가열 조리된 돼지고기의 Heterocyclic Amines 분석을 위한 Solid-phase 추출 방법의 비교 (Comparison of Different Solid-Phase Extraction Methods for the Analysis of Heterocyclic Amines from Pan-Fried Pork Meat)

  • 식품 중 HCAs을 검출하기 위한 최적 전처리 방법을 확립하기 위해 가열 조리된 돼지고기 패티를 이용하여 서로 다른 4가지 SPE(solid-phase extraction)방법을 비교하였다. 4가지 전처리 방법을 통해 얻은 15가지 HCAs 회수율은 3.0%(방법 A, Tri-P-1)-74.7%(방법A, Tri-P-2)이었다. 그 중 $MeA{\alpha}C$가 평균적으로 73.4%로 가장 높은 회수율을 보였으며 Tri-MeIQx가 15.2%의 가장 낮은 회수율을 보였다. 4가지 전처리 방법 중 방법A와 방법D가 가장 높은 회수율과 검출빈도를 보였으며 방법B와 방법C는 Harman(14.8%)을 제외하고는 전혀 검출되지 않았고 회수율을 구할 수 없었다 발암 가능 물질인 IQ, $A{\alpha}C$, $MeA{\alpha}C$, Glu-P-1, Glu-P-2, MeIQ, MeIQx, PhIP 등을 검출하는 방법에는 방법A(48.7-74.6%)가 4가지 방법 중 가장 유리하다. HCAs는 극성에 따라 polar amines과 less-polar amines으로 구분할 수 있는데 방법A는 less-polar amines검출에 유리하고 방법D는 polar amines검출에 유리하다. 방법A와 방법D의 chromatogram을 비교한 결과 방법A와 방법D 모두 15가지 HCAs가 깨끗하게 분리되었지만 Tri-MeIQx 등을 검출하는데 방법A가 더욱 유리하다. SPE 전처리 방법 및 LC/MS 분석방법에 대한 유효성을 확인하기 위해 LOD(0.2-2.1 ng/mL)와 LOQ(0.8-9.7 ng/mL), 표준 편차(0.2-8.6)를 구하였다.

남극 세종기지에서의 대기 중 PCB 모니터링 (Year-round Monitoring of Atmospheric Polychlorinated Biphenyls (PCBs) at the King Sejong Station in the Antarctic)

  • Atmospheric levels of polychlorinated biphenyls (PCBs) at the King Sejong station were monitored for one year using passive air samplers. Low-chlorinated PCB homologues were predominant in all samples. PCB levels were observed to decrease with distance from the station, which may indicate that a significant part of PCBs could be of local origin. Although the level of PCBs at the King Sejong station is very low (${\Sigma}_9PCB$ (18, 52, 101, 118, 128, 138, 153, 180, 187): $2.3\;pg\;m^{-3}$) probably due to decrease in the global PCB emissions, it is one order of magnitude higher than a background level in the Antarctic. Based on this preliminary study, more interpretation on PCB data and meteorological conditions is required.

  • Potatoes were first introduced outside the Andes region four centuries ago, and have become an integral part of much of the world's food. Potatoes were first introduced into Europe in the 16th century and Korea in the early 19th century. Potatoes have a short growing season, high production per unit area, relatively strong environmental adaptability, and are cultivated in more than 130 countries around the world. It is the world's fourth-largest crop, following rice, wheat, bean and maize. In the nutritional aspects, potatoes contain abundant vitamins and minerals, as well as an assortment of phytochemicals such as carotenoids and natural phenols. Due to the high content of potato functional compounds, it has known that potatoes are effective in the prevention of various human diseases. In particular, the potato contains a large amount of polar compounds, including the saponin in the polar compounds, and the physiological activity of the saponins, such as immunity enhancement, antioxidant and anti-inflammatory is known. In this study, the antioxidative activity of polar compounds from five potatoes was examined by cell based anti-oxidation assay. The smallest amount of ROS(Reactive oxygen species) was generated when the compound was derived from 'Haryung' and 'hongyoung' and strong SOD(Superoxide dismutase) activity was observed in 'Sumi' and 'Jayoung'. The results of this study reveal the antioxidative effect of polar compounds extracted from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers

Engineering and Characterization of the Isolated C-Terminal Domain of 5-Enolpyruvylshikimate-3-phosphate (EPSP) Synthase

  • 5-Enolpyruvylshikimate-3-phosphate (EPSP) synthase catalyzes the formation of EPSP and inorganic phosphate from shikimate-3-phosphate (S3P) and phosphoenolpyruvate (PEP) in the biosynthesis of aromatic amino acids. To delineate the domain-specific function, we successfully isolated the discontinuous C-terminal domain (residues 1-21, linkers, 240-427) of EPSP synthase (427 residues) by site-directed mutagenesis. The engineered C-terminal domains containing no linker (CTD), or with gly-gly ($CTD^{GG}$) and gly-ser-ser-gly ($CTD^{GSSG}$) linkers were purified and characterized as having distinct native-like secondary and tertiary structures. However, isothermal titration calorimetry (ITC), $^{15}N-HSQC$,\;and\;^{31}P-NMR$ revealed that neither its substrate nor inhibitor binds the isolated domain. The isolated domain maintained structural integrity, but did not function as the half of the full-length protein.

수분이 식품성분과 인쇄 용제와의 분배계수에 미치는 영향 (Effect of Water Content on Partitioning Behavior of Printing Ink Solvent on Food Ingredients Before and After Baking)

  • The partitioning behavior of five printing ink solvents was studied in various cookie ingredients before and after baking which had different water content and different structure. Solvents were ethyl acetate, hexane, isopropanol, methyl ethyl ketone, and toluene which represent different characteristic functional groups. Gas chromatography (G.C.) was used to measure partitioning values at $25^{\circ}C$ on each raw and baked cookie ingredients. Baking condition of cookie ingredients was $260^{\circ}C$ for 10 min. In cookie ingredients, decreases in water content generally affected Kp of polar solvents, but did not affect that of the non-polar solvents. However, as water content decreased in the cookie ingredient, the Kp of the non-polar and polar solvents showed mixed results.

Origins and Paleoceanographic Significance of Layered Diatom Ooze from Bransfield Strait in the Northern Antarctic Peninsula around 2.5 kyrs BP

  • We used diatom and porewater data of two piston cores from the central subbasin and one from the western subbasin in the Bransfield Strait in the northern Antarctic Peninsula to elucidate the depositional mechanism of the layered diatom ooze. The layered diatom ooze is characterized by an abundance of organic carbon, biogenic silica, sulfde sulfur, and lower porewater sulfate concentration. This lack of pore-water sulfate concentration in the diatom ooze interval may reflect development of reducing micro-environment in which bacterially mediated sulfate reduction occurred. The negative relationship between the total organic carbon and sulfate contents, however, indicates that sulfate reduction was partly taking place but does not control organic carbon preservation in this unit. Rather, well-preserved Chaetoceros resting spores in the layered diatom ooze indicate a rapid sedimentation of the diatom as a result of repetitive iceedge blooms on the Bransfield shelf during the cold period (around 2500 yrs BP) when the permanent seaice existed on the shelf, During this period, it is expected that the downslope-flowing cold and dense water was also formed on the Bransfield shelf as a result of sea ice formation, playing an important role for the formation of layered diatom ooze in the Bransfield subbasins.

동해고유수의 생성가능해역 I. 극전선에 의한 해역구분 (Possible Formation Area of the Japan Sea Proper Water I. Subareas by the Polar Front)

  • Based on the Results of Marine Meteorological and Oceanographical Observations during 1966∼1987 and the Ten-day Marine Report during 1970∼1989 by Japan Meteorological Agency, the possible area where the Japan Sea Proper Water (JSPW) can be formed is investigated by analyzing the distribution of water types in the Japan Sea. The Japan Sea can be divided into three subareas of Northern Cold Water(NCW), Polar Front(PF) and Tsushima Warm Current (TWC) by the Polar Front identified by a 6℃ isothermal line at the sea surface in vinter. Mean position of the Polar Front is approximately parallel to the latitude 39∼40。N. The standard deviation of the Polar Front from the mean position of about 130km width is the smallest in the region between 136。E and 138。E where the Polar Front is very stable, because the branches of the Tsushima Current are converging in this region. However, standard deviations are about 180∼250km near the Korean peninsula and the Tsugaru Strait due to greater variability of warm currents. In the NCW area north of 40∼30。N and west of 138。E, the water types of the sea surface to the loom depth are similar to those of the JSPW. This fact indicates that the surface layer of the NCW area is the possible region of the JSPW formation in winter.

영지버섯(Ganoderma lucidum)으로 부터 분리한 이차대사산물 및 이들의 항 염증 활성 (Secondary Metabolites Isolated from Ganoderma lucidum and Their Anti-inflammatory Activity)

  • Phytochemical study of Ganoderma lucidum (Polyporaceae) let to the isolation of five lanostane triterpenoid (1-5), along with two nitrogen derived phenolic compounds, N-phenylethylformamide (6) and N-acetylphenethylamine (7). The structures of the compounds were determined by 1D and 2D NMR, and MS experiments, as well as by comparison of their data with published values. Compounds 6 and 7 were isolated for the first time from the genus Ganoderma and this species. All the compounds were evaluated for cancer chemopreventive potential based on their ability to inhibit nitric oxide (NO) production induced by lipopolysaccharide (LPS) in mouse macrophage RAW 264.7 cells in vitro. Among the isolates, compounds 2 and 3 showed moderate inhibitory activity against NO production.

Observations of the Aurora by Visible All-Sky Camera at Jang Bogo Station, Antarctica

  • The auroral observation has been started at Jang Bogo Station (JBS), Antarctica by using a visible All-sky camera (v-ASC) in 2018 to routinely monitor the aurora in association with the simultaneous observations of the ionosphere, thermosphere and magnetosphere at the station. In this article, the auroral observations are introduced with the analysis procedure to recognize the aurora from the v-ASC image data and to compute the auroral occurrences and the initial results on their spatial and temporal distributions are presented. The auroral occurrences are mostly confined to the northern horizon in the evening sector and extend to the zenith from the northwest to cover almost the entire sky disk over JBS at around 08 MLT (magnetic local time; 03 LT) and then retract to the northeast in the morning sector. At near the magnetic local noon, the occurrences are horizontally distributed in the northern sky disk, which shows the auroral occurrences in the cusp region. The results of the auroral occurrences indicate that JBS is located most of the time in the polar cap near the poleward boundary of the auroral oval in the nightside and approaches closer to the oval in the morning sector. At around 08 MLT (03 LT), JBS is located within the auroral oval and then moves away from it, finally being located in the cusp region at the magnetic local noon, which indicates that the location of JBS turns out to be ideal to investigate the variabilities of the poleward boundary of the auroral oval from long-term observations of the auroral occurrences. The future plan for the ground auroral observations near JBS is presented.
