Antioxidative Effect of Pine, Oak, and Lily Pollen Extracts (송화분, 참나무 및 백합화분 추출물의 항산화 효능)

  • Antioxidative activities of pine, oak, and lily pollen extracts were evaluated based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging ability and inhibition of lipid peroxidation in animal tissues. Each pollen was extracted with 50% ethanol, 100% ethanol or water. DPPH radical-scavenging capacity of 50% ethanol extract ($EC_{50}$ 40.0 mg/mL) of pine pollen was higher than those of water (46.8 mg/mL) and 100% ethanol (131.2 mg/mL) extracts of pollen. Fifty percent ethanol (3,2 mg/mL) was also better than 100% ethanol (4.5 mg/mL) and water (8.3 mg/mL) for extraction of oak pollen. For preparation of lily pollen extracts, 100% ethanol was most effective (14.0 mg/mL), followed by water (18.8 mg/mL) and 50% ethanol (24.0 mg/mL). Oak pollen showed higher DPPH radical-scavenging activity than others. Lipid peroxidation in rat brain homogenate induced by ascorbate-Fe3+-EDTA and rat kidney homogenate were inhibited by water extracts of all pollens in dose-dependent manner. Extracts of oak and lily pollen showed higher lipid peroxidation inhibition than pine pollen extract. Polyphenol content was highest in oak pollen extract $(32.5{\pm}0.7\;{\mu}g/mg\;pollen)$, followed by lily extract $(25.9{\pm}1.4\;{\mu}g/mg\;pollen)$ and pine extract $(9.3{\pm}0.7\;{\mu}g/mg\;pollen)$.

  • The chemical components of Korean pollens and pollen extracts were analysed to provide fundamental data for pollen processing and products development. The contents of crude protein, crude lipid, crude ash, crude fiber and carbohydrate of pollens were $14.0{\sim}22.3%,\;3.0{\sim}8.8%,\;2.2{\sim}3.3%,\;0{\sim}37.5%,\;and\;31.9{\sim}64.4%$ respectively, showing the specially high crude fiber content (37.5%) in Pine pollen. Almost all the carbohydrate, lipid, protein and ash components except fiber were extracted by water and n-hexane. Free sugars identified in water extracts $(10\;^{\circ}Bx)$ of pollens were fructose and glucose as major component and maltose as minor component. The proportions of three free sugars to total soluble solid content of Mixed, Acorn, and Pine water extracts were 73.6%, 85.4% and 47.7% respectively. Potassium and phosphorus content in pollens and pollen water extracts were high, but Ca, Na, Mg, Fe and Zn were not major mineral components. The essential amino acids such as leucine, phenylalanine, methionine, lysine, valine, isoleucine and threonine were contained richly in pollens and those were almost completely extracted by water. Vitamin $B_1$ and C were not detected, but only small amount of vitamin $B_2$ was detected in the pollens and pollen extracts tested. The essential fatty acids such as linoleic acid and linolenic acid were contained richly in pollens. The proportion of unsaturated fatty acids to total fatty acid content in Mixed, Acorn and Darae pollen except Pine was higher than 50%.

Identification of Leucine Aminopeptidase in Legume-Pollen Extracts and the Isozymes (콩과식물화분의 Leucine Aminopeptidase 검출과 그 Isozyme에 대하여)

  • Identification and observations of leucine aminopeptidase (LAP) and multiple molecular forms of the enzyme, isozymes, were made with a technique of starch-gel electrophoresis for various legume pollen. Plants tested other than Leguminosae demonstrated either no indication of the presence or at least tract of enzymes and the isozymes, although all legume pollen tested showed strong LAP patterns. The electrophoretic patterns of LAP failed to be shown if the extracts were heated or otherwise denatured. Extent of zymogrammatic appearance of LAP and the isozymes were characteristic of a species.

Effect of Proteases on the Extraction of Crude Protein and Reducing Sugar in Pollen (화분에서의 조단백질 및 환원당 추출시 단백질 분해효소가 미치는 영향)

  • This study was conducted to increase crude protein and reducing sugar contents in pollen extracts by proteases. Four commercial neutral proteases (Alcalase 2.4L, Protamex, Flavozyme and Protease A) and two alkaline proteases (Protease S and Protease P) were used to prepare acorn and Darae pollen extracts. Contents of moisture, ash, crude protein and crude fat of acorn pollen were 5.2%, 2.7%, 6.2% and 22.3%, respectively, while those of Darae pollen were 5.4%, 2.8%, 1.8% and 27.8%, respectively. Contents of crude protein and reducing sugar in pollen extracts were increased by proteases. Alcalase 2.4L was the most effective in increasing protein contents while Protease A in increasing reducing sugar contents. It is suggested the use of proteases is one of the potential methods for increasing the contents of crude protein and reducing sugar in preparation of pollen extracts.

  • Preventive effect of azalea pollen extracts against aniline-induced hepatic toxicity in mice was investigated in this experiment. When the biochemical and histological changes were measured, preventive effect was more striking by treatment with water extract. After treatment with azalea pollen extracts, hepatic microsomal aniline hydroxylase activity increased as compared to control. Whereas, aniline level in serum and liver significantly decreased. The Vmax value without affecting Km value increased by the water extract treatment, the results obtained suggest that the characteristics of increase in the aniline hydroxylase activity may include induction of enzyme proteins. These data indicate that the observed preventive effects of azalea pollen extracts against hepatotoxicity is due to the induction of aniline metabolizing enzyme.

Sensory Characteristics of Korean Wheat Noodles With Pine pollen and Antioxidant activities of Pine pollen Extracts (송화를 첨가한 우리밀 국수의 관능특성 및 송화추출물의 항산화성)

  • Sensory characteristics and antimicrobial activity of Korean wheat noodle with pine pollen powder and antioxidant activity of pine pollen extracts were investigated to develop the health promoting product. Pine pollen powder was extracted with water and 70% ethanol and extracts were tested electron donating ability (EDA) and nitrite scavenging ability (NSA). Sensory characteristics were evaluated by its color, flavor, moisture, softness, texture and taste. Microbiological quality was tested by total viable cells. EDA was highest in 1,000 ppm for both water extract (67% of EDA) and ethanol extract (74% of EDA). NSA was highest in pH 1.2 for both water extract(46% of NSA) and ethanol extract (48% of NSA). Antimicrobial activity of Korean wheat noodle with 3% pine pollen powder were 0.5 log $cycle{\sim}1$ log cycle lower than that of control at 5days of storage in total vival cells and at 4days of storage in total fungus. From sensory evaluation, Korean wheat noodle with 3% pine pollen powder had significantly higher of all scores.

  • In search for new drugs and methods of pharmacotherapy of liver damage , the pollen extracts was administrated to the experimental animals and ivnestigated its biochemical effects in the serum and liver when aromatic hepatotoxicants-phenybutazone, aniline and benzo(a) pyrene-was administered. As the results are the pretretment of azalea pollen was prevented the leakage of asartate aminotransaminase (AST), alanine aminotransfease(ALT), alkaline phosphatase (ALP) activity and bilirubin level to serum and liver, except serum lactate dehydrogenase (LDH) activity.

  • This study was conducted to assess the influence of hazard pattern in the surface structure of pollen grains of Campsis grandiflora, and cytotoxicity of different part extracts and nectar on RAW264.7 macrophages. The pollen grains were medium sized ($21.8{\mu}m$) with tricolpate aperture type. In equatorial view, the pollens were prolate (P/E=1.8) and the exine pattern was smooth and reticulate. This result contradict with the rumor of having a hook-shaped protuberance that can damage the cornea because we couldn't observed any protuberance on the surface of the outer wall. Furthermore, we investigated the 70% MeOH extracts (flower, leaf, stem) and nectar of C. grandiflora for their cell viability in temporal basis via MTT analysis on RAW264.7 macrophage cells. There was no significant difference in the cytotoxicity among the MeOH extracts and nectar of C. grandiflora after 24 h. However, nectar showed the dosedependent cytotoxicity on RAW264.7 macrophage cells after 48 h.

A Systematic Study on the Selected Amentiferous Plant Taxa -On the Genus Quercus- (유이화서 식물군의 계통분류학적 연구 -참나무과를 중심으로-)

  • The concept of natural grouping of plant designated as the "Amentiferous" is no longer given serious credence, and many of the families included in this grouping have been dispersed in diverse order. Because a review of taxonomic treatments of amentiferous taxa reveals diverse classifications, it has become necessary to investigate new characteristics and attempt to determine the significance of these characteristics in terms of amentiferous taxonomy. Protein analyses by isoelectrofocusing(IEF) and rocket immunoelectrophoresis(RIE) have proved to be useful in the delicitation of Quercus taxa using pollen extracts from selected taxa. When Quercus pollen extracts were separated by electrophoresis based on their isoelectric points in a stable pH gradient and substrates for estrase activity were stained, ten bands were revealed between pH 5-14. Within Lepidobalanus grouping, a great diversity in the pollen protein zymograms was observed with some segregation corresponding to the designated taxonomic sections. Two taxa of Cyclobalanopsis produced a zymogram that is somewhat similar to taxa included within the section Prinus of Lepidobalanus, and less similar to taxa within the section Cerris of the same subgenus. Three tested taxa of the Cerris are in the similar zymogram each other, being segregable from the taxa of Prinus. Quantitative and qualitative analyses for serological relationships within and among th Quercus were also employed. To calculate the degree of protein similarity, total rocket heights obtained from RIE provided an index of serological correspondence(SC). It is reconfirmed that the Quercus is distantly separated from the Fagus according to SC. Comparative data from rocket number and SC in the tested taxa of Quercus also indicate that Lepidobalanus is separable from Cyclobalanopsis. Within the Lepidobalanus Q. acutissima and Q. acutissima x variabilis are almost homogeneors and distinguishable from the other tested taxa of same subgenus. Although the number of taxa tested has been limited, the overall serological evidence best reflects the classification proposed by Redher(1940) and Melchior (1964), having the genus Quercus subdivided into three subgunera: Erythrobalanus, Lepidobalanus, and Cyclobalanopsis.alanopsis.

Effects of Tongryeong-san and Constituents Extract in Cultured Rat Myocardial Cells (통령산과 구성약물 추출물이 배양 심근세포에 미치는 영향)

  • To certify the protective effect of herbal medicine against oxygen free radical-induced myocardiotoxicity, cytotoxicity was measured using TBARS assay and Beating rate in the presence of Tongryeong-san(TRS) extracts or single constituents of this prescription. Myocardial toxicity was evaluated in neonatal rat myocardiocytes in cultures. In the present study, xanthine oxidase/hypoxanthine (XO/HX) resulted in a increase in lipid peroxidation and decreases in beating rate in cultured myocardial cells. In the effect of TRS extract, it showed the prevention from the XO/HX-induced cardiotoxicity by the increases of beating rate as well as the decrease of lipid peroxidation, In the protective effect of Faeces Trogopterori(FT), Pollen Typhae(PT), Caulis Akebiae(CA) and Radix Paeoniae Rubra(PRR), all the extracts were significantly effective in the protection of XO/HX-induced cardiotoxocity in cultured myocardial cells by the increase of beating rate as well as th decrease of lipid peroxidation. From these results, they show that XO/HX is cardiotoxic in cultured myocardial cells derived from neonatal rat, and it suggests that TRS, FT, PT, CA and PRR extracts are positively effective in the blocking in XO/HX-induced cardiotoxicity.
