To improve the productivity of Pleurotus ostreatus, different conditions of liquid spawn culture were tested. The optimum culture conditions were potato dextrose broth, incubation temperature of $22^{\circ}C$, and pH 6. Fifteen different media ('A' to 'O') containing 0.3% soybean meal (SM) were prepared by varying sugar and glucose contents. The cultures were propagated in SM media for 14 days, at $22^{\circ}C$, and pH 6. Their absorbances were higher after 14 days of incubation in the media containing both sugar and glucose. In particular, the absorbance of media containing 5 to 20% of glucose alone ('C', 'D', 'E', and 'F') tended to increase in the incubation period. Dry cell weight was lower in media containing less than 20% sugar or glucose alone than in media containing 30% sugar ('A') or 30% glucose ('B') alone. In sawdust media, in 900 mL-bottle, the optimum inoculation volume of liquid spawn was 15 to 20 mL. The texture of the mushroom cultivated with the liquid spawn was superior to that cultivated in the solid spawn.
The two parts of chestnut peels, outer and inner layers, were collected and investigated for cultivation of various mushrooms, i.e., Pleurotus ostreatus, Lentinus edodes, Hericium ramosum and the others. The inner layer of chestnut peels was contaminated with the mycelia of the blue fungi when collected, consisted of 17~20 % holocellulose, being a good material for growths of the above fungi. The outer layer was considered to be poor materials for fungal growth, because of lower amounts of minerals. Both mushrooms of Pleurotus ostreatus and Hericium ramosum grow well, and were produced fruiting bodies on the solid media containing only inner layers of chestnut peels. The Imhyup No.6 and Imhyup No.7 varieties of Lentinus edodes grow well, but produced few fruiting bodies in the solid media containing only inner layers of chestnut peels. The growth patterns of P. ostreatus and L. edodes were evaluated by production of $CO_2$. and considered to be different according to fungi. The inner layers of chestnut peels as agricultural by-products were considered to be a good material for mushroom cultivation, and outer layers could be used as being added with the other additives. The chestnut peels, being the environmental pollutants in rural areas, was considered to be a good substrate for mushroom cultivation.
This study was carried out to develop the best medium of new oyster mushroom for bottle culture. The new oyster mushroom cv. Miso is a Pleurotus ostreatus developed at the Chungnam Agricultural Research & Extension Services. For the bottle culture media, poplar sawdust+beet pulp+cottonseed meal (5:3:2), poplar sawdust+wheat husk meal (8:2), poplar sawdust+beet pulp+cottonseed meal (6:2:2), poplar sawdust+beet pulp+cottonseed meal (4:4:2), and poplar sawdust+beet pulp+wheat husk meal (7:1:2) were used in 850cc PP bottle. The pH was 5~6 and the C/N ratio 19.7~28.3 in bottle culture media. The time of pinhead formation was 5 to 6 days. For the fruiting body formation after inoculation took 29~31 days. The yield of fruiting body of poplar sawdust+beet pulp+wheat husk meal (7:1:2) medium was the highest at 110.4g/bottle compared to other media. Therefore, such cultivation medium would be appropriate for the commercial production of bottle culture in the new oyster mushroom 'Miso'.
The traditional kochujang supplemented with mushrooms (Pleurotus ostreatus and Lentinus edodes) was prepared, and changes in quality characteristics during fermentation at ambient temperature were investigated. The moisture content of kochujang increased by addition of mushroom. The titratable acidity of mushroom-added kochujang was gradually increased during fermentation. Ethanol contents of mushroom-added kochujang decreased with the addition of oyster mushroom, but increased with the addition of oak mushroom. The amino-nitrogen contents increased gradually during fermentation, resulting in significantly higher contents of 9% mushroom-added kochujang than others. The a value of kochujang decreased greatly during fermentation, but L, a and b values showed significantly higher than the control. The growth of bacteria in kochujang was delayed by the addition of mushrooms. Bacterial and yeast counts at the end of fermentation were not influenced by the addition of mushrooms. Sensory evaluation test revealed that color and flavour of kochujang were improved by the addition of mushrooms, and higher score was obtained from the addition of oyster mushroom compared to oak mushroom.
The study was conducted to investigate the effects of post-incubation period and temperature treatment conditions during incubation on the uniform primordia formation and cultural characteristics of oyster mushroom (Pleurotus ostreatus). Three kinds post-incubation period; 25, 30, 35 days and control were applied for 30 days while two kinds incubation room temperature $23^{\circ}C$ and $26^{\circ}C$ and control were used $20^{\circ}C$. The substrate temperature during pre-incubation was of 'Suhan No. 1' and 'Gonji No. 7'. Oyster mushroom varieties tended to increase between $24^{\circ}C$ to $26^{\circ}C$ at 11 to 15 days after inoculation and then they were maintained in treatment temperature during post -incubation period. The $CO_2$ occurrence was at the highest at 6,500 ppm for 'Suhan No. 1' and 5,800 ppm for 'Gonji No. 7' at the time of the highest temperature increase. The ratio of un-uniformal primordia formation and the ratio of non-commercial fruit body were reduced by 40%, 10.5%, respectively compared to control for 'Suhan No. 1' when in the post-incubation temperature was $26^{\circ}C$, and incubated for 10 days and 15 days treatment. Also, 'Gonji No. 7' was reduced by 19%, 9.5%, respectively when in the post-incubation temperature was $26^{\circ}C$, and incubated for 10 days treatment. Therefore, the higher post-incubation temperature of room and longer post-incubation period resulted in the higher percentage of primordia formation of two cultivars.
This study was carried out to improve standardization of agricultural products and investigate quality changes during storage at different treatments. The standardization does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Therefore, modification of these standards is required to fit farmhouse situations. Average pileus diameter and thickness of A grades was 38.9 mm and 4.5 mm at shelf culture and 22.2 mm and 3.9 mm at bottle culture. Average stipe length of A grades was 49.6 mm at shelf culture and 66.7 mm at bottle culture, and stipes thick was the highest in A grades of shelf culture. The diameter of pileus of Pleurotus ostreatus stored for 14 days at $4^{\circ}C$ and $10^{\circ}C$ showed no significant difference. but the thickness of pileus decreased rapidly after 7 days. L-value and hardness of stipes and pileus were higher at shelf culture than bottle culture.
The study was conducted to investigate the effect of shading treatment on the uniform primordium formation and development of fruit body during incubation of major cultivated oyster mushrooms (Pleurotus ostreatus). Oyster mushrooms cultivars Suhan No. 1 and Gonji No. 7 were incubated while shading during different times in the incubation room wherein temperature of $20^{\circ}C$ was maintained. Substrate temperature during incubation was increased to $24{\sim}25^{\circ}C$ in 11~12 days for Suhan No. 1 and to $25{\sim}26^{\circ}C$ in 11~12 days for Gonji No. 7 regardless of shading times. The $CO_2$ generation was the highest as 9~11% for Suhan No. 1 and 9~10% for Gonji No. 7 at the time of temperature increase the highest, while $O_2$contents became decreased, thus constructed good environment for the growth of hyphae. Therefore, ratio of ununiformal primordia formation was reduced by 20% as against control for Suhan No. 1 and by 13% for Gonji No. 7 when shading was applied from $10^{th}$ day of inoculation with no difference in fruit body yield.
The oyster mushroom is a wide cultivar among cultivated edible mushrooms in Korea. But, due to the excess of domestic production, the price has been falling. This study has been conducted to develope new variety oyster mushroom(Pleurotus ostreatus) which have a long term storage to export in foreign market as well as domestic. 'Gonji-7ho', a new variety of oyster mushroom, for the bottle culture, was bred by mating with monokaryons isolated from 'Nongmin-59ho' and 'MT07156'. In the characteristics of fruit body, pilei were round type and gray and stipes were white color and soft. The fruit body growth was vital and uniform. When fruit-body was stored at 4 degrees after packing with plastic vinyl, storage period was extended 7 days longer than 28 day of chunchu-2ho. The yield was 166 g per a bottle(¢65, 900 ml).
The purpose of this study was to investigate the optimal mixture ratio for the mycelial culture of the Pleurotus ostreatus. The chief cultural media in this study were cotton hull, sawdust and rice straw and the supplemental media were zeolite, corn cob, defatted rice bran, white cotton, tobacco trash powder, rice hull and peat. The results of this study were as follows; the optimal mixture ratio of the chief cultural media were effective in 6 : 3 : 1(V/V, %), and the mycelial growth and density in the supplemental media were considerably better 1% zeolite,3% corn cob, 5% defatted rice bran, 1% white cotton, 1% tobacco trash powder, 7% rice hull in good order. The optimal mixture ratio be to the mixed supplemental media in the chief cultural media were as follows ; 2 : 2(V/V, %) at the conditions of mixed zeolite and corn cob; 3 : 2(V/V, %) at the conditions of mixed defatted rice bran and white cotton; 1 : 3(V/V, %) at the conditions of mixed tobacco trash powder and rice hull. At the conditions of the whole cultural media mixed, the mycelial growth and density were in good conditions ; cotton hull, sawdust, rice straw, zeolite, corn cob, defatted rice bran, white cotton, tobacco trash powder, rice hull, and peat were mixed 43.0 : 17.2 : 25.8 : 2.0 : 2.0 : 3.0 : 2.0 : 1.0 : 3.0 : 1.0 (V/V, %).
Oyster mushroom, Pleurotus ostreatus, was exposed to 0.625, 1.25, 2.5 and 5.0 ppm $SO_2$ for 15, 30, 60, 120 and 180 minutes at the tiny button stage in order to study the effect of sulfur dioxide on growth and cap discoloration of the mushroom. The cap color was changed from grey to greyish blue at low dosage of sulfur dioxide, and with the lapse of exposing time and rasing sulfur dioxide concentration up, the color was changed to light brown and dark brown. The cap dicoloration was begun by the exposure of sulfur dioxide to 0.625 ppm for 120 minutes, to 1.25 ppm for 30 minutes and to 2.5 ppm for 15 minutes. By the exposure of sulfur dioxide to 1.25 ppm for 120 minutes, to 2.5 ppm for 30 minutes, its growth was inhibited, but the fruit body was died by the exposure of the gas to 2.5 ppm for 180 minutes and to 5.0 ppm for 60 minutes. Sulfur dioxide inhibited conspicuously the development of basidia and basidiospores of the mushroom and 20 to 25 percent of basidiospores formed were abnormal. Ethylene production by mushroom exposed to sulfur dioxide was not recognized.
