부가 정자의 수정능 획득(capacitation)에 필요한 최적의 장소임을 의미한다. (3) 동면후기인 3월의 수란관 미측협부에는 정자들이 관찰되지 않았다. 이는 배란기인 4월에 난자를 만나기 위해 정자들이 수정부위인 팽대부 쪽으로 이행하였음을 의미한다. 이상의 결과로 미루어 보아, 긴 동면기 동안의 자성 생식도관 내에서의 정자의 장기저장과 생존여부 및 정자이동은 수정을 위한 일종의 mechanism이라 여겨진다.
