Effects of Dietary Enteromorpha compressa on Growth and Blood Properties in Israeli Strain of Common Carp (Cyprinus carpio) (파래첨가 사료가 이스라엘계 잉어(Cyprinus carpio)의 성장 및 혈액성상에 미치는 영향)

  • In order to study the effects of dietary E. compresa on the growth and blood properties in young Israeli strain of common carp (mean. 8 g BW.). Fish were fed the commercial diet (control) and three experimental diets, $1\%,\;5\%\;and\;10\%$ of E. compressa powder were added to the control diet. After four months of feeding trial fish were bled by cardiac puncture and some hematological parameters were analyzed as physiological indices. 1) Following supplementation, positive responses in plasma albumin and glucose levels, and decreased body weight gain, feed efficiency and hematocrit. 2) Condition factor (fatness) in creased up to $50\%$, but decreased with $10\%$ supplementation. 3) Hb, GOT and GPT did not show any remarkable difference among all diet groups.

The Effects of Treadmill Exercise on Blood components, Antioxidant enzymes and Reactive Oxygen in Hyperlipidemic Rats (트레드밀 운동이 고지혈증 흰쥐의 혈액 성분과 항산화효소 및 활성산소에 미치는 영향)

  • PURPOSE: The purpose of this study was to investigate the effects of treadmill exercise on blood components and antioxidant system in hyperlipidemic rats. METHODS: Three weeks old male rats were randomly assigned into General diet(GD, n=10), High fat Diet(HD, n=10), and High fat diet+Treadmill exercise(HDT, n=10) groups. Treadmill exercise consisted of the treadmill running 5 times per week during 6 weeks(30 min/time for first 3 weeks and 60 min/time the other 3 weeks). RESULTS: The levels of triglyceride and total cholesterol were increased in HD group compared with GD group, and recovered to level of GD group by treadmill exercise(p<.05). Plasma glucose and insulin concentrations were increased in HD group compared with GD group, and recovered to level of GD group by treadmill exercise(p<.05). Glutathione(GSH) and glutathione reductase(GRD) concentrations were decreased in HD group compared with GD group, and these decreases returned to the level of GD group by treadmill exercise(p<.05). Xanthine oxidase(XO) and malondialdehyyde (MDA) concentrations were increased in HD group compared with GD group, and these increases retuned to the level of GD group by treadmill exercise(p<.05). CONCLUSION: This study showed that treadmill exercise application were effective treatment strategy on hyperlipidemia. Therefore, it could be considered as a treatment method in the patients with hyperlipidemia disease. Treadmill exercise, Hyperlipidemia, Blood components.

Alterations in Hematological Parameters and Antioxidant Responses in the Biofloc-reared Flatfish Paralichthys olivaceus Following Ammonia Exposure (암모니아 급성노출에 의한 바이오플락 사육 넙치(Paralichthys olivaceus)의 혈액학적 성상 및 항산화반응의 변화)

  • Flatfish Paralichthys olivaceus raised in biofloc system (mean length $27.6{\pm}3.1cm$, mean weight $280.4{\pm}26.5g$) were exposed for to different concentrations of ammonia (0, 8, 16, 32, and $64mg\;{NH_4}^+/L$) for 7 days. Following ammonia exposure, hematological parameters such as hemoglobin and hematocrit were significantly reduced. Plasma components such as magnesium, glucose, aspartate aminotransferase (AST), and alanine aminotransaminase (ALT) were significantly altered by ammonia exposure, whereas there were no significant changes in calcium, cholesterol, or total protein. Antioxidant responses, such as superoxide dismutase (SOD) and catalase (CAT) levels were significantly elevated following ammonia exposure. The results of this study indicate that ammonia exposure induces significant changes in hematological parameters and antioxidant responses in biofloc-reared Paralichthys olivaceus as a toxic response.

An Exploratory Study of Electrochemical Skin Conductance for the Deficiency Pattern Identification in Diabetic Patients (당뇨병 환자의 허증별 전기전도도 특성에 대한 탐색적 관찰 연구)

  • Objectives The objective of this study is to examine the interpretability of the questionnaire-based pattern identification in terms of biosignals. For this purpose, we investigate the relationship between electrochemical skin conductance (ESC) and Qi-Blood-Yin-Yang Deficiency Questionnaire (QBYY-Q) in diabetic patients. Methods A total of 40 patients with diabetes mellitus answered the QBYY-Q and their ESC were measured by SUDOSCAN device (a diabetes screening device, France). To analyze the relationship between QBYY-Q and ESC, ANOVA analysis and Scheffe test were performed and Pearson correlation coefficients were obtained. Results Of the 40 diabetic patients, 23 (57.5%) were males and 17 (42.5%) were females. According to the QBYY-Q, 9 patients were classified into Qi deficiency pattern (QD), 9 patients were Blood deficiency pattern (BD), 10 patients were Yin deficiency pattern (YiD) and 12 patients were Yang deficiency pattern (YaD). Demographic information (age, body mass index, duration of illness, etc.), signs of vitality (blood pressure, body temperature, etc.), fasting plasma glucose and glycated hemoglobin were not significantly different in each deficiency pattern. The ESC of the right leg was significantly lower in the BD group compared to the YiD group (p<0.022). Pearson's correlation coefficient was negatively correlated with the BD questionnaire score (r=-0.343, p <0.05). Finally, ESC showed a positive correlation with hemoglobin and erythrocyte levels in all limbs (r=0.483, p<0.01). Conclusions We showed that ESC could be used to classify the Deficiency pattern identifications in diabetic patients. Especially, the ESC was significantly lower in the BD group and was negatively correlated with the BD scores. It implies the potential utility of the ESC to understand the BD in terms of modern biosignals.

  • Rapid increase of diabetic population is a major health concern in Korea. In a trial to develop food components which can prevent and/or cure diabetes, we investigated the anti-diabetic activity of curcumin in high fat diet (HFD)-induced type 2 diabetes mellitus (T2DM) animal model. C57BL/6 mice were divided into three groups: normal diet (ND), high fat diet (HFD), and curcumin (CUR, HFD+0.02% curcumin). Mice were fed each diet for 16 weeks. CUR significantly reduced body weight gain, the levels of plasma glucose, insulin, total-cholesterol (T-C), and LDL-C, whereas increased HDL-C compared to those of HFD group. Notably, insulin signaling pathway was activated by CUR. This suggests that CUR improves obesity-associated T2DM by overcoming insulin resistance in part.

Obtaining Reference Intervals of HbA1c by Immunoturbidimetry (면역비탁법에 의한 HbA1c의 참고범위 설정)

  • HbA1c is the major fraction of glycated hemoglobin, and used primarily to identify the average plasma glucose concentration over prolonged periods of time, mostly 2-3 months. It is used as markers for the diagnosis and monitoring of diabetic patients and increasingly used a marker in health screening check up for general populations. In this study, HbA1c was measured with Cobas integra 800 (Roche Diagnostics, Mannheim, Germany), using immunoturbidimetry principles. We established the reference interval for HbA1c with Cobas integra 800 and evaluated its significance. The study subjects were 36,140 (male 57.5%, and female 42.2%) who visited the Health Promotion Center of a tertiary care center in Seoul for health checkup from January to September, 2008. HbA1c levels were measured with immunoturbidimetric method. Statistical evaluation was done with SPSS. Comparison between male and female was checked with Mann-Whitney test, and among age groups with Kruskal-Wallis test. Reference interval for HbA1c was from 4.8% to 6.1%. There was no significant difference between male and female with Mann-Whitney test (P=0.539). There was significant difference among age groups with Kruskal-Wallis test (P<0.05). Reference interval for HbA1c with Cobas integra 800 was 4.8~6.1%, which was different from conventional one, 4.4-6.4%. Establishment of reference interval for each principle is needed.

Antioxidant activities of brown teff hydrolysates produced by protease treatment

  • Purpose: Various plants, herbal medicines, and marine foodstuffs have been used in kimchi preparation to improve its overall quality. Teff, which is rich in minerals and starches, facilitates stable blood glucose levels and is well-suited for use in gluten-free products; hence, it can be used to reinforce the mineral composition of kimchi. In this study, we probed the antioxidant activities of hydrolysates prepared by treatment of brown teff with three proteases under different conditions. Methods: The mineral composition of brown teff was determined by inductively coupled plasma spectrophotometry-mass spectrometry, and we established optimal hydrolysis conditions by determining the total phenol and flavonoid contents of teff hydrolysates obtained using three different proteases (protamax, flavourzyme, and alcalase), two different protease concentrations (1 and 3 wt%), and three different incubation times (1, 2, and 4 h). The antioxidant activity of the hydrolysates was further investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, total antioxidant capacity (TAC), and ferrous reducing antioxidant power (FRAP) assays. Results: Brown teff was rich in I, K, Mg, and Ca, and the highest total phenol content ($24.16{\mu}g/mL$), total flavonoid content ($69.08{\mu}g/mL$), and TAC were obtained for 1 wt% protamax treatment. However, the highest DPPH scavenging activity and FRAP values were observed for hydrolysates produced by alcalase and flavourzyme treatments, respectively. Conclusion: Treatment of brown teff with proteases affords hydrolysates with significantly increased antioxidant activities and high total phenol and flavonoid contents, and these antioxidant activities of teff hydrolysates have the potential to enhance the quality and functionality of kimchi in future applications.

Study on Antioxidant Effect of Hoveniae Semen cum Fructus and Hoveniae Ramulus on Liver Cells Isolated from Oxidatively Stressed Rat (산화동물 모델 흰쥐 간세포에 대한 지구자(枳椇子)와 지구지(枳椇枝)의 항산화 효과 연구)

  • Objectives : The Objective of this study is to compare the anti-oxidative effect of the Hoveniae Semen cum Fructus (HScF) to that of Hoveniae Ramulus (HR) in vivo. Methods : The extracts of HScF and HR were tested in vivo for their anti-oxidative effects. SD Rats were injected with the AAPH (2.2'-azobis (2-aminodinopropane) hydrochloride) to induce the acute oxidization stress, and the oxidized rats were administered with the HScF and HR decoction orally. The RBC, WBC and PLT populations in plasma, and the blood chemistry levels of the rats were measured to observe the overall changes in the blood. The anti-oxidative effects of the HScF and HR decoctions were investigated on the liver tissues of the oxidated rats. Results : 1. Thrombocyte decreased significantly in the group medicated with HScF. 2. Blood glucose increased significantly in the group medicated with HR. 3. SOD activity increased significantly in both the group medicated with HScF and that with HR. 4. NO concentration decreased significantly in both the group medicated with HScF and that with HR. 5. The catalase content increased significantly in both the group medicated with HScF and that with HR. Conclusions : The HScF and HR extracts have an anti-oxidative effect.

Bifidobacterium bifidum DS0908 and Bifidobacterium longum DS0950 Culture-Supernatants Ameliorate Obesity-Related Characteristics in Mice with High-Fat Diet-Induced Obesity

  • Probiotic supplements have promising therapeutic effects on chronic diseases. In this study, we demonstrated the anti-obesity effects of two potential probiotics, Bifidobacterium bifidum DS0908 (DS0908) and Bifidobacterium longum DS0950 (DS0950). Treatment with DS0908 and DS0950 postbiotics significantly induced the expression of the brown adipocyte-specific markers UCP1, PPARγ, PGC1α, PRDM16 and beige adipocyte-specific markers CD137, FGF21, P2RX5, and COX2 in C3H10T1/2 mesenchymal stem cells (MSCs). In mice with high-fat diet (HFD)-induced obesity, both potential probiotics and postbiotics noticeably reduced body weight and epididymal fat accumulation without affecting food intake. DS0908 and DS0950 also improved insulin sensitivity and glucose use in mice with HFD-induced obesity. In addition, DS0908 and DS0950 improved the plasma lipid profile, proved by reduced triglyceride, low-density lipoprotein, and cholesterol levels. Furthermore, DS0908 and DS0950 improved mitochondrial respiratory function, confirmed by the high expression of oxidative phosphorylation proteins, during thermogenesis induction in the visceral and epididymal fat in mice with HFD-induced obesity. Notably, the physiological and metabolic changes were more significant after treatment with potential probiotic culture-supernatants than those with the bacterial pellet. Finally, gene knockdown and co-treatment with inhibitor-mediated mechanistic analyses showed that both DS0908 and DS0950 exerted anti-obesity-related effects via the PKA/p38 MAPK signaling activation in C3H10T1/2 MSCs. Our observations suggest that DS0908 and DS0950 could potentially alleviate obesity as dietary supplements.

Effects of Green Tea Polyphenol and Vitamin C on Type 2 Diabetic Rats Induced by Low Dose Streptozotocin Following High Fat Diet (고지방식이와 저용량 스트렙토조토신으로 유도된 2형 당뇨병 흰쥐에서 녹차 폴리페놀과 비타민 C 병합 투여 효과)

  • This study investigated the effects of green tea polyphenol and vitamin C on type 2 diabetes mellitus by administering polyphenon 60 (P60) and sodium ascorbate (SA) to diabetic rats induced by high fat diet/low-dose streptozotocin. The experimental group was divided into five different groups: non-diabetic control group (NC), diabetes control group (DC), diabetes+P60 group (DM+P60), diabetes+SA group (DM+SA), and diabetes+P60+SA group (DM+P60+SA). P60 and SA were dissolved in 0.1% drinking water. After P60 and SA were administered for 16 weeks, fasting blood glucose, plasma insulin, serum triglyceride, blood urea nitrogen (BUN), and creatinine levels as well as kidney alkaline phosphatase (AP) and ${\gamma}$-glutamyltranspeptidase (GGT) activities were measured. Fasting blood glucose level increased 5-fold in the DC group compared to the NC group. In the DM+P60 group, fasting blood glucose level decreased by 14%. In the DM+P60+SA group, fasting blood glucose level decreased by 28% compared to the DC group, whereas the DM+SA group did not show any significant difference. The homeostasis model assessment for insulin resistance index increased in the DC group and decreased in the DM+P60+SA group compared to the DC group. Serum creatinine level increased in the DC group, but decreased by 17% in the DM+P60 group and by 43% in the DM+P60+SA group compared to the DC group. The serum BUN level increased in the DC group, but decreased by 41% in the DM+P60+SA group compared to the DC group. Kidney GGT and AP activities decreased in the DC group compared to the NC group; however, they were reversed by DM+P60+SA group. These results show that combined administration of both green tea polyphenol and vitamin C had better effects on improving blood glucose level, insulin resistance, serum triglyceride level, and protecting kidneys than administration of either green tea polyphenol or vitamin C alone in the context of type 2 diabetes.
