  • Platelets play a very important role in nomal hemostasis and their functions are more enhanced in various pathogenic states than in normal state. Especially it has been postulated that abnormal platelet and endothelium function might be major factors of microcirculatory disturbance in diabetes mellitus. Hydroxybrazilin, a phenolic constituent of Hematoxylon campechianum has been examined for its inhibitory effect on platelet aggregation. Its antiaggregatory effect might be mediated through the decrease of ATP release from dense granule and those of thromboxane $A_2$ production in normal and streptozotocin induced diabetic rats. The present study was undertaken to gain insight into the mechanism that hydroxybrazilin inhibited thromboxane $A_2$ production in platelets. Thus we measured the effect of hydroxybrazilin on phospholipase $A_2$, a rate limiting step of thromboxane $A_2$ production, in normal and streptozotocin induced diabetic rats. Hydroxybrazilin significantly inhibited the platelet phospholipase $A_2$ activity in normal and streptozotocin induced diabetic rats.

  • [ ${\alpha}-Al_2O_3$ ] precursor was synthesised by sol-gel method using aluminum sulfate, sodium sulfate and sodium carbonate as law materials. The flaky ${\alpha}-Al_2O_3$ crystals were prepared by heating using precursor about $1,050^{\circ}C$. In this study, the effect of some transition-metal sulfate ($FeSO_4,\;SnSO_4,\;ZnSO_4$) addition have been investigated. When iron sulfate was added, it could see that act on impurities in crystal growth process. In case of tin sulfate, distribution of Platelets was very broad. When flaky ${\alpha}-Al_2O_3$ crystals were prepared zinc sulfate addition, thickness, size, and distribution of platelets was suited to industrial application. The average diameter of flaky ${\alpha}-Al_2O_3$ crystals was about 20 $\mu$m, and its thickness was about 0.3 $\mu$m. Increasing addition of zinc sulfate, thickness of ${\alpha}-Al_2O_3$ platelet was decreased.

Inhibitory Effect of Clavicepitaceae on Serotonin Release out of Human Platelets and Human Platelet Aggregation

  • We have investigated the effects of hypha-water extracts (HWE), fruit body-water extracts (FWE) and cordycepin from Cordyceps militaris on serotonin release out of human platelets and human platelet aggregation. HWE and FWE inhibited the release of [$^3H$]-serotonin from human platelet stimulated by thrombin (2 U/ml) or collagen (20$\mu$g/ml) in a dose-dependent manner. Furthermore, cordycepin, a major component of Cordyceps militaris, inhibited the human platelet aggregation induced by collagen (10$\mu$g/ml) in a dose-dependent manner. These results suggest that cordycepin containing in HWE and FWE may inhibit the serotonin release by suppressing the collagen-induced human platelet aggregation. Accordingly, our data demonstrate that HWE and FWE containing much cordycepin might have antithrombotic and antimigrainous functions.

  • Purpose: The purpose of this pilot study was to investigate a potential of platelet concentrate obtained from blood bank(PCBB) in accelerating wound healing and to determine an effective treatment protocol by quantifying levels of platelet derived growth factor (PDGF)-BB in PCBB in vitro. Methods: The first study was designed to investigate quantity of PDGF-BB over stored time of the PCBB. The stored times for each PCBB were 1, 3, 5, 7, 9, 11 and 13 days. The second study was designed to determine efficacy of adding thrombin to stimulate release of PDGF-BB from the platelets of PCBB. The platelets were suspended and incubated in either with or without thrombin. On 30 minutes and days 1, 3, 5, 7 after incubation, the levels of PDGF-BB were measured. Results: PDGF-BB level showed a linear decrease over stored time of PCBB from the first day to the 13th day. Addition of thrombin increased PDGF-BB release from 30 minute through the 5th day. Conclusion: The results indicate that PCBB can provide sufficient amount of growth factors to stimulate wound healing and adding thrombin accelerate it.

황기건중탕이 알레르기에 미치는 영향 (Inhibitory Effects on the Type I hypersensitivity and Inflammatory Reaction of Hwanggigunjung-tang)

  • HGJT has been used for the treatment of general weakness, digestive organ disease and so on. This study was carried out for the purpose of knowing the inhibitory effect on the Type I hypersensitivity and Inflammatory reactions of Hwanggigunjungtang(HGJT). The reasults were obtained as follows: HGJT(0.1. 0.5. 1, 2mg/g) concentration of dependently inhibited compound48/80 induced anaphylaxis reaction in mice. HGJT(2mg/g) also inhibited permeability of evans blue into peritoneal cavity in mice. HGJT reduced IgE, CRP. WBC and Platelets on egg albumin induced hypersensitivity. But serum NO was grown. According to above results, HGJT may be beneficial in the type I hypersensitivity and Inflammatory reactions by inhibition of histamine release from mast cells.

  • Synthetic and biosynthetic vascular grafts of small diameter have long been considered to be prone to thrombosis, ultimately leading to the complete graft occlusion. Endothelial cell seeding onto synthetic blood-contacting surfaces has been suggested to be an ideal means to solve this problem. This study described a culture method of bovine endothelial cells and evaluated blood-compatibility and seeding efficiency of cultured endothelial cells. Bovine pulmonary artery endothelial cells were harvested enzymatically and grown to confluence on polystyrene culture flask surfaces using established techniques. The identification of endothelial cells was made through the demonstration of expression of factor VIII R:Ag by immunofluorescent technique. To quantitate the effect of improvement in blood-compatibility of viable endothelial cells, endothelial monolayers were exposed to blood containing $\^$111/In-oxine labeled platelets. Viable endothelial monolayers retained less labeled platelets than control surfaces. The Indium-labeled endothelial cells were seeded onto three different blood-contacting surfaces of Dacron vascular graft immobilized in specially equipped wells and incubated for specific time intervals (t=15, 30, 60, 120 minutes). Longer incubation times showed improved cell adherence in collagen-coated and fibrin-coated Dacron vascular graft groups. However in untreated Dacron grafts, no direct relationship was observed between incubation time and endothelial cell seeding efficiency. This may be due to leakage of endothelial cells through porosity of Dacron grafts in this in-vitro experimental condition.

Hypolipidemic and Antithrombotic Effects of Increasing Intake of Linolenic Acid Derived from Perilla Oil in Rats

  • This study investigated the hypolipidemic and antithrombotic effects of linolenic acid derived from Korean perilla oil. The experimental rats(male, Sprague-Dawley) were divided into 5 groups using a Randomized Complete Block Design and fed one of the five following diets : DO*/O#. D4/O, D4/4, D4/8, or D4/20(D*/# represents the ratio of linoleic to linoenic acid as the percentage of total dietary energy intake) for 4 or 8 months. Bleeding time and whole blood clotting time were determined and the composition of serum and platelet lipids analyzed. Comparisons from the DO/O to the D4/20 group showed that serum lipids (total lipid, triglyceride, total cholesterol, and HDL-cholesterol) gradually decreased with increasing linolenic acid intake - the hypolipidemic effect. The composition of platelet fatty acids[the ratio of eicosapentaenoic acid(EPA)/arachidonci aci(AA)] increased gradually with increasing linolenic acid intake. Higher linolenic acid intake increased bleeding time and whole blood clotting time, and decreased malondialdehyde(MDA) production in the platelets, though no significant differences. These results suggest that linolenic acid derived from perilla oil appears to suppress the conversion of linoleic acid to AA and the EPA transformed from linolenic acid appears to suppress the conversion of AA to TXA2. Since TXA2 is a platelet-aggregating and vasoconstricting agent, the redulction of TXA2 released by platelets with increasing intake of perilla oil containing a lot of linolenic acid confers an antithrombotic effect.

Sulfonated Poly(ethylene glycol) Containing Methacrylate Copolymer Surfaces; Preparation, Characterization and In Vitro Biocompatibility

  • Poly(ethylene glycol) (PEG1K) and sulfonated PEG (PEG1K-SO$_3$) methacrylate (MA) copolymers have been prepared and characterized. The structures of the synthesized copolymers were confirmed by $^1$H and $^{13}$ C NMR spectroscopy and elemental analysis. The bulk characteristics of the copolymers were evaluated by viscosity and thermal analysis. The surface properties of the copolymers were investigated using dynamic contact angle measurements and electron spectroscopy for chemical analysis. The hydrophilicity of the surfaces modified with PEG1KMA or PEG1K-SO$_3$MA increased, possibly as a result of the orientation of the hydrophilic PEG1KMA/PEG1K-SO$_3$MA chains into the water phase. Platelets adhered less to the surfaces of the copolymers than they did to a polyurethane control. In addition, adhesion of platelets to the copolymer surfaces decreased upon increasing the chain density of PEG1KMA and sulfonated PEG1KMA in the copolymers. Both bacterial adhesion and protein adsorption were significantly reduced on the copolymer surfaces and their levels differ depending on the kind of surface or media.
