Studies on the Blood Steroid Hormone Levels during the Reproductive Stage in Cheju Pony Mares II.   • This stduy was conducted to find out the changes of estradiol-17$\beta$ and progesterone levels in the plasma of Cheju mares during gestatation period and before and after parturition with 12 heads of Cheju ponies. The results are as follows : 1. The estradiol-17$\beta$ and progesterone levels on the 1~4 months showed 61.6~134.2pg/ml and 7.74~9.20ng/ml respectively, but the estradiol-17$\beta$ levels rapidly increased to 426.4~1772.9pg/ml, and the progesterone levels decreased to 1.42~5.43ng/ml on the 5~10 months of gestation period. 2. The progesterone levels of the pregnant and the non-pregnant mares showed 4.80ng/ml and 0.04ng/ml in winter, respectively, but both of the two appeared 1,211.5pg/ml and 99.4pg/ml of estradiol-17$\beta$, respectively. Therefore the diagnosis of pregnancy might be more accurate with the levels of estrdiol-17$\beta$ in autumn. 3. The progesterone levels showed 3.20ng/ml the day before parturition and less than 1ng/ml on 2 days after parturition. The estradiol-17$\beta$ levels, showing the rising trend, and 90.1~162.4pg/ml up to 10 days.

Serial Ultrasonographic Appearance of Normal Uterus during Estrous Cycle in Miniature Schnauzer Dogs

  • Serial ultrasonography was performed to measure the normal appearance of uterine during estrous cycle and to determine whether the unterine appearance was related to the sex hormone, progesterone and estrogen. The uterine appearances, shape, diameter and echogenicity were daily monitored with ultrasonography in 9 Miniature Schnauzer dogs undergoing II estrous cycles. During proestrus and estrous, the uterus became hypoechoic but developed hyperechoic luminal echo. In the longitudinal view, the shape of the uterus occasionally changed from rectangular to coiled or serpentine, compared to other stages of the cycle. The diameter of the uterus during proestrus and estrous was larger (range: 0.60${\sim}$0.86 mm) than other stages (range: 0.48${\sim}$0.62 mm) of the cycle. The rising estrogen concentrations (range: 14.51${\sim}$16.86 pg/ml) in plasma during proestrus correlated with changes in the uterus (p<0.05). Progesterone concentrations were 0.08${\sim}$0.15 ng/ml at the onset of proestrus, but rose 1.06${\sim}$1.26 ng/ml at the end of proestrus. There was no relation to progesterone concentration from onset of estrus (p>0.05). There was dramatical changes in normal uterus and sex hormone during estrous cycle. Especially, the appearance, shape and diameter of uterus were related to plasma estrogen concentration during proestrus, correlated with other stages of the cycle.

  • In many fish species, including Nile tilapia (Oreochromis niloticus), gonadal development occurs at the expense of stored energy and nutrients. Therefore, reproductive systems are inhibited by limited food supply. It has been well established that reproductive function is highly sensitive to both metabolic status and energy balance. Nothing is known about the possible mediated connection between energy balance and reproduction. Kisspeptin, a neuropeptide product of the Kiss gene has emerged as an essential gatekeeper of reproduction and may be possibly be linked to energy balance and reproduction in non-mammalians. Thus, in this study, the effect of fasting (10 days) on the expression of kisspeptin and the gonadotropin-releasing hormone (GnRH) gene were assessed in Nile tilapia (male and female) using qRT-PCR. In addition, plasma levels of estradiol-$17{\beta}$ ($E_2$) and 11-ketotestosterone (11-KT) in adult tilapia were measured by ELISA. In male tilapia, fasting reduced Kiss2 and GnRH I mRNA expression in the brain and 11-KT level in comparison with the fed tilapia (p < 0.05). In females, however, there were no significant differences in GnRH I mRNA expression and $E_2$ between fish subjected to fasting and those fed (p > 0.05). These data indicate the impact of nutritional states on kisspeptin as a potential regulatory mechanism for the control of reproduction in male Nile tilapia.

Effects of Altered Calcium Metabolism on Cardiac Parameters in Primary Aldosteronism

  • Background: Increasing evidence supports interplay between aldosterone and parathyroid hormone (PTH), which may aggravate cardiovascular complications in various heart diseases. Negative structural cardiovascular remodeling by primary aldosteronism (PA) is also suspected to be associated with changes in calcium levels. However, to date, few clinical studies have examined how changes in calcium and PTH levels influence cardiovascular outcomes in PA patients. Therefore, we investigated the impact of altered calcium homeostasis caused by excessive aldosterone on cardiovascular parameters in patients with PA. Methods: Forty-two patients (mean age $48.8{\pm}10.9$ years; 1:1, male:female) whose plasma aldosterone concentration/plasma renin activity ratio was more than 30 were selected among those who had visited Severance Hospital from 2010 to 2014. All patients underwent adrenal venous sampling with complete access to both adrenal veins. Results: The prevalence of unilateral adrenal adenoma (54.8%) was similar to that of bilateral adrenal hyperplasia. Mean serum corrected calcium level was $8.9{\pm}0.3mg/dL$ (range, 8.3 to 9.9). The corrected calcium level had a negative linear correlation with left ventricular end-diastolic diameter (LVEDD, ${\rho}=-0.424$, P=0.031). Moreover, multivariable regression analysis showed that the corrected calcium level was marginally associated with the LVEDD and corrected QT (QTc) interval (${\beta}=-0.366$, P=0.068 and ${\beta}=-0.252$, P=0.070, respectively). Conclusion: Aldosterone-mediated hypercalciuria and subsequent hypocalcemia may be partly involved in the development of cardiac remodeling as well as a prolonged QTc interval, in subjects with PA, thereby triggering deleterious effects on target organs additively.

Effects of Oral Administration of Thyroid Hormone on Physiological Activity and Growth of Black Porgy Reared in Freshwater or Seawater (담수 및 해수사육 감성돔, Acanthopagrus schlegeli의 생리활성과 성장에 미치는 갑상선 호르몬의 효과)

  • Stress responses, osmoregulation and growth of black porgy reared in freshwater or seawater were investigated by supplying the feed (10 mg/kg feed) treated with the $T_3$ (3,5,3'-triiodo-L-thyronine) for 90 days. Plasma $T_3$ in black porgy reared for 60 days in freshwater with (FT) or without (FN) $T_3$, treatment was lower than that in black porgy reared in seawater with (ST) or without (SN) $T_3$ treatment. The concentration of $T_3$ in FT was significantly more than that in FN. Plasma cortisol in FT, at the same day, was 4 times higher than that in FN, whereas the differences of the hormone between ST and SN was not significant. Although the levels of plasma glucose in FN and FT tended to be lower than that in ST and SN, no differences were found between fish reared in freshwater and seawater after $T_3$ treatment. Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) of black porgy reared in seawater showed no difference throughout the experimental period. However, at the beginning stages of the experiment, the plasma levels of these enzymes in FN and FT were higher than in SN and ST. Recovering of AST level to the level of SN or FT was laster in FT than in FN. However, no effects of the exogenous $T_3$ treatment on osmoregulatory capacity and growth of black porgy reared in freshwater or seawater were observed in this study.

  • The changes of thyroid hormone levels in plasma, eggs and yolk-sac larvae in female rockfish (Sebastes schiegeli) were examined after maternal injection of 3,5,3'-triiodo-L-thyronine ($T_3$) with a dose of 20 mg/kg fish wt. There were no differences of L-thyronine ($T_4$) level in maternal plasma and yolk-sac larvae between $T_3$ injection and vehicle-injected control. The significant decrease of $T_4$ level was found in eggs from the $T_3$ injected mother fish compared with that of control. $T_3$ levels in maternal plasma, eggs, and yolk-sac larvae were significantly elevated by $T_3$ injection. Therefore $T_4$ and $T_3$ in maternal circulation may be transferred into oocytes and subsequently into the yolk-sac larvae. These findings suggest that thyroid hormones may play some roles on physiological metabolism during early larval development of rockfish.

Effects of Supplemental Glutamine on Growth Performance, Plasma Parameters and LPS-induced Immune Response of Weaned Barrows after Castration

  • Two experiments were conducted to investigate the effects of supplemental glutamine on growth performance, plasma parameters and LPS-induced immune response of weaned barrows after castration. In experiment 1, forty-eight weaned male piglets were used and fed maize and soybean meal diets supplemented with 0 (Control) or 2% L-Gln (Gln+) for 25 days. The results indicated that the Gln+ group tended to increase average daily gain compared to control in stages of days 7 to 14 and 0 to 25. The Gln+ had significantly better feed efficiency than the control group did during days 14 to 25 and 0 to 25. The plasma blood urea nitrogen and alkaline phosphatase contents of Gln+ group were higher than those of the control group on day 14 post-weaning. In experiment 2, sixteen weaned male piglets were injected with E. coli K88+ lipopolysaccharide (LPS) on day 14 post-weaning. The results showed that the Gln+ group had lower concentrations of plasma adrenocorticotrophic hormone and cortisol than the control group on day 14 pre-LPS challenge. In addition, Gln+ group had higher plasma IgG concentration than the control group for pre- or post-LPS challenged on day 14 post-weaning. In summary, dietary supplementation of Gln was able to alleviate the stressful condition and inflammation associated with castration in weaned barrows, and to improve their immunity and growth performance in the early starter stage.

Effects of 3,5,3'-triiodo-L-thyronine ($T^3$) on Growth of GR-transgenic Coho Salmon, Oncorhynchus Kitsutch

  • GH-transgenic coho salmon (Oncorhynchus kitsutch) juveniles in tGH*T$_3$and tGH*PTU were fed with the diets containing 1 ug/g fish of 3,5,3'-triiodo-L-thyronine (T$_3$) and 30 ug/g fish of 6-n-propyl-2- thiouracil (PTU), respectively, to assess the effect of these drugs on the change of physiological activity, growth and survival rate in comparison with normal transgenic (tGH*C) and nontransgenic coho salmon (Wild) for 90 days. Although the daily food intakes of all transgenic (tGH)-groups were higher than Wild, the amount was reduced by exogenous PTU supply. The fred efficiencies of tGH-groups were lower than Wild, but the efficiency was reduced both by T$_3$and PTU. The survival rate of tGH-group was significantly higher than that of Wild, but there was no significant difference among tGH-groups. Although the growth of tGH-coho salmon was faster than Wild. the growth rate of transgenic salmon was increased by exogenous T$_3$, but was reduced by PTU Plasma TT$_4$levels of tGH-groups was approximately 2-fold higher relative to Wild, but there were no difference of plasma TT$_4$levels among tGH-groups. plasma TT$_3$level or tGH-coho salmon was increased by exogenous T$_3$administration, but was reduced by exogenous PTU. In addition, although plasma GH levels of all tGH-groups were higher than that of Wild, the GH level in plasma of transgenic coho salmon was increased by exogenous T$_3$and reduced by exogenous PTU. In the meantime, the transgenic fishes also displayed head, jaw and opercular abnormalities typical of the offsets of this gene construct in coho salmon, indicating that some imbalance in growth processes has been induced. However, the abnormalities of transgenic coho salmon was reduced following exogenous PTU administration.
