All studies on date palm somatic embryogenesis have focused on germination in the presence of light while neglecting germination in darkness, which mimics the germination process of zygotic embryos within seeds. To improve the date palm micropropagation protocol, we investigated the effects of light and darkness incubation on the germination of indirect date palm somatic embryos and their subsequent conversion into plantlets. Darkness incubation emerged as a pivotal factor in the germination of indirect date palm somatic embryos and their successful conversion into plantlets. Darkness incubation significantly decreased the time required for the conversion of indirect somatic embryos into plantlets, halving the duration from 24 weeks to only 12 weeks. The micropropagation protocol was modified, consolidating the previous two distinct stages of germination and elongation under light incubation into a single stage under darkness incubation. These findings modified the protocol and significantly reduced the overall duration of the date palm micropropagation protocol.

High productivity of ginger (Zingiber officinale Roscoe) was obtained from the rhizome produced by shoot-tip culture with Korean native variety, Seosanjong. Seed rhizomes induced by shoot-tip culture were successfully established in the field. The rhizomes induced by both plant or rhizome were higher in emergence rate and faster in days to emergence than those of home seed production. The seed rhizome production induced by shoot-tip culture was two times heavier than that of home seed production. These results suggest that shoot-tip culture might be one of mass propagation methods in seed rhizome of ginger plant.

Plant regeneration has been optimized increasingly by organogenesis and somatic embryogenesis using a range of explants with tissue culture improvements focusing on factors, such as the age of the explant, genotype, media supplements and $Agrobacterium$ co-cultivation. The production of haploids and doubled haploids using microspores has accelerated the production of homozygous lines in Brassicaceae crop plants. Somatic cell fusion has facilitated the development of interspecific and intergeneric hybrids in sexually incompatible species of $Brassica$. Crop improvement using somaclonal variation has also been achieved. Transformation technologies are being exploited routinely to elucidate the gene function and contribute to the development of novel enhanced crops. The $Agrobacterium$-mediated transformation is the most widely used approach for the introduction of transgenes into Brassicaceae, and $in$$vitro$ regeneration is a key factor in developing an efficient transformation method in plants. Although many other Brassicaceae are used as model species for improving plant regeneration and transformation systems, this paper focuses on the recent technologies used to regenerate the most important Brassicaceae crop plants.
Pongamia pinnata Pierre is a tree legume, having potential in production of raw material for biodiesel. A protocol for in wk propagation of this plant was standardized using seedling explants. Growth regulators (GR) including gibberellic acid $(GA_3),\;N^6-benzylaminopurine(BA)$, thidiazuron (TDZ), and Adenine sulphate (Ads) were tested for optimum germination of seeds. Removal of seed coat prior to germination, controlled fungal growth partially but enhanced bacterial growth. Antibiotic cefotaxime was ineffective in controlling bacterial contamination. Seedling derived nodal explants and cotyledon nodes with attached cotyledons were excised and cultured for induction of shoots. Optimum sprouting and multiplication of shoot buds were obtained in MS medium supplemented with $8.88{\mu}M$ BA. These buds differentiated and rooted on medium devoid of GR. Optimum growth of Pongamia seedling was obtained in cotton plugged culture vessels. Reculturing of the cotyledon node explants produced more shoots from the same site. This process of removing shoots and reculturing of cotyledon node was followed for eight passages yielding 4 to 8 shoots in each cycle. The shoots (75%) rooted on half strength MS basal medium supplemented with 0.22% charcoal. All plants survived on transfer to soil. This is the first report on in vitro regeneration of Pongamia pinnata. This report demonstrates the possibility of coupling more than one parameter in single experiment to hasten the process of standardization. The process of cycling the nodal explant repeatedly for production of large number of shoots from single meristem may find application in genetic transformation experiments wherein meristems are used for transformation.

Nerine was originated from South Africa and around 30 kinds of species are distributed in worldwide. In present study we have characterized the condition of storage and growing conditions on the bulb propagation of Nerine by the tissue culture. Result indicated that the percentage of decrement, decay and sprouting of bulblets in the tissue culture was high at the higher temperature compare to lower temperature. The optimum conditions for storage and time were 12 weeks at $15^{\circ}C$ which were having 86.4% sprouting rate. The sprouting rate at different temperature and acclimation time, using bulblets in the tissue culture, were optimum at early June when temperature is relatively higher($22^{\circ}C$) that facilitate to breaking the dormancy. Growth characteristics and capacity of photosynthesis of Nerine were better at red and blue mixed lightness.
Experiments initiated 30 years ago to obtain selectable markers have led to a series of studies of Trp biosynthesis and anthranilate synthase (AS) the control enzyme using largely plant tissue cultures since they have experimental properties that can be readily exploited. Enzymological and compound feeding studies provided evidence that AS is the control point in the Trp biosynthesis branch and that altering the AS feedback control by the selection of mutants resistant to the Trp analog 5-methyl-tryptophan (5MT) can lead to the overproduction of this important amino acid. Plants regenerated from these Trp overproducing lines of most species also had high free Trp levels but Nicotiana tabaum (tobacco) plants expressed the feedback altered AS only in cultured cells and not in the regenerated plants. further tests by transient and stable expression of the cloned promoter for the naturally occurring tobacco feedback-insensitive AS, denoted ASA2, confirmed the tissue culture specific nature of the expression control. The 5MT caused by the expression of a feedback-insensitive AS from tobacco has been used to select protoplast fusion hybrids with several species since the resistance is expressed dominantly. Recently the ASA2 gene has been used successfully as a selectable marker to select transformed Astragalus sinicus and Glycine max hairy roots induced by Agrobactetium rhizogenes. These results show that the ASA2y-subunit can interact with the y-subunit of another species to form active feedback-insensitive enzyme that may be useful for selecting transformed cells. Plastid DNA transformation of tobacco has also effectively expressed ASA2 in the compartment in which Trp biosynthesis is localized in the cell.
Two potato (Solanum tuberosum L.) cultivars were transformed by Agrobacterium tumefaciens harboring cauliflower mosaic virus (CaMV) 35S promoter and $\beta$-glucuronidase (GUS) gene. Expression patterns of the CaMV 35S promoter according to tissue types and developmental stages, and genetic stability of GUS gene were investigated in the clonal progenies of transgenic potatoes. Kanamycin-resistant shoot emerged from tuber disc after 4 weeks of culture, and root was induced 6 weeks after culture on the selection medium. Shooting frequency of cvs. Superior and Dejima were 43% and 27%, respectively. Mature transformants and their clonal progenies showed no phenotypical abnormality. GUS activity was expressed primarily at parenchymatous cells of phloem tissue around the vascular cambium in the stem and root, and higher activity was found at the apical meristem of shoot, root and adventious shoot bud. GUS activity was higher at tubers of young explants than at stored tubers. These facts indicate that expression level of the CaMV 35S promoter differed according to tissue types and developmental stages of the organs. The GUS gene was stably inherited to each clonal progeny and normally expressed.
In Japan, propagation of cyclamen is mainly from seedlings. However, seeds are expensive and germination is slow and non..uniform. Therefore, to achieve genetically uniform propagation, multiplication must be vegetative. The rooted tuberous stems without shoots as sources of explants were cultured on the media containing BA and sucrose. After 30 days cultivation, tuberous roots were produced from the root ends attached to a tuberous stem and its capability was dependent on the type of media. The highest percentage of tuberous root formation was observed in Culture on the medium of 1/3 MS containing 0.05mgL$^{-1}$ NAA, 0.5mg L$^{-1}$ BA and 5% sucrose. Growth rates of the tuberous roots were greatly influenced by the cutting positions of a root in explants. The highest growth of was observed if small amount of root end was cut at initiation of tissue culture.

The development of tissue culture systems for Populus species, and the utilization of tissue culture biotechnology will be reviewed and discussed. Special emphasis will be placed on prospects for genetic transformation by Agrobacterium-mediated gene transfer methods.
Aloe in vitro culture was attempted to induce callus and regeneration ability from different explant sources onto MS medium with 0.5mg/l NAA plus 1.0mg/l BA. Anthers that no developed any callus and plant regeneration, while only four out of 274 filament explants induced calli at cut edge without regenerated plants. Twenty ovary explants regenerated four direct plantlets without via callus from the base of epidermal tissues. Regenerated plants on the root tip gave 2n=14 of chromosome numbers.
