Anti-carcinogenic effects of non-polar components containing licochalcone A in roasted licorice root

  • BACKGROUND/OBJECTIVE: Licorice has been shown to possess cancer chemopreventive effects. However, glycyrrhizin, a major component in licorice, was found to interfere with steroid metabolism and cause edema and hypertension. The roasting process of licorice modifies the chemical composition and converts glycyrrhizin to glycyrrhetinic acid. The purpose of this study was to examine the anti-carcinogenic effects of the ethanol extract of roasted licorice (EERL) and to identify the active compound in EERL. MATERIALS/METHODS: Ethanol and aqueous extracts of roasted and un-roasted licorice were prepared. The active fraction was separated from the methylene chloride (MC)-soluble fraction of EERL and the structure of the purified compound was determined by nuclear magnetic resonance spectroscopy. The anti-carcinogenic effects of licorice extracts and licochalcone A was evaluated using a MTT assay, Western blot, flow cytometry, and two-stage skin carcinogenesis model. RESULTS: EERL was determined to be more potent and efficacious than the ethanol extract of un-roasted licorice in inhibiting the growth of DU145 and MLL prostate cancer cells, as well as HT-29 colon cancer cells. The aqueous extracts of un-roasted and roasted licorice showed minimal effects on cell growth. EERL potently inhibited growth of MCF-7 and MDA-MB-231 breast, B16-F10 melanoma, and A375 and A2058 skin cancer cells, whereas EERL slightly stimulated the growth of normal IEC-6 intestinal epithelial cells and CCD118SK fibroblasts. The MC-soluble fraction was more efficacious than EERL in inhibiting DU145 cell growth. Licochalcone A was isolated from the MC fraction and identified as the active compound of EERL. Both EERL and licochalcone A induced apoptosis of DU145 cells. EERL potently inhibited chemically-induced skin papilloma formation in mice. CONCLUSIONS: Non-polar compounds in EERL exert potent anti-carcinogenic effects, and that roasted rather than un-roasted licorice should be favored as a cancer preventive agent, whether being used as an additive to food or medicine preparations.

CARBON ISOTOPE ANALYSES OF INDIVIDUAL HYDROCARBON MOLECULES IN BITUMINOUS COAL, OIL SHALE, AND MURCHISON METEORITE

  • To study the origin of organic matter in meteorite, terrestrial rocks which contain or-ganic compounds similar to the ones found in carbonaceous chondrites are studied and compared with Muchison meteorite. Hydrocarbon molecules were extracted by benzene and methanol from bituminous coal and oil shale and the extracts were partitioned into aliphatic, aromatic, and polar fractions by silica gel column chromatography. Carbon isotopic ratios in each fractions were analysed by GC-C-IRMS. Molec-ular compound identifications were carried by GC-MS Engine. Bituminous coal and oil shale show the organic compound composition similar to that of meteorite. Oil shale has a wide range of ${\delta}^{13}C,-20.1%_0~-54.4%_0$ compared to bituminous coal, $-25.2%_0~34.3%_0$. Delta values of several molecular compounds in two terrestrial samples are different. They show several distinct distributions in isotopic ratios compared to those of meteorite; Murchison meteorite has a range of ${\delta}^13C\;from\;-13%_0\;to\;+30%_0$. These results provide interpretation for the source and the formation condition of each rock, in particular alteration and migration processes of organic matter. Especially, they show an important clue whether some hydrocarbon molecules observed in meteorite are indigenous or not.

Antitumor Constituents from Anthriscus Sylvestris (L.) Hoffm

  • Bioassay-guided chemical investigation of the roots of Anthriscus sylvestris (L.) Hoffm. resulted in the isolation of nine compounds, whose structures were determined by spectroscopic methods. Compound 1 was isolated from this plant for the first time and compounds 3 and 9 were first found from this genus. Different polar fractions of A. sylvestris extract and compounds 1, 6-8 and 9 were evaluated for antitumor activities against HepG2 (human hepatocellular carcinoma), MG-63 (human osteosarcoma cells), B16 (melanoma cells) and HeLa (human cervical carcinoma cells) lines by the MTT method. The petroleum ether fraction of A. sylvestris extract exhibited excellent inhibitory activity with an $IC_{50}$ value of $18.3{\mu}g/ml$. Among the isolates from the petroleum ether fraction, compound 7 showed significant inhibition against the growth of the four tumor cells with $IC_{50}$ values ranging from $12.2-43.3{\mu}g/ml$.

수산건제품의 갈변에 관한 연구 붕장어육 및 유의 산화, 갈변 물질의 성상 (Studies in Browning Reaction in Dried Fish Lipid Oxidative Browning in Dried Conger eel and Properties of Browning Products)

  • This studies was carried out in order to investigate the browning reaction of lipid originated compound with nitrogenous compound in dried conger eel. The major fatty acids were $C_{16:0},\;C_{16:1},\;C_{18:1},\;C_{20:5}\;and\;C_{22:6}$. The nonpolar lipid contained the highest percentage of $C_{16:0}$, while the polar lipid contained the highest percentage of $C_{22:6}$. The browning reaction there was a rapidly developed with the beginning of the decline in carbonyl value and remarkable decrease in polyunsaturated fatty acids such as $C_{20:5},\;C_{22:5},\;C_{22:6}$ compared with the other fatty acid, in the water soluble fraction of the browning product obtained from tile fish was detected some antioxidation activity but in the lipid soluble fraction which covers most of the browning reactions in the fish meat antioxidation activity was not detected. In the test of conger eel oil, the phosphatidylcholine was largest in quantity and browning products provided in this experiment showed very low reducing activity.

Molecular Networking-based De-replication Strategy Leads to the Isolation of a New Chromone from Pleosporales sp.

  • A new chromone analogue (1) was isolated from an EtOAc-extract of Pleosporales sp. culture medium, together with five known chromones (2 - 6). The isolation workflow was guided by a Molecular Networking-based dereplication strategy. The chemical structure of the new compound was elucidated using NMR and MS spectroscopy, and the absolute configuration was established by the Mosher's method. All isolated compounds were evaluated for their inhibitory effects on lipopolysaccharide-induced nitirc oxide production in RAW 264.7 macrophages. Compound 1 showed marginal inhibitory activity with an IC50 value of 118.7 μM.

유기오염물질의 복합독성이 요각류 사망률에 미치는 영향 (Compound Effect of Persistent Organic Pollutants on the Mortality of Marine Copepods)

  • This study investigated primarily the toxic effects of bis(tributyltin)oxide (TBT) and DDT (Dichlorodiphenyltrichloroethane) on the mortality of adult Acartia omorii and barnacle nauplii as well as the hatching rate of A. omorii. Subsequently, compound effects of TBT and DDT on the mortality of immature copepods were tested in order to assess whether or not synergistic influence existed in the mixture of sublethal concentration of two pollutants. Mortality of adult A. omorii increased as exposure concentration of DDT increased in the range of from 0.0001 to 1ppm. Egg hatching rate of the copepod showed no distinctive difference in the range between 0.1 and 10ppm, while barnacle nauplii showed abnormal motility of their appendages in the range of 0.0001 to 1 ppm. Mortality of adult A. omorii increased as TBT concentration increased within the range of 1 and 10 ppb, whereas egg hatching rate of the copepod showed no linear response to the same exposure range. Moreover, copepod nauplii were almost motionless even though copepod eggs hatched under the exposure condition of TBT $(0{\sim}10 ppb)$ and DDT $(0{\sim}10 ppm)$, respectively, suggesting that the nauplii are hard to develop into adult stage. On the basis of the sublethal concentration less than the 24-h $LC_{50}$, 0.001 ppm (DDT) and 2 ppb (TBT) were selected to confirm the compound effects of two pollutants on the mortality of immature copepods. Mortality of immature copepods under the condition of mixture of the two pollutants was higher than that in the single exposure condition. This result seems to indicate that synergistic effects of sublethal toxicants can make a more hazardous effect on the survival of immature copepods even though the concentration of single toxicant is not lethal to copepods in the marine environment.

Isolation of 6,6'-Bieckol from Grateloupia elliptica and its Antioxidative and Anti-Cholinesterase Activity

  • During the search for anticholinesterase compounds from marine organisms, we were able to isolate 6,6'-bieckol from a red alga, Grateloupia elliptica. This compound showed moderate acetylcholinesterase (AChE) inhibitory activity in a micromole range ($IC_{50}$ $44.5{\mu}M$). However, for butyrylcholinesterase (BuChE), a new target for the treatment of Alzheimer's disease (AD), it showed particularly potent inhibitory activity ($IC_{50}$ $27.4{\mu}M$), which is more potent compared to AChE. It also inhibits BACE-1, a new target for reducing the generation of ${\beta}-amyloid$.

Preparation of New Polyenaminonitriles Containing Cyclic and Methylene Units and Their Thermal Properties

  • Polyenaminonitriles containing cycloaliphatic and aliphatic units were prepared by interfacial or solution polymerization reaction of p-bis(1-chloro-2,2-dicyanovinyl) benzene (1) with 4-aminobenzyl-amine, 1-(2-aminoethyl)piperazine, 2-(aminomethyl)pyrrolidine and 4-(aminomethyl)piperidine. The chemical structure of the polymers was confirmed through a syntheses of the model compound. The resulting polymers possessed inherent viscosities of 0.29∼0.62 dL/g and they were easily soluble in polar aprotic solvents and common organic solvents. Thermal properties of the polymers such as curability and stability were studied by differential scanning calorimetry (DSC), thermogravimetric analysis (TGA) and infrared spectroscopy. The polymers exhibited a large exotherm in DSC analyses and underwent a curing reaction around 340-370$\^{C}$ to form insoluble materials. The polymers showed 70-80% residual weight at 600 $\^{C}$ under nitrogen.

Bioactivity of two medicinal plant Xylocarpus granatum Koen. (Meliaceae) and Sarcolobus globosus Wall. (Asclepiadaceae) of Sundarbans mangrove forest

  • Two medicinal plant of Sundarbans mangrove forest has been tested for the evaluation of growth inhibitory and antibacterial activity. The methanol extract of Xylocarpus granatum stem bark showed potent wheat rootlet ($IC_{50}=0.01{\mu}g/ml$) and shoot ($IC_{50}=0.23{\mu}g/ml$) growth inhibitory activity in a concentration related manner. The growth inhibitory activity was markedly decreased in residual methanol extract. The methanol extract showed antibacterial activity (MIC > 3 mg/ml) against Bacillus subtilis, Staphylococcus aureous and Proteus vulgaris. The residual methanol extract did not show any antibacterial activity. The results suggest the bioactive principle(s) of Xylocarpus granatum may be relatively non polar compound(s). The methanol extract and residual methanol extract of Sarcolobus globosus stem showed poor wheat rootlet and shoot growth inhibitory activity and no antibacterial activity.

  • The fluorescence properties and photoisomerization behavior of 1-(9-anthryl)-2-(n-quinolinyl)ethene (n-AQE, n=2-4) have been investigated in various solvents. t-3-AQE is strongly fluorescent, but does not accomplish photoisomerization, similar to parent hydrocarbon compound, t-1-(9-anthryl)-2-phenylethene (t-9-APE) or t-1-(9-anthryl)-2-(1-naphthyl)ethene (t-1-ANE). Fluorescence and photoisomerization oft-2-AQE and t-4-AQE are strongly affected by solvent polarity. Dependence of fluorescence quantum yield on the solvent polarity is moderate for t-2-AQE and large for t-4-AQE. In nonpolar solvent (in n-hexane), they exhibit relatively strong fluorescence, but do not isomerize to cis isomer on irradiation, even if inefficient isomerization is observed for t-4-AQE. However, as solvent polarity increases, their fluorescences become weak with efficient photoisomerization to corresponding cis isomer. Intramolecular charge-transfer excited state is presumed to contribute to photoisomerization. The S$_1$ decay parameters were found to be solvent-dependent due to the charge-transfer character of lowest S$_1$ state. In polar solvents, the activation barrier to twisting is reduced enhancing the isomerization of r-2-AQE and t-4-AQE in the singlet manifold.
